The paraventricular nucleus of the hypothalamus (PVH) plays a critical role in the regulation of autonomic, neuroendocrine, and behavioral activities. This understanding has come from extensive characterization of the PVH in rats, and for this mammalian species we now have a robust model of basic PVH neuroanatomy and function. However, in mice, whose use as a model research animal has burgeoned with the increasing sophistication of tools for genetic manipulation, a comparable level of PVH characterization has not been achieved. To address this, we employed a variety of fluorescent tract tracing and immunostaining techniques in several different combinations to determine the neuronal connections and cyto- and chemoarchitecture of the PVH in the commonly used C57BL/6J male mouse. Our findings reveal a distinct organization in the mouse PVH that is substantially different from the PVH of male rats. The differences are particularly evident with respect to the spatial relations of two principal neuroendocrine divisions (magnocellular and parvicellular) and three descending preautonomic populations in the PVH. We discuss these data in relation to what is known about PVH function and provide the work as a resource for further studies of the neuronal architecture and function of the mouse PVH.

Le noyau paraventriculaire de l&#x26;#39;hypothalamus (PVH) joue un r&#xF4;le essentiel dans la r&#xE9;gulation des activit&#xE9;s autonomes, neuroendocriniennes et comportementales. Cette compr&#xE9;hension est venu de la caract&#xE9;risation approfondie de la PVH chez les rats, et pour cette esp&#xE8;ce de mammif&#xE8;res, nous avons maintenant un mod&#xE8;le solide de PVH neuroanatomie de base et la fonction. Cependant, chez la souris, dont l&#x26;#39;utilisation comme un animal mod&#xE8;le de recherche a connu un essor avec la sophistication croissante des outils pour la manipulation g&#xE9;n&#xE9;tique, un niveau comparable de la caract&#xE9;risation des PVH n&#x26;#39;a pas &#xE9;t&#xE9; atteint. Nos r&#xE9;sultats r&#xE9;v&#xE8;lent une organisation distincte dans le PVH souris qui est sensiblement diff&#xE9;rente de la PVH chez des rats m&#xE2;les. Les diff&#xE9;rences sont particuli&#xE8;rement &#xE9;videntes en ce qui concerne les relations spatiales des deux principales divisions neuroendocrines (magnocellulaire et parvicellular) et trois descendants des populations preautonomic dans le PVH. Der Kern paraventricularis des Hypothalamus (PVH) spielt eine wichtige Rolle bei der Regulation des autonomen, neuroendokrine und Verhaltens-Aktivit&auml;ten. Dieses Verst&auml;ndnis hat umfangreiche Charakterisierung von PVH bei Ratten stammen und f&uuml;r diese Arten von S&auml;ugetieren haben wir nun ein stabiles Modell der grundlegenden PVH Neuroanatomie und Funktion. Bei M&auml;usen, deren Verwendung als Modell Forschung Tier mit die zunehmende Komplexit&auml;t der Tools f&uuml;r Genmanipulation Filmstars hat, hat man sich jedoch nicht mit den eine vergleichbare PVH-Charakterisierung erreicht. Um dieses Problem anzugehen, haben wir eine Vielzahl von fluoreszierenden Trakt Ablaufverfolgung und Paraffinschnitten Techniken in mehrere verschiedene Kombinationen zur Bestimmung der neuronalen Verbindungen und Zyto- und Chemoarchitecture von der PVH in der h&auml;ufig verwendeten m&auml;nnlichen C57BL/6J-Maus. Questa comprensione &egrave; venuto dall&#39;ampia caratterizzazione di PVH nei ratti, e per questa specie di mammiferi ora abbiamo un robusto modello di base di neuroanatomia PVH e funzione. Ectopic bone formation is a unique biologic entity--distinct from other areas of skeletal biology. Animal research models of ectopic bone formation most often employ rodent models and have unique advantages over orthotopic (bone) environments, including a relative lack of bone cytokine stimulation and cell-to-cell interaction with endogenous (host) bone-forming cells. This allows for relatively controlled in vivo experimental bone formation. A wide variety of ectopic locations have been used for experimentation, including subcutaneous, intramuscular, and kidney capsule transplantation. The method, benefits and detractions of each method are summarized in the following review. Briefly, subcutaneous implantation is the simplest method. However, the most pertinent concern is the relative paucity of bone formation in comparison to other models. Intramuscular implantation is also widely used and relatively simple, however intramuscular implants are exposed to skeletal muscle satellite progenitor cells. Thus, distinguishing host from donor osteogenesis becomes challenging without cell-tracking studies. The kidney capsule (perirenal or renal capsule) method is less widely used and more technically challenging. It allows for supraphysiologic blood and nutrient resource, promoting robust bone growth. In summary, ectopic bone models are extremely useful in the evaluation of bone-forming stem cells, new osteoinductive biomaterials, and growth factors; an appropriate choice of model, however, will greatly increase experimental success.

