Despite the successful outcome of anti-D prophylaxis program, alloimmunization still occurs. The aim of this study was to examine the specificity and isotype of anti-Rh antibodies in plasma samples of Rh negative alloimmunized individuals and to study the same parameters in lymphoblastoid cell lines (LCLs) generated from the same donors. Specificity of anti-Rh antibodies was determined in plasma of nine alloimmunized subjects by direct hemagglutination using a panel of known RBC genotypes and isotype of specific antibodies were identified by an antigen specific ELISA. Similar methods were employed to determine specificity and isotype of antibodies produced by Rh specific LCLs established from four donors. LCLs were generated by Epstein-Barr virus transformation of peripheral blood mononuclear cells isolated from each donor followed by their culture over a feeder of human fetal fibroblasts. Upon emergence of lymphoblastoid cells, culture supernatants were assayed for presence of Rh specific antibody by hemagglutination assay. Anti-D was the predominant antibody in both plasma samples and among the 128 established LCLs; however, antibodies to other Rh specificities namely C and E were also produced. The isotype of anti-Rh antibody in all plasma samples was found to be IgG, predominantly IgG1, combined in 7 samples with IgM. Similarly 76%, 9.2% and 14.8% of LCLs were determined to produce antibody of IgG, IgM and of both isotypes, respectively. The data supported that the D antigen is the immunodominant component of the Rh system as indicated by the in vitro and in vivo profiles of Rh specificities in our alloimmunized subjects.

Specificity and isotype of Rh specific antibodies produced by human B-cell lines established from alloimmunized Rh negative women.

Malgr&eacute; la r&eacute;ussite du programme de prophylaxie anti-d, allo-immunisation persiste. Le but de cette &eacute;tude &eacute;tait d&#39;examiner la sp&eacute;cificit&eacute; et l&#39;isotype d&#39;anticorps anti-Rh dans les &eacute;chantillons de plasma des individus Rh alloimmunized n&eacute;gatif et d&#39;&eacute;tudier les m&ecirc;mes param&egrave;tres dans des lign&eacute;es cellulaires lymphoblastiques (L.i.c.) produites des m&ecirc;mes donneurs. Sp&eacute;cificit&eacute; des anticorps anti-Rh a &eacute;t&eacute; d&eacute;termin&eacute;e dans le plasma des neuf alloimmunized sujets par h&eacute;magglutination directe utilisant un panel de g&eacute;notypes connus de RBC et isotype d&#39;anticorps sp&eacute;cifiques ont &eacute;t&eacute; identifi&eacute;s par une technique ELISA sp&eacute;cifique de l&#39;antig&egrave;ne. Des m&eacute;thodes similaires ont &eacute;t&eacute; utilis&eacute;es pour d&eacute;terminer la sp&eacute;cificit&eacute; et l&#39;isotype d&#39;anticorps produits par Rh L.i.c. sp&eacute;cifique &eacute;tabli &agrave; partir de quatre bailleurs de fonds. L.i.c. ont &eacute;t&eacute; g&eacute;n&eacute;r&eacute;s par le virus Epstein - Barr transformation des cellules mononucl&eacute;aires de sang p&eacute;riph&eacute;rique isol&eacute;e de chaque donateur suivie de leur culture sur une mangeoire de fibroblastes foetus humains. D&egrave;s l&#39;apparition de cellules lymphoblasto&iuml;des, surnageants de culture ont &eacute;t&eacute; analys&eacute;s pour la pr&eacute;sence des anticorps sp&eacute;cifiques Rh par test d&#39;h&eacute;magglutination. Anti-d a &eacute;t&eacute; l&#39;anticorps pr&eacute;dominant dans les deux &eacute;chantillons de plasma et parmi les 128 L.i.c. &eacute;tablis ; Toutefois, les anticorps d&#39;autres sp&eacute;cificit&eacute;s Rh&eacute;sues &agrave; savoir C et E ont &eacute;galement &eacute;t&eacute; produites. L&#39;isotype d&#39;anticorps anti-Rh dans tous les &eacute;chantillons de plasma s&#39;est av&eacute;r&eacute; pour &ecirc;tre des IgG, surtout IgG1, combin&eacute; dans 7 &eacute;chantillons avec IgM. De m&ecirc;me, 76 %, 9,2 % et 14,8 % des L.i.c. ont &eacute;t&eacute; d&eacute;termin&eacute;s pour produire des anticorps d&#39;IgG, IgM et des deux isotypes, respectivement. Les donn&eacute;es prises en charge que l&#39;antig&egrave;ne D est le composant d&#39;immunodominante du syst&egrave;me Rh, comme indiqu&eacute; par les profils in vitro et in vivo des sp&eacute;cificit&eacute;s Rh&eacute;sues dans nos sujets alloimmunized.

