Prostaglandin (PG) D2 is well known as a mediator of inflammation. Hematopoietic PGD synthase (HPGDS) is responsible for the production of PGD2 involved in inflammatory responses. Microglial activation and astrogliosis are commonly observed during neuroinflammation, including that which occurs during demyelination. Using the genetic demyelination mouse twitcher, a model of human Krabbe's disease, we discovered that activated microglia expressed HPGDS and activated astrocytes expressed the DP1 receptor for PGD2 in the brain of these mice. Cultured microglia actively produced PGD2 by the action of HPGDS. Cultured astrocytes expressed two types of PGD2 receptor, DP1 and DP2, and showed enhanced GFAP production after stimulation of either receptor with its respective agonist. These results suggest that PGD2 plays an important role in microglia/astrocyte interaction. We demonstrated that the blockade of the HPGDS/PGD2/DP signaling pathway using HPGDS- or DP1-null twitcher mice, and twitcher mice treated with an HPGDS inhibitor, HQL-79 (4-benzhydryloxy-1-[3-(1H-tetrazol-5-yl)-propyl]piperidine), resulted in remarkable suppression of astrogliosis and demyelination, as well as a reduction in twitching and spasticity. Furthermore, we found that the degree of oligodendroglial apoptosis was also reduced in HPGDS-null and HQL-79-treated twitcher mice. These results suggest that PGD2 is the key neuroinflammatory molecule that heightens the pathological response to demyelination in twitcher mice.

D2 de prostaglandines (PG) est bien connu comme un m&eacute;diateur de l&#39;inflammation. H&eacute;matopo&iuml;&eacute;tique PGD synthase (HPGDS) est responsable de la production de PGD2 impliqu&eacute;s dans les r&eacute;actions inflammatoires. Astrogliosis et activation microgliale sont couramment observ&eacute;s au cours de la neuro-inflammation, y compris celle qui se produit au cours de la d&eacute;my&eacute;linisation. Prostaglandine (PG) D2 ist bekannt als Vermittler der Entz&uuml;ndung. H&auml;matopoetische PGD-Synthase (HPGDS) ist verantwortlich f&uuml;r die Produktion von PGD2 entz&uuml;ndliche Reaktionen beteiligt. Microglial Aktivierung und Astrogliosis sind allgemein beobachtet, w&auml;hrend Neuroinflammation, einschlie&szlig;lich das, was w&auml;hrend der Demyelinisierung auftritt. D2 de prostaglandina (PG) es conocida como un mediador de la inflamaci&oacute;n. Hematopoy&eacute;tica PGD sintasa (HPGDS) es responsable de la producci&oacute;n de PGD2 implicados en las respuestas inflamatorias. La activaci&oacute;n Microglial y astrogliosis com&uacute;nmente se observan durante la neuroinflamaci&oacute;n, la que se produce durante la desmielinizaci&oacute;n incluida. Human cytomegalovirus (CMV) is a leading congenital infectious agent in developed countries. In the past, the incidence of congenital infection has been rather low in Japan because a high seroprevalence of CMV present in young women. However, this seroprevalence has been decreasing in recent years, so that the incidence of congenital CMV infection in Japanese neonates may increase and approach the level seen in other developed countries. The method was used for detecting CMV DNA reported by Barbi et al. [Barbi et al. (1996): Clin Diagn Virol 6:27-32] using a dried blood spot on filter paper, to diagnose congenital CMV infection in Japanese neonates. This method is effective and less laborious than virus isolation both for epidemiological studies and for identifying asymptomatic infected babies. Japanese neonates (1,176) were examined; two of who were asymptomatic were found to be infected.

