Les odeurs sont repr&eacute;sent&eacute;s par des mod&egrave;les sp&eacute;cifiques d&#39;activit&eacute; spatio-temporelle dans le bulbe olfactif des vert&eacute;br&eacute;s et son analogue d&#39;insecte, le lobe antennaire. Dans les circuits inhibiteurs d&#39;abeilles dans l&#39;AL sont impliqu&eacute;s dans le traitement des odeurs aux r&eacute;ponses de forme odeur aff&eacute;rente. GABA est connu comme un transmetteur inhibiteur dans le lobe antennaire, mais pas tous les interneurones sont GABAergiques. C&#39;est pourquoi nous avons cherch&eacute; &agrave; analyser le r&ocirc;le fonctionnel de l&#39;histamine transmetteur inhibiteur pour le traitement des odeurs chez l&#39;abeille AL.

Ger&uuml;che werden durch spezifische raumzeitlichen Aktivit&auml;tsmuster in den Riechkolben der Wirbeltiere und seine Insekt Analog, der Kopf Lappen dargestellt. Honigbienen hemmenden Schaltungen in der AL engagieren sich in der Verarbeitung von Ger&uuml;chen auf die Reaktionen der Shape-afferenten Geruch. GABA ist bekannt als einen hemmenden Transmitter in den Kopf Lappen, aber nicht alle Interneuronen sind GABAergic. Wir wollten daher die funktionale Rolle der hemmenden Transmitter Histamin f&uuml;r die Verarbeitung von Ger&uuml;chen in die Honigbiene AL zu analysieren.

Odors are represented by specific spatio-temporal activity patterns in the olfactory bulb of vertebrates and its insect analogue, the antennal lobe. In honeybees inhibitory circuits in the AL are involved in the processing of odors to shape afferent odor responses. GABA is known as an inhibitory transmitter in the antennal lobe, but not all interneurons are GABAergic. Therefore we sought to analyze the functional role of the inhibitory transmitter histamine for the processing of odors in the honeybee AL.

Los olores son representados por los patrones de actividad espec&iacute;fica de spatio-temporal en el bulbo olfativo de los vertebrados y su an&aacute;logo de insecto, l&oacute;bulo antenal. En circuitos inhibitorios de las abejas en AL est&aacute;n implicadas en el procesamiento de los olores a respuestas de forma aferente olor. GABA es conocido como un transmisor inhibitorio en el l&oacute;bulo antenal, pero no todas interneuronas son GABA&eacute;rgicas. To understand how odor information is represented and processed in the antennal lobe (AL) of Drosophila melanogaster, we have optically recorded glomerular calcium responses to single odors and odor mixtures from olfactory sensory neurons (OSNs) and projection neurons (PNs). Odor mixtures offer a good tool to analyze odor processing because experimental results can be tested against clear predictions. At the level of the OSNs, the representation of odor mixtures could be predicted from the response patterns of the components in most cases. PN responses to mixtures, however, provide evidences of interglomerular inhibition. Application of picrotoxin (PTX), an antagonist of GABA(A)-like receptors, enhanced odor responses, modified their temporal course, and eliminated mixture suppression at the PN level. Our results can be best explained by postulating the existence of at least two local networks in the fly AL: a glomerulus specific network, which includes excitatory and inhibitory connections and a PTX sensitive inhibitory global network that acts on all glomeruli with proportional strength to the global AL input.

Para entender c&oacute;mo informaci&oacute;n de olor es representado y procesado en el l&oacute;bulo antenal (AL) de Drosophila melanogaster, hemos registrado &oacute;pticamente las respuestas de calcio glomerular a olores individuales y mezclas de olor de neuronas sensoriales olfativas (OSNs) y las neuronas de proyecci&oacute;n (PNs). Mezclas de olor ofrecen una buena herramienta para analizar el procesamiento de olor porque los resultados experimentales pueden probarse contra predicciones claras. En el plano de las OSNs, la representaci&oacute;n de mezclas de olor puede predecirse de los patrones de respuesta de los componentes en la mayor&iacute;a de los casos. Las respuestas de la PN a mezclas, sin embargo, proporcionan evidencias de inhibici&oacute;n de la interglomerular. Aplicaci&oacute;n de picrotoxin (PTX), un antagonista del GABA (A)-como receptores, mejora las respuestas de olor, modificado su curso temporal y eliminado la supresi&oacute;n de la mezcla en el nivel de la PN. When an animal smells an odor, olfactory sensory neurons generate an activity pattern across olfactory glomeruli of the first sensory neuropil, the insect antennal lobe or the vertebrate olfactory bulb. Here, several networks of local neurons interact with sensory neurons and with output neurons--insect projection neurons, or vertebrate mitral/tufted cells. The extent and form of information processing taking place in these local networks has been subject of controversy. To investigate the role of local neurons in odor information processing we have used the calcium sensor G-CaMP to perform in vivo recordings of odor-evoked spatiotemporal activity patterns in five genetically defined neuron populations of the antennal lobe of Drosophila melanogaster: three distinct populations of local neurons (two GABAergic and one cholinergic), as well as sensory neurons and projection neurons. Odor-specific and concentration dependent spatiotemporal response patterns varied among neuron populations. Activity transfer differed along the olfactory pathway for different glomerulus-odor combinations: we found cases of profile broadening and of linear and complex transfer. Moreover, the discriminability between the odors also varied across neuron populations and was maximal in projection neurons. Discriminatory power increased with higher odor concentrations over a wide dynamic range, but decreased at the highest concentration. These results show the complexity and diversity of odor information processing mechanisms across olfactory glomeruli in the fly antennal lobe.