Nonostante il buon esito del programma di profilassi anti-d, alloimmunizzazione si verifica ancora. Lo scopo di questo studio era di esaminare la specificit&agrave; e l&#39;isotipo di anticorpi anti-Rh in campioni di plasma di individui Rh negativi alloimmunized e di studiare gli stessi parametri in linee cellulari linfoblastoidi (LCLs) generati da donatori stessi. Specificit&agrave; degli anticorpi anti-Rh &egrave; stato determinato nel plasma di nove soggetti alloimmunized di emoagglutinazione diretta usando un pannello di genotipi di RBC noti e isotype degli anticorpi specifici sono stati individuati da un antigene specifico ELISA. Metodi simili sono stati impiegati per determinare la specificit&agrave; e l&#39;isotipo di anticorpi prodotti da Rh LCLs specifici stabiliti da quattro donatori. LCLs sono stati generati dal virus Epstein - Barr trasformazione di cellule mononucleari di sangue periferico isolato da ogni donatore seguita dalla loro cultura sopra un alimentatore di fibroblasti fetali umani. All&#39;emersione delle cellule linfoblastoidi, supernatanti di cultura sono stati analizzati per la presenza di anticorpi specifici Rh test di emoagglutinazione. Anti-d &egrave; stato l&#39;anticorpo predominante in entrambi i campioni di plasma e tra i 128 LCLs stabilito; Tuttavia, gli anticorpi di altre specificit&agrave; Rh, vale a dire la C e la E anche sono state prodotte. L&#39;isotipo dell&#39;anticorpo RH in tutti i campioni di plasma &egrave; stato trovato per essere IgG, prevalentemente IgG1, combinato in 7 campioni con IgM. A pesar del &eacute;xito del programa de profilaxis anti-d, aloinmunizaci&oacute;n todav&iacute;a ocurre. El objetivo de este estudio era examinar la especificidad y el isotipo de anticuerpos anti-Rh en muestras de plasma de los individuos Rh negativo alloimmunized y estudiar los mismos par&aacute;metros en l&iacute;neas de c&eacute;lulas linfoblastoides (LCLs) generados a partir de los donantes de la mismos. Especificidad de los anticuerpos anti-Rh se determin&oacute; en plasma de nueve temas alloimmunized por hemoaglutinaci&oacute;n directa usando un panel de genotipos conocidos de RBC e isotipo de anticuerpos espec&iacute;ficos fueron identificados por un ELISA espec&iacute;fica de ant&iacute;geno. Se emplearon m&eacute;todos similares para determinar la especificidad y el isotipo de anticuerpos producidos por Rh LCLs espec&iacute;ficos establecidos de cuatro donantes. LCLs fueron generados por el virus Epstein - Barr transformaci&oacute;n de c&eacute;lulas mononucleares de sangre perif&eacute;rica aislada de cada donante, seguido por su cultura sobre un comedero de fibroblastos fetales humanos. Sobre la aparici&oacute;n de c&eacute;lulas linfoblastoides, sobrenadante de cultivo fueron analizada para la presencia de anticuerpos espec&iacute;ficos Rh por hemaglutinaci&oacute;n. Anti-d fue el anticuerpo predominante en ambas muestras de plasma y entre los 128 LCLs establecidos; sin embargo, tambi&eacute;n se produjeron anticuerpos contra otras especificidades Rh, es decir, C y E. El isotipo de anticuerpo anti-Rh en todas las muestras de plasma fue encontrado para ser IgG, predominante IgG1, combinado en 7 muestras con IgM. Asimismo, 76%, 9,2% y 14,8% de LCLs determinaron para producir anticuerpos de IgG, IgM y de ambos isotipos, respectivamente. Los datos de apoyaron que el ant&iacute;geno D es el componente inmunodominante del sistema Rh seg&uacute;n lo indicado por los perfiles in vitro e in vivo de Rh especificidades en nuestras materias alloimmunized.
