Neuropeptide S (NPS) is the endogenous ligand of a formerly orphan G protein-coupled receptor (GPCR). The NPS receptor (NPSR) belongs to the subfamily of peptide GPCRs and is widely expressed in the brain. NPS promotes arousal and induces anxiolytic-like effects after central administration in rodents. Previously, we have reported that the N107I polymorphism in the human NPS receptor results in a gain-of-function characterized by an increase in agonist potency without changing agonist binding affinity. We have extended our findings by investigating pharmacological and biochemical consequences of mutations in the vicinity of position 107. Alanine substitutions were made for D105 and N101, and stable clones were analyzed for agonist-induced changes of intracellular Ca(2+). Receptor protein expression was monitored by Western blot and flow cytometry. The mutation D105A produced receptors that have a approximately 200-fold higher EC(50) despite elevated total receptor protein and surface expression compared to cell lines expressing the parental receptor NPSR-N107. The mutation N101A resulted in slightly reduced agonist potency without affecting the ability of the protein to form functional receptors. Stable NPSR-A101 clones show little expression of the fully glycosylated form. However, NPSR-A101 receptors are expressed on the cell surface and are functional, suggesting that full glycosylation is not required for receptor function. Our studies suggest that N-linked glycosylation is not important for receptor biogenesis or function, and that residue D105 might be critical for receptor binding.

Neuropeptide S (NPS) est le ligand endog&egrave;ne d&#39;un autrefois orphelin r&eacute;cepteur de coupl&eacute;s &agrave; la prot&eacute;ine de G (RCPG). Le r&eacute;cepteur NPS (NPSR) appartient &agrave; la sous-famille des peptides RCPG et est largement exprim&eacute; dans le cerveau. NPS favorise l&#39;excitation et induit des effets anxiolytiques apr&egrave;s administration centrale chez les rongeurs. Auparavant, nous avons signal&eacute; que le polymorphisme de N107I dans le r&eacute;cepteur humain de NPS se traduit par une gain de fonction caract&eacute;ris&eacute;e par une augmentation de puissance agoniste sans changer l&#39;affinit&eacute; agoniste. La mutation que n101a a l&eacute;g&egrave;rement r&eacute;duit la puissance agoniste sans affecter la capacit&eacute; de la prot&eacute;ine pour former des r&eacute;cepteurs fonctionnels. Stables NPSR-A101 clones montrent peu d&#39;expression de la forme glycosyl&eacute;e enti&egrave;rement. Zuvor haben wir berichtet, dass die N107I-Polymorphismus in der menschlichen NPS-Rezeptor ein Gain von Function zeichnet sich durch eine Steigerung in Agonist Potenz ohne Agonist Bindungsaffinit&auml;t f&uuml;hrt. Neurop&eacute;ptido S (NPS) es el ligando end&oacute;geno de un receptor antes hu&eacute;rfano acoplado a prote&iacute;nas G (GPCR). El receptor NPS (NPSR) pertenece a la subfamilia de p&eacute;ptido GPCRs y ampliamente se expresa en el cerebro. NPS promueve la excitaci&oacute;n e induce efectos ansiol&iacute;ticos como despu&eacute;s de la administraci&oacute;n central en roedores. Previamente, nos han informado que el polimorfismo N107I en el receptor NPS humano resulta en una ganancia de funci&oacute;n caracterizada por un aumento en la potencia del agonista sin cambiar la afinidad de uni&oacute;n del agonista. Hemos ampliado nuestros resultados por investigar consecuencias farmacol&oacute;gicas y bioqu&iacute;micas de las mutaciones en las cercan&iacute;as de posici&oacute;n 107. Sustituciones de alanina fueron hechas para D105 y N101, y clones estables se analizaron para cambios inducidos por agonistas de CA intracelular. Expresi&oacute;n de la prote&iacute;na del receptor se control&oacute; mediante citometr&iacute;a de flujo y la mancha blanca /negra occidental. La mutaci&oacute;n que N101A dio lugar a ligeramente reducida potencia agonista sin afectar la capacidad de la prote&iacute;na para formar receptores funcionales. Estables NPSR-A101 clones muestran poca expresi&oacute;n de la forma totalmente glicosilada. Sin embargo, NPSR-A101 receptores se expresan en la superficie celular y son funcionales, sugiriendo que glicosilaci&oacute;n completo no es necesario para la funci&oacute;n del receptor. Hier berichten wir, dass normale hPSC--Linien sind auch in induzierte pluripotente--karyotypic Mosaik der euploid Zellen mit vielen Zellen zeigen nicht-klonale Aneuploidien durch Chromosom z&auml;hlen gemischt, spektrale Chromosomenanalyse (Himmel) und Fluoreszenz in Situ Hybridisierung (FISH) Zwischenphase/nicht-mitotischen Zellen. Dieses Mosaik-Aneuploidie &auml;hnelt in Vorl&auml;uferzellen der entwickelnden Gehirn und Pr&auml;implantationsdiagnostik Embryonen, was darauf hindeutet, dass es eine normale, sondern als pathologische, Funktion von Stammzelllinien beobachtet. Die karyotypic Heterogenit&auml;t von Mosaik Aneuploidie generiert kann zu der gemeldeten funktionale und ph&auml;notypischen Heterogenit&auml;t der hPSCs Linien, als auch ihre therapeutische Wirksamkeit und Sicherheit nach Transplantation beitragen.

