A diferencia de la beta-c&eacute;lulas aisladas, que generalmente producen picos continuas o arranques r&aacute;pidos e irregulares, el&eacute;ctricamente acoplados beta-c&eacute;lulas pueden exhibir robustos estallido potenciales de acci&oacute;n. Consideramos el ruido inducido por fluctuaciones termales, as&iacute; como por canal-que bloquean la estocasticidad y examinar sus efectos en el comportamiento del potencial de acci&oacute;n del modelo de c&eacute;lulas beta. Num&eacute;ricamente se observa que dichos sonidos en general ayuda a las c&eacute;lulas para producir una variedad de actividades el&eacute;ctricas. Adem&aacute;s, nosotros tambi&eacute;n sonda acoplamiento mediante uniones comunicantes entre c&eacute;lulas vecinas, con heterogeneidad inducida por el ruido, para encontrar que realza r&aacute;fagas regulares.

Unlike isolated beta-cells, which usually produce continuous spikes or fast and irregular bursts, electrically coupled beta-cells are apt to exhibit robust bursting action potentials. We consider the noise induced by thermal fluctuations as well as that by channel-gating stochasticity and examine its effects on the action potential behavior of the beta-cell model. It is observed numerically that such noise in general helps single cells to produce a variety of electrical activities. In addition, we also probe coupling via gap junctions between neighboring cells, with heterogeneity induced by noise, to find that it enhances regular bursts.

Contrairement aux b&ecirc;ta-cellules isol&eacute;es, qui produisent g&eacute;n&eacute;ralement des pointes continus ou salves rapides et irr&eacute;guli&egrave;res, b&ecirc;ta-cellules &eacute;lectriquement coupl&eacute;s sont port&eacute;s &agrave; pi&egrave;ce robustes rupture potentiels d&#39;action. Nous consid&egrave;rent le bruit induit par les fluctuations thermiques ainsi que celle de cloisonnement canal stochasticit&eacute; et examiner ses effets sur le comportement du potentiel d&#39;action du mod&egrave;le des cellules b&ecirc;ta. Num&eacute;riquement, on a observ&eacute; que ces bruits permet en g&eacute;n&eacute;ral des cellules individuelles pour produire une vari&eacute;t&eacute; d&#39;activit&eacute;s &eacute;lectriques. Pancreatic beta-cells are clustered in islets of Langerhans, which are typically a few hundred micrometers in a variety of mammals. In this study, we propose a theoretical model for the growth of pancreatic islets and derive the islet size distribution, based on two recent observations: First, the neogenesis of new islets becomes negligible after some developmental stage. Second, islets grow via a random process, where any cell in an islet proliferates with the same rate regardless of the present size of the islet. Our model predicts either log-normal or Weibull distributions of the islet sizes, depending on whether cells in an islet proliferate coherently or independently. To confirm this, we also measure the islet size by selectively staining islets, which are exposed from exocrine tissues in mice after enzymatic treatment. Indeed revealed are skewed distributions with the peak size of approximately 100 cells, which fit well to the theoretically derived ones. Interestingly, most islets turned out to be bigger than the expected minimal size (approximately 10 or so cells) necessary for stable synchronization of beta-cells through electrical gap-junction coupling. The collaborative behavior among cells is known to facilitate synchronized insulin secretion and tends to saturate beyond the critical (saturation) size of approximately 100 cells. We further probe how the islets change as normal mice grow from young (6 weeks) to adult (5 months) stages. It is found that islets may not grow too large to maintain appropriate ratios between cells of different types. Our results implicate that growing of mouse islets may be regulated by several physical constraints such as the minimal size required for stable cell-to-cell coupling and the upper limit to keep the ratios between cell types. Within the lower and upper limits the observed size distributions of islets can be faithfully regenerated by assuming random and uncoordinated proliferation of each beta-cell at appropriate rates.

