La maturation des ovocytes, f&#xE9;condation, le d&#xE9;veloppement embryonnaire pr&#xE9;coce se produisent en l&#x26;#39;absence de transcription du g&#xE8;ne. Par cons&#xE9;quent, il est essentiel de comprendre &#xE0; un niveau global des &#xE9;v&#xE9;nements post-transcriptionnels qui sont &#xE0; l&#x26;#39;origine de ces transitions. Ici nous avons utilis&#xE9; une approche syst&#xE9;mique en combinant le profilage ARNm polysome et la bioinformatique pour identifier les ARN motifs de liaison dans les ARNm qui soit entrent ou sortent de la piscine pendant la maturation des ovocytes polysome souris. Association de l&#x26;#39;ARNm avec les polysomes en corr&#xE9;lation avec la traduction active. Oocyte maturation, fertilization, and early embryonic development occur in the absence of gene transcription. Therefore, it is critical to understand at a global level the post-transcriptional events that are driving these transitions. Here we used a systems approach by combining polysome mRNA profiling and bioinformatics to identify RNA-binding motifs in mRNAs that either enter or exit the polysome pool during mouse oocyte maturation. Association of mRNA with the polysomes correlates with active translation. Using this strategy, we identified highly specific patterns of mRNA recruitment to the polysomes that are synchronized with the cell cycle. A large number of the mRNAs recovered with translating ribosomes contain motifs for the RNA-binding proteins DAZL (deleted in azoospermia-like) and CPEB (cytoplasmic polyadenylation element-binding protein). Although a Dazl role in early germ cell development is well established, no function has been described during oocyte-to-embryo transition. We demonstrate that CPEB1 regulates Dazl post-transcriptionally, and that DAZL is essential for meiotic maturation and embryonic cleavage. In the absence of DAZL synthesis, the meiotic spindle fails to form due to disorganization of meiotic microtubules. Therefore, Cpeb1 and Dazl function in a progressive, self-reinforcing pathway to promote oocyte maturation and early embryonic development.

Maturazione degli ovociti, fecondazione e sviluppo embrionale precoce si verificano in assenza di trascrizione del gene. Pertanto, &egrave; fondamentale capire a livello globale l&#39;espresione eventi che determinano queste transizioni. Qui abbiamo utilizzato un approccio di sistema combinando Polisoma mRNA profilatura e bioinformatica per identificare i motivi di RNA-binding in mRNA che entra o uscita il pool Polisoma durante la maturazione degli ovociti del mouse. La maduraci&#xF3;n de ovocitos, fecundaci&#xF3;n y desarrollo embrionario temprano se producen en ausencia de la transcripci&#xF3;n de genes. Por lo tanto, es fundamental para entender a un nivel global de los eventos post-transcripcionales que est&#xE1;n impulsando estas transiciones. Aqu&#xED; hemos utilizado un enfoque de sistemas mediante la combinaci&#xF3;n de perfiles de mRNA polisomas y la bioinform&#xE1;tica para identificar los motivos de uni&#xF3;n al ARN en el ARNm que, o bien entrar o salir de la piscina polisomas durante la maduraci&#xF3;n de los ovocitos de rat&#xF3;n. Asociaci&#xF3;n de ARNm con los polisomas se correlaciona con la traducci&#xF3;n activa. Con esta estrategia, hemos identificado patrones muy espec&#xED;ficos de la contrataci&#xF3;n del mRNA de los polisomas que se sincronizan con el ciclo celular. Un gran n&#xFA;mero de los mRNAs recuperados con ribosomas traducen contienen motivos para la ARN-prote&#xED;nas de uni&#xF3;n DAZL (suprimido en azoospermia-like) y CPEB (poliadenilaci&#xF3;n citoplasm&#xE1;tica elemento de uni&#xF3;n a la prote&#xED;na). Aunque un papel DAZL en el desarrollo temprano de las c&#xE9;lulas germinales est&#xE1; bien establecida, ninguna funci&#xF3;n ha sido descrito durante la transici&#xF3;n de ovocitos a embri&#xF3;n. Se demuestra que regula CPEB1 DAZL post-transcripcional, y que DAZL es esencial para la maduraci&#xF3;n mei&#xF3;tica y la escisi&#xF3;n embrionaria. En ausencia de la s&#xED;ntesis de DAZL, el huso mei&#xF3;tico no se forma debido a la desorganizaci&#xF3;n de los microt&#xFA;bulos mei&#xF3;ticas. The final stages of female gamete maturation occur in the virtual absence of transcription, with gene expression driven by a program of selective unmasking, translation, and degradation of maternal mRNAs. Here we demonstrate that the timing of  mRNA translation in mouse oocytes is dependent on the presence of transcripts with different 3' untranslated regions (UTRs). This 3' UTR heterogeneity directs distinct temporal patterns of translational activation or repression. Inclusion or exclusion of -acting elements is responsible for these divergent regulations. Our findings reveal an additional layer of translation control through alternative polyadenylation usage required to fine-tune the timing of meiosis progression.

With the progress in our understanding of germ cell development, there is an emerging need to investigate the mechanisms of mRNA translation functioning in these cells. Indeed, posttranscriptional regulations of gene expression drive the most important transitions of the germ cell life cycle. Here we describe a strategy to measure mRNA translation in the oocyte, taking advantage of an approach originally developed to identify the transcriptome of a subgroup of cells in a complex cell mixture. This technique takes advantage of the "RiboTag" approach to express an HA-tag on the large ribosomal subunit of the ribosomes in the oocyte. Immunoprecipitation of the extracts followed by qPCR or RNAseq is used to identify mRNAs actively translated.
