Oncolytic viruses constitute a promising therapy against malignant gliomas (MGs). However, virus-induced type I IFN greatly limits its clinical application. The kinase mammalian target of rapamycin (mTOR) stimulates type I IFN production via phosphorylation of its effector proteins, 4E-BPs and S6Ks. Here we show that mouse embryonic fibroblasts and mice lacking S6K1 and S6K2 are more susceptible to vesicular stomatitis virus (VSV) infection than their WT counterparts as a result of an impaired type I IFN response. We used this knowledge to employ a pharmacoviral approach to treat MGs. The highly specific inhibitor of mTOR rapamycin, in combination with an IFN-sensitive VSV-mutant strain (VSV(DeltaM51)), dramatically increased the survival of immunocompetent rats bearing MGs. More importantly, VSV(DeltaM51) selectively killed tumor, but not normal cells, in MG-bearing rats treated with rapamycin. These results demonstrate that reducing type I IFNs through inhibition of mTORC1 is an effective strategy to augment the therapeutic activity of VSV(DeltaM51).

Vesicular stomatitis virus oncolysis is potentiated by impairing mTORC1-dependent type I IFN production.

Les virus oncolytiques constitue une th&#xE9;rapie prometteuse contre les gliomes malins (MSG). Toutefois, induite par le virus de type I IFN limite grandement son application clinique. La cible mammalienne de la rapamycine kinase (mTOR) stimule la production d&#x26;#39;IFN de type I via la phosphorylation de ses prot&#xE9;ines effectrices, 4E-BP et S6Ks. Ici, nous montrons que les fibroblastes embryonnaires de souris et souris d&#xE9;pourvues S6K1 et S6K2 sont plus sensibles aux virus de la stomatite v&#xE9;siculeuse (VSV) une infection que leurs homologues WT &#xE0; la suite d&#x26;#39;un type avec facult&#xE9;s affaiblies, je la r&#xE9;ponse IFN. Nous avons utilis&#xE9; ces connaissances pour utiliser une approche pharmacoviral pour traiter MG. L&#x26;#39;inhibiteur hautement sp&#xE9;cifique de mTOR rapamycine, en combinaison avec un IFN-sensible VSV-souche mutante (VSV (DeltaM51)), consid&#xE9;rablement accrue &#xE0; la survie des rats immunocomp&#xE9;tentes portant MG. Plus important encore, VSV (DeltaM51) tu&#xE9; de fa&#xE7;on s&#xE9;lective la tumeur, mais pas les cellules normales, dans MG-porteurs les rats trait&#xE9;s avec de la rapamycine. Ces r&#xE9;sultats d&#xE9;montrent que la r&#xE9;duction de type I IFN par l&#x26;#39;inhibition de mTORC1 est une strat&#xE9;gie efficace pour augmenter l&#x26;#39;activit&#xE9; th&#xE9;rapeutique du VSV (DeltaM51).

Onkolytische Viren bilden eine vielversprechende Therapie gegen maligne Gliome (MGs). Jedoch Virus-induzierte Typ h IFN Beschr&auml;nkungen erheblich seine klinische Anwendung. Stimuliert das Kinase S&auml;ugetier-Ziel von angiomiolipomas (mTOR) Typ I IFN-Produktion &uuml;ber Phosphorylierung der Effektor-Proteine, 4E-BPs und S6Ks. Hier zeigen wir, dass M&auml;useembryonalen und M&auml;usen fehlt S6K1 und S6K2 anf&auml;lliger f&uuml;r vesikul&auml;re Stomatitis-Virus (VSV) Infektion sind, als Gegenst&uuml;cke WT durch eine gest&ouml;rte Typ I IFN Antwort. Wir nutzten dieses Wissen um einen Pharmacoviral Ansatz zur Behandlung von MGs. Der hochspezifische Inhibitor des mTOR-angiomiolipomas, in Kombination mit einem IFN-Sensitive VSV-Mutant-Stamm (VSV(DeltaM51)), drastisch erh&ouml;ht das &Uuml;berleben von immunkompetenten Ratten mit MGs. Noch wichtiger ist, get&ouml;tet VSV(DeltaM51) selektiv Tumor, aber keine normalen Zellen, in MG-Lager-Ratten, die mit angiomiolipomas behandelt. Diese Ergebnisse zeigen, dass Typ I IFNs durch Hemmung der mTORC1 ist eine wirksame Strategie zur erg&auml;nzen der therapeutischen T&auml;tigkeit des VSV(DeltaM51).

