Ab&ecirc;talipoprot&eacute;in&eacute;mie (ABL) est une affection rare mend&eacute;lienne du m&eacute;tabolisme lipidique due &agrave; une carence g&eacute;n&eacute;tique dans la prot&eacute;ine de transfert microsomal triglyceride (PSG). Elle est associ&eacute;e aux d&eacute;fauts de transfert lipidique induite par le PSG sur apolipoprot&eacute;ine B (APOB) et troubles de la s&eacute;cr&eacute;tion des lipoprot&eacute;ines contenant l&#39;APOB. R&eacute;cemment, PSG a &eacute;t&eacute; montr&eacute; pour r&eacute;glementer la famille CD1 de mol&eacute;cules pr&eacute;sentatrices de lipides, mais peu est connu sur la fonction immunitaire chez les patients de l&#39;ABL. Ici, nous avons montr&eacute; que ABL est caract&eacute;ris&eacute;e par l&#39;abri d&eacute;fauts affectant la pr&eacute;sentation de soi et antig&egrave;nes microbiens lipidique par groupe 1 (CD1a, CD1b, CD1c) et groupe 2 mol&eacute;cules CD1 (CD1d). Dans les cellules dendritiques isol&eacute;es de patients de l&#39;ABL, carence du PSG a &eacute;t&eacute; associ&eacute; &agrave; la d&eacute;gradation de l&#39;activit&eacute; accrue des mol&eacute;cules CD1 groupe 1. Bien que CD1d &eacute;chapp&eacute; &agrave; la d&eacute;gradation, il n&#39;a pas pu charger les antig&egrave;nes et pr&eacute;sentait des d&eacute;fauts fonctionnels semblables &agrave; celles qui affectent les mol&eacute;cules CD1 de groupe 1. La r&eacute;duction en fonction du CD1 entra&icirc;n&eacute; activation alt&eacute;r&eacute;e de restriction CD1 T et les lymphocytes T (iNKT) tueuses naturels invariants et un nombre r&eacute;duit et des alt&eacute;rations ph&eacute;notypiques des cellules iNKT compatibles avec les centraux et p&eacute;riph&eacute;riques CD1 d&eacute;fauts in vivo. Ces donn&eacute;es soulignent le PSG comme un r&eacute;gulateur unique des humains voies m&eacute;taboliques et immunitaires et r&eacute;v&egrave;lent que l&#39;ABL est non seulement un trouble du m&eacute;tabolisme des lipides, mais aussi une maladie immunitaire impliquant le CD1.

Abetalipoproteinemia (ABL) is a rare Mendelian disorder of lipid metabolism due to genetic deficiency in microsomal triglyceride transfer protein (MTP). It is associated with defects in MTP-mediated lipid transfer onto apolipoprotein B (APOB) and impaired secretion of APOB-containing lipoproteins. Recently, MTP was shown to regulate the CD1 family of lipid antigen-presenting molecules, but little is known about immune function in ABL patients. Here, we have shown that ABL is characterized by immune defects affecting presentation of self and microbial lipid antigens by group 1 (CD1a, CD1b, CD1c) and group 2 (CD1d) CD1 molecules. In dendritic cells isolated from ABL patients, MTP deficiency was associated with increased proteasomal degradation of group 1 CD1 molecules. Although CD1d escaped degradation, it was unable to load antigens and exhibited functional defects similar to those affecting the group 1 CD1 molecules. The reduction in CD1 function resulted in impaired activation of CD1-restricted T and invariant natural killer T (iNKT) cells and reduced numbers and phenotypic alterations of iNKT cells consistent with central and peripheral CD1 defects in vivo. These data highlight MTP as a unique regulator of human metabolic and immune pathways and reveal that ABL is not only a disorder of lipid metabolism but also an immune disease involving CD1.

Abetalipoprotein&auml;mie (ABL) ist eine seltene Mendelian St&ouml;rung des Fettstoffwechsels durch genetische Mangel in mikrosomale Triglycerid-Transferprotein (MTP). Es ist verbunden mit defekten im MTP-vermittelte Lipid-Transfer auf Apolipoprotein B (APOB) und Sekretion von APOB-haltigen Lipoproteinen beeintr&auml;chtigt. Vor kurzem MTP wurde gezeigt, dass die CD1-Familie von Antigen-pr&auml;sentierenden Lipid-Molek&uuml;le regulieren, aber wenig &uuml;ber Immunfunktion bei ABL-Patienten bekannt ist. Hier haben wir gezeigt, dass ABL zeichnet sich durch immun M&auml;ngel, die sich auf Darstellung des selbst und der mikrobiellen Lipid-Antigene von Gruppe 1 (CD1a, CD1b, CD1c) und Gruppe 2 (CD1d) CD1-Molek&uuml;le. In dendritischen Zellen von ABL Patienten isoliert wurde MTP-Mangel erh&ouml;hte Proteasomal Verschlechterung der Gruppe 1 CD1-Molek&uuml;le zugeordnet. CD1d Abbau entziehen, es konnte Antigene laden und funktionelle M&auml;ngel betreffen die Gruppe 1-CD1-Molek&uuml;le &auml;hnlich. Der R&uuml;ckgang der CD1-Funktion f&uuml;hrte zu gest&ouml;rter Aktivierung von CD1-eingeschr&auml;nkte T und invarianten nat&uuml;rliche M&ouml;rder T (iNKT) Zellen und erm&auml;&szlig;igte Zahlen und ph&auml;notypische Ver&auml;nderungen der iNKT-Zellen mit zentralen und peripheren CD1 M&auml;ngel in-vivo. Diese Daten markieren MTP als einzigartige Regulator der menschliche Stoffwechsel- und immun-Wege und zeigen, dass ABL ist nicht nur eine St&ouml;rung des Fettstoffwechsels, sondern auch ein Imundefekt mit CD1.

