clk-1 encodes a demethoxyubiquinone (DMQ) hydroxylase that is necessary for ubiquinone biosynthesis. When Caenorhabditis elegans clk-1 mutants are grown on bacteria that synthesize ubiquinone (UQ), they are viable but have a pleiotropic phenotype that includes slowed development, behaviors, and aging. However, when grown on UQ-deficient bacteria, the mutants arrest development transiently before growing up to become sterile adults. We identified nine suppressors of the missense mutation clk-1(e2519), which harbors a Glu-to-Lys substitution. All suppress the mutant phenotypes on both UQ-replete and UQ-deficient bacteria. However, each mutant suppresses a different subset of phenotypes, indicating that most phenotypes can be uncoupled from each other. In addition, all suppressors restore the ability to synthesize exceedingly small amounts of UQ, although they still accumulate the precursor DMQ, suggesting that the presence of DMQ is not responsible for the Clk-1 phenotypes. We cloned six of the suppressors, and all encode tRNA(Glu) genes whose anticodons are altered to read the substituted Lys codon of clk-1(e2519). To our knowledge, these suppressors represent the first missense suppressors identified in any metazoan. The pattern of suppression we observe suggests that the individual members of the tRNA(Glu) family are expressed in different tissues and at different levels.

To date, two maternal-effect genes have been shown to have causative roles in recurrent hydatidiform moles (RHMs); NLRP7 that is mutated in 48-60% of patients with RHMs and C6orf221 (HUGO-approved nomenclature is now KHDC3L), a recently identified gene, that is mutated in 14% of patients with RHMs who are negative for NLRP7 mutations. We sequenced KHDC3L in 97 patients with RHMs and reproductive loss who are mostly negative for NLRP7 mutations. We identified three unrelated patients, each homozygous for one of the two protein-truncating mutations, a novel 4-bp deletion resulting in a frameshift, c.299_302delTCAA, p.Ile100Argfs*2, and a previously described 4-bp deletion, c.322_325delGACT, p.Asp108Ilefs*30, transmitted on a shared haplotype to three patients from different populations. We show that five HM tissues from one of these patients are diploid and biparental similar to HMs from patients with two defective NLRP7 mutations. Using immunofluorescence, we show that KHDC3L protein displays a juxta perinuclear signal and colocalizes with NLRP7 in lymphoblastoid cell lines from normal subjects. Using cell lines from patients, we demonstrate that the KHDC3L mutations do not change the subcellular localization of the protein in hematopoietic cells. Our data highlight the similarities between the two causative genes for RHMs, KHDC3L and NLRP7, in their subcellular localization, the parental contribution to the HM tissues caused by them, and the presence of several founder mutations and variants in both of them indicating positive selection and adaptation.European Journal of Human Genetics advance online publication, 12 December 2012; doi:10.1038/ejhg.2012.274.

Report of four new patients with protein-truncating mutations in C6orf221/KHDC3L and colocalization with NLRP7.

A ce jour, deux g&egrave;nes effet maternel montrent jouent un r&ocirc;le causal dans la Mole hydatiforme r&eacute;currentes (RHMs) ; NLRP7 qui est mut&eacute; dans les 48 &agrave; 60 % des personnes atteintes de RHMs et C6orf221 (approuv&eacute; par HUGO nomenclature est maintenant KHDC3L), un g&egrave;ne identifi&eacute; r&eacute;cemment, qui est mut&eacute; dans 14 % des diab&eacute;tiques RHMs qui sont n&eacute;gatifs pour les mutations NLRP7. Hasta la fecha, dos genes de efecto materno se ha demostrado que tienen un papel causal en hidatiformes recurrente (RHMs); NLRP7 que es transformado en 48-60% de los pacientes con RHMs y C6orf221 (nomenclatura aprobada por HUGO es ahora KHDC3L), un gen recientemente identificado, que es transformado en el 14% de los pacientes con RHMs que son negativos para las mutaciones NLRP7. Ordenamos KHDC3L en 97 pacientes con RHMs y p&eacute;rdida reproductiva que en su mayor&iacute;a son negativos para las mutaciones NLRP7. Se identificaron a tres pacientes sin relaci&oacute;n, cada uno homocig&oacute;ticos para una de las dos mutaciones de truncamiento de la prote&iacute;na, una canceladura de novela 4 puntos de ebullici&oacute;n, dando por resultado un frameshift, c.299_302delTCAA, p.Ile100Argfs*2 y una canceladura de 4 puntos de ebullici&oacute;n previamente descrito, c.322_325delGACT, p.Asp108Ilefs*30, transmitido en un haplotipo compartido a tres pacientes de diferentes poblaciones. Demostramos que los tejidos de cinco HM de uno de estos pacientes son diploides y biparental similar al HMs de pacientes con dos mutaciones de NLRP7 defectuosos. Mediante inmunofluorescencia, demostramos que la prote&iacute;na de KHDC3L muestra una se&ntilde;al de juxta perinuclear y colocalizes con NLRP7 en las l&iacute;neas de celulares linfoblastoides de sujetos normales. Utilizando l&iacute;neas celulares de pacientes, demostramos que las mutaciones KHDC3L no cambian la localizaci&oacute;n subcelular de la prote&iacute;na en c&eacute;lulas hematopoy&eacute;ticas.
