The extraordinary cellular heterogeneity of the mammalian nervous system has largely hindered the molecular analysis of neuronal identity and diversity. In order to uncover mechanisms involved in neuronal differentiation and diversification, we have monitored the expression profiles of individual neurons and progenitor cells collected from dissociated tissue or captured from intact slices. We demonstrate that this technique provides a sensitive and reproducible representation of the single-cell transcriptome. In the olfactory system, hundreds of transcriptional differences were identified between olfactory progenitors and mature sensory neurons, enabling us to define the large variety of signaling pathways expressed by individual progenitors at a precise developmental stage. Finally, we show that regional differences in gene expression can be predicted from transcriptional analysis of single neuronal precursors isolated by laser capture from defined areas of the developing brain.

La straordinaria eterogeneit&agrave; cellulare del sistema nervoso dei mammiferi in gran parte ha ostacolato l&#39;analisi molecolare di un neurone identit&agrave; e diversit&agrave;. Al fine di scoprire i meccanismi coinvolti nella differenziazione neuronale e diversificazione, abbiamo monitorato i profili di espressione dei singoli neuroni e cellule progenitrici raccolte dal tessuto dissociata o catturato da fette intatti. La extraordinaria heterogeneidad celular del sistema nervioso mam&iacute;fero ha dificultado en gran medida el an&aacute;lisis molecular de identidad neuronal y la diversidad. In order to gain insights into the regulatory control of neuronal diversity in the mammalian olfactory system, we have identified the transcriptional profile of individual olfactory neurons. A single cell microarray strategy was performed to search for candidate genes involved in the molecular specification of dorso-ventral zones of olfactory receptor (OR) expression. Several transcripts were identified that display differential expression in distinct OR zones, including a novel family of genes, the Lozenge-like (Lzl) genes which share sequence consensus motifs with Lozenge, a transcription factor involved in the patterning of the Drosophila olfactory and visual systems.

Al fine di acquisire conoscenze nel controllo normativo della diversit&agrave; neuronale nel sistema olfattivo dei mammiferi, abbiamo individuato il profilo trascrizionale dei singoli neuroni olfattivi. Una strategia di microarray di singola cellula &egrave; stata eseguita per la ricerca di geni candidati coinvolti nella specifica molecolare delle zone dorso-ventrale dell&#39;espressione dei recettori olfattivi (OR). Diverse trascrizioni sono state identificate che visualizzano espressione differenziale in distinte o zone, tra cui una romanzo famiglia di geni, i geni simili a losanga (Lzl) che condividono i motivi di sequenza consenso con losanga, un fattore di trascrizione coinvolto nella modellatura dei sistemi olfattivi e visivi di Drosophila.

MicroRNAs (miRNAs) comprise 1 to 3% of all vertebrate genes, but their in vivo functions and mechanisms of action remain largely unknown. Zebrafish miR-430 is expressed at the onset of zygotic transcription and regulates morphogenesis during early development. By using a microarray approach and in vivo target validation, we find that miR-430 directly regulates several hundred target messenger RNA molecules (mRNAs). Most targets are maternally expressed mRNAs that accumulate in the absence of miR-430. We also show that miR-430 accelerates the deadenylation of target mRNAs. These results suggest that miR-430 facilitates the deadenylation and clearance of maternal mRNAs during early embryogenesis.

As many as 10% of humans suffer chronic sleep disturbances, yet the genetic mechanisms that regulate sleep remain essentially unknown. It is therefore crucial to develop simple and cost-effective vertebrate models to study the genetic regulation of sleep. The best characterized mammalian sleep/wake regulator is hypocretin/orexin (Hcrt), whose loss results in the sleep disorder narcolepsy and that has also been implicated in feeding behavior, energy homeostasis, thermoregulation, reward seeking, addiction, and maternal behavior. Here we report that the expression pattern and axonal projections of embryonic and larval zebrafish Hcrt neurons are strikingly similar to those in mammals. We show that zebrafish larvae exhibit robust locomotive sleep/wake behaviors as early as the fifth day of development and that Hcrt overexpression promotes and consolidates wakefulness and inhibits rest. Similar to humans with insomnia, Hcrt-overexpressing larvae are hyperaroused and have dramatically reduced abilities to initiate and maintain rest at night. Remarkably, Hcrt function is modulated by but does not require normal circadian oscillations in locomotor activity. Our zebrafish model of Hcrt overexpression indicates that the ancestral function of Hcrt is to promote locomotion and inhibit rest and will facilitate the discovery of neural circuits, genes, and drugs that regulate Hcrt function and sleep.

