Cyanobacteria that grow above seawater salinity at temperatures above 45 degrees C have rarely been studied. Cyanobacteria of this type of thermo-halophilic extremophile were isolated from siliceous crusts at 40-45 degrees C in a geothermal seawater lagoon in southwest Iceland. Iceland Clone 2e, a Leptolyngbya morphotype, was selected for further study. This culture grew only at 45-50 degrees C, in medium ranging from 28 to 94 g L(-1) TDS, It showed 3 doublings 24 h(-1) under continuous illumination. This rate at 54 degrees C was somewhat reduced, and death occurred at 58 degrees C. A comparison of the 16S rDNA sequence with all others in the NCBI database revealed 2 related Leptolyngbya isolates from a Greenland hot spring (13-16 g L(-1) TDS). Three other similar sequences were from Leptolyngbya isolates from dry, endolithic habitats in Yellowstone National Park. All 6 formed a phylogenetic clade, suggesting common ancestry. These strains shared many similarities to Iceland Clone 2e with respect to temperature and salinity ranges and optima. Two endolithic Leptolyngbya isolates, grown previously at 23 degrees C in freshwater medium, grew well at 50 degrees C but only in saline medium. This study shows that limited genotypic similarity may reveal some salient phenotypic similarities, even when the related cyanobacteria are from vastly different and remote habitats.

Ecosystems can be conceptually thought of as interconnected environmental and metabolic systems, in which small molecules to macro-molecules interact through diverse networks. State-of-the-art technologies in post-genomic science offer ways to inspect and analyze this biomolecular web using omics-based approaches. Exploring useful genes and enzymes, as well as biomass resources responsible for anabolism and catabolism within ecosystems will contribute to a better understanding of environmental functions and their application to biotechnology. Here we present ECOMICS, a suite of web-based tools for ECosystem trans-OMICS investigation that target metagenomic, metatranscriptomic, and meta-metabolomic systems, including biomacromolecular mixtures derived from biomass. ECOMICS is made of four integrated webtools. E-class allows for the sequence-based taxonomic classification of eukaryotic and prokaryotic ribosomal data and the functional classification of selected enzymes. FT2B allows for the digital processing of NMR spectra for downstream metabolic or chemical phenotyping. Bm-Char allows for statistical assignment of specific compounds found in lignocellulose-based biomass, and HetMap is a data matrix generator and correlation calculator that can be applied to trans-omics datasets as analyzed by these and other web tools. This web suite is unique in that it allows for the monitoring of biomass metabolism in a particular environment, i.e., from macromolecular complexes (FT2DB and Bm-Char) to microbial composition and degradation (E-class), and makes possible the understanding of relationships between molecular and microbial elements (HetMap). Microbial mats containing the filamentous anoxygenic photosynthetic bacterium Chloroflexus aggregans develop at Nakabusa hot spring in Japan. Under anaerobic conditions in these mats, interspecies interaction between sulfate-reducing bacteria as sulfide producers and C. aggregans as a sulfide consumer has been proposed to constitute a sulfur cycle; however, the electron donor utilized for microbial sulfide production at Nakabusa remains to be identified. In order to determine this electron donor and its source, ex situ experimental incubation of mats was explored. In the presence of molybdate, which inhibits biological sulfate reduction, hydrogen gas was released from mat samples, indicating that this hydrogen is normally consumed as an electron donor by sulfate-reducing bacteria. Hydrogen production decreased under illumination, indicating that C. aggregans also functions as a hydrogen consumer. Small amounts of hydrogen may have also been consumed for sulfur reduction. Clone library analysis of 16S rRNA genes amplified from the mats indicated the existence of several species of hydrogen-producing fermentative bacteria. Among them, the most dominant fermenter, Fervidobacterium sp., was successfully isolated. This isolate produced hydrogen through the fermentation of organic carbon. Dispersion of microbial cells in the mats resulted in hydrogen production without the addition of molybdate, suggesting that simultaneous production and consumption of hydrogen in the mats requires dense packing of cells. We propose a cyclic electron flow within the microbial mats, i.e., electron flow occurs through three elements: S (elemental sulfur, sulfide, sulfate), C (carbon dioxide, organic carbon) and H (di-hydrogen, protons).

Tapetes microbianos que contiene la bacteria fotosint&eacute;tica anoxig&eacute;nica filamentosos Chloroflexus aggregans se convierten en Nakabusa termales en Jap&oacute;n. Bajo condiciones anaer&oacute;bicas en estas esteras, interacci&oacute;n entre las especies de Sulfato-reducci&oacute;n bacterias como productores de sulfuro y C. aggregans como un consumidor de sulfuro se ha propuesto constituir un ciclo de azufre; sin embargo, el donante del electr&oacute;n utilizado para la producci&oacute;n de sulfuro microbiana en Nakabusa queda identificarse. Para determinar este donante del electr&oacute;n y su fuente, ex situ experimental fue explorado incubaci&oacute;n de esteras. En presencia de molibdato, que inhibe la reducci&oacute;n del sulfato biol&oacute;gica, gas de hidr&oacute;geno fue liberado de estera muestras, indicando que este hidr&oacute;geno se consume normalmente como donante de electrones por Sulfato-reducci&oacute;n bacterias. Disminuci&oacute;n de la producci&oacute;n de hidr&oacute;geno bajo iluminaci&oacute;n, indicando aggregans C. tambi&eacute;n funciones como un consumidor de hidr&oacute;geno. Peque&ntilde;as cantidades de hidr&oacute;geno pueden se tambi&eacute;n han consumido para la reducci&oacute;n de azufre. Clon biblioteca an&aacute;lisis de 16S rRNA genes amplificados de las esteras indic&oacute; la existencia de varias especies de bacterias fermentativas producci&oacute;n de hidr&oacute;geno. Entre ellos, el fermentador m&aacute;s dominante, Fervidobacterium SP., fue correctamente aislado. Este aislante producido hidr&oacute;geno mediante la fermentaci&oacute;n de carbono org&aacute;nico. Dispersi&oacute;n de las c&eacute;lulas microbianas en las esteras dio lugar a la producci&oacute;n de hidr&oacute;geno sin la adici&oacute;n de molibdato, sugiriendo que la producci&oacute;n simult&aacute;nea y consumo de hidr&oacute;geno en las esteras requiere embalaje densa de las c&eacute;lulas. Proponemos un flujo c&iacute;clico de electrones en los Tapetes microbianos, es decir, flujo de electrones se produce a trav&eacute;s de tres elementos: S (azufre elemental, sulfuro, sulfato), C (di&oacute;xido de carbono, el carbono org&aacute;nico) y H (di-hidr&oacute;geno, protones).