Jedoch hat diese Seropr&auml;valenz in den letzten Jahren r&uuml;ckl&auml;ufig damit die H&auml;ufigkeit von angeborenen CMV-Infektion bei japanischen Neugeborenen erh&ouml;hen und das Niveau in anderen entwickelten L&auml;ndern zu n&auml;hern kann. Die Methode wurde verwendet f&uuml;r die Erkennung von CMV-DNA berichtet von Barbi Et Al. [Barbi Et Al. (1996): Clin Diagn Virol 06:27-32] mit einem getrocknetem Blut vor Ort auf Filterpapier, angeborene CMV-Infektion bei japanischen Neugeborenen zu diagnostizieren. Citomegalovirus humano (CMV) es un agente infeccioso cong&eacute;nito l&iacute;der en los pa&iacute;ses desarrollados. En el pasado, la incidencia de la infecci&oacute;n cong&eacute;nita ha sido m&aacute;s bien baja en Jap&oacute;n porque una alta seroprevalencia de CMV en mujeres j&oacute;venes. Sin embargo, este seroprevalencia ha disminuido en los &uacute;ltimos a&ntilde;os, por lo que puede aumentar la incidencia de la infecci&oacute;n cong&eacute;nita de CMV en reci&eacute;n nacidos japoneses y se acercan al nivel visto en otros pa&iacute;ses desarrollados. Se utiliz&oacute; el m&eacute;todo para la detecci&oacute;n de ADN de CMV por Barbi et al [Barbi et al (1996): Clin Diagn Virol 6:27-32] utilizando una mancha de sangre seca en papel de filtro, para diagnosticar la infecci&oacute;n cong&eacute;nita de CMV en reci&eacute;n nacidos japoneses. Este m&eacute;todo es eficaz y menos laborioso que el aislamiento del virus para estudios epidemiol&oacute;gicos y para identificar a los fetos infectados asintom&aacute;ticos. Prostaglandin D2 (PGD) is synthesized by hematopoietic PGD synthase (HPGDS) or lipocalin-type PGDS (L-PGDS), depending on the organ in which it is produced, and binds specifically to either DP1 or DP2 receptors. We investigated the role of PGD2 in the pathogenesis of hypoxic-ischemic encephalopathy (HIE) in neonatal mice at postnatal day 7. In wild-type mice, hypoxia-ischemia increased PGD2 production in the brain up to 90-fold compared with the level in sham-operated brains at 10 min after cessation of hypoxia. Whereas the size of the infarct was not changed in L-PGDS or DP2 knock-out mouse brains compared with that in the wild-type HIE brains, it was significantly increased in HPGDS-L-PGDS double knock-out or DP1 knock-out mice. The PGD2 level in L-PGDS, HPGDS, and HPGDS-L-PGDS knock-out mice at 10 min of reoxygenation was 46, 7, and 1%, respectively, of that in the wild-type ones, indicating the infarct size to be in inverse relation to the amount of PGD2 production. DP1 receptors were exclusively expressed in endothelial cells after 1 h of reoxygenation, and cerebral blood flow decreased more rapidly after the onset of hypoxia and did not return to the baseline level after reoxygenation in HPGDS-L-PGDS knock-out mice. Endothelial cells were severely damaged in HPGDS-L-PGDS and DP1 knock-out mice after 1 h of reoxygenation. In the human neonatal HIE brain, HPGDS-positive microglia were increased in number. In conclusion, it is probable that PGD2 protected the neonatal brain from hypoxic-ischemic injury mainly via DP1 receptors by preventing endothelial cell degeneration.

Prostaglandina D2 (DGP) es sintetizada por hematopoy&eacute;tico PGD sintasa (HPGDS) o tipo lipocalina DGPS (L-DGPS), dependiendo del &oacute;rgano en el que se produce y que se une espec&iacute;ficamente a receptores DP1 o DP2. Investigamos el papel de PGD2 en la patogenia de la encefalopat&iacute;a hip&oacute;xica-isqu&eacute;mica (EHI) en ratones neonatales en d&iacute;a postnatal 7. En los ratones de tipo salvaje, hipoxia-isquemia aument&oacute; producci&oacute;n de PGD2 en el cerebro hasta 90-fold en comparaci&oacute;n con el nivel en cerebros de farsa-operado en 10 minutos despu&eacute;s de la cesaci&oacute;n de la hipoxia. Considerando que el tama&ntilde;o del infarto no fue cambiado en cerebros de rat&oacute;n knock-out L-DGPS o DP2 con respecto a los cerebros de la encefalopat&iacute;a hip&oacute;xico-isqu&eacute;mica de tipo salvaje, aument&oacute; significativamente en HPGDS-L-DGPS doble knock-out o ratones knock-out de DP1. El nivel de PGD2 en L-DGPS, HPGDS y HPGDS-L-DGPS ratones knock-out a 10 min de reoxygenation era 46, 7 y 1%, respectivamente, de en los de tipo salvaje, que indica el tama&ntilde;o del infarto que in inverse relation to la cantidad de producci&oacute;n de PGD2. En el cerebro humano de encefalopat&iacute;a hip&oacute;xico-isqu&eacute;mica neonatal, HPGDS-positivo microglia aumentaron en n&uacute;mero. En conclusi&oacute;n, es probable que PGD2 hab&iacute;a protegido el cerebro neonatal de lesi&oacute;n hip&oacute;xico-isqu&eacute;mica principalmente a trav&eacute;s de receptores de DP1 previniendo la degeneraci&oacute;n de c&eacute;lulas endoteliales.

