A novel approach to high-throughput sequence deconvolution of on-bead small peptides (MW < 2000 Da) using on-target MALDI-TOF/TOF instrumentation is presented. Short peptides of pentamer and octamer length, covalently attached to TentaGel polystyrene beads through a photolabile linker, were placed onto the MALDI target, apportioned with suitable matrix (2,5-dihydroxybenzoic acid) and then hit with the instrument laser (Nd : YAG, 355 nm). This induced easy and highly reproducible photochemical cleavage, desorption (MS mode) and fragmentation (MS/MS mode). Peptide fragments were identified with a mass accuracy of 0.1 Da of the expected values. This technique significantly accelerates the sequence determination of positive peptide hits obtained from random combinatorial libraries when screening against biological targets, paving the way for a rapid and efficient method to identify molecular imaging ligands specific to pathological targets in cancer and other diseases.

Ein neuartiger Ansatz f&uuml;r Hochdurchsatz-Sequenz Entwindung der auf-Bead kleine Peptide (MW &lt; 2000 Da) mit zielgerechte MALDI-TOF/TOF Instrumentation wird vorgestellt. Kurze Peptide mit Pentamers und Octamer L&auml;nge, kovalent gebunden, TentaGel Polystyrol Perlen durch einen Linker Photolabile wurden auf MALDI vorbei, mit geeigneten Matrix (2, 5-Dihydroxybenzoic Acid) aufgeteilt und schlagen Sie dann mit dem Instrument-Laser (Nd: YAG, 355 nm). Dies induziert einfach und hoch reproduzierbare photochemische Spaltung, Desorption (MS-Modus) und Fragmentierung (MS/MS-Modus). Un nuovo approccio alla deconvoluzione di alto-rendimento sequenza di piccoli peptidi sul tallone (MW &lt; Da 2000) utilizzando strumentazione MALDI-TOF/TOF sul bersaglio &egrave; presentato. Brevi peptidi di lunghezza pentamero e octamer, covalentemente associata a TentaGel perle di polistirolo tramite un linker photolabile, sono stati collocati sul bersaglio MALDI, ripartiti con matrice adatto (2,5-dihydroxybenzoic acid) e poi ha colpiti con il laser strumento (Nd: YAG, 355 nm). Un nuevo enfoque de la deconvoluci&#xF3;n de la secuencia de alto rendimiento de grano en peque&#xF1;os p&#xE9;ptidos (PM &#x26;lt;2000 Da) con destino en la instrumentaci&#xF3;n MALDI-TOF/TOF se presenta. In pursuit of a single molecule with potential applications for both in vitro fluorescence microscopy and in vivo PET imaging, novel targeted (69/71)Ga/(68)Ga-protoporphyrin IX (PPIX) probes were developed. Optical analysis, fluorescence microscopy and radiolabeling with gallium-68 were performed to confirm potential applications. The use of a targeted-PPIX ring, in conjunction with (69/71)Ga or (68)Ga, eliminates the need for the attachment of multiple imaging tags, allowing for either in vitro fluorescent-based evaluation or in vivo nuclear-based imaging.