En effet r&eacute;v&eacute;l&eacute; sont distributions asym&eacute;triques avec la taille maximale d&#39;environ 100 cellules, qui correspondent bien &agrave; celles th&eacute;oriquement d&eacute;riv&eacute;es. Fait int&eacute;ressant, la plupart des &icirc;lots s&#39;est av&eacute;r&eacute; pour &ecirc;tre plus grand que la taille minimale attendue (environ 10 ou si les cellules), n&eacute;cessaire pour la synchronisation stable des b&ecirc;ta-cellules par couplage &eacute;lectrique gap-junction. Le comportement de collaboration entre les cellules est connu pour faciliter la s&eacute;cr&eacute;tion d&#39;insuline synchronis&eacute; et a tendance &agrave; saturer au-del&agrave; de la taille critique (saturation) d&#39;environ 100 cellules. Nous sonder plus loin comment les &icirc;lots changent que les souris normales poussent de jeunes (6 semaines) au stade adulte (5 mois). Il se trouve que les &icirc;lots peuvent poussent pas trop grands &agrave; entretenir des rapports appropri&eacute;s entre les cellules de diff&eacute;rents types. Nos r&eacute;sultats indiquent que cette croissance d&#39;&icirc;lots peuvent &ecirc;tre r&eacute;glement&eacute;es par plusieurs contraintes physiques comme la taille minimal requis pour le couplage stable de cellule-cellule et la limite sup&eacute;rieure de garder les rapports entre les types de cellules de souris. Dans les limites inf&eacute;rieures et sup&eacute;rieures les distributions observ&eacute;es taille des &icirc;lots peuvent &ecirc;tre r&eacute;g&eacute;n&eacute;r&eacute;es fid&egrave;lement en supposant une prolif&eacute;ration al&eacute;atoire et non coordonn&eacute;e de chaque cellule b&ecirc;ta au taux appropri&eacute;s.

Pankreatische Betazellen gruppiert sind in der Langerhans-Inseln, die in der Regel ein paar Hundert Mikrometern in einer Vielzahl von S&auml;ugetieren sind. In dieser Studie, die wir vorschlagen, ein theoretisches Modell f&uuml;r das Wachstum des Pankreas Inseln und Inselchen Korngr&ouml;&szlig;enverteilung, basierend auf zwei j&uuml;ngsten Beobachtungen ableiten: zuerst wird der Neogenesis neue Inseln nach einige Entwicklungsstadiums vernachl&auml;ssigbar. Zweitens wachsen Inselchen &uuml;ber einen zuf&auml;lligen Prozess, in dem eine beliebige Zelle in eine kleine Insel mit der gleichen Rate unabh&auml;ngig von der derzeitigen Gr&ouml;&szlig;e der Insel wuchert. Unser Modell prognostiziert Log-Normal &quot;oder&quot; Weibull-Verteilungen der Inselchen Gr&ouml;&szlig;en, je nachdem, ob die Zellen in eine kleine Insel zusammenh&auml;ngend oder selbstst&auml;ndig vermehren. Wir Pr&uuml;fpunkt weiter, wie die Inselchen zu &auml;ndern, wie normale M&auml;use von jung (6 Wochen) f&uuml;r Erwachsene (5 Monate) Stufen wachsen. Es wird festgestellt, dass die Inseln nicht zu gro&szlig; entsprechende Relationen zwischen Zellen unterschiedlicher Typen beibehalten werden k&ouml;nnen. Unsere Ergebnisse implizieren, dass Anbau von Maus, die Inseln von mehreren physikalischen Einschr&auml;nkungen z. B. der minimale Gr&ouml;&szlig;e f&uuml;r stabile Zell-Zell-Kopplung und die Obergrenze f&uuml;r die Relationen zwischen Zelltypen zu halten geregelt werden k&ouml;nnen. In den unteren und oberen Grenzen k&ouml;nnen die beobachteten Gr&ouml;&szlig;e-Distributionen Inselchen getreulich regeneriert werden, durch die Annahme von zuf&auml;lligen und unkoordinierte Verbreitung der einzelnen Beta-Zellen zu angemessenen Preisen.