Lipocalin-type prostaglandin (PG) D synthase (L-PGDS) is up-regulated in oligodendrocytes (OLs) in mouse models for genetic neurological disorders including globoid cell leukodystrophy (twitcher) and GM1 and GM2 gangliosidoses and in the brain of patients with multiple sclerosis. Since L-PGDS-deficient twitcher mice undergo extensive neuronal death, we concluded that L-PGDS functions protectively against neuronal degeneration. In this study, we investigated whether L-PGDS is also up-regulated in acute and massive brain injury resulting from neonatal hypoxic-ischemic encephalopathy (HIE). Analysis of brains from human neonates who had died from HIE disclosed that the surviving neurons in the infarcted lesions expressed L-PGDS. Mouse models for neonatal HIE were made on postnatal day (PND) 7. Global infarction in the ipsilateral hemisphere was evident at 24h after reoxygenation in this model. Intense L-PGDS immunoreactivity was already observed at 10 min after reoxygenation in apparently normal neurons in the cortex, and thereafter, in neurons adjacent to the infarcted area. Quantitative RT-PCR revealed that the L-PGDS mRNA level of the infarcted hemisphere was 33-fold higher than that of the sham-operated mouse brains at 1h after reoxygenation and that it decreased to the normal level by 24h thereafter. Furthermore, in both human and mouse brains, many of L-PGDS-positive cells were also immunoreactive for p53; and some of these expressed cleaved caspase-3. The expression of L-PGDS in degenerating neurons implies that L-PGDS functions as an early stress protein to protect against neuronal death in the HIE brain.

Tipo lipocalina (PG) de la prostaglandina D sintasa (L-DGPS) est&aacute; regulado hasta en oligodendrocitos (OLs) en modelos de rat&oacute;n para trastornos neurol&oacute;gicos gen&eacute;ticos como gangliosidosis GM1 y GM2 Leucodistrofia de c&eacute;lulas globoides (twitcher) y en el cerebro de pacientes con esclerosis m&uacute;ltiple. Pues la muerte neuronal extensa ratones de twitcher L-DGPS-deficientes, concluimos que el L-DGPS funciones protectora contra la degeneraci&oacute;n neuronal. En este estudio, investigamos si L-DGPS tambi&eacute;n est&aacute; regulado, hasta en lesi&oacute;n cerebral aguda y masiva resultantes neonatal encefalopat&iacute;a hip&oacute;xica-isqu&eacute;mica (EHI). An&aacute;lisis de cerebros de humanos reci&eacute;n nacidos fallecidos por HIE divulg&oacute; que las neuronas supervivientes en las lesiones infartadas expresaran L-DGPS. Modelos de rat&oacute;n para HIE neonatal se realizaron en d&iacute;a postnatal (PND) 7. Infarto global en el hemisferio ipsilateral era evidente a las 24 h despu&eacute;s de reoxygenation en este modelo. Intensa inmunoreactividad de L-DGPS se observ&oacute; ya en 10 min despu&eacute;s de reoxygenation en las neuronas al parecer normales en la corteza y despu&eacute;s de eso, en las neuronas adyacentes a la zona infartada. RT-PCR cuantitativa revel&oacute; que el nivel de L-DGPS mRNA del hemisferio infartado era 33-fold mayor que el de los cerebros de rat&oacute;n funciona simulada a 1 h despu&eacute;s de reoxygenation y que disminuy&oacute; al nivel normal por 24 h despu&eacute;s de eso. Adem&aacute;s, en ambos humanos y cerebro de rat&oacute;n, muchas de las c&eacute;lulas L-DGPS-positivas tambi&eacute;n eran immunoreactive para p53; y algunas de ellas expresadas exfoliados caspasa-3. La expresi&oacute;n de L-DGPS en neuronas en degeneraci&oacute;n implica que L-DGPS funciona como una prote&iacute;na de estr&eacute;s temprano para proteger contra la muerte neuronal en el cerebro de la encefalopat&iacute;a hip&oacute;xico-isqu&eacute;mica.