A theoretical inverse relationship exists between osteogenic (bone forming) and adipogenic (fat forming) mesenchymal stem cell (MSC) differentiation. This inverse relationship in theory partially underlies the clinical entity of osteoporosis, in which marrow MSCs have a preference for adipose differentiation that increases with age. Two pro-osteogenic cytokines have been recently studied that each also possesses antiadipogenic properties: Sonic Hedgehog (SHH) and NELL-1 proteins. In the present study, we assayed the potential additive effects of the biologically active N-terminus of SHH (SHH-N) and NELL-1 protein on osteogenic and adipogenic differentiation of human primary adipose-derived stromal cell (hASCs). We observed that both recombinant SHH-N and NELL-1 protein significantly enhanced osteogenic differentiation and reduced adipose differentiation across all markers examined (alkaline phosphatase, Alizarin red and Oil red O staining, and osteogenic gene expression). Moreover, SHH-N and NELL-1 directed signaling produced additive effects on the pro-osteogenic and antiadipogenic differentiation of hASCs. NELL-1 treatment increased Hedgehog signaling pathway expression; coapplication of the Smoothened antagonist Cyclopamine reversed the pro-osteogenic effect of NELL-1. In summary, Hedgehog and Nell-1 signaling exert additive effects on the pro-osteogenic and antiadipogenic differentiation of ASCs. These studies suggest that the combination cytokines SHH-N+NELL-1 may represent a viable future technique for inducing the osteogenic differentiation of MSCs.

Una relaci&oacute;n inversa te&oacute;rica existe entre osteog&eacute;nico (formadoras de hueso) y diferenciaci&oacute;n de c&eacute;lulas madre mesenquimales (MSC) adipogenic (formaci&oacute;n de grasa). Esta relaci&oacute;n inversa en teor&iacute;a parcialmente subyace la entidad cl&iacute;nica de osteoporosis, en que m&eacute;dula MSCs tienen una preferencia por diferenciaci&oacute;n adiposa que aumenta con la edad. Dos citoquinas pro-osteog&eacute;nicos han sido estudiadas recientemente que cada uno posee tambi&eacute;n propiedades antiadipogenic: prote&iacute;nas Sonic Hedgehog (SHH) y NELL-1. En el presente estudio, analizan los posibles efectos aditivos del N-terminal biol&oacute;gicamente activo de SHH (SHH-N) y NELL-1 prote&iacute;na en osteog&eacute;nico y adipogenic diferenciaci&oacute;n de humano c&eacute;lulas estromales adiposo derivado primario (hASCs). Observamos que tanto recombinante SHH-N y prote&iacute;na NELL-1 significativamente mayor diferenciaci&oacute;n osteog&eacute;nica y reducci&oacute;n la diferenciaci&oacute;n adiposa a trav&eacute;s de todos los marcadores examinados (fosfatasa alcalina, rojo de alizarina y aceite rojo O tinci&oacute;n y expresi&oacute;n g&eacute;nica osteog&eacute;nico). Por otra parte, SHH-N y NELL-1 dirigido se&ntilde;alizaci&oacute;n producen efectos aditivos en la diferenciaci&oacute;n osteog&eacute;nica-pro y antiadipogenic de hASCs. NELL-1 tratamiento aument&oacute; Hedgehog se&ntilde;alizaci&oacute;n expresi&oacute;n del camino; coapplication de la antagonista de alisado Ciclopamina invierte el efecto pro-osteog&eacute;nico de NELL-1. En Resumen, erizo y se&ntilde;alizaci&oacute;n ejercer efectos aditivos en la diferenciaci&oacute;n osteog&eacute;nica-pro y antiadipogenic de ASCs Nell-1. Estos estudios sugieren que las citoquinas de combinaci&oacute;n SHH-N + NELL-1 pueden representar una t&eacute;cnica futura viable para inducir la diferenciaci&oacute;n osteog&eacute;nica de MSCs.
