Sensory signal transduction of the inner ear afferent neurons and hair cells (HCs) requires numerous ionic conductances. The KCNQ4 voltage-gated M-type potassium channel is thought to set the resting membrane potential in cochlear HCs. Here we describe the spatiotemporal expression patterns of Kcnq4 and the associated alternative splice forms in the HCs of vestibular labyrinth. Whole mount immunodetection, qualitative and quantitative RT-PCR were performed to characterize the expression patterns of Kcnq4 transcripts and proteins. A topographical expression and upregulation of Kcnq4 during development was observed and indicated that Kcnq4 is not restricted to either a specific vestibular structure or cell type, but is present in afferent calyxes, vestibular ganglion neurons, and both type I and type II HCs. Of the four alternative splice variants, Kcnq4_v1 transcripts were the predominant form in the HCs, while Kcnq4_v3 was the major variant in the vestibular neurons. Differential quantitative expression of Kcnq4_v1 and Kcnq4_v3 were respectively detected in the striolar and extra-striolar regions of the utricle and saccule. Analysis of gerbils and rats yielded results similar to those obtained in mice, suggesting that the spatiotemporal expression pattern of Kcnq4 in the vestibular system is conserved among rodents. Analyses of vestibular HCs of Bdnf conditional mutant mice, which are devoid of any innervation, demonstrate that regulation of Kcnq4 expression in vestibular HCs is independent of innervation.

Transduction du signal sensoriel de l&#x26;#39;oreille interne des neurones aff&#xE9;rents et les cellules cili&#xE9;es (HCS) n&#xE9;cessite de nombreuses conductances ioniques. Le KCNQ4 voltage-d&#xE9;pendants des canaux potassiques de type M est pens&#xE9; pour d&#xE9;finir le potentiel de membrane de repos dans cochl&#xE9;aire HC. Nous d&#xE9;crivons ici les profils d&#x26;#39;expression spatio-temporelle des KCNQ4 et les formes associ&#xE9;es d&#x26;#39;&#xE9;pissage alternatif dans le HC de labyrinthe vestibulaire. Immunod&#xE9;tection montage entier, qualitative et quantitative de RT-PCR ont &#xE9;t&#xE9; r&#xE9;alis&#xE9;es afin de caract&#xE9;riser les profils d&#x26;#39;expression de KCNQ4 transcriptions et des prot&#xE9;ines. Une expression topographique et une r&#xE9;gulation positive de KCNQ4 au cours du d&#xE9;veloppement a &#xE9;t&#xE9; observ&#xE9;e et a indiqu&#xE9; que KCNQ4 ne se limite pas &#xE0; une structure sp&#xE9;cifique soit vestibulaire ou type de cellule, mais elle est pr&#xE9;sente dans les calices aff&#xE9;rents, neurones du ganglion vestibulaire, et les deux de type I et type II HC. Parmi les quatre variantes d&#x26;#39;&#xE9;pissage alternatif, Kcnq4_v1 transcriptions ont &#xE9;t&#xE9; la forme pr&#xE9;dominante dans le HC, tandis que la variante Kcnq4_v3 &#xE9;tait majeur dans les neurones vestibulaires. Sensorische Signaltransduktion des Innenohrs afferenten Neuronen und Haarzellen (HCs) erfordert zahlreiche Ionische Conductances. KCNQ4 Spannung-Gating M-Typ Kaliumkanals ist vermutlich die ruhende Membran Potenzial bei cochlear HCs festgelegt. Hier beschreiben wir die raumzeitliche Ausdrucksmuster Kcnq4 und den zugeh&ouml;rigen alternative Splei&szlig;-Formen in die HCs vestibul&auml;ren Labyrinths. Von den vier alternative Splice-Varianten waren Kcnq4_v1 Abschriften die vorherrschende Form in der HCs, Kcnq4_v3 die gr&ouml;&szlig;ere Variante in den vestibul&auml;ren Neuronen war. Differenzielle quantitativer Ausdruck von Kcnq4_v1 und Kcnq4_v3 wurden jeweils in den Regionen Striolar und Extra-Striolar der Schlauch und Gleichgewichtsorgan entdeckt. Analyse der Rennm&auml;use und Ratten ergab Ergebnisse &auml;hnlich denen bei M&auml;usen, was darauf hindeutet, dass die raumzeitliche Ausdrucksmuster von Kcnq4 im vestibul&auml;ren System unter Nager erhalten bleibt erhalten. Analysen der vestibul&auml;ren HCs Bdnf bedingte Mutante M&auml;use, die frei von jeder Innervation, zeigen, dass Regulierung von Kcnq4 Ausdruck im vestibul&auml;ren HCs unabh&auml;ngig von Innervation.

