We have created an immunology-related microarray chip containing primarily known genes with well-studied functional properties. By looking at known genes rather than expressed sequence tags, we hope to gain a better understanding of immunological pathways and how they work. The immunology gene chip contains genes from the following functional categories: T cell genes; B cell genes; dendritic cell genes; chemokine and cytokine genes; apoptosis genes; cell cycle genes; cell interaction genes; general hematology and immunology genes; and adhesion genes. We have also developed a novel three-color cDNA array platform in which arrays are directly visualized before hybridization, which allows us to select only high-quality chips for our experiments. In an effort to provide quantitative quality control for each array element as well as the entire chip, we have developed Matarray, a software package for image processing and data acquisition. With Matarray, we have built a quantitative data filtering and normalization scheme that has proved to be more efficient than the existing methods. Abbiamo creato un chip di microarray immunologia-correlati contenenti geni conosciuti principalmente con propriet&agrave; funzionali ben studiata. Guardando noti geni piuttosto che expressed sequence tags, speriamo di acquisire una migliore comprensione delle vie immunologiche e come funzionano. Hemos creado un chip de microarray relacionados con la inmunolog&iacute;a que contienen genes principalmente conocidos con propiedades funcionales muy estudiados. Mirando de genes conocidos en lugar de expressed sequence tags, esperamos obtener una mejor comprensi&oacute;n de v&iacute;as inmunol&oacute;gicas y c&oacute;mo funcionan. El chip de gene de Inmunolog&iacute;a contiene genes de las siguientes categor&iacute;as funcionales: genes de la c&eacute;lula T; Genes de c&eacute;lulas b; genes de c&eacute;lulas dendr&iacute;ticas; genes de quimioquinas y citoquinas; genes de la apoptosis; genes de ciclo celular; genes de la interacci&oacute;n de la c&eacute;lula; Hematolog&iacute;a general y genes de la inmunolog&iacute;a; y genes de adherencia. Tambi&eacute;n hemos desarrollado una plataforma de matriz de cDNA tricolor novela en la que las matrices se visualizan directamente antes de la hibridaci&oacute;n, que nos permite seleccionar los chips de alta calidad para nuestros experimentos. En un esfuerzo para proporcionar control cuantitativo de la calidad para cada elemento de la matriz, as&iacute; como el chip todo, hemos desarrollado Matarray, un paquete de software para la adquisici&oacute;n de datos y procesamiento de im&aacute;genes. Con Matarray, hemos construido un sistema de filtrado y normalizaci&oacute;n de datos cuantitativos que ha demostrado para ser m&aacute;s eficaz que los m&eacute;todos existentes. Type 1 diabetes (T1D) is a T-cell mediated autoimmune disease targeting the insulin-producing pancreatic beta cells. Naturally occurring FOXP3(+)CD4(+)CD25(high) regulatory T cells (T(regs)) play an important role in dominant tolerance, suppressing autoreactive CD4(+) effector T cell activity. Previously, in both recent-onset T1D patients and beta cell antibody-positive at-risk individuals, we observed increased apoptosis and decreased function of polyclonal T(regs) in the periphery. Our objective here was to elucidate the genes and signaling pathways triggering apoptosis in T(regs) from T1D subjects.

Apoptosis of CD4+ CD25(high) T cells in type 1 diabetes may be partially mediated by IL-2 deprivation.

Diab&#xE8;te de type 1 (DT1) est un T-cell disease auto-immune ciblant les cellules b&#xEA;ta productrices d&#x26;#39;insuline du pancr&#xE9;as. FOXP3 d&#x26;#39;origine naturelle (+) CD4 (+) CD25 (&#xE9;lev&#xE9;) des cellules T r&#xE9;gulatrices (T (regs)) jouent un r&#xF4;le important dans la tol&#xE9;rance dominante, la suppression CD4 autor&#xE9;actifs (+) l&#x26;#39;activit&#xE9; des cellules T effectrices. Typ 1 Diabetes (T1D) ist ein T-Zell-vermittelte Autoimmunerkrankung, die Ausrichtung der Insulin produzierenden pankreatischen Betazellen. Nat&uuml;rlich vorkommende FOXP3(+)CD4(+)CD25(high) regulatorische T-Zellen (T(regs)) spielen eine wichtige Rolle im dominanten Toleranz, autoreaktive CD4(+) Effektor T-Zellaktivit&auml;t unterdr&uuml;cken. Zuvor, in den letzten beginnende T1D Patienten und Beta-Zelle Antik&ouml;rper-positiven gef&auml;hrdeten Personen, wir beobachteten erh&ouml;hten Apoptose und verringerte Funktion der polyklonalen T(regs) in der Peripherie. Hier unser Ziel war es, die Gene und die Signalwege, die Ausl&ouml;sung von Apoptose im T(regs) von T1D Probanden zu erhellen.

