The RNA polymerase (pol) II and III human small nuclear RNA (snRNA) genes have very similar promoters and recruit a number of common factors. In particular, both types of promoters utilize the small nuclear RNA activating protein complex (SNAP(c)) and the TATA box binding protein (TBP) for basal transcription, and are activated by Oct-1. We find that SNAP(c) purified from cell lines expressing tagged SNAP(c) subunits is associated with Yin Yang-1 (YY1), a factor implicated in both activation and repression of transcription. Recombinant YY1 accelerates the binding of SNAP(c) to the proximal sequence element, its target within snRNA promoters. Moreover, it enhances the formation of a complex on the pol III U6 snRNA promoter containing all the factors (SNAP(c), TBP, TFIIB-related factor 2 (Brf2), and B double prime 1 (Bdp1)) that are sufficient to direct in vitro U6 transcription when complemented with purified pol III, as well as that of a subcomplex containing TBP, Brf2, and Bdp1. YY1 is found on both the RNA polymerase II U1 and the RNA polymerase III U6 promoters as determined by chromatin immunoprecipitations. Thus, YY1 represents a new factor that participates in transcription complexes formed on both pol II and III promoters.

La ARN polimerasa (pol) II y III humanos peque&ntilde;os nucleares (snRNA) genes de ARN tienen promotores muy similares y reclutan un n&uacute;mero de factores comunes. En particular, ambos tipos de promotores utilizan el peque&ntilde;o arn nuclear compleja prote&iacute;na activadora (SNAP(c)) y el TATA caja prote&iacute;na de uni&oacute;n (TBP) transcripci&oacute;n basal y se activan por Oct-1. Encontramos que SNAP(c) purificada a partir de l&iacute;neas celulares expresando etiquetados subunidades SNAP(c) se asocia con Yin Yang-1 (YY1), un factor implicado en la activaci&oacute;n y represi&oacute;n de la transcripci&oacute;n. YY1 recombinante acelera la Uni&oacute;n de SNAP(c) para el elemento de la secuencia proximal, su objetivo dentro de snRNA promotores. Por otra parte, mejora la formaci&oacute;n de un complejo en el promotor de snRNA pol III U6 que contiene todos los factores (SNAP(c), TBP, TFIIB relacionadas con el factor 2 (Brf2) y el primer doble de B 1 (Bdp1)) que son suficientes para dirigir la transcripci&oacute;n in vitro del U6 cuando se complementa con purificada pol III, as&iacute; como la de un subcomplex que contiene PDD, Brf2, y Bdp1. YY1 se encuentra en la ARN polimerasa II U1 y la ARN polimerasa III U6 promotores seg&uacute;n lo determinado por la cromatina immunoprecipitations. As&iacute;, YY1 representa un nuevo factor que participa en la transcripci&oacute;n complejos formados en ambos promotores de pol II y III.

Whereas the zebrafish retina has long been an important model system for developmental and genetic studies, little is known about the responses of the inner retinal neurons. Here we report single-unit ganglion cell recordings from 5- to 6-day-old zebrafish larvae. In wild-type larvae we identify at least five subtypes of ganglion cell responses to full-field illumination, with ON-OFF and ON-type cells predominating. In the nrc mutant retina, in which the photoreceptor terminals develop abnormally, we observe normal OFF responses but abnormal ON-OFF responses and no ON responses. Previously characterized as blind, these mutants lack an optokinetic reflex (OKR), but in another behavioral assay nrc mutant fish have near-normal responses to the offset of light and slow and sluggish responses to the onset of light. Pharmacological block of the ON pathway mimics most of the nrc visual defects. We conclude that the abnormal photoreceptor terminals in nrc mutants predominantly perturb the ON pathway and that the ON pathway is necessary to drive the OKR in larval zebrafish.

OFF ganglion cells cannot drive the optokinetic reflex in zebrafish.

