Oxidative stress is the main etiological factor behind the pathogenesis of various diseases including inflammation, cancer, cardiovascular and neurodegenerative disorders. Due to the spin trapping abilities and various pharmacological properties of nitrones, their application as therapeutic agent has been gaining attention. Though the antioxidant properties of the nitrones are well known, the mechanism by which they modulate the cellular defense machinery against oxidative stress is not well investigated and requires further elucidation. Here, we have investigated the mechanisms of cytoprotection of the nitrone spin traps against oxidative stress in bovine aortic endothelial cells (BAEC). Cytoprotective properties of both the cyclic nitrone 5,5-dimethyl-pyrroline N-oxide (DMPO) and linear nitrone Î±-phenyl N-tert-butyl nitrone (PBN) against H(2)O(2)-induced cytotoxicity were investigated. Preincubation of BAEC with PBN or DMPO resulted in the inhibition of H(2)O(2)-mediated cytotoxicity and apoptosis. Nitrone-treatment resulted in the induction and restoration of phase II antioxidant enzymes via nuclear translocation of NF-E2-related factor 2 (Nrf-2) in oxidatively-challenged cells. Furthermore, the nitrones were found to inhibit the mitochondrial depolarization and subsequent activation of caspase-3 induced by H(2)O(2). Significant down-regulation of the pro-apoptotic proteins p53 and Bax, and up-regulation of the anti-apoptotic proteins Bcl-2 and p-Bad were observed when the cells were preincubated with the nitrones prior to H(2)O(2)-treatment. It was also observed that Nrf-2 silencing completely abolished the protective effects of nitrones. Hence, these findings suggest that nitrones confer protection to the endothelial cells against oxidative stress by modulating phase II antioxidant enzymes and subsequently inhibiting mitochondria-dependent apoptotic cascade.

Le stress oxydatif est le principal facteur &eacute;tiologique derri&egrave;re la pathogen&egrave;se des maladies diverses, notamment : inflammation, cancer, cardiovasculaire et neurod&eacute;g&eacute;n&eacute;ratives. En raison de la capacit&eacute; de pi&eacute;geage de spin et de diverses propri&eacute;t&eacute;s pharmacologiques des nitrones, leur application comme agent th&eacute;rapeutique a gagn&eacute; l&#39;attention. Bien que les propri&eacute;t&eacute;s antioxydantes de la nitrones sont bien connues, le m&eacute;canisme par lequel ils modulent la machinerie cellulaire de d&eacute;fense contre le stress oxydatif n&#39;est pas bien &eacute;tudi&eacute; et exige d&#39;autres &eacute;lucidation. Ici, nous avons &eacute;tudi&eacute; les m&eacute;canismes de la cytoprotection de pi&egrave;ges &agrave; spin les nitrone contre le stress oxydatif dans les cellules endoth&eacute;liales aortiques bovines (CEAB). Propri&eacute;t&eacute;s cytoprotectrice des deux la nitrone cyclique 5,5-dim&eacute;thyl-pyrroline N-oxyde (ODMP) et lin&eacute;aire nitrone &alpha;-ph&eacute;nyl N-tert-butyl nitrone (PBN) contre H (2) O (2)-cytotoxicit&eacute; induite ont &eacute;t&eacute; &eacute;tudi&eacute;s. La pr&eacute;incubation de BAEC a Nea ou DMPO conduit &agrave; l&#39;inhibition de H (2) O (2)-m&eacute;diation de cytotoxicit&eacute; et l&#39;apoptose. Nitrone-traitement conduit &agrave; l&#39;induction et la restauration des enzymes de phase II antioxydant par translocation nucl&eacute;aire du facteur NF-E2-associ&eacute;s 2 (Nrf-2) dans les cellules par oxydation-contest&eacute;. En outre, les nitrones trouv&eacute;es pour inhiber la d&eacute;polarisation mitochondrique et activation subs&eacute;quente de la caspase-3 induite par le H(2)O(2). R&eacute;gulation &agrave; la baisse significative de la pro-apoptotic prot&eacute;ines p53 et Bax et r&eacute;gulation des prot&eacute;ines anti-apoptotiques Bcl-2 et p-Bad ont &eacute;t&eacute; observ&eacute;es lorsque les cellules ont &eacute;t&eacute; pr&eacute;incub&eacute;es avec les nitrones avant 2 H (2) O traitement. On a aussi observ&eacute; que la Nrf-2 silencieux abolit compl&egrave;tement les effets protecteurs des nitrones. Par cons&eacute;quent, ces r&eacute;sultats sugg&egrave;rent que nitrones conf&egrave;rent une protection aux cellules endoth&eacute;liales contre le stress oxydatif par modulation des enzymes antioxydantes de phase II et ensuite inhibant la cascade apoptotic de mitochondries-d&eacute;pendante.

