The vascular component of trees possesses some of the most specialized processes active in the formation of roots, stems, and branches, and its wood component continues to be of primary importance to our daily lives. The molecular mechanisms of wood development, however, remain poorly understood with few well-characterized regulatory genes. We have identified a vascular tissue-specific MADS-box gene, Populus tremuloides MADS-box 5 (PTM5) that is expressed in differentiating primary and secondary xylem and phloem. Phylogenetic analysis has shown that PTM5 is a member of the SOC1/TM3 class of MADS-box genes. Temporal expression analysis of PTM5 in staged vascular cambium and other tissues indicated that PTM5 expression is seasonal and is limited to spring wood formation and rapidly expanding floral catkins. Spatial expression analysis using in situ hybridization revealed that PTM5 expression is localized within a few layers of differentiating vascular cambium and xylem tissues as well as the vascular bundles of expanding catkins. Since many MADS-box genes are known to act as transcription factors, these results suggest that the coordinated expression of PTM5 with other vascular developmental genes may be a hallmark of the complex events that lead to the formation of the woody plant body.

La componente vascolare di alberi possiede alcuni dei processi pi&ugrave; specializzati attivi nella formazione di radici, fusti e rami, e la componente legno continua ad essere di primaria importanza per la nostra vita quotidiana. I meccanismi molecolari di sviluppo legno, tuttavia, rimangono scarsamente comprensibili con pochi geni regolatori ben caratterizzati. Abbiamo identificato un gene MADS-box specifico tessuto vascolare, Populus tremuloides-MADS-box 5 (PTM5) che &egrave; espresso nel differenziare il floema e xilema primario e secondario. One of the most important processes to the survival of a species is its ability to reproduce. In plants, SEPALLATA-class MADS-box genes have been found to control the development of the inner whorls of flowers. However, while much is known about floral development in herbaceous plants, similar systems in woody trees remain poorly understood. Populus tremuloides (trembling aspen) is a widespread North American tree having important economic value, and its floral development differs from that of well-studied species in that the flowers have only two whorls and are truly unisexual. Sequence based analyses indicate that PTM3 (Populus tremuloides MADS-box 3), and a duplicate gene PTM4, are related to the SEPALLATA1-and 2-class of MADS-box genes. Another gene, PTM6, is related to SEP3, and each of these genes has a counterpart in the poplar genomic database along with additional members of the A, B, C, D, and E-classes of MADS-box genes. PTM3/4 and 6 are expressed in all stages of male and female aspen floral development. However, PTM3/4 is also expressed in the terminal buds, young leaves, and young stems. In situ RNA localization identified PTM3/4 and 6 transcripts predominantly in the inner, sexual whorl, within developing ovules of female flowers and anther primordia of male flowers. Tree researchers often use heterologous systems to help study tree floral development due to the long juvenile periods found in most trees. We found that the participation of PTM3/4 in floral development is supported by transgenic experiments in both P. tremuloides and heterologous systems such as tobacco and Arabidopsis. However, phenotypic artifacts were observed in the heterologous systems. Together the results suggest a role for poplar SEP-class genes in reproductive viability.