Alors que la r&#xE9;tine du poisson z&#xE8;bre a longtemps &#xE9;t&#xE9; un important syst&#xE8;me mod&#xE8;le pour les &#xE9;tudes sur le d&#xE9;veloppement et g&#xE9;n&#xE9;tiques, on sait peu sur les r&#xE9;ponses des neurones internes de la r&#xE9;tine. Nous rapportons ici les enregistrements unitaires des cellules ganglionnaires de 5 - pour les larves de poisson z&#xE8;bre 6-day-old. Dans les larves de type sauvage, nous identifier au moins cinq sous-types de r&#xE9;ponses des cellules ganglionnaires &#xE0; plein-champ d&#x26;#39;illumination, avec ON-OFF et ON-type pr&#xE9;dominant des cellules. Dans la r&#xE9;tine nrc mutant, dans lequel les terminaux photor&#xE9;cepteurs se d&#xE9;veloppent anormalement, nous observons normale OFF r&#xE9;ponses anormales mais ON-OFF r&#xE9;ponses et pas sur les r&#xE9;ponses. Auparavant caract&#xE9;ris&#xE9; aussi aveugle, ces mutants ne pas avoir d&#x26;#39;optocin&#xE9;tique r&#xE9;flexe (OKR), mais dans un autre test comportemental nrc mutant poissons ont pr&#xE8;s de la normale r&#xE9;ponses &#xE0; l&#x26;#39;offset des r&#xE9;ponses claires et lent et l&#xE9;thargique &#xE0; l&#x26;#39;apparition de la lumi&#xE8;re. Bloc pharmacologique de la voie ON imite la plupart des d&#xE9;fauts visuels du CNRC. Nous concluons que les bornes photor&#xE9;ceptrices anormaux chez des mutants du CNRC principalement perturber la voie ON et que la voie ON est n&#xE9;cessaire pour conduire l&#x26;#39;OKR chez le poisson z&#xE8;bre larvaire.

W&auml;hrend die Zebrafisch-Netzhaut ist langem ein wichtiges Modellsystem f&uuml;r Entwicklungsbiologie und genetische Studien seit, ist wenig bekannt &uuml;ber die Antworten der inneren Netzhaut Neuronen. Hier berichten wir Einzel-Einheit Ganglion Zelle Aufnahmen von 5 auf 6-Tage alten Zebrafisch-Larven. Im Wildtyp Larven identifizieren wir mindestens f&uuml;nf Untertypen des Ganglion Zelle Antworten zu Vollfeld-Beleuchtung, mit ON-OFF und ON-Typ-Zellen dominieren. Der nrc mutant Netzhaut, in der die Photorezeptoren-Terminals au&szlig;erplanm&auml;&szlig;ig entwickeln beobachten wir normale Antworten aber abnormale ON-OFF-Antworten und keine ON-Antworten. Vorher gekennzeichnet als Blind, diese Mutanten fehlt ein Optokinetischer Reflex (OKR), aber in einem anderen verhaltensbasierten Assay nrc mutant Fisch haben ann&auml;hernd normale Reaktionen auf den Offset des Licht und langsam und schwerf&auml;llig Antworten zum Ausbruch von Licht. Pharmakologische Block des Signalweges ON ahmt die meisten der nrc-Sehst&ouml;rungen. Wir schlie&szlig;en, dass die abnorme Photorezeptor-Terminals in nrc Mutanten &uuml;berwiegend die ON-Pathway verl&auml;ngern und die ON-Pathway die OKR in larvalen Zebrafisch fahren notwendig ist.

OFF Ganglion fahren nicht Zellen den Optokinetischer Reflex im Zebrafisch.

Considerando che la retina di zebrafish &egrave; stata a lungo un importante modello sistema per gli studi di genetici e di sviluppo, poco si sa circa le risposte dei neuroni della retina interni. Qui riportiamo le registrazioni delle cellule gangliari unit&agrave; singola da larve di zebrafish 5 - a 6-day-old. Nel selvaggio-tipo larve identifichiamo almeno cinque sottotipi delle risposte delle cellule gangliari a pieno campo di illuminazione, con ON-OFF e ON-tipo cellule nelle quali predomina. Nella retina mutante nrc, in cui i terminali fotorecettore sviluppano anormalmente, osserviamo normale fuori risposte ma risposte anomale ON-OFF e non ON. Precedentemente caratterizzati come ciechi, questi mutanti mancano un riflesso optocinetico (OKR), ma in un altro pesce mutante nrc a test comportamentali hanno quasi normale risposte all&#39;offset del leggero e lento e pigro responses to l&#39;insorgenza della luce. Blocco farmacologico del percorso ON imita la maggior parte dei difetti visivi del nrc. Possiamo concludere che i terminali fotorecettore anormale in mutanti nrc prevalentemente perturbare il pathway ON e che il percorso ON &egrave; necessario per guidare il OKR nello zebrafish larvale.

The Life Sciences-Howard Hughes Medical Institute Outreach Program at Harvard University supports high school science education by offering an on-campus program for students and their teachers to participate in investigative, hands-on laboratory sessions. The outreach program has recently designed and launched a successful zebrafish embryology protocol that we present here. The main objectives of this protocol are to introduce students to zebrafish as a model research organism and to provide students with direct experience with current techniques used in embryological research. The content of the lab is designed to generate discussions on embryology, genetics, fertilization, natural selection, and animal adaptation. The protocol produces reliable results in a time-efficient manner using a minimum of reagents. The protocol presented here consists of three sections: observations of live zebrafish larvae at different developmental stages, cartilage staining of zebrafish larvae, and a mutant hunt involving identification of two zebrafish mutants (nacre and chokh). Here, we describe the protocol, show the results obtained for each section, and suggest possible alternatives for different lab settings.