Oxidativer Stress ist der wichtigste &auml;tiologische Faktor hinter der Pathogenese von verschiedenen Krankheiten, einschlie&szlig;lich Entz&uuml;ndung, Krebs, Herz-Kreislauf- und neurodegenerativen Erkrankungen. Aufgrund der Drehbeschleunigung-Trapping-F&auml;higkeiten und verschiedenen pharmakologischen Eigenschaften der Nitrones wurde ihre Anwendung als Therapeutikum Aufmerksamkeit gewinnt. Obwohl die antioxidativen Eigenschaften von den Nitrones bekannt sind, wird der Mechanismus, mit dem sie die Maschinerie der zellul&auml;ren Abwehr gegen oxidativen Stress modulieren, ist nicht gut untersucht und erfordert weitere Aufkl&auml;rung. Hier haben wir die Mechanismen der Cytoprotection der Nitrone Spin Traps gegen oxidativen Stress in bovine Aorten Endothelzellen (BAEC) untersucht. Cytoprotective Eigenschaften der beiden die zyklische Nitrone 5,5-Dimethyl-Pyrroline N-oxid (DMPO) und lineare Nitrone &alpha;-Phenyl-N-Tert-Butyl-Nitrone (PBN) gegen H (2) O (2)-induzierte Zytotoxizit&auml;t untersucht wurden. Preincubation von BAEC mit PBN oder DMPO f&uuml;hrte die Hemmung der H (2) O (2)-vermittelte Zytotoxizit&auml;t und Apoptose. Nitrone-Behandlung f&uuml;hrte die Induktion und Restaurierung der Phase II antioxidative Enzyme &uuml;ber nukleare Translokation des NF-E2-bezogenen Faktor 2 (Nrf-2) in den Zellen oxidativ gehandicapte. Dar&uuml;ber hinaus wurden die Nitrones gefunden, die mitochondriale Depolarisation und anschlie&szlig;ende Aktivierung von Caspase-3 induziert durch H(2)O(2) zu hemmen. Bedeutende Herunterregulation der Pro-apoptotischen Proteine p53 und Bax und bis-Regulierung der Anti-apoptotischen Proteine Bcl-2 und p-Bad wurden beobachtet, wenn die Zellen mit den Nitrones vor H (2) O 2-Behandlung preincubated waren. Es wurde auch beobachtet, dass die sch&uuml;tzende Wirkung von Nitrones Nrf-2 Unterdr&uuml;ckender vollst&auml;ndig abgeschafft werden. Diese Erkenntnisse empfehlen daher Nitrones Schutz zu den endothelialen Zellen gegen oxidativen Stress &uuml;bertragen, indem modulierenden Phase II antioxidative Enzyme und anschlie&szlig;end Hemmung Mitochondrien-abh&auml;ngige Apoptotic Kaskade.