Modelli di risposta spatiotemporal dipendente odore specifico e la concentrazione variavano tra le popolazioni del neurone. Differiva per trasferimento attivit&agrave; lungo il percorso olfattivo per combinazioni differenti glomerulo-odore: abbiamo trovato casi di ampliare il profilo di trasferimento lineare e complesso. Inoltre, la discriminability tra gli odori anche variato attraverso popolazioni neurone ed era massima nei neuroni di proiezione. Potere discriminatorio aumentato con alte concentrazioni di odore sopra un&#39;ampia gamma dinamica, ma &egrave; diminuito alla massima concentrazione. Cuando un animal huele un olor, neuronas sensoriales olfativas generan un patr&oacute;n de actividad a trav&eacute;s de los glom&eacute;rulos olfativos del primero neuropil sensorial, el l&oacute;bulo antenal insecto o vertebrado bulbo olfativo. Aqu&iacute;, varias redes de neuronas locales interact&uacute;an con neuronas sensoriales y las neuronas de salida--proyecci&oacute;n insectos neuronas, o c&eacute;lulas mitrales/copetudo vertebradas. El alcance y la forma de tomar lugar en estas redes locales de procesamiento de informaci&oacute;n ha sido tema de controversia. Para investigar el papel de las neuronas locales en el procesamiento de la informaci&oacute;n del olor hemos utilizado el sensor de calcio G-campamento para realizar grabaciones en vivo de patrones de actividad spatiotemporal olor-evocado en cinco poblaciones de neuronas gen&eacute;ticamente definidos del l&oacute;bulo antenal de Drosophila melanogaster: tres poblaciones distintas de neuronas locales (dos GABA&eacute;rgico y uno colin&eacute;rgicos), as&iacute; como las neuronas sensoriales y las neuronas de proyecci&oacute;n. Patrones de respuesta spatiotemporal dependiente de olor espec&iacute;fico y concentraci&oacute;n variaron entre las poblaciones de la neurona. Transferencia de actividad diferenci&oacute; a lo largo de la v&iacute;a olfativa para combinaciones diferentes glom&eacute;rulo-olor: encontramos casos de ampliaci&oacute;n de perfil y de transferencia lineal y compleja. Por otra parte, la discriminabilidad entre los olores tambi&eacute;n vari&oacute; entre las poblaciones de neuronas y fue m&aacute;xima en las neuronas de proyecci&oacute;n. Poder discriminatorio aumentado con concentraciones m&aacute;s altas de olor en un amplio rango din&aacute;mico, pero disminuy&oacute; en la concentraci&oacute;n m&aacute;s alta. Trace conditioning is a form of classical conditioning, where a neutral stimulus (conditioned stimulus, CS) is associated with a following appetitive or aversive stimulus (unconditioned stimulus, US). Unlike classical delay conditioning, in trace conditioning there is a stimulus-free gap between CS and US, and thus a poststimulus neural representation (trace) of the CS is required to bridge the gap until its association with the US. The properties of such stimulus traces are not well understood, nor are their underlying physiological mechanisms. Using behavioral and physiological approaches, we studied appetitive olfactory trace conditioning in honeybees. We found that single-odor presentation created a trace containing information about odor identity. This trace conveyed odor information about the initial stimulus and was robust against interference by other odors. Memory acquisition decreased with increasing CS-US gap length. The maximum learnable CS-US gap length could be extended by previous trace-conditioning experience. Furthermore, acquisition improved when an additional odor was presented during the CS-US gap. Using calcium imaging, we tested whether projection neurons in the primary olfactory brain area, the antennal lobe, contain a CS trace. We found odor-specific persistent responses after stimulus offset. These post-odor responses, however, did not encode the CS trace, and perceived odor quality could be predicted by the initial but not by the post-odor response. Our data suggest that olfactory trace conditioning is a less reflexive form of learning than classical delay conditioning, indicating that odor traces might involve higher-level cognitive processes.