Los principales objetivos del presente Protocolo est&aacute;n introducir a los alumnos a pez cebra como un organismo modelo de investigaci&oacute;n y proporcionar a los estudiantes con experiencia directa con las t&eacute;cnicas actuales utilizadas en la investigaci&oacute;n embriol&oacute;gica. El contenido del laboratorio est&aacute; dise&ntilde;ado para generar discusiones sobre embriolog&iacute;a, gen&eacute;tica, fertilizaci&oacute;n, selecci&oacute;n natural y adaptaci&oacute;n del animal. El protocolo produce resultados confiables de manera eficiente con un m&iacute;nimo de reactivos. El protocolo que presentamos consta de tres secciones: observaciones de larvas de pez cebra vivo en diferentes etapas de desarrollo, cart&iacute;lago tinci&oacute;n de larvas de pez cebra y una caza mutante que implica la identificaci&oacute;n de dos mutantes de pez cebra (n&aacute;car y chokh). Darkness serves as a stimulus for vertebrate photoreceptors; they are actively depolarized in the dark and hyperpolarize in the light. Here, we show that larval zebrafish essentially turn off their visual system at night when they are not active. Electroretinograms recorded from larval zebrafish show large differences between day and night; the responses are normal in amplitude throughout the day but are almost absent after several hours of darkness at night. Behavioral testing also shows that larval zebrafish become unresponsive to visual stimuli at night. This phenomenon is largely circadian driven as fish show similar dramatic changes in visual responsiveness when maintained in continuous darkness, although light exposure at night partially restores the responses. Visual responsiveness is decreased at night by at least two mechanisms: photoreceptor outer segment activity decreases and synaptic ribbons in cone pedicles disassemble.

La oscuridad sirve como un est&#xED;mulo para los fotorreceptores de vertebrados, sino que son activamente despolarizado en la oscuridad y hiperpolarizar a la luz. Aqu&#xED;, mostramos que larvas de pez cebra en esencia apagar su sistema visual en la noche cuando no est&#xE1;n activos. Electrorretinogramas registrados a partir de larvas de pez cebra muestran grandes diferencias entre el d&#xED;a y la noche, las respuestas son normales en amplitud durante todo el d&#xED;a, pero est&#xE1;n casi ausentes despu&#xE9;s de varias horas de oscuridad en la noche. Studies in several vertebrate species have shown that visual sensitivity and a number of other retinal phenomena are regulated by circadian mechanisms. For example, ultra-structural studies of 5 day old zebrafish larvae have shown that synaptic ribbons in photoreceptor terminals undergo dramatic diurnal alterations. These synaptic ribbons are very prominent during the day, but are almost completely absent at night. The implications of this circadian driven process on visual function are not well understood. We recently showed that larval zebrafish essentially lose visual responsiveness at night. This shut-down of retinal function at night is regulated by at least two mechanisms: the disassembly of synaptic ribbons in cone pedicles and a decrease of outer segment activity. Here, we summarize our recently reported observations and further discuss our hypothesis on how this phenomenon of shutting-down retinal function at night may provide a means for zebrafish larvae to conserve energy.

Diese synaptischen B&auml;nder sind sehr prominent tags&uuml;ber, jedoch fast v&ouml;llig abwesend in der Nacht. Die Auswirkungen dieser circadian angetriebenen Prozess auf visuelle Funktion nicht gut verstanden. Vor kurzem zeigten wir die larvalen Zebrafisch visuelle Reaktionsf&auml;higkeit im Wesentlichen in der Nacht zu verlieren. Diese Abschaltung der Netzhaut-Funktion in der Nacht wird durch mindestens zwei Mechanismen: die Demontage der synaptischen Multifunktionsleisten in Kegel Pedicles und eine Abnahme der &auml;u&szlig;eren Segment-Aktivit&auml;t. En un estudio reciente gen&#xF3;mico, Leung et al. utiliz&#xF3; un an&#xE1;lisis factorial de microarrays para identificar Smarca4 (Brg1) de los genes regulados en la micro-disecci&#xF3;n retinas de pez cebra. Doscientos cincuenta y nueve genes se agrupan en modelos de ANOVA de tres v&#xED;as que llevaron a la retina el cambio m&#xE1;s espec&#xED;fico. Para validar los resultados de microarrays y para dilucidar los patrones celulares de expresi&#xF3;n de los genes importantes para la caracterizaci&#xF3;n adicional, 32 genes que se sabe fueron seleccionados al azar de este grupo. La hibridaci&#xF3;n in situ de estos genes se realiz&#xF3; en los mismos tipos de muestras (de tipo salvaje (WT) y smarca4a50/a50 (Yng) mutante) en las mismas etapas (36 y 52 horas despu&#xE9;s de la fertilizaci&#xF3;n (HPF)) como en el microarray estudio.