Lo stress ossidativo &egrave; il fattore eziologico principale dietro la patogenesi di varie malattie tra cui infiammazioni, cancro, cardiovascolare e malattie neurodegenerative. Dovuto l&#39;abilit&agrave; di spin trapping e varie propriet&agrave; farmacologiche di nitroni, loro applicazione come agente terapeutico &egrave; stato guadagnando attenzione. Anche se le propriet&agrave; antiossidanti dei Nitroni sono ben note, il meccanismo mediante il quale essi modulano il meccanismo di difesa cellulare contro lo stress ossidativo non &egrave; ben studiato e richiede ulteriore delucidazione. Qui, abbiamo studiato i meccanismi della cytoprotection delle trappole spin nitrone contro lo stress ossidativo nelle cellule endoteliali aortiche bovina (BAEC). Propriet&agrave; citoprotettivo di entrambi il nitrone ciclico 5,5-dimetil-pirrolina N-ossido (DMPO) e nitrone lineare &alpha;-fenil-N-t-butil nitrone (PBN) contro H (2) O (2)-citotossicit&agrave; indotta sono state studiate. Preincubazione di BAEC con PBN o DMPO ha provocato l&#39;inibizione di H (2) O (2)-mediata citotossicit&agrave; e l&#39;apoptosi. Nitrone-trattamento ha portato l&#39;induzione e il restauro di enzimi antiossidanti di fase II via traslocazione nucleare del fattore NF-E2-correlati 2 (Nrf-2) nelle cellule agli sfidati. Inoltre, sono stati trovati i Nitroni per inibire la depolarizzazione mitocondriale e la successiva attivazione di caspase-3 indotta da H(2)O(2). Quindi, questi risultati suggeriscono che Nitroni conferiscono protezione alle cellule endoteliali contro lo stress ossidativo modulando enzimi antiossidanti di fase II e successivamente inibendo la cascata apoptotica mitocondrio-dipendente.

El estr&eacute;s oxidativo es el factor etiol&oacute;gico principal detr&aacute;s de la patogenia de varias enfermedades incluyendo la inflamaci&oacute;n, c&aacute;ncer, cardiovascular y enfermedades neurodegenerativas. Debido a las capacidades de captura de vuelta y varias propiedades farmacol&oacute;gicas de nitrones, su aplicaci&oacute;n como agente terap&eacute;utico ha ido ganando atenci&oacute;n. Aunque las propiedades antioxidantes de la nitrones son bien conocidas, el mecanismo por el cual modulan los mecanismos de defensa celular contra el estr&eacute;s oxidativo no se investiga bien y requiere mayor aclaraci&oacute;n. Aqu&iacute;, hemos investigado los mecanismos de citoprotecci&oacute;n de las trampas de la vuelta de nitrona contra el estr&eacute;s oxidativo en c&eacute;lulas endoteliales a&oacute;rticas bovinas (BAEC). Propiedades de citoprotector de ambos el nitrona c&iacute;clica 5, 5-dimetil-pyrroline n-&oacute;xido (DMPO) y nitrona lineal &alpha;-fenil N-tert-butil nitrona (PBN) contra H (2) O (2)-investigaron la citotoxicidad inducida. Preincubaci&oacute;n de BAEC con PBN o DMPO dio lugar a la inhibici&oacute;n de la H (2) O (2)-mediada la citotoxicidad y la apoptosis. Nitrona-tratamiento dio lugar a la inducci&oacute;n y la restauraci&oacute;n de enzimas de fase II antioxidantes via translocaci&oacute;n nuclear del factor NF-E2-related 2 (Nrf-2) en c&eacute;lulas de problemas insolubles. Adem&aacute;s, los nitrones encontradas para inhibir la despolarizaci&oacute;n mitocondrial y la subsecuente activaci&oacute;n de la caspasa-3 inducida por h. Se observ&oacute; significativa abajo-regulaci&oacute;n de la pro-apoptosis prote&iacute;nas p53 y Bax y para arriba-regulaci&oacute;n de las prote&iacute;nas anti-apopt&oacute;ticas Bcl-2 y p-Bad cuando las c&eacute;lulas se preincubated con el nitrones antes de 2 H (2) O tratamiento.