Abetalipoproteinemia (ABL) &egrave; una rara malattia mendeliana del metabolismo lipidico da deficit genetico nella proteina di trasporto dei trigliceridi microsomiali (MTP). &Egrave; associato con difetti nel trasferimento dei lipidi MTP-mediata su apolipoproteina B (APOB) e alterata secrezione delle lipoproteine contenenti APOB. Recentemente, MTP &egrave; stato dimostrato che regolano la famiglia CD1 di molecole lipidiche presentanti l&#39;antigene, ma poco &egrave; noto sulla funzione immunitaria nei pazienti ABL. Qui, abbiamo dimostrato che ABL &egrave; caratterizzata da difetti immuni che riguardano la presentazione di s&eacute; e degli antigeni microbici lipidi dal gruppo 1 (CD1a, CD1b, CD1c) e gruppo 2 molecole CD1 (CD1d). In cellule dendritiche isolate da pazienti ABL, deficit di MTP &egrave; stato associato con proteosomale aumentata degradazione delle molecole CD1 gruppo 1. Sebbene CD1d sfuggito degrado, fu in grado di caricare gli antigeni ed esposto i difetti funzionali analoghi a quelle che colpiscono le molecole CD1 gruppo 1. La riduzione della funzione CD1 ha portato alterata attivazione del CD1-limitato T e invarianti naturali killer cellule T (iNKT) e riduzione del numero e alterazioni fenotipiche delle cellule iNKT coerente con centrali e periferici CD1 difetti in vivo. Questi dati evidenziano MTP come unico regolatore delle vie metaboliche e sistema immunitario umani e rivelano che ABL &egrave; non solo un disturbo del metabolismo lipidico, ma anche una malattia immunitaria che coinvolgono CD1.

Abetalipoproteinemia (ABL) es un raro desorden mendeliano de metabolismo de los l&iacute;pidos debido a deficiencia gen&eacute;tica en la prote&iacute;na de transferencia de triglic&eacute;ridos micros&oacute;micos (MTP). Se asocia con defectos en la transferencia de l&iacute;pidos mediada por MTP sobre Apolipoprote&iacute;na B (APOB) y alteraci&oacute;n de la secreci&oacute;n de lipoprote&iacute;nas que contienen APOB. Recientemente, MTP fue demostrado para regular la familia CD1 de mol&eacute;culas de l&iacute;pidos presentadoras de ant&iacute;geno, pero poco se sabe sobre la funci&oacute;n inmune en pacientes ABL. Aqu&iacute;, hemos demostrado que la ABL se caracteriza por defectos inmunes que afectan a la presentaci&oacute;n de uno mismo y ant&iacute;genos microbianos l&iacute;pidos por grupo 1 (CD1a, CD1b, CD1c) y grupo 2 mol&eacute;culas CD1 (CD1d). En c&eacute;lulas dendr&iacute;ticas aisladas de pacientes de la ABL, deficiencia MTP se asoci&oacute; con degradaci&oacute;n proteasomal aumento de mol&eacute;culas CD1 de grupo 1. Aunque CD1d escapado de degradaci&oacute;n, se pudo cargar ant&iacute;genos y defectos funcionales similares a las que afectan a las mol&eacute;culas del CD1 de grupo 1. La reducci&oacute;n en la funci&oacute;n de CD1 dio lugar a la activaci&oacute;n deteriorada de T CD1-restringido e invariantes c&eacute;lulas de asesinas naturales del T (tinta) y n&uacute;meros reducidos y alteraciones fenot&iacute;picas de las c&eacute;lulas de tinta compatibles con defectos CD1 centrales y perif&eacute;ricas en vivo. Estos datos destacan MTP como un regulador &uacute;nico de rutas metab&oacute;licas e inmunes humanas y revelan que la ABL es no s&oacute;lo un trastorno del metabolismo de l&iacute;pidos, sino una enfermedad inmune involucrando CD1.

Organic aerosols (OAs) play important roles in multiple atmospheric processes, including climate change, and can impact human health. The physico-chemical properties of OAs are important for all these processes and can evolve through reactions with various atmospheric components, including oxidants. The dynamic nature of these reactions makes it challenging to obtain a true representation of their composition and surface chemistry. Here we investigate the microscopic viscosity of the model OA composed of squalene, undergoing chemical aging. We employ Fluorescent Lifetime Imaging Microscopy (FLIM) in conjunction with viscosity sensitive probes termed molecular rotors, in order to image the changes in microviscosity in real time during oxidation with ozone and hydroxyl radicals, which are two key oxidising species in the troposphere. We also recorded the Raman spectra of the levitated particles to follow the reactivity during particle ozonolysis. The levitation of droplets was achieved via optical trapping that enabled simultaneous levitation and measurement via FLIM or Raman spectroscopy and allowed the true aerosol phase to be probed. Our data revealed a very significant increase in viscosity of the levitated squalene droplets upon ozonolysis, following their transformation from the liquid to solid phase that was not observable when the oxidation was carried out on coverslip mounted droplets. FLIM imaging with sub-micron spatial resolution also revealed spatial heterogeneity in the viscosity distribution of oxidised droplets. Overall, a combination of molecular rotors, FLIM and optical trapping is able to provide powerful insights into OA chemistry and the microscopic structure that enables the dynamic monitoring of microscopic viscosity in aerosol particles in their true phase.