Human pluripotent stem cell (hPSC) lines have been considered to be homogeneously euploid. Here we report that normal hPSC--including induced pluripotent--lines are karyotypic mosaics of euploid cells intermixed with many cells showing non-clonal aneuploidies as identified by chromosome counting, spectral karyotyping (SKY) and fluorescent in situ hybridization (FISH) of interphase/non-mitotic cells. This mosaic aneuploidy resembles that observed in progenitor cells of the developing brain and preimplantation embryos, suggesting that it is a normal, rather than pathological, feature of stem cell lines. The karyotypic heterogeneity generated by mosaic aneuploidy may contribute to the reported functional and phenotypic heterogeneity of hPSCs lines, as well as their therapeutic efficacy and safety following transplantation.

For some highly endangered species there are too few reproductively capable animals to maintain adequate genetic diversity, and extraordinary measures are necessary to prevent extinction. We report generation of induced pluripotent stem cells (iPSCs) from two endangered species: a primate, the drill, Mandrillus leucophaeus and the nearly extinct northern white rhinoceros, Ceratotherium simum cottoni. iPSCs may eventually facilitate reintroduction of genetic material into breeding populations.

Rapid and dependable methods for isolating human pluripotent stem cell (hPSC) populations are urgently needed for quality control in basic research and in cell-based therapy applications. Using lectin arrays, we analyzed glycoproteins extracted from 26 hPSC samples and 22 differentiated cell samples, and identified a small group of lectins with distinctive binding signatures that were sufficient to distinguish hPSCs from a variety of non-pluripotent cell types. These specific biomarkers were shared by all the 12 human embryonic stem cell and the 14 human induced pluripotent stem cell samples examined, regardless of the laboratory of origin, the culture conditions, the somatic cell type reprogrammed, or the reprogramming method used. We demonstrated a practical application of specific lectin binding by detecting hPSCs within a differentiated cell population with lectin-mediated staining followed by fluorescence microscopy and flow cytometry, and by enriching and purging viable hPSCs from mixed cell populations using lectin-mediated cell separation. Global gene expression analysis showed pluripotency-associated differential expression of specific fucosyltransferases and sialyltransferases, which may underlie these differences in protein glycosylation and lectin binding. Taken together, our results show that protein glycosylation differs considerably between pluripotent and non-pluripotent cells, and demonstrate that lectins may be used as biomarkers to monitor pluripotency in stem cell populations and for removal of viable hPSCs from mixed cell populations.

Los m&#xE9;todos r&#xE1;pidos y fiables para el aislamiento de c&#xE9;lulas madre pluripotentes humanas (hPSC) las poblaciones se necesitan con urgencia para el control de la calidad en la investigaci&#xF3;n b&#xE1;sica y en aplicaciones de terapia a base de c&#xE9;lulas. El uso de matrices de lectina, se analizaron las glicoprote&#xED;nas extra&#xED;das de 26 muestras hPSC y 22 muestras de c&#xE9;lulas diferenciadas, e identific&#xF3; un peque&#xF1;o grupo de lectinas con distintivos de firmas obligatorias que eran suficientes para distinguir hPSCs de una variedad de tipos de c&#xE9;lulas no pluripotentes. Estos biomarcadores espec&#xED;ficos eran compartidos por todos los 12 de c&#xE9;lulas madre de embriones humanos y de las 14 muestras humanas pluripotentes inducidas de c&#xE9;lulas madre analizadas, independientemente del laboratorio de origen, las condiciones de cultivo, el tipo de c&#xE9;lulas som&#xE1;ticas reprogramadas, o el m&#xE9;todo de reprogramaci&#xF3;n utilizado. Hemos demostrado una aplicaci&#xF3;n pr&#xE1;ctica de lectina de uni&#xF3;n espec&#xED;ficos mediante la detecci&#xF3;n de hPSCs dentro de una poblaci&#xF3;n de c&#xE9;lulas diferenciadas con lectina mediada por tinci&#xF3;n seguido por microscop&#xED;a de fluorescencia y citometr&#xED;a de flujo, y por enriquecer y purga hPSCs viables a partir de poblaciones de c&#xE9;lulas mixtas utilizando lectina mediada por separaci&#xF3;n de c&#xE9;lulas. An&#xE1;lisis global de la expresi&#xF3;n g&#xE9;nica mostraron pluripotencia asociada a la expresi&#xF3;n diferencial de fucosyltransferases espec&#xED;ficos y sialiltransferasas, que puede ser la base de estas diferencias en la glicosilaci&#xF3;n de prote&#xED;nas y la lectina de uni&#xF3;n a.
