Mutations in sarcoplasmic/endoplasmic reticulum calcium ATPase 2 (SERCA2) underlie Darier disease (DD), a dominantly inherited skin disorder characterized by loss of keratinocyte adhesion (acantholysis) and abnormal keratinization (dyskeratosis) resulting in characteristic mucocutaneous abnormalities. However, the molecular pathogenic mechanism by which these changes influence keratinocyte adhesion and viability remains unknown. We show here that SERCA2 protein is extremely sensitive to endoplasmic reticulum (ER) stress, which typically results in aggregation and insolubility of the protein. Depletion of ER calcium stores is not necessary for the aggregation but accelerates the progression. Systematic analysis of diverse mutants identical to those found in DD patients demonstrated that the ER stress initiator is the SERCA2 mutant protein itself. These SERCA2 proteins were found to be less soluble, to aggregate and to be more polyubiquitinylated. After transduction into primary human epidermal keratinocytes, mutant SERCA2 aggregates elicited ER stress, caused increased numbers of cells to round up and detach from the culture plate, and induced apoptosis. These mutant induced events were exaggerated by increased ER stress. Furthermore, knockdown SERCA2 in keratinocytes rendered the cells resistant to apoptosis induction. These features of SERCA2 and its mutants establish a mechanistic base to further elucidate the molecular pathogenesis underlying acantholysis and dyskeratosis in DD.

Protein aggregation of SERCA2 mutants associated with Darier disease elicits ER stress and apoptosis in keratinocytes.

Des mutations dans sarcoplasmique / endoplasmique de calcium du r&#xE9;ticulum ATPase 2 (SERCA2) sous-tendent la maladie de Darier (DD), une maladie de peau h&#xE9;r&#xE9;ditaire dominante caract&#xE9;ris&#xE9;e par une perte d&#x26;#39;adh&#xE9;rence des k&#xE9;ratinocytes (acantholyse) et une k&#xE9;ratinisation anormale (dysk&#xE9;ratose) r&#xE9;sultant de caract&#xE9;ristiques des anomalies cutan&#xE9;o-muqueuses. Mutationen im sarcoplasmic/endoplasmic Reticulum Kalzium ATPase-2 (SERCA2) unterliegen Darier-Krankheit (DD), eine dominant vererbte Hauterkrankung, gekennzeichnet durch Verlust von Keratinozyten Adh&auml;sion (Akantholyse) und abnorme Keratinisierung (Dyskeratosis) in charakteristische mukokutane Anomalien. Die molekulare pathogene Mechanismen, durch die diese Ver&auml;nderungen Keratinozyten Adh&auml;sion und Lebensf&auml;higkeit beeinflussen, bleibt jedoch unbekannt. Hier zeigen wir, dass SERCA2 Protein extrem empfindlich gegen&uuml;ber Endoplasmatisches Reticulum (ER) Spannung, was in der Regel Aggregation und Unl&ouml;slichkeit des Proteins f&uuml;hrt. Ersch&ouml;pfung der ER Kalzium-Speicher ist nicht notwendig f&uuml;r die Aggregation aber beschleunigt das Fortschreiten. Systematischer Analyse der vielf&auml;ltigen Mutanten, die identisch mit denen in DD-Patienten zeigte, dass ER Stress Initiator der SERCA2-mutant-Protein selbst ist. Diese SERCA2 Proteine wurden, weniger gut l&ouml;slich, greifbar sein, weitere Polyubiquitinylated werden gefunden. Nach Transduktion in prim&auml;ren menschlichen epidermalen Keratinozyten mutant SERCA2 Aggregate entlockt ER Spannung, verursacht erh&ouml;hten Zahl von Zellen zu vervollst&auml;ndigen und l&ouml;sen sich von der Kultur-Platte und induzierten Apoptose. Diese Mutante induzierten Ereignisse wurden durch erh&ouml;hte ER Stress &uuml;bertrieben. Dar&uuml;ber hinaus gerendert Niederschlag SERCA2 in Keratinozyten die Zellen resistent gegen Apoptose-Induktion. Diese Features des SERCA2 und seine Mutanten schaffen eine mechanistische Basis um weiter zu erhellen, die molekulare Pathogenese zugrunde liegenden Akantholyse und Dyskeratosis im Format TT.