Uno de los procesos m&aacute;s importantes para la supervivencia de una especie es su capacidad para reproducirse. En las plantas, se han encontrado genes MADS-box de SEPALLATA-clase para controlar el desarrollo de las espiras interiores de flores. Sin embargo, aunque se sabe mucho sobre desarrollo floral en plantas herb&aacute;ceas, sistemas similares en le&ntilde;osas, &aacute;rboles siguen siendo mal entendidos. Populus tremuloides (temblor aspen) es un &aacute;rbol norteamericano generalizado con importante valor econ&oacute;mico, y su desarrollo floral de especies estudiadas difiere en que las flores tienen s&oacute;lo dos verticilos y son verdaderamente unisexuales. Secuencia basada en an&aacute;lisis indican que PTM3 (Populus tremuloides MADS-cuadro 3), y un gen duplicado PTM4, est&aacute;n relacionados con la SEPALLATA1- y 2-clase de genes MADS-box. Se relaciona con otro gen, PTM6, SEP3, y cada uno de estos genes tiene su contraparte en la base de datos gen&oacute;micos de &aacute;lamo junto con miembros adicionales de la A, B, C, D y E-clases de genes MADS-box. PTM3/4 y 6 se expresan en todas las etapas de desarrollo floral de &aacute;lamo tembl&oacute;n masculino y femenino. Sin embargo, PTM3/4 tambi&eacute;n se expresa en los cogollos, hojas j&oacute;venes y tallos j&oacute;venes. Localizaci&oacute;n de RNA in situ hab&iacute;a identificado transcripciones PTM3/4 y 6 predominante en el verticilo interno, sexual, en desarrollo de &oacute;vulos de las flores femeninas y los primordios de la antera de flores masculinas. Los investigadores de &aacute;rbol a menudo utilizan sistemas heter&oacute;logos para ayudar a estudiar el desarrollo floral &aacute;rbol debido a los per&iacute;odos mucho menores en la mayor&iacute;a de los &aacute;rboles. In a past article, our lab described the identification and characterization of a novel vegetative MADS-box gene from quaking aspen trees, Populus tremuloides MADS-box 5 (PTM5). PTM5 was shown to be a member of the SOC1/TM3 class of MADS-box genes with a seasonal expression pattern specific to developing vascular tissues including the vascular cambium, the precursor to all woody branches, stems, and roots. Since the proper function of MADS-box proteins is dependent on specific interactions with other regulatory proteins, we further examined PTM5 protein-protein interactions as a means to better understand its function. Through yeast two-hybrid analyses, it was demonstrated that, like other SOC1/TM3 class proteins, PTM5 is capable of interacting with itself as well as other MADS-box proteins from aspen. In addition, yeast two-hybrid library screening revealed that PTM5 interacts with two non-MADS proteins, an actin depolymerizing factor (PtADF) and a novel leucine-rich repeat protein (PtLRR). In situ RNA localization was used to verify the overlapping expression patterns of these genes, and transgenic studies showed that over-expression of PTM5 in aspen causes alterations in root vasculature and root biomass development consistent with the cell growth and expansion functions of related ADF and LRR genes. These results suggest that the interaction of vegetative MADS-box genes with specific protein cofactors is a key step in the mechanisms that control woody tissue development in trees.

Dans un article pass&eacute;, notre laboratoire a d&eacute;crit l&#39;identification et la caract&eacute;risation d&#39;un g&egrave;ne MADS-box v&eacute;g&eacute;tative provenant d&#39;arbres de tremble, Populus tremuloides MADS-box 5 (PTM5). PTM5 s&#39;est av&eacute;r&eacute; &ecirc;tre un membre de la classe SOC1/TM3 de g&egrave;nes MADS-box avec un mod&egrave;le d&#39;expression saisonni&egrave;re sp&eacute;cifique au d&eacute;veloppement des tissus vasculaires, y compris le cambium vasculaire, le pr&eacute;curseur de toutes les branches ligneuses, tiges et racines. En un art&iacute;culo pasado, nuestro laboratorio describe la identificaci&oacute;n y caracterizaci&oacute;n de un gene nuevo de MADS-box vegetativo de &aacute;rboles quaking aspen, Populus tremuloides MADS-box 5 (PTM5). With the completion of the poplar tree genome database, Populus species have become one of the most useful model systems for the study of woody plant biology. Populus tremuloides (quaking aspen) is the most wide-spread tree species in North America, and its rapid growth generates the most abundant wood-based biomass out of any other plant species. To study such beneficial traits, there is a need for easier and more efficient transformation procedures that will allow the study of large numbers of tree genes. We have developed transformation procedures that are suitable for high-throughput format transformations using either Agrobacterium tumefaciens to produce transformed trees or Agrobacterium rhizogenes to generate hairy roots. Our method uses Agrobacterium inoculated aspen seedling hypocotyls followed by direct thidiazuron (TDZ)-mediated shoot regeneration on selective media. Transformation was verified through beta-glucuronidase (GUS) reporter gene expression in all tree tissues, PCR amplification of appropriate vector products from isolated genomic DNA, and northern hybridization of incorporated and expressed transgenes. The hairy root protocol follows the same inoculation procedures and was tested using GUS reporter gene integration and antibiotic selection. The benefit of these procedures is that they are simple and efficient, requiring no maintenance of starting materials and allowing fully formed transgenic trees (or hairy roots) to be generated in only 3-4 months, rather than the 6-12 months required by more traditional methods. Likewise, the fact that the protocols are amenable to high-throughput formats makes them better suited for large-scale functional genomics studies in poplars.

