Leptin is an adipose tissue-derived secretory hormone that suppresses appetite by inhibition of neuropepeptide Y (NPY) gene expression in arcuate nucleus (ARC) in the hypothalamus. To investigate the transcriptional regulation of NPY gene by leptin, we carried out a luciferase assay using NPY gene promotor plasmid (NPY-luc) in NPY expressing cells such as N18TG2, NG108-15, and PC12 cells. In these cells, the NPY gene was transactivated by leptin through activation of leptin receptor. Leptin-induced transactivation was mediated through the 221-bp region of the NPY gene promotor, which possesses two putative STAT3 binding sites. To investigate the mechanism of in vivo suppression of NPY gene transcription in ARC by leptin, the effect of SOCS members on the leptin-induced transactivation of NPY gene was studied. In vivo SOCS2 and SOCS3 mRNAs were induced in mouse hypothalamus by leptin. Although leptin (125 ng/ml) induced significant increase in NPY gene transcriptional activity in mock-transfected cells, the leptin-induced NPY gene transcriptional activity was completely abolished in SOCS3-transfected cells. SOCS3 also suppressed the basal NPY gene transcription. These finding suggested that leptin inhibits NPY gene transcription in the hypothalamus in vivo and SOCS3 is a negative regulator of the NPY gene.

Transcriptional regulation of neuronal genes and its effect on neural functions: transcriptional regulation of neuropeptide Y gene by leptin and its effect on feeding.

La leptine est une hormone de s&eacute;cr&eacute;tion provenant du tissu adipeux qui supprime l&#39;app&eacute;tit par inhibition de la neuropepeptide l&#39;expression des g&egrave;nes Y (NPY) dans le noyau arqu&eacute; (ARC) dans l&#39;hypothalamus. Afin d&#39;&eacute;tudier la r&eacute;gulation transcriptionnelle du g&egrave;ne NPY de leptine, nous avons effectu&eacute; une analyse de luciferase utilisant le plasmide de promoteur de g&egrave;ne NPY (NPY-luc) de NPY exprimant des cellules comme les cellules PC12 N18TG2 et NG108-15. Dans ces cellules, le g&egrave;ne NPY a &eacute;t&eacute; transactivated par la leptine par l&#39;interm&eacute;diaire de l&#39;activation du r&eacute;cepteur de la leptine. La leptina &egrave; un ormone secretivo derivate da tessuto adiposo che sopprime l&#39;appetito di inibizione dell&#39;espressione del gene Y (NPY) nel nucleo arcuato (ARC) nell&#39;ipotalamo neuropepeptide. Per studiare la regolazione trascrizionale del gene NPY dalla leptina, abbiamo effettuato un&#39;analisi di luciferase usando plasmide di promotor gene NPY (NPY-luc) in NPY che esprimono le cellule come N18TG2, NG108-15 e le cellule PC12. In queste cellule il gene NPY &egrave; stato transactivated dalla leptina attraverso l&#39;attivazione del recettore della leptina. La leptina es una hormona secretora derivados de tejido adiposo que suprime el apetito por la inhibici&oacute;n de la expresi&oacute;n del gen Y (NPY) en el n&uacute;cleo arcuato (ARC) en el hipot&aacute;lamo de neuropepeptide. Para investigar la regulaci&oacute;n transcripcional del gen NPY por leptina, llevamos a cabo un ensayo de luciferasa utilizando pl&aacute;smido promotor de gene NPY (NPY-luc) en NPY expresando las c&eacute;lulas como las c&eacute;lulas PC12, NG108-15 y N18TG2. En estas c&eacute;lulas, el gene NPY fue transactivated por la leptina a trav&eacute;s de la activaci&oacute;n del receptor de la leptina. Transactivaci&oacute;n leptina-inducida fue mediado por la regi&oacute;n de 221-bp del promotor del gen NPY, que posee dos sitios de uni&oacute;n de STAT3 putativos. Para investigar el mecanismo de supresi&oacute;n in vivo de la transcripci&oacute;n de genes NPY en arco de leptina, se estudi&oacute; el efecto de los miembros de la Soc en la leptina-inducida de transactivaci&oacute;n del gen NPY. SOCS2 en vivo y SOCS3 fueron inducidos en el hipot&aacute;lamo de rat&oacute;n por la leptina. In Japan, the "bedridden state" is one of the most serious problems the aged face, and it is becoming a social problem. The main causes of the bedridden state are cerebrovascular disorders and bone fractures following falls. The purpose of this study was to predict risk factors for falls and resultant bone fracture due to osteoporosis. We explored mobility parameters for possible fall prevention. In order to examine the correlation between the risk of falling and resultant bone fracture due to osteoporosis, logistic regression analysis was performed between bone mass (independent variable) and various factors dependent variables: body mass index [BMI], body fat percentage, atherogenic index, presence of transformation-related osteoarthritis of knee, presence of transformation-related osteoarthritis of spine, maximum step length, single-leg stance with open eyes, and hip-joint flexion motion angle); predictive factors were then examined. Predictive factors were determined by the stepwise method. Subjects who could not perform the "single-leg stance with open eyes" test had a risk of falling and bone fracture 2.49 times as large as that of subjects who could. The "single-leg stance with open eyes" test may be considered a useful method for the early detection of the risk of falling and bone fracture associated with osteoporosis. CS (chondroitin sulfate) is a glycosaminoglycan species that is widely distributed in the extracellular matrix. To understand the physiological roles of enzymes involved in CS synthesis, we produced CSGalNAcT1 (CS N-acetylgalactosaminyltransferase 1)-null mice. CS production was reduced by approximately half in CSGalNAcT1-null mice, and the amount of short-chain CS was also reduced. Moreover, the cartilage of the null mice was significantly smaller than that of wild-type mice. Additionally, type-II collagen fibres in developing cartilage were abnormally aggregated and disarranged in the homozygous mutant mice. These results suggest that CSGalNAcT1 is required for normal CS production in developing cartilage.