In questo studio, proponiamo un modello teorico per la crescita di isole pancreatiche e derivare la distribuzione di dimensione di isolotto, basata su due recenti osservazioni: in primo luogo, neogenesis nuovi isolotti diventa trascurabile dopo qualche fase dello sviluppo. In secondo luogo, isolotti crescono attraverso un processo casuale, dove ogni cella in un&#39;isoletta prolifera con lo stesso tasso indipendentemente dalle dimensioni del presente dell&#39;isoletta. Il nostro modello predice o log-normale o distribuzioni di Weibull di dimensioni l&#39;isolotto, a seconda se le cellule in un isolotto proliferano coerentemente o indipendentemente. Per confermare questo, misuriamo anche la dimensione dell&#39;isolotto macchiando selettivamente isolotti, che vengono esposti dai tessuti esocrino nei topi dopo il trattamento enzimatico. Infatti rivelato sono inclinate distribuzioni con le dimensioni di picco di circa 100 cellule, che si adattano bene a quelli teoricamente derivati. &Egrave; interessante notare che, la maggior parte delle isolette si rivel&ograve; per essere pi&ugrave; grande di dimensione minima prevista (circa 10 o cos&igrave; cellule) necessaria per la sincronizzazione stabile di cellule beta tramite accoppiamento elettrico gap-junction. Abbiamo ulteriormente sonda come gli isolotti cambiano come topi normali crescono da giovane (6 settimane) a fasi di adulto (5 mesi). Si &egrave; trovato che isolotti non possono crescere troppo grande per mantenere appropriati rapporti tra cellule di diversi tipi. I nostri risultati implicano che cresce del mouse isolotti possono essere regolati da numerosi vincoli fisici quali la dimensione minima necessaria per accoppiamento stabile di cellula-cellula e il limite superiore per mantenere i rapporti tra i tipi di cella. Entro i limiti inferiori e superiori le distribuzioni osservate dimensione di isolotti possono essere rigenerate fedelmente assumendo casuale e scoordinata proliferazione di ogni cellula beta a tariffe adeguate.

Las c&eacute;lulas beta pancre&aacute;ticas se agrupan en islotes de Langerhans, que suelen ser unos cien micr&oacute;metros en una variedad de mam&iacute;feros. En este estudio, proponemos un modelo te&oacute;rico para el crecimiento de los islotes pancre&aacute;ticos y derivan de la distribuci&oacute;n de tama&ntilde;o de islote, basada en observaciones recientes dos: primero, la neog&eacute;nesis de islotes nuevos llega a ser insignificante despu&eacute;s de cierta etapa de desarrollo. En segundo lugar, islotes crecen mediante un proceso aleatorio, donde cualquier c&eacute;lula en un islote prolifera con la misma tarifa sin importar el tama&ntilde;o actual del islote. Nuestro modelo predice log-normal o distribuciones Weibull de los tama&ntilde;os del islote, dependiendo de si las c&eacute;lulas en un islote proliferan coherente o independientemente. Para confirmarlo, tambi&eacute;n medimos el tama&ntilde;o del islote manchando selectivamente de islotes, que se exponen los tejidos exocrinos en ratones despu&eacute;s de tratamiento enzim&aacute;tico. De hecho revel&oacute; son distribuci&oacute;n sesgada con el tama&ntilde;o m&aacute;ximo de aproximadamente 100 c&eacute;lulas, que se ajuste bien a los que te&oacute;ricamente derivados. Curiosamente, la mayor&iacute;a islotes resultaron para ser m&aacute;s grande que el tama&ntilde;o m&iacute;nimo esperado (aproximadamente 10 o m&aacute;s c&eacute;lulas) necesario para la sincronizaci&oacute;n estable de c&eacute;lulas beta a trav&eacute;s de acoplamiento el&eacute;ctrico gap-junction. El comportamiento de la colaboraci&oacute;n entre las c&eacute;lulas se conoce para facilitar la secreci&oacute;n de insulina sincronizada y tiende a saturar m&aacute;s all&aacute; del tama&ntilde;o de la cr&iacute;tica (saturaci&oacute;n) de aproximadamente 100 c&eacute;lulas. Nos sondeo m&aacute;s c&oacute;mo los islotes cambian como ratones normales crecen desde joven (6 semanas) a las etapas de adulto (5 meses). Se encuentra que los islotes no crezca demasiado grandes para mantener las proporciones apropiadas entre c&eacute;lulas de diversos tipos. Nuestros resultados implican cultivo de islotes pueden regularse por varias limitaciones f&iacute;sicas como el tama&ntilde;o m&iacute;nimo requerido para que acoplamiento estable de c&eacute;lula a c&eacute;lula y el l&iacute;mite superior para mantener las proporciones entre tipos de c&eacute;lulas de rat&oacute;n. Dentro de los l&iacute;mites superiores e inferiores las distribuciones de tama&ntilde;o observado de islotes pueden ser regeneradas fielmente asumiendo la proliferaci&oacute;n al azar y falta de coordinaci&oacute;n de cada c&eacute;lula beta a precios adecuados.