Nous avons d&eacute;velopp&eacute; des proc&eacute;d&eacute;s de transformation qui se pr&ecirc;tent &agrave; des transformations de format haut d&eacute;bit utilisant soit Agrobacterium tumefaciens pour produire des arbres transform&eacute;s ou Agrobacterium rhizogenes pour g&eacute;n&eacute;rer des racines velues. Mit der Fertigstellung der Pappel Baum Genom Datenbank sind Populus-Arten die n&uuml;tzlichsten Modellsysteme f&uuml;r die Erforschung der waldreichen Pflanzenbiologie geworden. Populus Tremuloides (Zitterpappeln) ist die am meisten verbreiteten Baumarten in Nordamerika und das rasche Wachstum generiert die am h&auml;ufigsten vorkommenden Holz-Biomasse aus jeder anderen Pflanzenarten. Um solche vorteilhafte Merkmale zu studieren, gibt es muss einfacher und effizienter-Transformation, die die Studie einer gro&szlig;en Anzahl von Genen, Baum erm&ouml;glichen. Wir haben Transformation Verfahren entwickelt, die f&uuml;r Hochdurchsatz-Format-Transformationen, die mithilfe von entweder Agrobacterium Tumefaciens an transformierten B&auml;ume oder Agrobacterium Rhizogenes behaarte Wurzeln erzeugen produzieren geeignet sind. Unsere Methode verwendet Agrobacterium beimpften Espe S&auml;mling Hypocotyls von direkten Thidiazuron (TDZ gefolgt)-Shoot Regenerierung auf selektiven Medien vermittelt. Transformation wurde durch Beta-Glukuronidase (GUS) Reporter-Gen-Ausdrucks in allen Baum Geweben, PCR-Verst&auml;rkung der entsprechenden Vektor-Produkte von isolierten genomic DNA, verifiziert und n&ouml;rdlichen Hybridation der aufgenommen und transgenen ausgedr&uuml;ckt. Das behaarte Stamm-Protokoll folgt die gleichen Impfung-Verfahren und die mit GUS Reporter-gen Integration und Antibiotikum getestet wurde. Der Vorteil dieser Verfahren ist, dass sie einfach und effizient, erfordern keine Wartung der Ausgangsstoffe und erm&ouml;glicht vollst&auml;ndig transgene B&auml;ume (oder behaarte Wurzeln) generiert werden, in nur 3 Monaten, anstatt die 6-12 Monate ben&ouml;tigt, die durch traditionelle Methoden gebildet. Ebenso formatiert die Tatsache, dass die Protokolle f&uuml;r Hochdurchsatz-zug&auml;nglich macht, die sie besser f&uuml;r gro&szlig; angelegte funktionelle Genomik Studien in Pappeln geeignet.

Con il completamento del database del genoma albero di pioppo, specie di Populus sono diventati uno dei pi&ugrave; utili sistemi modello per lo studio della biologia della pianta legnosa. Populus tremuloides (quaking aspen) &egrave; la specie di albero pi&ugrave; diffuso in Nord America, e la sua rapida crescita genera il pi&ugrave; abbondante biomassa di legno fuori qualsiasi altra specie di pianta. Per lo studio di tali caratteristiche benefiche, c&#39;&egrave; una necessit&agrave; per le procedure di trasformazione pi&ugrave; facile e pi&ugrave; efficiente che permetter&agrave; lo studio di un gran numero di geni di albero. Abbiamo sviluppato le procedure di trasformazione che sono adatte per le trasformazioni di formato ad alta velocit&agrave; utilizzando sia Agrobacterium tumefaciens per produrre alberi trasformati o Agrobacterium rhizogenes per generare radici pelose. Il nostro metodo utilizza Agrobacterium inoculati semenzale aspen ipocotili seguite da thidiazuron diretto (TDZ)-mediata rigenerazione sparare su terreni selettivi. Trasformazione &egrave; stata verificata attraverso la beta-glucuronidasi (GUS) espressione del gene reporter in tutti i tessuti di albero, di appropriati prodotti vettoriali da DNA genomic isolato, l&#39;amplificazione di PCR e l&#39;ibridazione nordica di incorporato ed espresso transgeni. Il protocollo di radice pelosa segue le stesse procedure di inoculazione ed &egrave; stato testato utilizzando GUS reporter integrazione e antibiotico selezione genetica. Il vantaggio di queste procedure &egrave; che essi sono semplici ed efficienti, che richiedono nessuna manutenzione dei materiali di partenza e permettendo completamente formata alberi transgenici (o pelose radici) per essere generato in solo 3-4 mesi, piuttosto che i 6-12 mesi richiesti dai metodi pi&ugrave; tradizionali. Allo stesso modo, il fatto che i protocolli sono suscettibili di alto-rendimento formati rende che loro pi&ugrave; adatti per studi di genomica funzionale su larga scala in pioppi.

