TAL1/SCL is a master regulator of haematopoiesis whose expression promotes opposite outcomes depending on the cell type: differentiation in the erythroid lineage or oncogenesis in the T-cell lineage. Here, we used a combination of ChIP sequencing and gene expression profiling to compare the function of TAL1 in normal erythroid and leukaemic T cells. Analysis of the genome-wide binding properties of TAL1 in these two haematopoietic lineages revealed new insight into the mechanism by which transcription factors select their binding sites in alternate lineages. Our study shows limited overlap in the TAL1-binding profile between the two cell types with an unexpected preference for ETS and RUNX motifs adjacent to E-boxes in the T-cell lineage. Furthermore, we show that TAL1 interacts with RUNX1 and ETS1, and that these transcription factors are critically required for TAL1 binding to genes that modulate T-cell differentiation. Thus, our findings highlight a critical role of the cellular environment in modulating transcription factor binding, and provide insight into the mechanism by which TAL1 inhibits differentiation leading to oncogenesis in the T-cell lineage.

Differential genomic targeting of the transcription factor TAL1 in alternate haematopoietic lineages.

TAL1/SCL est un ma&#xEE;tre r&#xE9;gulateur de l&#x26;#39;h&#xE9;matopo&#xEF;&#xE8;se dont l&#x26;#39;expression favorise des r&#xE9;sultats oppos&#xE9;s selon le type de cellule: diff&#xE9;renciation dans la lign&#xE9;e &#xE9;rythro&#xEF;de ou l&#x26;#39;oncogen&#xE8;se dans la lign&#xE9;e des lymphocytes T. Ici, nous avons utilis&#xE9; une combinaison de s&#xE9;quen&#xE7;age et de ChIP profil d&#x26;#39;expression g&#xE9;nique de comparer la fonction de TAL1 dans des conditions normales et leuc&#xE9;miques &#xE9;rythro&#xEF;de des cellules T. L&#x26;#39;analyse des propri&#xE9;t&#xE9;s du g&#xE9;nome entier de liaison de TAL1 dans ces deux lign&#xE9;es h&#xE9;matopo&#xEF;&#xE9;tiques a r&#xE9;v&#xE9;l&#xE9; un nouvel &#xE9;clairage sur le m&#xE9;canisme par lequel les facteurs de transcription de s&#xE9;lectionner leurs sites de liaison dans des lign&#xE9;es de substitution. Notre &#xE9;tude montre un chevauchement limit&#xE9; dans le profil TAL1-liaison entre les deux types de cellules avec une pr&#xE9;f&#xE9;rence inattendu pour ETS et des motifs RUNX adjacentes &#xE0; E-boxes dans la lign&#xE9;e des lymphocytes T. TAL1/SCL ist ein master Regulator der Blutbildung, deren Ausdruck entgegengesetzte Ergebnisse je nach Zelle f&ouml;rdert: Differenzierung in der Erythroid Abstammung oder Onkogenese in der T-Zell-Linie. Hier haben wir verwendet eine Kombination von ChIP-Sequenzierung und Genexpression Profilerstellung, um die Funktion der TAL1 in normalen Erythroid und leuk&auml;mischen T-Zellen zu vergleichen. Analyse der genomweiten Bindungseigenschaften TAL1 in diesen zwei h&auml;matopoetischen Linien ergab neue Einblicke in den Mechanismus durch die, den Transkription Faktoren ihre Bindungsstellen in Alternative Linien ausw&auml;hlen. Unsere Studie zeigt begrenzte &Uuml;berlappung im Profil TAL1-Bindung zwischen den beiden Zelltypen mit einem unerwarteten Vorliebe f&uuml;r ETS und RUNX Motive neben E-Boxen in der T-Zell-Linie. Dar&uuml;ber hinaus zeigen wir, dass RUNX1 und ETS1 TAL1 interagiert und diese Transkriptionsfaktoren f&uuml;r TAL1 Bindung an Gene kritisch erforderlich sind, die T-Zell-Differenzierung zu modulieren. &#x4EE3;&#x66FF;&#x9020;&#x8840;&#x7CFB;&#x7D71;&#x306B;&#x304A;&#x3051;&#x308B;&#x8EE2;&#x5199;&#x56E0;&#x5B50;&#x306E;TAL1&#x306E;&#x30BF;&#x30FC;&#x30B2;&#x30C6;&#x30A3;&#x30F3;&#x30B0;&#x5DEE;&#x30B2;&#x30CE;&#x30E0;

TAL1/SCL es un regulador principal de la hematopoyesis cuya expresi&#xF3;n promueve resultados opuestos, dependiendo del tipo de c&#xE9;lula: la diferenciaci&#xF3;n en el linaje eritroide o oncog&#xE9;nesis en el linaje de c&#xE9;lulas T. En este caso, se utiliz&#xF3; una combinaci&#xF3;n de secuenciaci&#xF3;n de CHIP y perfiles de expresi&#xF3;n g&#xE9;nica para comparar la funci&#xF3;n de TAL1 en condiciones normales de eritroides y c&#xE9;lulas T leuc&#xE9;micas. An&#xE1;lisis de las propiedades de uni&#xF3;n en todo el genoma de TAL1 en estos dos linajes hematopoy&#xE9;ticos revel&#xF3; una nueva visi&#xF3;n del mecanismo por el cual los factores de transcripci&#xF3;n seleccionar a sus sitios de uni&#xF3;n en los linajes alternativos. Nuestro estudio demuestra la poca superposici&#xF3;n en el perfil de TAL1 vinculante entre los dos tipos de c&#xE9;lulas con una preferencia inesperada para ETS y motivos Runx adyacentes a E-boxes en el linaje de c&#xE9;lulas T. Por otra parte, nos muestran que TAL1 interact&#xFA;a con RUNX1 y ETS1, y que estos factores de transcripci&#xF3;n son cr&#xED;ticamente necesarios para la uni&#xF3;n de los genes que modulan la diferenciaci&#xF3;n de c&#xE9;lulas T TAL1. Por lo tanto, nuestros resultados ponen de relieve un papel cr&#xED;tico del ambiente celular en el factor de transcripci&#xF3;n de modulaci&#xF3;n obligatoria, y proporcionar informaci&#xF3;n sobre el mecanismo por el cual TAL1 inhibe la diferenciaci&#xF3;n que conduce a la oncog&#xE9;nesis en el linaje de c&#xE9;lulas T.