Duchenne muscular dystrophy is a fatal muscle wasting disease that is characterized by a deficiency in the protein dystrophin. Previously, we reported that the expression of hematopoietic prostaglandin D synthase (HPGDS) appeared in necrotic muscle fibers from patients with either Duchenne muscular dystrophy or polymyositis. HPGDS is responsible for the production of the inflammatory mediator, prostaglandin D(2). In this paper, we validated the hypothesis that HPGDS has a role in the etiology of muscular necrosis. We investigated the expression of HPGDS/ prostaglandin D(2) signaling using two different mouse models of muscle necrosis, that is, bupivacaine-induced muscle necrosis and the mdx mouse, which has a genetic muscular dystrophy. We treated each mouse model with the HPGDS-specific inhibitor, HQL-79, and measured both necrotic muscle volume and selected cytokine mRNA levels. We confirmed that HPGDS expression was induced in necrotic muscle fibers in both bupivacaine-injected muscle and mdx mice. After administration of HQL-79, necrotic muscle volume was significantly decreased in both mouse models. Additionally, mRNA levels of both CD11b and transforming growth factor beta1 were significantly lower in HQL-79-treated mdx mice than in vehicle-treated animals. We also demonstrated that HQL-79 suppressed prostaglandin D(2) production and improved muscle strength in the mdx mouse. Our results show that HPGDS augments inflammation, which is followed by muscle injury. Furthermore, the inhibition of HPGDS ameliorates muscle necrosis even in cases of genetic muscular dystrophy.

Auparavant, nous avons signal&eacute; que l&#39;expression de la prostaglandine h&eacute;matopo&iuml;&eacute;tiques D synthase (HPGDS) est apparu dans les fibres musculaires n&eacute;crotiques de patients atteints de dystrophie musculaire de Duchenne ou de polymyosite. Distrofia muscular de Duchenne es una muscular fatal enfermedad que se caracteriza por una deficiencia en la prote&iacute;na distrofina. Previamente, inform&oacute; que la expresi&oacute;n de hematopoy&eacute;tico prostaglandina D sintasa (HPGDS) apareci&oacute; en fibras de m&uacute;sculo necr&oacute;ticas de pacientes con distrofia muscular de Duchenne o polimiositis. HPGDS es responsable de la producci&oacute;n del mediador inflamatorio, prostaglandina D(2). En este trabajo, validamos la hip&oacute;tesis de que el HPGDS tiene un papel en la etiolog&iacute;a de la necrosis muscular. Hemos investigado la expresi&oacute;n de HPGDS / prostaglandina D(2) se&ntilde;alizaci&oacute;n con dos modelos de rat&oacute;n diferente de la necrosis del m&uacute;sculo, que es, necrosis muscular inducida por bupivaca&iacute;na y el rat&oacute;n mdx, que tiene una gen&eacute;tica de la distrofia muscular. Tratamos a cada modelo de rat&oacute;n con el inhibidor espec&iacute;fico HPGDS, HQL-79, para medir el volumen de ambos m&uacute;sculo necr&oacute;tico y seleccionado los niveles de mRNA de citoquinas. Despu&eacute;s de la administraci&oacute;n de HQL-79, volumen del m&uacute;sculo necr&oacute;tico disminuy&oacute; significativamente en ambos modelos de rat&oacute;n. Adem&aacute;s, los niveles de mRNA de CD11b y beta1 de factor de crecimiento transformante fueron significativamente inferiores en ratones tratados con HQL y 79 mdx que en animales tratados con veh&iacute;culo. Neonatal hypoxic-ischemic encephalopathy (HIE) is a leading cause of severe and permanent neurologic disability after birth. The inducible cyclooxygenase COX-2, which along with COX-1 catalyzes the first committed step in prostaglandin (PG) synthesis, elicits significant brain injury in models of cerebral ischemia; however its downstream PG receptor pathways trigger both toxic and paradoxically protective effects. Here, we investigated the function of PGE(2) E-prostanoid (EP) receptors in the acute outcome of hypoxic-ischemic (HI) injury in the neonatal rat. We determined the temporal and cellular expression patterns of the EP1-4 receptors before and after HIE and tested whether modulation of EP1-4 receptor function could protect against cerebral injury acutely after HIE. All four EP receptors were expressed in forebrain neurons and were induced in endothelial cells after HIE. Inhibition of EP1 signaling with the selective antagonist SC-51089 or co-activation of EP2-4 receptors with the agonist misoprostol significantly reduced HIE cerebral injury 24 h after injury. These receptor ligands also protected brain endothelial cells subjected to oxygen glucose deprivation, suggesting that activation of EP receptor signaling is directly cytoprotective. These data indicate that the G-protein coupled EP receptors may be amenable to pharmacologic targeting in the acute setting of neonatal HIE.

