The hypothalamus is a central regulator of many behaviors that are essential for survival, such as temperature regulation, food intake and circadian rhythms. However, the molecular pathways that mediate hypothalamic development are largely unknown. To identify genes expressed in developing mouse hypothalamus, we performed microarray analysis at 12 different developmental time points. We then conducted developmental in situ hybridization for 1,045 genes that were dynamically expressed over the course of hypothalamic neurogenesis. We identified markers that stably labeled each major hypothalamic nucleus over the entire course of neurogenesis and constructed a detailed molecular atlas of the developing hypothalamus. As a proof of concept of the utility of these data, we used these markers to analyze the phenotype of mice in which Sonic Hedgehog (Shh) was selectively deleted from hypothalamic neuroepithelium and found that Shh is essential for anterior hypothalamic patterning. Our results serve as a resource for functional investigations of hypothalamic development, connectivity, physiology and dysfunction.

L&#x26;#39;hypothalamus est un r&#xE9;gulateur central de beaucoup de comportements qui sont essentiels pour la survie, tels que la r&#xE9;gulation de temp&#xE9;rature, l&#x26;#39;apport alimentaire et des rythmes circadiens. Nous avons identifi&#xE9; des marqueurs qui stablement &#xE9;tiquet&#xE9;s chaque noyau hypothalamique majeure sur l&#x26;#39;ensemble du cours de la neurogen&#xE8;se et construit un atlas d&#xE9;taill&#xE9; mol&#xE9;culaire de l&#x26;#39;hypothalamus en d&#xE9;veloppement. Comme une preuve de concept de l&#x26;#39;utilit&#xE9; de ces donn&#xE9;es, nous avons utilis&#xE9; ces marqueurs pour analyser le ph&#xE9;notype des souris chez lesquelles Sonic Hedgehog (Shh) a &#xE9;t&#xE9; s&#xE9;lectivement supprim&#xE9;s de neuro&#xE9;pith&#xE9;lium hypothalamique et a constat&#xE9; que Shh est essentiel pour la mod&#xE9;lisation hypothalamique ant&#xE9;rieure. Un Atlas g&#xE9;nomique de la souris hypothalamo d&#xE9;veloppement

Der Hypothalamus ist ein zentraler Regulator der viele Verhaltensweisen, die zum &Uuml;berleben, wie Temperaturregelung, Nahrungsaufnahme und Tagesrhythmus unverzichtbar sind. Die molekularen Bahnen, die hypothalamischen Entwicklung zu vermitteln sind jedoch weitgehend unbekannt. Um die Identifizierung von Genen, die in Entwicklungsl&auml;ndern Maus Hypothalamus ausgedr&uuml;ckt, f&uuml;hrten wir Microarray Analyse zu 12 verschiedenen Entwicklungsst&ouml;rungen Zeitpunkten. Wir haben dann Entwicklungsst&ouml;rungen in situ Hybridisierung f&uuml;r 1.045 Gene, die im Laufe der hypothalamischen Neurogenese dynamisch ausgedr&uuml;ckt wurden durchgef&uuml;hrt. Wir haben Markierungen, die stabil bezeichnet jede gro&szlig;e hypothalamischen Kern f&uuml;r den gesamten Verlauf der Neurogenese und konstruiert einen detaillierten molekularen Atlas des entwickelnden Hypothalamus identifiziert. Als Proof of Concept des Dienstprogramms dieser Daten haben wir diese Marker verwendet, um der Ph&auml;notyp von M&auml;usen zu analysieren, in denen Sonic Hedgehog (Shh) wurde selektiv aus hypothalamischen Neuroepithelium gel&ouml;scht und festgestellt, dass Shh f&uuml;r anterior Hypothalamus Musterung wichtig. L&#39;ipotalamo &egrave; un regolatore centrale di molti comportamenti che sono essenziali per la sopravvivenza, come la regolazione della temperatura, assunzione di cibo e ritmi circadiani. Tuttavia, le vie molecolari che mediano lo sviluppo dell&#39;ipotalamo sono in gran parte sconosciute. Per identificare i geni espressi nell&#39;ipotalamo di topo in via di sviluppo, abbiamo eseguito analisi microarray a 12 punti differenti di tempo dello sviluppo. El hipot&#xE1;lamo es un regulador central de muchos de los comportamientos que son esenciales para la supervivencia, como la regulaci&#xF3;n de la temperatura, la ingesta de alimentos y los ritmos circadianos. Sin embargo, las v&#xED;as moleculares que median en el desarrollo del hipot&#xE1;lamo son en gran parte desconocido. The anatomy of the mammalian thalamus is characterized by nuclei, which can be readily identified in postnatal animals. However, the molecular mechanisms that guide specification and differentiation of neurons in specific thalamic nuclei are still largely unknown, and few molecular markers are available for most of these thalamic subregions at early stages of development. We therefore searched for patterned gene expression restricted to specific mouse thalamic regions by in situ hybridization during the onset of thalamic neurogenesis (embryonic [E] days E10.5-E12.5). To obtain correct regional information, we used Shh as a landmark and compared spatial relationships with the zona limitans intrathalamica (Zli), the border of the p2 and p3 compartments of the diencephalon. We identified genes that are expressed specifically in the ventricular zone of the thalamic neuroepithelium and also identified a number of genes that already exhibited regional identity at E12.5. Although many genes expressed in the mantle regions of the thalamus at E12.5 showed regionally restricted patterns, none of these clearly corresponded to individual thalamic nuclei. We next examined gene expression at E15.5, when thalamocortical axons (TCAs) project from distinct regions of the thalamus and reach their targets in the cerebral cortex. Regionally restricted patterns of gene expression were again seen for many genes, but some regionally bounded expression patterns in the early postnatal thalamus had shifted substantially by E15.5. These findings reveal that nucleogenesis in the developing thalamus is associated with selective and complex changes in gene expression and provide a list of genes that may actively regulate the development of thalamic nuclei.