Distribuci&oacute;n de tama&ntilde;o de los islotes de Langerhans del rat&oacute;n.

PDC109 is a modular multi-domain protein with two fibronectin type II (Fn2) repeats joined by a linker. It plays a major role in bull sperm binding to the oviductal epithelium through its interactions with phosphorylcholines (PhCs), a head group of sperm cell membrane lipids. The crystal structure of the PDC109-PhC complex shows that each PhC binds to the corresponding Fn2 domain, while the two domains are on the same face of the protein. Long timescale explicit solvent molecular dynamics (MD) simulations of PDC109, in the presence and absence of PhC, suggest that PhC binding strongly correlates with the relative orientation of choline-phospholipid binding sites of the two Fn2 domains; unless the two domains tightly bind PhCs, they tend to change their relative orientation by deforming the flexible linker. The effective PDC109-PhC association constant of 28 M(-1), estimated from their potential of mean force is consistent with the experimental result. Principal component analysis of the long timescale MD simulations was compared to the significantly less expensive normal mode analysis of minimized structures. The comparison indicates that difference between relative domain motions of PDC109 with bound and unbound PhC is captured by the first principal component in the principal component analysis as well as the three lowest normal modes in the normal mode analysis. The present study illustrates the use of detailed MD simulations to clarify the energetics of specific ligand-domain interactions revealed by a static crystallographic model, as well as their influence on relative domain motions in a multi-domain protein.

Conformational dynamics and ligand binding in the multi-domain protein PDC109.

Social systems have recently attracted much attention, with attempts to understand social behavior with the aid of statistical mechanics applied to complex systems. Collective properties of such systems emerge from couplings between components, for example, individual persons, transportation nodes such as airports or subway stations, and administrative districts. Among various collective properties, criticality is known as a characteristic property of a complex system, which helps the systems to respond flexibly to external perturbations. This work considers the criticality of the urban transportation system entailed in the massive smart card data on the Seoul transportation network. Analyzing the passenger flow on the Seoul bus system during one week, we find explicit power-law correlations in the system, that is, power-law behavior of the strength correlation function of bus stops and verify scale invariance of the strength fluctuations. Such criticality is probed by means of the scaling and renormalization analysis of the modified gravity model applied to the system. Here a group of nearby (bare) bus stops are transformed into a (renormalized) "block stop" and the scaling relations of the network density turn out to be closely related to the fractal dimensions of the system, revealing the underlying structure. Specifically, the resulting renormalized values of the gravity exponent and of the Hill coefficient give a good description of the Seoul bus system: The former measures the characteristic dimensionality of the network whereas the latter reflects the coupling between distinct transportation modes. It is thus demonstrated that such ideas of physics as scaling and renormalization can be applied successfully to social phenomena exemplified by the passenger flow.
