The auditory sensory epithelium (organ of Corti), where sound waves are converted to electrical signals, comprises a highly ordered array of sensory receptor (hair) cells and nonsensory supporting cells. Here, we report that Sprouty2, which encodes a negative regulator of signaling via receptor tyrosine kinases, is required for normal hearing in mice, and that lack of SPRY2 results in dramatic perturbations in organ of Corti cytoarchitecture: instead of two pillar cells, there are three, resulting in the formation of an ectopic tunnel of Corti. We demonstrate that these effects are due to a postnatal cell fate transformation of a Deiters' cell into a pillar cell. Both this cell fate change and hearing loss can be partially rescued by reducing Fgf8 gene dosage in Spry2 null mutant mice. Our results provide evidence that antagonism of FGF signaling by SPRY2 is essential for establishing the cytoarchitecture of the organ of Corti and for hearing.

En este caso, nos informan de que Sprouty2, que codifica un regulador negativo de la se&#xF1;alizaci&#xF3;n a trav&#xE9;s de receptores de tirosina quinasas, se requiere de la audici&#xF3;n normal en ratones, y que la falta de resultados en SPRY2 perturbaciones dram&#xE1;ticas en &#xF3;rgano de Corti citoarquitectura: en lugar de dos c&#xE9;lulas de los pilares, hay tres, resultando en la formaci&#xF3;n de un t&#xFA;nel de Corti ect&#xF3;pico. Se demuestra que estos efectos se deben a una transformaci&#xF3;n del destino celular postnatal de c&#xE9;lulas de un Deiters en una celda de columna. Tanto este cambio de destino celular y la p&#xE9;rdida de la audici&#xF3;n puede ser parcialmente rescatado por la reducci&#xF3;n de la dosis g&#xE9;nica en Fgf8 Spry2 ratones mutantes nulos. Nuestros resultados proporcionan evidencia de que el antagonismo de los FGF se&#xF1;alizaci&#xF3;n por SPRY2 es esencial para establecer la citoarquitectura del &#xF3;rgano de Corti y de la audici&#xF3;n.

The auditory sensory epithelium is the specialized region of the cochlear epithelium that transduces sound. It is composed of a highly ordered, repeated array of mechanosensory hair cells and nonsensory supporting cells that run along the length of the cochlea. On the apical surface of the hair cells is a specialized structure called the hair bundle that deflects in response to sound vibration, resulting in depolarization of the hair cell and neurotransmitter release. Formation of the auditory sensory epithelium during embryogenesis involves strict control of both cell proliferation and cell patterning. Misregulation of these events can lead to congenital hearing loss, and damage to the auditory sensory epithelium during adult life can lead to adult-onset deafness. This paper reviews recent data on the formation of the auditory sensory epithelium during embryogenesis, the identification of components of the sound transduction apparatus, and advances in the treatment of hearing impairment.

Multiple signaling molecules, including Fibroblast Growth Factor (FGF) and Wnt, induce two patches of ectoderm on either side of the hindbrain to form the progenitor cell population for the inner ear, or otic placode. Here we report that in Spry1, Spry2 compound mutant embryos (Spry1â»/â»; Spry2â»/â» embryos), the otic placode is increased in size. We demonstrate that the otic placode is larger due to the recruitment of cells, normally destined to become cranial epidermis, into the otic domain. The enlargement of the otic placode observed in Spry1â»/â»; Spry2â»/â» embryos is preceded by an expansion of a Wnt8a expression domain in the adjacent hindbrain. We demonstrate that both the enlargement of the otic placode and the expansion of the Wnt8a expression domain can be rescued in Spry1â»/â»; Spry2â»/â» embryos by reducing the gene dosage of Fgf10. Our results define a FGF-responsive window during which cells can be continually recruited into the otic domain and uncover SPRY regulation of the size of a putative Wnt inductive center.