Mutazioni del calcio del reticolo sarcoplasmatico/endoplasmic ATPasi 2 (SERCA2) sottendono alla malattia di Darier (DD), una malattia cutanea ereditaria dominante caratterizzata da perdita di aderenza dei cheratinociti (acantolisi) e anomala cheratinizzazione (discheratosi) che risultano in anomalie mucocutanee caratteristiche. Tuttavia, il meccanismo molecolare di patogeno da cui queste modifiche influenzano vitalit&agrave; e adesione dei cheratinociti rimane sconosciuto. Mostriamo qui che la proteina SERCA2 &egrave; estremamente sensibile allo stress del reticolo endoplasmatico (ER), che in genere si traduce in aggregazione e insolubilit&agrave; della proteina. Deplezione dei depositi di calcio ER non &egrave; necessaria per l&#39;aggregazione ma accelera la progressione. Analisi sistematica di diversi mutanti identici a quelli trovati in pazienti DD ha dimostrato che l&#39;iniziatore dello stress ER &egrave; la proteina mutante SERCA2 stessa. Queste proteine SERCA2 sono state trovate per essere meno solubile, di aggregazione e di essere pi&ugrave; polyubiquitinylated. Dopo la trasduzione in cheratinociti epidermici umani primari, mutanti SERCA2 aggregati ha suscitato lo stress ER, ha causato aumento del numero delle cellule per arrotondare e staccare dalla piastra di coltura e induce apoptosi. Las mutaciones en sarcopl&#xE1;smico / ret&#xED;culo endoplasm&#xE1;tico de calcio ATPasa 2 (SERCA2) subyacen a la enfermedad de Darier (DD), un trastorno de la piel de herencia dominante caracterizada por la p&#xE9;rdida de la adhesi&#xF3;n de los queratinocitos (acant&#xF3;lisis) y queratinizaci&#xF3;n anormal (disqueratosis) que resulta en alteraciones mucocut&#xE1;neas caracter&#xED;sticas. Mutations in either EPM2A, the gene encoding a dual-specificity phosphatase named laforin, or NHLRC1, the gene encoding an E3 ubiquitin ligase named malin, cause Lafora disease in humans. Lafora disease is a fatal neurological disorder characterized by progressive myoclonus epilepsy, severe neurological deterioration and accumulation of poorly branched glycogen inclusions, called Lafora bodies or polyglucosan bodies, within the cell cytoplasm. The molecular mechanism underlying the neuropathogenesis of Lafora disease remains unknown. Here, we present data demonstrating that in the cells expressing low levels of laforin protein, overexpressed malin and its Lafora disease-causing missense mutants are stably polyubiquitinated. Malin and malin mutants form ubiquitin-positive aggregates in or around the nuclei of the cells in which they are expressed. Neither wild-type malin nor its mutants elicit endoplasmic reticulum stress, although the mutants exaggerate the response to endoplasmic reticulum stress. Overexpressed laforin impairs the polyubiquitination of malin while it recruits malin to polyglucosan bodies. The recruitment and activities of laforin and malin are both required for the polyglucosan body disruption. Consistently, targeted deletion of laforin in brain cells from Epm2a knockout mice increases polyubiquitinated proteins. Knockdown of Epm2a or Nhlrc1 in neuronal Neuro2a cells shows that they cooperate to allow cells to resist ER stress and apoptosis. These results reveal that a functional laforin-malin complex plays a critical role in disrupting Lafora bodies and relieving ER stress, implying that a causative pathogenic mechanism underlies their deficiency in Lafora disease.

Laforin is required for the functional activation of malin in endoplasmic reticulum stress resistance in neuronal cells.

Glycogen, the largest cytosolic macromolecule, is soluble because of intricate construction generating perfect hydrophilic-surfaced spheres. Little is known about neuronal glycogen function and metabolism, though progress is accruing through the neurodegenerative epilepsy Lafora disease (LD) proteins laforin and malin. Neurons in LD exhibit Lafora bodies (LBs), large accumulations of malconstructed insoluble glycogen (polyglucosans). We demonstrated that the laforin-malin complex reduces LBs and protects neuronal cells against endoplasmic reticulum stress-induced apoptosis. We now show that stress induces polyglucosan formation in normal neurons in culture and in the brain. This is mediated by increased glucose-6-phosphate allosterically hyperactivating muscle glycogen synthase (GS1) and is followed by activation of the glycogen digesting enzyme glycogen phosphorylase. In the absence of laforin, stress-induced polyglucosans are undigested and accumulate into massive LBs, and in laforin-deficient mice, stress drastically accelerates LB accumulation and LD. The mechanism through which laforin-malin mediates polyglucosan degradation remains unclear but involves GS1 dephosphorylation by laforin. Our work uncovers the presence of rapid polyglucosan metabolism as part of the normal physiology of neuroprotection. We propose that deficiency in the degradative phase of this metabolism, leading to LB accumulation and resultant seizure predisposition and neurodegeneration, underlies LD.

Laforin prevents stress-induced polyglucosan body formation and Lafora disease progression in neurons.

Bladder cancer is a common and deadly malignancy but its treatment has advanced little due to poor understanding of the factors and pathways that promote disease. ATDC/TRIM29 is a highly expressed gene in several lethal tumor types, including bladder tumors, but its role as a pathogenic driver has not been established. Here we show that overexpression of ATDC in vivo is sufficient to drive both noninvasive and invasive bladder carcinoma development in transgenic mice. ATDC-driven bladder tumors were indistinguishable from human bladder cancers, which displayed similar gene expression signatures. Clinically, ATDC was highly expressed in bladder tumors in a manner associated with invasive growth behaviors. Mechanistically, ATDC exerted its oncogenic effects by suppressing miR-29 and subsequent upregulation of DNMT3A, leading to DNA methylation and silencing of the tumor suppressor PTEN. Taken together, our findings established a role for ATDC as a robust pathogenic driver of bladder cancer development, identified downstream effector pathways, and implicated ATDC as a candidate biomarker and therapeutic target.