Espressione differenziale quantitativa di Kcnq4_v1 e Kcnq4_v3 sono stati rilevati rispettivamente nelle regioni di striolar ed extra-striolar dell&#39;utricolo e sacculo. Analisi di gerbilli e ratti ha dato risultati simili a quelli ottenuti nel topo, suggerendo che il pattern di espressione spatiotemporal di Kcnq4 del sistema vestibolare &egrave; conservato tra i roditori. Transducci&#xF3;n de se&#xF1;ales de las neuronas sensoriales del o&#xED;do interno aferentes y las c&#xE9;lulas ciliadas (HCS) exige numerosos conductancias i&#xF3;nicas. El KCNQ4 dependiente de voltaje del tipo M canal de potasio se considera para establecer el potencial de reposo de la membrana coclear en el HC. Aqu&#xED; se describen los patrones de expresi&#xF3;n espacio-temporales de KCNQ4 y las formas alternativas de empalme asociados en la HC del laberinto vestibular. Inmunodetecci&#xF3;n Todo el montaje, cualitativo y cuantitativo de RT-PCR se realiz&#xF3; para caracterizar los patrones de expresi&#xF3;n de KCNQ4 transcripciones y las prote&#xED;nas. Una expresi&#xF3;n topogr&#xE1;fica y de la regulaci&#xF3;n positiva KCNQ4 durante el desarrollo se observ&#xF3; e indic&#xF3; que KCNQ4 no se limita a cualquiera de una estructura espec&#xED;fica vestibular o tipo de c&#xE9;lulas, sino que est&#xE1; presente en los c&#xE1;lices aferentes, neuronas del ganglio vestibulares, y tanto de tipo I y tipo II HC. De las cuatro variantes de empalme alternativo, Kcnq4_v1 transcripciones fueron la forma predominante en el HC, mientras que Kcnq4_v3 fue la variante importante en las neuronas vestibulares. La expresi&#xF3;n diferencial cuantitativo de Kcnq4_v1 y Kcnq4_v3 se detectaron, respectivamente, en las regiones striolar y extra-striolar del utr&#xED;culo y el s&#xE1;culo. An&#xE1;lisis de los jerbos y ratas produjo resultados similares a los obtenidos en ratones, lo que sugiere que el patr&#xF3;n de expresi&#xF3;n espacio-temporal de KCNQ4 en el sistema vestibular se conserva entre los roedores. Los an&#xE1;lisis de HC vestibular de los ratones mutantes condicionales BDNF, que carecen por completo de la inervaci&#xF3;n, demuestran que la regulaci&#xF3;n de la expresi&#xF3;n en KCNQ4 vestibular HC es independiente de la inervaci&#xF3;n.

Expresi&#xF3;n del desarrollo de KCNQ4 en las neuronas vestibulares y el epitelio neurosensorial

Loss of neurosensory cells of the ear, caused by genetic and non-genetic factors, is becoming an increasing problem as people age, resulting in deafness and vestibular disorders. Unveiling useful mechanisms of cell cycle regulation may offer the possibility to generate new cells out of remaining ones, thus providing the cellular basis to induce new hair cell differentiation in the mammalian ear. Here, we provide an overview of cell cycle regulating genes in general and of those studied in the ear in particular. We categorize those genes into regulators that act upstream of the pocket proteins and into those that act downstream of the pocket proteins. The three members of the pocket protein family essentially determine, through interaction with the eight members of the E2F family, whether or not the cell cycle will progress to the S-phase and thus cell division. The abundant presence of one or more members of these families in adult hair cells supports the notion that inhibition of cell cycle progression through these proteins is a lifelong process. Indeed, manipulating some of those proteins, unfortunately, leads to abortive entry into the cell cycle. Combined with recent success to induce hair cell differentiation through molecular therapy, these approaches may provide a viable strategy to restore lost hair cells in the inner ear.

Pocket proteins and cell cycle regulation in inner ear development.

Adult mammalian auditory hair cells (HCs) and their associated supporting cells (SCs) do not proliferate, and HC death leads to irreversible neurosensory hearing loss and balance impairment. In nonmammalian vertebrates, loss of HCs induces mitotic proliferation of adjacent nonsensory SCs and/or direct SC transdifferentiation to generate replacement cells. This results in the structural and functional recovery of the nonmammalian sensory systems. Potential replacement of mammalian auditory HCs, either by transplanting cells or by transforming existing cells through molecular therapy, has long been proposed. However, HC replacement strategies with clear therapeutic potential remain elusive. The retinoblastoma (pRB) family of cell cycle regulators, Rb1, Rbl1 (p107), and Rbl2 (p130), regulate the G(1)- to S-phase transition in proliferating cells. In the inner ear, the biochemical and molecular pathways involving pRBs, particularly p107 and p130, are relatively unexplored and their therapeutic suitability is yet to be determined. In this study, we analyzed the cochleae of adult p130 knock-out (p130(-/-)) mice and showed that lack of the p130 gene results in extra rows of HCs and SCs in the more apical regions of the cochlea. No evidence of transdifferentiation of these supernumerary SCs into HCs was observed in the p130(-/-) mouse. Nevertheless, unscheduled proliferation of SCs in the adult p130(-/-) cochlea coupled to downregulation of bona fide cell cycle inhibitors provides a mechanistic basis for the role of p130 as a regulator of SC and HC mitotic quiescence in the more apical regions of the cochlea. Interestingly, p130(-/-) mice exhibited nearly normal peripheral auditory sensitivity.