Diese Ergebnisse zeigen, dass Nucleogenesis in Entwicklungsl&auml;ndern Thalamus verbunden mit selektiven ist und komplexe &Auml;nderungen der Genexpression und stellen eine Liste von Genen, die aktiv die Entwicklung der Thalamuskernen Regeln kann.

La anatom&#xED;a del t&#xE1;lamo mam&#xED;feros se caracteriza por los n&#xFA;cleos, que pueden ser f&#xE1;cilmente identificados en los animales despu&#xE9;s del parto. Sin embargo, los mecanismos moleculares que gu&#xED;a de especificaciones y la diferenciaci&#xF3;n de las neuronas en los n&#xFA;cleos tal&#xE1;micos espec&#xED;ficos a&#xFA;n no se conocen, y pocos marcadores moleculares est&#xE1;n disponibles para la mayor&#xED;a de estas subregiones tal&#xE1;micas en las primeras etapas de desarrollo. Por lo tanto, busc&#xF3; la expresi&#xF3;n de genes patr&#xF3;n restringido a determinadas regiones del t&#xE1;lamo del rat&#xF3;n por hibridaci&#xF3;n in situ durante el inicio de la neurog&#xE9;nesis del t&#xE1;lamo (embrionaria [E] d&#xED;a E10.5 E12.5-). Para obtener informaci&#xF3;n regional correcta, hemos utilizado Shh como un hito y se comparan las relaciones espaciales con el intrathalamica limitante zona (ZLI), la frontera de los compartimentos p2 y p3 del dienc&#xE9;falo. Se identificaron genes que se expresan espec&#xED;ficamente en la zona ventricular del neuroepitelio del t&#xE1;lamo y tambi&#xE9;n identific&#xF3; un n&#xFA;mero de genes que ya exhiben la identidad regional en E12.5. Previous studies in the developing mouse thalamus have demonstrated that regional identity is established during early stages of development (Suzuki-Hirano et al. J. Comp. Neurol. 2011;519:528-543). However, the developing thalamus often shows little resemblance to the anatomical organization of the postnatal thalamus, making it difficult to identify genes that might mediate the organization of thalamic nuclei. We therefore analyzed the expression pattern of genes that we have identified as showing regional expression in embryonic thalamus on postnatal days (P) 6-8 by using in situ hybridization. We also identified several genes expressed only in the postnatal thalamus with restricted expression in specific nuclei. We first demonstrated the selective expression of neurotransmitter-related genes (vGlut2, vGAT, D2R, and HTR2C), identifying the neurotransmitter subtypes of cells in this region, and we also demonstrated selective expression of additional genes in the thalamus (Steel, Slitrk6, and AI852580). In addition, we demonstrated expression of genes specific to somatosensory thalamic nuclei, the ventrobasal posterior nuclei (VP); a visual thalamic nucleus, the dorsal lateral geniculate nucleus (dLGN); and an auditory thalamic nucleus, the medial geniculate body (MGB) (p57Kip, Nr1d1, and GFRÎ±1). We also identified genes that are selectively expressed in multiple different nuclei (Foxp2, Chst2, and EphA8). Finally, we demonstrated that several bone morphogenetic proteins (BMPs) and their inhibitors are expressed in the postnatal thalamus in a nucleus-specific fashion, suggesting that BMPs play roles in the postnatal thalamus unrelated to their known role in developmental patterning. Our findings provide important information for understanding the mechanisms of nuclear specification and connectivity during development, as well as their maintenance in adult thalamus.

Region-specific gene expression in early postnatal mouse thalamus.