Vesikul&auml;re Stomatitis-Virus, die Oncolysis von mTORC1-abh&auml;ngige beeintr&auml;chtigen potenzierte ist Typ I IFN-Produktion.

Il bersaglio dei mammiferi della chinasi del rapamycin (mTOR) stimola la produzione di IFN tramite la fosforilazione delle proteine effettrici, 4E-BPs e S6Ks di tipo I. Qui vi mostriamo che fibroblasti embrionali di topo e topi privi di S6K1 e S6K2 sono pi&ugrave; suscettibili di stomatite vescicolare (VSV) infezione che le loro controparti WT come conseguenza di un&#39;alterata risposta IFN di tipo I. Abbiamo usato questa conoscenza per adottare un approccio pharmacoviral per il trattamento di MGs. L&#39;altamente specifico inibitore di mTOR rapamycin, in combinazione con un ceppo sensibile IFN VSV-mutante (VSV(DeltaM51)), drammaticamente aumentato la sopravvivenza dei topi immunitarie cuscinetto MGs. Oncolytic viruses (OVs) have been engineered or selected for cancer cell-specific infection however, we have found that following intravenous administration of vesicular stomatitis virus (VSV), tumor cell killing rapidly extends far beyond the initial sites of infection. We show here for the first time that VSV directly infects and destroys tumor vasculature in vivo but leaves normal vasculature intact. Three-dimensional (3D) reconstruction of infected tumors revealed that the majority of the tumor mass lacks significant blood flow in contrast to uninfected tumors, which exhibit relatively uniform perfusion. VSV replication in tumor neovasculature and spread within the tumor mass, initiates an inflammatory reaction including a neutrophil-dependent initiation of microclots within tumor blood vessels. Within 6 hours of intravenous administration of VSV and continuing for at least 24 hours, we observed the initiation of blood clots within the tumor vasculature whereas normal vasculature remained clot free. Blocking blood clot formation with thrombin inhibitors prevented tumor vascular collapse. Our results demonstrate that the therapeutic activity of an OV can go far beyond simple infection and lysis of malignant cells.

Targeting tumor vasculature with an oncolytic virus.

Les virus oncolytiques (OVS) ont &#xE9;t&#xE9; con&#xE7;us ou s&#xE9;lectionn&#xE9;s pour le cancer des cellules sp&#xE9;cifiques d&#x26;#39;infection cependant, nous avons constat&#xE9; que suite &#xE0; l&#x26;#39;administration intraveineuse de virus de la stomatite v&#xE9;siculeuse (VSV), la destruction des cellules tumorales s&#x26;#39;&#xE9;tend rapidement au-del&#xE0; des sites initiaux d&#x26;#39;infection. Nous montrons ici pour la premi&#xE8;re fois que le VSV infecte directement et il d&#xE9;truit la vascularisation tumorale in vivo, mais laisse intacte vascularisation normale. Tridimensionnelle (3D) des tumeurs infect&#xE9;es a r&#xE9;v&#xE9;l&#xE9; que la majorit&#xE9; de la masse tumorale n&#x26;#39;a pas la circulation sanguine importante, contrairement &#xE0; des tumeurs non infect&#xE9;s, qui pr&#xE9;sentent perfusion relativement uniforme. La r&#xE9;plication du VSV dans n&#xE9;ovascularisation tumorale et la diss&#xE9;mination au sein de la masse tumorale, initie une r&#xE9;action inflammatoire, y compris une initiation des neutrophiles d&#xE9;pendant du microclots dans les vaisseaux sanguins tumoraux. Dans les 6 heures suivant l&#x26;#39;administration intraveineuse de VSV et continue pendant au moins 24 heures, nous avons observ&#xE9; l&#x26;#39;initiation de la formation de caillots sanguins &#xE0; l&#x26;#39;int&#xE9;rieur de la vascularisation de la tumeur alors vascularisation normale est rest&#xE9; un caillot libre. Nos r&#xE9;sultats d&#xE9;montrent que l&#x26;#39;activit&#xE9; th&#xE9;rapeutique d&#x26;#39;un OV peut aller bien au-del&#xE0; simple infection et une lyse des cellules malignes.