Mit Kalzium-Bildgebung, testeten wir, ob Projektion Neuronen im Bereich prim&auml;ren olfaktorischen Gehirn, der Kopf Ohrl&auml;ppchen, eine CS-Ablaufverfolgung enthalten. Traccia condizionata &egrave; una forma di condizionamento classico, dove uno stimolo neutro (stimolo condizionato, CS) &egrave; associato con una seguente appetitiva o aversive stimolo (stimolo unconditioned, noi). A differenza di condizionamento classico ritardo, nella traccia di condizionamento c&#39;&egrave; un divario tra Stati Uniti e CS privo di stimolo e quindi una rappresentazione neurale poststimulus (traccia) del CS &egrave; necessaria per colmare il divario fino alla sua associazione con gli Stati Uniti. Le propriet&agrave; di tali tracce di stimolo non sono ben compresi, n&eacute; sono i loro meccanismi fisiologici sottostanti. Utilizzando approcci comportamentali e fisiologici, abbiamo studiato condizionata traccia olfattiva appetitiva nelle API. Abbiamo trovato quel singolo-odore presentazione creata una traccia contenente le informazioni sull&#39;identit&agrave; di odore. Questa traccia convogliata odore informazioni lo stimolo iniziale ed era robusta contro le interferenze di altri odori. Acondicionado de rastro es una forma de condicionamiento cl&aacute;sico, donde un est&iacute;mulo neutro (est&iacute;mulo condicionado, CS) se asocia a un siguiente apetitivo o est&iacute;mulo aversivo (est&iacute;mulo incondicionado, U.S.). A diferencia de acondicionado de retardo cl&aacute;sica, en el rastro acondicionado all&iacute; es un espacio libre de est&iacute;mulo entre CS y Estados Unidos, y as&iacute; una representaci&oacute;n poststimulus de los nervios (rastro) de la CS se requiere para cerrar la brecha hasta su asociaci&oacute;n con los Estados Unidos. Las propiedades de tales huellas de est&iacute;mulo no son bien entendidas, ni son sus mecanismos fisiol&oacute;gicos subyacentes. Utilizando los enfoques conductuales y fisiol&oacute;gicos, estudiamos apetitivo acondicionado de rastro olfativo de las abejas. Encontramos esa presentaci&oacute;n de olor solo creada un rastro que contiene informaci&oacute;n sobre la identidad de olor. Este rastro transmiti&oacute; informaci&oacute;n de olor sobre el est&iacute;mulo inicial y fue firme contra la injerencia de otros olores. Adquisici&oacute;n de memoria disminuye con el aumento de longitud de brecha de CS-Estados Unidos. La longitud m&aacute;xima puede aprender de CS-US brecha podr&iacute;a extenderse por la experiencia previa del rastro-acondicionado. Adem&aacute;s, adquisici&oacute;n mejor&oacute; cuando un olor adicional se present&oacute; durante la brecha de CS-Estados Unidos. To sense myriad environmental odors, animals have evolved multiple, large families of divergent olfactory receptors. How and why distinct receptor repertoires and their associated circuits are functionally and anatomically integrated is essentially unknown. We have addressed these questions through comprehensive comparative analysis of the Drosophila olfactory subsystems that express the ionotropic receptors (IRs) and odorant receptors (ORs). We identify ligands for most IR neuron classes, revealing their specificity for select amines and acids, which complements the broader tuning of ORs for esters and alcohols. IR and OR sensory neurons exhibit glomerular convergence in segregated, although interconnected, zones of the primary olfactory center, but these circuits are extensively interdigitated in higher brain regions. Consistently, behavioral responses to odors arise from an interplay between IR- and OR-dependent pathways. We integrate knowledge on the different phylogenetic and developmental properties of these receptors and circuits to propose models for the functional contributions and evolution of these distinct olfactory subsystems.

Wir haben diese Fragen durch umfassende vergleichende Analyse der Taufliege olfaktorischen Subsysteme gerichtet, die die ionotrope Rezeptoren (IRs) und Odorant-Rezeptoren (ORs) Ausdr&uuml;cken. Wir identifizieren die Liganden f&uuml;r die meisten IR-Neuron-Klassen, offenbart ihre Spezifit&auml;t f&uuml;r ausgew&auml;hlte Amine und S&auml;uren, die erg&auml;nzt, die breiteren tuning von ORs f&uuml;r Ester und Alkohole. Para detectar miles de olores del ambiente, los animales han evolucionado las familias m&#xFA;ltiples, grandes cantidades de receptores olfativos divergentes. &#xBF;C&#xF3;mo y por qu&#xE9; repertorios distintos de los receptores y sus circuitos asociados son funcional y anat&#xF3;mica integrada es esencialmente desconocida. Hemos abordado estas cuestiones a trav&#xE9;s del an&#xE1;lisis comparativo global de los subsistemas de Drosophila olfativos que expresan los receptores ionotr&#xF3;picos (IRS) y receptores de olor (RUP). Nos identificamos ligandos para la mayor&#xED;a de las clases de neuronas IR, revelando su especificidad para las aminas y &#xE1;cidos selectos, que complementa la afinaci&#xF3;n m&#xE1;s amplio de las RUP para los &#xE9;steres y alcoholes. O IR y las neuronas sensoriales muestran la convergencia glomerular en la segregaci&#xF3;n, aunque interconectadas, las zonas del centro olfatorio principal, pero estos circuitos son ampliamente interdigitados en las regiones superiores del cerebro. Constantemente, las respuestas de comportamiento a los olores surgen de una interacci&#xF3;n entre IR-O y dependiente de las v&#xED;as.