Con la terminaci&oacute;n de la base de datos del genoma &Aacute;lamo &aacute;rbol, especie de Populus se ha convertido en uno de los sistemas de modelo m&aacute;s &uacute;tiles para el estudio de la biolog&iacute;a de las plantas le&ntilde;osas. Populus tremuloides (quaking aspen) es la especie de &aacute;rbol m&aacute;s extenso en Am&eacute;rica del norte, y su r&aacute;pido crecimiento genera la m&aacute;s abundante biomasa con base de madera de cualquier otra especie de la planta. Para estudiar tales rasgos beneficiosos, hay una necesidad de procedimientos de transformaci&oacute;n m&aacute;s f&aacute;cil y eficiente que permitir&aacute; el estudio de un gran n&uacute;mero de genes del &aacute;rbol. Hemos desarrollado procedimientos de transformaci&oacute;n que son adecuados para transformaciones de formato de alto rendimiento utilizando cualquier Agrobacterium tumefaciens para producir &aacute;rboles transformados o Agrobacterium rhizogenes generar ra&iacute;ces pilosas. Nuestro m&eacute;todo utiliza Agrobacterium inoculados pl&aacute;ntula aspen hipocotilos seguidos thidiazuron directa (TDZ)-mediada por la regeneraci&oacute;n de brotes en medios selectivos. Transformaci&oacute;n se verific&oacute; a trav&eacute;s de la expresi&oacute;n del gen de beta-glucuronidasa (GUS) reportero en todos los tejidos de &aacute;rbol, amplificaci&oacute;n por PCR de productos apropiados del vector de ADN gen&oacute;mico aislada, y norte hibridaci&oacute;n de incorporado y expresa los transgenes. El protocolo de la ra&iacute;z melenuda sigue los mismos procedimientos de inoculaci&oacute;n y se evalu&oacute; mediante la selecci&oacute;n de integraci&oacute;n y antibi&oacute;tico de gen reportero GUS. La ventaja de estos procedimientos es que son simples y eficientes, que no requieren ning&uacute;n mantenimiento de materias primas y permitiendo completamente formado &aacute;rboles transg&eacute;nicos (o ra&iacute;ces pilosas) que se generen en s&oacute;lo 3-4 meses, en lugar de los 6-12 meses requeridos por los m&eacute;todos m&aacute;s tradicionales. Adem&aacute;s, el hecho de que los protocolos son susceptibles de alto rendimiento formatos hace de que ellos los m&aacute;s adecuados para los estudios de gen&oacute;mica funcional a gran escala en &Aacute;lamos.

This study compared the leaf transcription profiles, physiological characteristics and primary metabolites of two Populus tremuloides genotypes (clones 216 and 271) known to differ in their responses to long-term elevated [CO2] (e[CO2]) at the Aspen free-air CO2 enrichment site near Rhinelander, WI, USA. The physiological responses of these clones were similar in terms of photosynthesis, stomatal conductance and leaf area index under e[CO2], yet very different in terms of growth enhancement (0-10% in clone 216; 40-50% in clone 271). Although few genes responded to long-term exposure to e[CO2], the transcriptional activity of leaf e[CO2]-responsive genes was distinctly different between the clones, differentially impacting multiple pathways during both early and late growing seasons. An analysis of transcript abundance and carbon/nitrogen biochemistry suggested that the CO2-responsive clone (271) partitions carbon into pathways associated with active defense/response to stress, carbohydrate/starch biosynthesis and subsequent growth. The CO2-unresponsive clone (216) partitions carbon into pathways associated with passive defense (e.g. lignin, phenylpropanoid) and cell wall thickening. This study indicates that there is significant variation in expression patterns between different tree genotypes in response to long-term exposure to e[CO2]. Consequently, future efforts to improve productivity or other advantageous traits for carbon sequestration should include an examination of genetic variability in CO2 responsiveness.

Cette &eacute;tude a compar&eacute; les profils de transcription de feuille, les caract&eacute;ristiques physiologiques et les m&eacute;tabolites primaires des deux Populus tremuloides g&eacute;notypes (clones 216 et 271) qui diff&egrave;rent dans leurs r&eacute;ponses &agrave; long terme &eacute;lev&eacute;s [CO2] (e[CO2]) sur le site d&#39;enrichissement de CO2 Aspen &agrave; l&#39;air libre pr&egrave;s de Rhinelander, WI, USA.