Cibler la vascularisation des tumeurs avec un virus oncolytique

Onkolytische Viren (OVs) entwickelt oder f&uuml;r Krebs zellspezifische Infektion jedoch ausgew&auml;hlt wurden, wir haben festgestellt, dass nach intraven&ouml;ser Verabreichung vesikul&auml;re Stomatitis-Virus (VSV), Tumorzellen t&ouml;ten schnell weit &uuml;ber die ersten Seiten der Infektion hinausgeht. Wir zeigen hier zum ersten Mal, dass VSV direkt infiziert und Tumor Gef&auml;&szlig;system in-vivo zerst&ouml;rt jedoch normal Gef&auml;&szlig;system intakt. Dreidimensionale (3D) Rekonstruktion von infizierten Tumoren ergab, dass die Mehrheit der Tumor Masse ermangelt bedeutende Blut im Gegensatz zu nicht infizierten Tumoren, flie&szlig;en die relativ einheitliche Perfusion aufweisen. VSV-Replikation im Tumor Neovasculature und Ausbreitung der Tumor-Masse, initiiert eine entz&uuml;ndliche Reaktion, einschlie&szlig;lich einer Neutrophilen-abh&auml;ngige-Initiation des Microclots innerhalb von Tumor-Blutgef&auml;&szlig;en. Innerhalb von 6 Stunden intraven&ouml;se Verabreichung von VSV und f&uuml;r mindestens 24 Stunden fortgesetzt beobachteten wir die Einleitung von Blutgerinnseln in den Tumor-Gef&auml;&szlig;system, w&auml;hrend normale Gef&auml;&szlig;system Blutgerinnsel frei blieben. Blutgerinnsel Bildung mit Thrombin-Inhibitoren blockieren verhindert Tumor Kreislauf Zusammenbruch. Unsere Ergebnisse zeigen, dass die therapeutische T&auml;tigkeit von einem OV weit &uuml;ber einfache Infektion und Lyse von malignen Zellen hinausgehen kann.

Virus oncol&#xED;ticos (VO) han sido dise&#xF1;ados o seleccionados para el c&#xE1;ncer de c&#xE9;lulas espec&#xED;ficas de infecci&#xF3;n sin embargo, hemos encontrado que despu&#xE9;s de la administraci&#xF3;n intravenosa de virus de la estomatitis vesicular (VSV), muerte celular del tumor r&#xE1;pidamente se extiende m&#xE1;s all&#xE1; de los sitios iniciales de la infecci&#xF3;n. Se muestra aqu&#xED; por primera vez que VSV directamente infecta y destruye la vasculatura del tumor in vivo, pero deja intacto vasculatura normal. Tridimensional (3D) de reconstrucci&#xF3;n de tumores infectados revel&#xF3; que la mayor&#xED;a de la masa tumoral carece de flujo de sangre significativa en contraste con los tumores no infectados, que exhiben perfusi&#xF3;n relativamente uniforme. Replicaci&#xF3;n VSV en neovasculatura tumor y propagaci&#xF3;n en la masa tumoral, inicia una reacci&#xF3;n inflamatoria que incluye una iniciaci&#xF3;n de neutr&#xF3;filos dependiente de microco&#xE1;gulos dentro de los vasos sangu&#xED;neos del tumor. Dentro de 6 horas de la administraci&#xF3;n intravenosa de VSV y continuando por lo menos 24 horas, se observ&#xF3; el inicio de la formaci&#xF3;n de co&#xE1;gulos sangu&#xED;neos dentro de la vasculatura del tumor, mientras que vasculatura normal se mantuvo libre de co&#xE1;gulos. El bloqueo de la formaci&#xF3;n de co&#xE1;gulo de sangre con inhibidores de la trombina impedido colapso vascular del tumor. Nuestros resultados demuestran que la actividad terap&#xE9;utica de un OV puede ir m&#xE1;s all&#xE1; de simple infecci&#xF3;n y lisis de las c&#xE9;lulas malignas.