The rapid increase in the number of genetically modified mouse strains has produced a high demand for their frozen spermatozoa from laboratories and mouse banking facilities. Historically, plastic straws have been used preferentially as containers for frozen mammalian spermatozoa because spermatozoa frozen in plastic straws have a high survival rate after thawing. However, plastic straws are more fragile and are used less often than the cryotubes used for conventional cell freezing. In this study, we sought to develop a new protocol for sperm freezing using cryotubes as the container to increase the accessibility of mouse sperm cryopreservation. Epididymal spermatozoa were collected from mature ICR or C57BL/6J (B6) males and were suspended in 18% raffinose and 3% skim milk solution. We then optimized the following conditions using the sperm survival rate as an index: 1) distance of cryotubes from the surface of the liquid nitrogen at freezing, 2) volume of the sperm suspension in the cryotube and 3) temperature of warming sperm during thawing. The best result was obtained when cryotubes containing 10 Âµl of sperm suspension were immersed 1 cm below the surface of the liquid nitrogen and then thawed at 50 C. The fertilization rates using spermatozoa frozen and thawed using this method were 63.1% in ICR mice and 28.2% in B6 mice. The latter rate was increased to 62.3% by adding reduced glutathione to the fertilization medium. After embryo transfer, 68% and 62% of the fertilized oocytes developed into normal offspring in the ICR and B6 strains, respectively. These results show that cryotubes can be used for cryopreservation of mouse spermatozoa under optimized conditions. This protocol is easy and reproducible, and it may be used in laboratories that do not specialize in sperm cryopreservation.

El r&#xE1;pido aumento en el n&#xFA;mero de cepas de ratones gen&#xE9;ticamente modificados ha producido una gran demanda de sus espermatozoides congelados de los laboratorios e instalaciones de rat&#xF3;n bancarias. Hist&#xF3;ricamente, las pajitas de pl&#xE1;stico se han utilizado preferentemente como contenedores de espermatozoides congelados y los mam&#xED;feros, porque los espermatozoides congelados en pajillas de pl&#xE1;stico tienen una alta tasa de supervivencia despu&#xE9;s de la descongelaci&#xF3;n. Sin embargo, pajitas de pl&#xE1;stico son m&#xE1;s fr&#xE1;giles y se utilizan con menos frecuencia que los criotubos utilizados para la congelaci&#xF3;n celular convencional. Because the genetic diversity of the laboratory mouse (Mus musculus) is very limited, wild-derived strains from this genus could provide invaluable experimental models for studies of mouse genetics and epigenetics such as quantitative trait locus analysis. However, such strains generally show poor reproductive performance under conventional husbandry conditions so their use for large-scale analyses has been limited. This study was undertaken to devise assisted reproductive technologies (ARTs) for the efficient production of offspring in two wild-derived strains, MSM/Ms and JF1/Ms (Mus musculus molossinus). First, as females of these strains are poor responders to equine chorionic gonadotropin (eCG) stimulation, we examined the efficiency of superovulation by injecting anti-inhibin serum followed by human chorionic gonadotropin (hCG). About four to six times more oocytes were ovulated compared with eCG-hCG treatment in both strains, reaching ~25-30 oocytes per female. Consequently, the procedures for in vitro fertilization using these superovulated oocytes and cryopreservation of embryos and spermatozoa could be optimized for both of the wild-derived strains. However, MSM/Ms embryos, but not JF1/Ms embryos, failed to develop to term after embryo transfer because of intrauterine death at mid- to late-gestation. We could overcome this obstacle by cotransfer of these embryos with those from laboratory strains combined with treatment of recipient females with an immunosuppressant (cyclosporin A). Thus, a series of ARTs essential for efficient production and preservation of the wild-derived strains were devised successfully. These technologies will facilitate systematic studies on mouse genetics and epigenetics using a wider range of genetic diversity than currently available in the genus Mus.

Efficient Production of Offspring from Japanese Wild-Derived Strains of Mice (Mus musculus molossinus) by Improved Assisted Reproductive Technologies.

Debido a la diversidad gen&#xE9;tica de los ratones de laboratorio (Mus musculus) es muy limitado, silvestres derivados de cepas de este g&#xE9;nero podr&#xED;an proporcionar valiosos modelos experimentales para el estudio de la gen&#xE9;tica del rat&#xF3;n y la epigen&#xE9;tica, como el an&#xE1;lisis cuantitativo rasgo de legitimaci&#xF3;n. Sin embargo, estas cepas muestran en general pobre desempe&#xF1;o reproductivo en las condiciones de cr&#xED;a convencionales por lo que su uso para el an&#xE1;lisis a gran escala ha sido limitada. Este estudio se realiz&#xF3; para dise&#xF1;ar tecnolog&#xED;as de reproducci&#xF3;n asistida (ART) para la producci&#xF3;n eficiente de los hijos en dos silvestres derivados de cepas, HSH / M y JF1/Ms (Mus musculus molossinus). En primer lugar, que las hembras de estas cepas son una respuesta pobre a la gonadotropina cori&#xF3;nica equina (eCG) la estimulaci&#xF3;n, se analiz&#xF3; la eficiencia de superovulaci&#xF3;n mediante la inyecci&#xF3;n de suero anti-inhibina seguido de gonadotropina cori&#xF3;nica humana (hCG).
