Isoflurane is currently the most common volatile anaesthetic used in laboratory mice, whereas in human medicine the more modern sevoflurane is often used for inhalation anaesthesia. This study aimed to characterize and compare the clinical properties of both anaesthetics for inhalation anaesthesia in mice. In an approach mirroring routine laboratory conditions (spontaneous breathing, gas supply via nose mask, preventing hypothermia by a warming mat) a 50 min anaesthesia was performed. Anaesthetics were administered in oxygen as carrier gas at standardized dosages of 1.5 minimum alveolar concentrations, which was 2.8% for isoflurane and 4.9% for sevoflurane. Both induction and recovery from anaesthesia proceeded quickly, within 1-2 min. During anaesthesia, all reflex testing was negative and no serious impairment of vital functions was found; all animals survived. The most prominent side-effect during anaesthesia was respiratory depression with hypercapnia, acidosis and a marked decrease in respiration rate. Under anaesthesia, heart rate and core body temperature remained within the normal range, but were significantly increased for 12 h after anaesthesia. Locomotor activity, daily food and water consumption and body weight progression showed no abnormalities after anaesthesia. No significant difference was found between the two anaesthetics. In conclusion, isoflurane and sevoflurane provided an equally reliable anaesthesia in laboratory mice.

L&#x26;#39;isoflurane est l&#x26;#39;anesth&#xE9;sique actuellement la plus courante volatile utilis&#xE9;e dans des souris de laboratoire, alors que dans la m&#xE9;decine humaine du s&#xE9;voflurane plus moderne est souvent utilis&#xE9; pour l&#x26;#39;anesth&#xE9;sie par inhalation. Cette &#xE9;tude visait &#xE0; caract&#xE9;riser et de comparer les propri&#xE9;t&#xE9;s cliniques des deux anesth&#xE9;siques pour l&#x26;#39;anesth&#xE9;sie par inhalation chez la souris. Dans une approche refl&#xE9;tant des conditions de laboratoire de routine (respiration spontan&#xE9;e, l&#x26;#39;approvisionnement en gaz par l&#x26;#39;interm&#xE9;diaire d&#x26;#39;un masque nez, pour pr&#xE9;venir l&#x26;#39;hypothermie par un tapis de r&#xE9;chauffement) de 50 min d&#x26;#39;anesth&#xE9;sie a &#xE9;t&#xE9; r&#xE9;alis&#xE9;e. Anesth&#xE9;siques ont &#xE9;t&#xE9; administr&#xE9;s en oxyg&#xE8;ne comme gaz porteur &#xE0; des doses standardis&#xE9;es de 1,5 concentrations minimales alv&#xE9;olaires, qui &#xE9;tait de 2,8% pour l&#x26;#39;isoflurane et 4,9% pour le s&#xE9;voflurane. L&#x26;#39;induction de l&#x26;#39;anesth&#xE9;sie et la r&#xE9;cup&#xE9;ration s&#x26;#39;est faite rapidement, dans les 1-2 min. Isofluran ist derzeit die h&auml;ufigste volatile An&auml;sthetikum in Labor-M&auml;use, verwendet, w&auml;hrend in der Humanmedizin die moderneren Sevofluran oft f&uuml;r Einatmen An&auml;sthesie verwendet wird. Ziel dieser Studie waren charakterisieren und die klinischen Eigenschaften der beiden An&auml;sthetika f&uuml;r Einatmen An&auml;sthesie bei M&auml;usen zu vergleichen. In einem Ansatz Spiegelung Routine Laborbedingungen (Spontane Atmung, Gasversorgung &uuml;ber Nase Maske, Verhinderung von Hypothermie durch eine w&auml;rmende Matte) wurde ein 50 min-An&auml;sthesie durchgef&uuml;hrt. An&auml;sthetika wurden in Sauerstoff als Tr&auml;gergas bei standardisierten Dosierungen von 1,5 minimale alveol&auml;re Konzentration verabreicht, die war 2,8 % f&uuml;r Isofluran und Sevofluran 4,9 %. Sowohl die Induktion als auch die Verwertung von An&auml;sthesie verlief schnell, innerhalb von 1-2 min. W&auml;hrend der An&auml;sthesie alle reflex Tests negativ war und keine ernsthafte Beeintr&auml;chtigung der lebenswichtigen Funktionen gefunden wurde; alle Tiere &uuml;berlebten. Die bekannteste Nebenwirkung w&auml;hrend der An&auml;sthesie war Atemdepression mit Hyperkapnie, Azidose und eine deutliche Abnahme der Atmung-Tarif. In Narkose Herzfrequenz und K&ouml;rperkerntemperatur blieb im normalen Bereich, aber stieg signifikant f&uuml;r 12 h nach An&auml;sthesie. Lokomotorische Aktivit&auml;t, t&auml;gliche Nahrung und Wasser-Verbrauch und K&ouml;rper Gewicht Verlauf zeigte keine Auff&auml;lligkeiten nach An&auml;sthesie. Kein signifikanter Unterschied ergab sich zwischen den beiden An&auml;sthetika. Isoflurano &egrave; attualmente il pi&ugrave; comune anestetico volatile utilizzato nei topi di laboratorio, mentre in medicina umana il sevoflurano pi&ugrave; moderna &egrave; spesso usato per anestesia di inalazione. Questo studio finalizzato a caratterizzare e comparare le propriet&agrave; cliniche di entrambi anestetici per anestesia inalazione nei topi. In un approccio mirroring condizioni di laboratorio di routine (respiro spontaneo, la fornitura di gas tramite maschera di naso, prevenire l&#39;ipotermia da un tappetino riscaldante) &egrave; stata eseguita un&#39;anestesia 50 min. Anestetici sono stati amministrati in ossigeno come gas di trasporto a dosaggi standardizzati di 1,5 concentrazione minime alveolare, che era di 2,8% per isoflurano e 4,9% per sevoflurano. Induzione e il recupero dall&#39;anestesia procedette rapidamente, entro 1-2 min. Durante l&#39;anestesia, tutte le reflex test era negativo ed &egrave; stato trovato nessun grave compromissione delle funzioni vitali; tutti gli animali sopravvissero. Detection of persistent pain of a mild-to-moderate degree in laboratory mice is difficult because mice do not show unambiguous symptoms of pain or suffering using standard methods of short-term observational or clinical monitoring. This study investigated the potential use of burrowing performance - a spontaneous and highly motivated behavior - as a measure of post-operative pain in laboratory mice. The influence of minor surgery on burrowing was investigated in adult C57BL/6J mice of both genders in a modified rodent burrowing test (displacement of food pellets from a pellet-filled tube) within the animal's home cage. Almost all (98%) healthy mice burrowed (mean latency 1.3â€‰h, SEM 0.5â€‰h). After surgery without pain treatment, latency of burrowing was significantly prolonged (mean Î” latency 10â€‰h). Analgesic treatment using the anti-inflammatory drug carprofen (5â€‰mg/kg bodyweight) decreased latency of burrowing after surgery (mean Î” latency 5.5â€‰h) to the level found in mice that had been anesthetized (mean Î” latency 5.4â€‰h) or had received anesthesia and analgesia (mean Î” latency 4.6â€‰h). Analgesia during surgery was associated with a significantly earlier onset of burrowing compared to surgery without pain treatment. A distinct gradation in burrowing performance was found ranging from the undisturbed pre-operative status to the intermediate level following anesthesia/analgesia and surgery with analgesia, to the pronounced prolongation of latency to burrow after surgery without pain relief. In conclusion, post-surgical impairment of general condition, probably mainly attributable to pain, can be conveniently assessed in laboratory mice on the basis of the burrowing test.