Le meilleur r&eacute;gulateur/veille mammif&egrave;res caract&eacute;ris&eacute; est hypocretin/orexine (Hcrt), dont la perte entra&icirc;ne la narcolepsie de trouble du sommeil et qui a &eacute;galement &eacute;t&eacute; impliqu&eacute;e dans le comportement alimentaire, l&#39;hom&eacute;ostasie &eacute;nerg&eacute;tique, thermor&eacute;gulation, r&eacute;compense qui cherchent, addiction et comportement maternel. Nous montrons que les larves de poisson z&egrave;bre manifester des comportements de robuste locomotive sommeil/&eacute;veil, d&egrave;s le cinqui&egrave;me jour du d&eacute;veloppement et que la surexpression de Hcrt favorise et consolide l&#39;&eacute;veil et inhibe le reste. Tanto como el 10% de los seres humanos sufren trastornos del sue&ntilde;o cr&oacute;nico, sin embargo, los mecanismos gen&eacute;ticos que regulan el sue&ntilde;o sigue siendo esencialmente desconocidos. Por lo tanto es crucial para desarrollar modelos de vertebrados simples y rentables para estudiar la regulaci&oacute;n gen&eacute;tica del sue&ntilde;o. El mejor regulador de sue&ntilde;o-vigilia mam&iacute;feros caracterizado es hipocretina/orexin (Hcrt), cuya p&eacute;rdida resulta en la narcolepsia trastorno de sue&ntilde;o y que tambi&eacute;n se ha implicado en la alimentaci&oacute;n de comportamiento, la homeostasis energ&eacute;tica, termorregulaci&oacute;n, b&uacute;squeda de recompensa, adicci&oacute;n y el comportamiento maternal. Aqu&iacute; se reporta que el patr&oacute;n de expresi&oacute;n y proyecciones axonal de pez cebra embrionario y larval Hcrt neuronas son sorprendentemente similares a los mam&iacute;feros. Demostramos que las larvas de pez cebra exhiben comportamientos robusta locomotora de sue&ntilde;o/vigilia tan pronto como el quinto d&iacute;a de desarrollo y que la sobreexpresi&oacute;n Hcrt promueve y consolida la vigilia e inhibe el resto. Similar a los seres humanos con insomnio, larvas de Hcrt-overexpressing son hyperaroused y han reducido espectacularmente la capacidad para iniciar y mantener el resto en la noche. Notable, funci&oacute;n de Hcrt es modulada por pero no requiere oscilaciones circadianas normales actividad locomotriz. Whereas the zebrafish retina has long been an important model system for developmental and genetic studies, little is known about the responses of the inner retinal neurons. Here we report single-unit ganglion cell recordings from 5- to 6-day-old zebrafish larvae. In wild-type larvae we identify at least five subtypes of ganglion cell responses to full-field illumination, with ON-OFF and ON-type cells predominating. In the nrc mutant retina, in which the photoreceptor terminals develop abnormally, we observe normal OFF responses but abnormal ON-OFF responses and no ON responses. Previously characterized as blind, these mutants lack an optokinetic reflex (OKR), but in another behavioral assay nrc mutant fish have near-normal responses to the offset of light and slow and sluggish responses to the onset of light. Pharmacological block of the ON pathway mimics most of the nrc visual defects. We conclude that the abnormal photoreceptor terminals in nrc mutants predominantly perturb the ON pathway and that the ON pathway is necessary to drive the OKR in larval zebrafish.

OFF ganglion cells cannot drive the optokinetic reflex in zebrafish.

Hier berichten wir Einzel-Einheit Ganglion Zelle Aufnahmen von 5 auf 6-Tage alten Zebrafisch-Larven. Im Wildtyp Larven identifizieren wir mindestens f&uuml;nf Untertypen des Ganglion Zelle Antworten zu Vollfeld-Beleuchtung, mit ON-OFF und ON-Typ-Zellen dominieren. Der nrc mutant Netzhaut, in der die Photorezeptoren-Terminals au&szlig;erplanm&auml;&szlig;ig entwickeln beobachten wir normale Antworten aber abnormale ON-OFF-Antworten und keine ON-Antworten. Vorher gekennzeichnet als Blind, diese Mutanten fehlt ein Optokinetischer Reflex (OKR), aber in einem anderen verhaltensbasierten Assay nrc mutant Fisch haben ann&auml;hernd normale Reaktionen auf den Offset des Licht und langsam und schwerf&auml;llig Antworten zum Ausbruch von Licht. Pharmakologische Block des Signalweges ON ahmt die meisten der nrc-Sehst&ouml;rungen. Wir schlie&szlig;en, dass die abnorme Photorezeptor-Terminals in nrc Mutanten &uuml;berwiegend die ON-Pathway verl&auml;ngern und die ON-Pathway die OKR in larvalen Zebrafisch fahren notwendig ist.

Possiamo concludere che i terminali fotorecettore anormale in mutanti nrc prevalentemente perturbare il pathway ON e che il percorso ON &egrave; necessario per guidare il OKR nello zebrafish larvale.