N&#xE9;onatale enc&#xE9;phalopathie hypoxique-isch&#xE9;mique (EHI) est une cause majeure de handicap neurologique grave et permanente apr&#xE8;s la naissance. Le cyclo-oxyg&#xE9;nase inductible COX-2, qui, avec la COX-1 catalyse la premi&#xE8;re &#xE9;tape engag&#xE9;e dans la prostaglandine (PG) de synth&#xE8;se, suscite une l&#xE9;sion c&#xE9;r&#xE9;brale importante dans les mod&#xE8;les d&#x26;#39;isch&#xE9;mie c&#xE9;r&#xE9;brale, mais son r&#xE9;cepteur aval PG voies de d&#xE9;clenchement des effets toxiques et, paradoxalement, de protection. Die durch Induktion Cyclooxygenase COX-2, die zusammen mit der COX-1 den ersten Foul Schritt in der Synthese der Prostaglandine (PG) katalysiert, entlockt erhebliche-Hirn-Trauma in Modellen der zerebralen Isch&auml;mie; aber seine nachgelagerten PG-Rezeptor-Wege sowohl toxische als auch paradoxerweise sch&uuml;tzende Effekte ausl&ouml;sen. Hier untersuchten wir die Funktion des PGE(2) E-Eikosanoide (EP)-Rezeptoren in dieser akuten hypoxic isch&auml;mische (HI) Verletzungen bei neugeborenen Ratten. Neonatal, encefalopat&#xED;a hip&#xF3;xico-isqu&#xE9;mica (EHI) es la principal causa de discapacidad neurol&#xF3;gica severa y permanente despu&#xE9;s del nacimiento. El inducible ciclooxigenasa COX-2, que junto con la COX-1 cataliza el primer paso comprometido en la prostaglandina (PG) s&#xED;ntesis, provoca una lesi&#xF3;n cerebral significativa en modelos de isquemia cerebral, sin embargo su receptor aguas abajo PG v&#xED;as gatillo ambos efectos t&#xF3;xicos y de protecci&#xF3;n parad&#xF3;jicamente. En este caso, se investig&#xF3; la funci&#xF3;n de los PGE (2) E-prostanoides (PE), los receptores en el resultado de la aguda hip&#xF3;xico-isqu&#xE9;mica (HI), una lesi&#xF3;n en la rata neonatal. Se determinaron los patrones de expresi&#xF3;n temporal y celulares de los receptores EP1-4 antes y despu&#xE9;s de HIE y probaron si la modulaci&#xF3;n de la funci&#xF3;n del receptor EP1-4 podr&#xED;a proteger contra la lesi&#xF3;n cerebral aguda despu&#xE9;s de la HIE. Los cuatro receptores EP se expresan en las neuronas del cerebro anterior y se induce en las c&#xE9;lulas endoteliales despu&#xE9;s de la HIE. La inhibici&#xF3;n de la EP1 de se&#xF1;alizaci&#xF3;n con el antagonista selectivo SC-51089 o co-activaci&#xF3;n de los receptores EP2-4 con el misoprostol agonista redujo significativamente lesi&#xF3;n cerebral HIE 24 h despu&#xE9;s de la lesi&#xF3;n. Estos ligandos del receptor tambi&#xE9;n se protegen las c&#xE9;lulas endoteliales del cerebro sometido a la privaci&#xF3;n de ox&#xED;geno la glucosa, lo que sugiere que la activaci&#xF3;n de la se&#xF1;alizaci&#xF3;n del receptor EP es directamente citoprotector. Estos datos indican que la prote&#xED;na G-receptores acoplados a EP pueden ser susceptibles al tratamiento farmacol&#xF3;gico dirigido en la fase aguda de la EHI neonatal.