Analgesie w&auml;hrend der Operation war verbunden mit einer deutlich fr&uuml;heren Beginn der grabende verglichen mit Chirurgie ohne Schmerzbehandlung. Refined vaccines and adjuvants are urgently needed to advance immunization against global infectious challenges such as HIV, hepatitis C, tuberculosis and malaria. Large-scale screening efforts are ongoing to identify adjuvants with improved efficacy profiles. Reactogenicity often represents a major hurdle to the clinical use of new substances. Yet, irrespective of its importance, this parameter has remained difficult to screen for, owing to a lack of sensitive small animal models with a capacity for high throughput testing. Here we report that continuous telemetric measurements of heart rate, heart rate variability, body core temperature and locomotor activity in laboratory mice readily unmasked systemic side-effects of vaccination, which went undetected by conventional observational assessment and clinical scoring. Even minor aberrations in homeostasis were readily detected, ranging from sympathetic activation over transient pyrogenic effects to reduced physical activity and apathy. Results in real-time combined with the potential of scalability and partial automation in the industrial context suggest multiparameter telemetry in laboratory mice as a first-line screen for vaccine reactogenicity. This may accelerate vaccine discovery in general and may further the success of vaccines in combating infectious disease and cancer.

Vacunas y adyuvantes refinados se necesitan con urgencia para avanzar en la inmunizaci&#xF3;n contra los problemas mundiales infecciosas como el VIH, la hepatitis C, la tuberculosis y la malaria. Los grandes esfuerzos de detecci&#xF3;n est&#xE1;n llevando a cabo para identificar adyuvantes con los perfiles de eficacia mejorada. Reactogenicidad a menudo representa un obst&#xE1;culo importante para el uso cl&#xED;nico de nuevas sustancias. Sin embargo, independientemente de su importancia, este par&#xE1;metro se ha mantenido dif&#xED;cil de detectar, debido a la falta de modelos animales sensibles a peque&#xF1;os con una capacidad para realizar pruebas de alto rendimiento. Aqu&#xED; mostramos que las mediciones continuas de telemetr&#xED;a de la frecuencia card&#xED;aca, variabilidad del ritmo card&#xED;aco, la temperatura corporal y la actividad locomotora en ratones de laboratorio f&#xE1;cilmente desenmascarado efectos secundarios sist&#xE9;micos de la vacunaci&#xF3;n, que no fue detectado por la evaluaci&#xF3;n convencional de observaci&#xF3;n y de puntuaci&#xF3;n cl&#xED;nica. Incluso las aberraciones de menor importancia en la homeostasis se detecta f&#xE1;cilmente, que van desde la activaci&#xF3;n simp&#xE1;tica sobre los efectos transitorios pirog&#xE9;nicas de reducci&#xF3;n de la actividad f&#xED;sica y la apat&#xED;a. Resultados en tiempo real combinadas con el potencial de escalabilidad y automatizaci&#xF3;n parcial en el contexto industrial sugieren telemetr&#xED;a multiparam&#xE9;trico en ratones de laboratorio como una pantalla de primera l&#xED;nea para la reactogenicidad de la vacuna. Esto puede acelerar el descubrimiento de la vacuna, en general, y puede favorecer el &#xE9;xito de las vacunas en la lucha contra las enfermedades infecciosas y el c&#xE1;ncer.