En la retina NRC mutante, en el que los terminales fotorreceptoras desarrollan anormalmente, se observa normal, pero OFF respuestas anormales respuestas de ON-OFF y no sobre las respuestas. Anteriormente caracteriza por ser ciego, estos mutantes no tienen un reflejo de optocin&#xE9;tico (okr), pero en otro ensayo de conducta NRC peces mutantes tienen casi normales las respuestas a la compensaci&#xF3;n de las respuestas de la luz y lento y perezoso a la aparici&#xF3;n de la luz. Bloque farmacol&#xF3;gico de la v&#xED;a ON imita la mayor&#xED;a de los defectos visuales NRC. Llegamos a la conclusi&#xF3;n de que los terminales del fotorreceptor anormales en mutantes NRC predominantemente perturbar la v&#xED;a ON y que la v&#xED;a ON es necesaria para impulsar el okr en larvas de pez cebra.

Transient receptor potential (TRP) ion channels have been implicated in detecting chemical, thermal, and mechanical stimuli in organisms ranging from mammals to Caenorhabditis elegans. It is well established that TRPA1 detects and mediates behavioral responses to chemical irritants. However, the role of TRPA1 in detecting thermal and mechanical stimuli is controversial. To further clarify the functions of TRPA1 channels in vertebrates, we analyzed their roles in zebrafish. The two zebrafish TRPA1 paralogs are expressed in sensory neurons and are activated by several chemical irritants in vitro. High-throughput behavioral analyses of trpa1a and trpa1b mutant larvae indicate that TRPA1b is necessary for behavioral responses to these chemical irritants. However, TRPA1 paralogs are not required for behavioral responses to temperature changes or for mechanosensory hair cell function in the inner ear or lateral line. These results support a role for zebrafish TRPA1 in chemical but not thermal or mechanical sensing, and establish a high-throughput system to identify genes and small molecules that modulate chemosensation, thermosensation, and mechanosensation.

A major obstacle for the discovery of psychoactive drugs is the inability to predict how small molecules will alter complex behaviors. We report the development and application of a high-throughput, quantitative screen for drugs that alter the behavior of larval zebrafish. We found that the multidimensional nature of observed phenotypes enabled the hierarchical clustering of molecules according to shared behaviors. Behavioral profiling revealed conserved functions of psychotropic molecules and predicted the mechanisms of action of poorly characterized compounds. In addition, behavioral profiling implicated new factors such as ether-a-go-go-related gene (ERG) potassium channels and immunomodulators in the control of rest and locomotor activity. These results demonstrate the power of high-throughput behavioral profiling in zebrafish to discover and characterize psychotropic drugs and to dissect the pharmacology of complex behaviors.

Darkness serves as a stimulus for vertebrate photoreceptors; they are actively depolarized in the dark and hyperpolarize in the light. Here, we show that larval zebrafish essentially turn off their visual system at night when they are not active. Electroretinograms recorded from larval zebrafish show large differences between day and night; the responses are normal in amplitude throughout the day but are almost absent after several hours of darkness at night. Behavioral testing also shows that larval zebrafish become unresponsive to visual stimuli at night. This phenomenon is largely circadian driven as fish show similar dramatic changes in visual responsiveness when maintained in continuous darkness, although light exposure at night partially restores the responses. Visual responsiveness is decreased at night by at least two mechanisms: photoreceptor outer segment activity decreases and synaptic ribbons in cone pedicles disassemble.

Zebrafish has emerged in the past 5 years as a model for the study of sleep and wake behaviors. Experimental evidence has shown that periods of behavioral quiescence in zebrafish larvae and adults are sleep-like states, as these rest bouts are regulated by the circadian cycle, are associated with decreases in arousal, and are increased following rest deprivation. Furthermore, zebrafish share with mammals a hypocretin/orexin system that promotes wakefulness, and drugs that alter mammalian sleep have similar effects on zebrafish rest. In this chapter, we review the zebrafish sleep literature and describe a long-term, high-throughput monitoring system for observing sleep and wake behaviors in larval zebrafish.

The larval zebrafish has emerged as a vertebrate model system amenable to small molecule screens for probing diverse biological pathways. Two large-scale small molecule screens examined the effects of thousands of drugs on larval zebrafish sleep/wake and photomotor response behaviors. Both screens identified hundreds of molecules that altered zebrafish behavior in distinct ways. The behavioral profiles induced by these small molecules enabled the clustering of compounds according to shared phenotypes. This approach identified regulators of sleep/wake behavior and revealed the biological targets for poorly characterized compounds. El pez cebra larval se ha convertido en un sistema modelo vertebrados susceptible de pantallas de mol&eacute;cula peque&ntilde;a para sondear diversas v&iacute;as biol&oacute;gicas. Dos pantallas de mol&eacute;cula peque&ntilde;a a gran escala examinan los efectos de miles de f&aacute;rmacos en comportamientos de respuesta de sue&ntilde;o/vigilia y fotomotor larvas de pez cebra. Ambas pantallas identifican cientos de mol&eacute;culas que alteraci&oacute;n el comportamiento del pez cebra en formas distintas. Los perfiles conductuales inducidos por estas peque&ntilde;as mol&eacute;culas permitieron el agrupamiento de compuestos seg&uacute;n fenotipos compartidos. Este enfoque identifica los reguladores del comportamiento del sue&ntilde;o/vigilia y revel&oacute; los objetivos biol&oacute;gicos compuestos mal caracterizado.