Des &#xE9;tudes ant&#xE9;rieures dans le thalamus de souris en d&#xE9;veloppement ont d&#xE9;montr&#xE9; que l&#x26;#39;identit&#xE9; r&#xE9;gionale est &#xE9;tabli durant les premiers stades de d&#xE9;veloppement (Suzuki-Hirano et al J. Toutefois, le thalamus en d&#xE9;veloppement montre souvent peu de ressemblance avec l&#x26;#39;organisation anatomique du thalamus postnatale, ce qui rend difficile d&#x26;#39;identifier les g&#xE8;nes qui pourraient influer sur l&#x26;#39;organisation de noyaux thalamiques. Nous avons donc analys&#xE9; le profil d&#x26;#39;expression des g&#xE8;nes que nous avons identifi&#xE9;s comme montrant l&#x26;#39;expression r&#xE9;gionale dans le thalamus embryonnaire sur jours apr&#xE8;s la naissance (P) 6-8 en utilisant l&#x26;#39;hybridation in situ. Fr&uuml;here Studien in Entwicklungsl&auml;ndern Maus Thalamus haben gezeigt, dass regionaler Identit&auml;t in fr&uuml;hen Phasen der Entwicklung festgestellt wird (Suzuki-Hirano Et Al. J. Komp. Neurol. 2011; 519:528-543). Entwickelnden Thalamus zeigt jedoch oft wenig &Auml;hnlichkeit mit der anatomischen Organisation der postnatalen Thalamus, macht es schwierig, die Gene zu identifizieren, die die Organisation der Thalamuskernen vermitteln k&ouml;nnte. Wir analysierten daher die Muster des Ausdrucks der Gene, die wir identifiziert haben, als die regionale Ausdruck in embryonalen Thalamus an postnatalen Tagen (P) 6-8 mithilfe von in situ-Hybridisierung. Wir wurden auch Gene, die selektiv in mehreren verschiedenen Kernen (Foxp2, Chst2 und EphA8) ausgedr&uuml;ckt werden. Regionsspezifische Genexpression in fr&uuml;hen postnatalen Maus Thalamus.

Precedenti studi nel talamo di topo in via di sviluppo hanno dimostrato che l&#39;identit&agrave; regionale &egrave; stabilito durante le prime fasi dello sviluppo (Suzuki-Hirano et al., J. COMP. Neurol. 2011; 519:528-543). Tuttavia, il talamo in via di sviluppo spesso dimostra poca rassomiglianza all&#39;organizzazione anatomica del talamo postnatale, rendendo difficile identificare i geni che potrebbero mediare l&#39;organizzazione dei nuclei talamici. Abbiamo anche identificato geni che sono espressi in modo selettivo nei pi&ugrave; diversi nuclei (Foxp2, Chst2 ed EphA8). Infine, abbiamo dimostrato che molti bone morphogenetic proteins (BMP) e loro inibitori sono espressi nel talamo postnatale in modo specifico del nucleo, suggerendo che BMPs svolgono ruoli nel talamo postnatale non correlato al loro ruolo conosciuto nella serie evolutiva. Estudios previos en el t&#xE1;lamo del rat&#xF3;n en desarrollo han demostrado que la identidad regional se establece durante las primeras etapas de desarrollo (Suzuki-Hirano y otros J. Comp Neurol 2011;... 519:528-543). Sin embargo, el t&#xE1;lamo en desarrollo a menudo se muestra poco parecido con la organizaci&#xF3;n anat&#xF3;mica del t&#xE1;lamo despu&#xE9;s del parto, lo que hace dif&#xED;cil identificar los genes que podr&#xED;an mediar en la organizaci&#xF3;n de los n&#xFA;cleos del t&#xE1;lamo. Eso, hemos analizado el patr&#xF3;n de expresi&#xF3;n de los genes que hemos identificado como muestra la expresi&#xF3;n regional en el t&#xE1;lamo embrionaria en los d&#xED;as postnatales (P) 6-8 mediante el uso de la hibridaci&#xF3;n in situ. Tambi&#xE9;n se identificaron varios genes expresados &#x200B;&#x200B;s&#xF3;lo en el t&#xE1;lamo postnatal con expresi&#xF3;n restringida en n&#xFA;cleos espec&#xED;ficos. En primer lugar, demostraron la expresi&#xF3;n selectiva de genes relacionados con el neurotransmisor (vGlut2, vGAT, D2R, y HTR2C), la identificaci&#xF3;n de los subtipos de neurotransmisores de las c&#xE9;lulas en esta regi&#xF3;n, y tambi&#xE9;n ha demostrado la expresi&#xF3;n selectiva de genes adicionales en el t&#xE1;lamo (Acero, Slitrk6, y AI852580). Tambi&#xE9;n se identificaron los genes que se expresan selectivamente en los n&#xFA;cleos m&#xFA;ltiples distinto (FOXP2, Chst2 y EphA8). Por &#xFA;ltimo, hemos demostrado que varias prote&#xED;nas morfogen&#xE9;ticas &#xF3;seas (BMP) y sus inhibidores se expresan en el t&#xE1;lamo despu&#xE9;s del parto de una manera espec&#xED;fica de n&#xFA;cleo, lo que sugiere que las BMP juegan un papel en el t&#xE1;lamo despu&#xE9;s del parto no relacionado con su papel conocido en los patrones de desarrollo.