The transcription factor Foxp3 is essential for the development of functional, natural Treg (nTreg), which plays a prominent role in self-tolerance. Suppressive Foxp3(+) Treg cells can be generated from naÃ¯ve T cells ex vivo, following TCR and TGF-beta1 stimulations. However, the molecular contributions from the different arms of these pathways leading to Foxp3 expression are not fully understood. TGF-beta1-activated Smad3 plays a major role in the expression of Foxp3, since TGF-beta1-induced-Treg generation from Smad3(-/-) mice is markedly reduced and abolished by inactivating Smad2. In the TCR pathway, deletion of Bcl10, which activates NF-kappaB, markedly reduces both IL-2 and Foxp3 production. However, partial rescue of Foxp3 expression occurs on addition of exogenous IL-2. TGF-beta1 significantly attenuates NF-kappaB binding to the Foxp3 promoter, while inducing Foxp3 expression. Furthermore, deletion of p50, a NF-kappaB subunit, results in increased Foxp3 expression despite a decline in the IL-2 production. We posit several TCR-NF-kappaB pathways, some increasing (Bcl10-IL-2-Foxp3) while others decreasing (p50-Foxp3) Foxp3 expression, with the former predominating. A better understanding of Foxp3 regulation could be useful in dissecting the cause of Treg dysfunction in several autoimmune diseases and for generating more potent TGF-beta1-induced-Treg cells for therapeutic purposes.

The role of NF-kappaB and Smad3 in TGF-beta-mediated Foxp3 expression.

TGF-beta1-activ&#xE9; Smad3 joue un r&#xF4;le majeur dans l&#x26;#39;expression de Foxp3, depuis TGF-beta1-induite Treg g&#xE9;n&#xE9;ration de Smad3 (- / -) des souris est nettement r&#xE9;duits et supprim&#xE9;s par l&#x26;#39;inactivation Smad2. Dans la voie du TCR, la suppression de Bcl10, qui active NF-kB, r&#xE9;duit de fa&#xE7;on marqu&#xE9;e &#xE0; la fois l&#x26;#39;IL-2 et de la production Foxp3. Cependant, le sauvetage partielle d&#x26;#39;expression Foxp3 se produit sur les plus de IL-2 exog&#xE8;ne. Unterdr&uuml;ckende Foxp3(+) Treg-Zellen k&ouml;nnen von naiven T-Zellen ex Vivo, nach TCR und TGF-beta1 Erregungen erzeugt werden. Allerdings sind die molekularen Beitr&auml;ge aus den verschiedenen Armen der diese &Uuml;bertragungswege bei Foxp3 Ausdruck nicht vollst&auml;ndig verstanden. TGF-beta1-aktiviert Smad3 spielt eine wichtige Rolle in der Auspr&auml;gung der Foxp3, da TGF-beta1-induzierte-Treg-Generation von Smad3(-/-) M&auml;usen ist deutlich reduziert und von inaktiven Smad2 abgeschafft. In die TCR-Pathway reduziert die Streichung des Bcl10, dem NF-KappaB aktiviert wird, deutlich Il und Foxp3 Produktion. Allerdings tritt teilweise Rettung der Foxp3 Ausdruck &uuml;ber den Zusatz von exogenen Il. CD4+CD25+(high) regulatory T cells (Tregs) play a pivotal role in the control of the immune response. A growing body of evidence suggests the reduced function of these cells in autoimmune diseases, including type 1 diabetes (T1D). Restoration of their function can potentially delay further disease development. In the present study, we have converted conventional effector T cells into induced Tregs (iTregs) in recent-onset (RO) T1D (n=9) and compared them with the same cells generated in controls (n=12) and in long-standing (LS) T1D subjects (n=9). The functional potential of in-vitro-generated Tregs was measured by using an in vitro proliferation assay. We noted that the suppressive potential of iTregs exceeded that of natural regulatory T cells (nTregs) only in the RO T1D subjects. We showed that iTregs from RO T1D subjects had increased expression of Foxp3, E3 ubiquitin ligase (ITCH) and TGF-beta-inducible early gene 1 (TIEG1) compared with control and LS T1D subjects. We also expanded natural, thymically derived Tregs (nTregs) and compared the functional ability of these cells between subject groups. Expanded cells from all three subject groups were suppressive. RO T1D subjects were the only group in which both iTregs and expanded Tregs were functional, suggesting that the inflammatory milieu impacts in vitro Treg generation. Future longitudinal studies should delineate the actual contribution of the stage of disease to the quality of in-vitro-generated Tregs.

RO T1D Themen waren die einzige Gruppe, in der sowohl die iTregs als auch die erweiterten Tregs funktional waren, was darauf hindeutet, dass das entz&uuml;ndliche Milieu, in-vitro-Treg-Generierung auswirkt. En el presente estudio, hemos convertido convencionales c&#xE9;lulas T efectoras en c&#xE9;lulas T reguladoras inducidas (iTregs) en el reciente comienzo (RO) DT1 (n = 9) y los compar&#xF3; con las mismas c&#xE9;lulas generadas en los controles (n = 12) y en el largo permanentes (LS) T1D sujetos (n = 9). El potencial funcional de in-vitro generado Tregs se midi&#xF3; mediante un ensayo in vitro de la proliferaci&#xF3;n.
