Hyperpolarization-activated and cyclic nucleotide-gated (HCN) channels contribute to pacemaker activity, and co-determine the integrative behaviour of neurons and shape their response to synaptic stimulation. Four channel isoforms, HCN1-4, have been described in mammals. Recent studies showed particularly strong expression of HCN1 channels in rods and cones of the rat retina, suggesting that HCN1 channels are involved in the shaping of light responses in both types of photoreceptors. Therefore, the loss of HCN1 channels should lead to pronounced changes in light-induced electrical responses under both scotopic and photopic conditions. This was tested using a mouse transgenic approach. We used immunohistochemistry and patch-clamp recording to study the distribution of HCN1 channels in the mouse retina. HCN1 channels were strongly expressed in rod and cone photoreceptors, as well as in some bipolar, amacrine and ganglion cell types. In electroretinograms (ERGs) from animals in which the HCN1 channel gene had been knocked out, the b-wave amplitudes were unaltered (scotopic conditions) or somewhat reduced (photopic conditions), whereas the duration of both scotopic and photopic ERG responses was strikingly prolonged. Our data suggest that in visual information processing, shortening and shaping of light responses by activation of HCN1 at the level of the photoreceptors is an important step in both scotopic and photopic pathways.

Quatre canaux isoformes, HCN1-4, ont &eacute;t&eacute; d&eacute;crites chez les mammif&egrave;res. Des &eacute;tudes r&eacute;centes ont montr&eacute; une expression particuli&egrave;rement forte de canaux HCN1 en b&acirc;tonnets et les c&ocirc;nes de la r&eacute;tine de rat, sugg&eacute;rant que les cha&icirc;nes HCN1 sont impliqu&eacute;s dans la mise en forme des r&eacute;ponses clair dans les deux types de photor&eacute;cepteurs. Par cons&eacute;quent, la perte de canaux HCN1 devrait conduire &agrave; des changements importants dans les r&eacute;ponses &eacute;lectriques induites par la lumi&egrave;re, dans des conditions photopiques et scotopiques. Hyperpolarisation aktiviert und zyklische Nukleotid-Gating (HCN) Kan&auml;le tragen zur Herzschrittmacher-Aktivit&auml;t, und mitentscheiden das integrative Verhalten der Neuronen und ihre Reaktion auf synaptische Stimulation zu gestalten. Vierkanal-Isoformen, HCN1-4, wurden bei S&auml;ugetieren beschrieben. Neuere Studien zeigten besonders starken Ausdruck HCN1 Kan&auml;le in St&auml;bchen und Zapfen der Netzhaut Ratte, darauf hindeutet, dass HCN1 Kan&auml;le f&uuml;r die Gestaltung der leichten Reaktionen in beide Arten von Photorezeptoren beteiligt sind. Deshalb sollte der Verlust der HCN1 Kan&auml;le zu ausgepr&auml;gten Ver&auml;nderungen in Licht-induzierten elektrischen Antworten sowohl die Augeninnendruckmessung als auch die Photopische Bedingungen f&uuml;hren. Dies wurde getestet, mit einem transgenen Maus-Ansatz. Wir verwendeten Immunhistochemie und Patch-Clamp-Aufnahme um die Verteilung der HCN1 Kan&auml;le in der Netzhaut Maus zu studieren. HCN1 Kan&auml;le wurden stark in Rute und Kegel Photorezeptoren sowie einige bipolaren, Amacrine und Ganglion Zelltypen ge&auml;u&szlig;ert. Im Elektroretinogramm (ERG) von Tieren, die in denen HCN1 Kanal gen ausgeschlagen worden hatte, die b-Wellen-Amplituden wurden unver&auml;ndert (Augeninnendruckmessung Bedingungen) oder etwas reduziert (Photopische Bedingungen), w&auml;hrend die Dauer der sowohl die Augeninnendruckmessung als auch die Photopische ERG Antworten auffallend verl&auml;ngert wurde. Unsere Daten zeigen, dass bei der visuellen Informationsverarbeitung, Verk&uuml;rzung und Gestaltung der leichten Antworten durch Aktivierung von HCN1 auf der Ebene der die Photorezeptoren ein wichtiger Schritt in sowohl die Augeninnendruckmessung als auch die Photopische Wege ist.

Leichte Antworten in der Netzhaut Maus sind auf gezielte L&ouml;schung des Gens Kanal HCN1 verl&auml;ngert.

Studi recenti hanno mostrato particolarmente forte espressione di canali HCN1 in coni e bastoncelli della retina di topo, suggerendo che canali HCN1 sono coinvolti nella formazione delle risposte luce in entrambi i tipi di fotorecettori. Pertanto, la perdita dei canali HCN1 dovrebbe portare a cambiamenti pronunciati nelle risposte elettriche indotta da luce scotopica e fotopica in condizioni. Questo &egrave; stato testato utilizzando un approccio di mouse transgenici. In electroretinograms (ERG) da animali in cui il gene del canale HCN1 era stato eliminato, le ampiezze d&#39;onda b erano inalterate (condizioni scotopica) o ridotte (condizioni di visione fotopica), considerando che la durata delle risposte ERG scotopica e fotopica &egrave; stata straordinariamente prolungata. I nostri dati suggeriscono che nell&#39;elaborazione delle informazioni visive, accorciamento e modellamento delle risposte luce mediante l&#39;attivazione di HCN1 a livello dei fotorecettori &egrave; un passo importante in percorsi scotopica e fotopica.

Risposte luce nella retina del mouse sono prorogate su mirati delezione del gene del canale del HCN1.

Canales (HCN) nucle&oacute;tido-bloqueados activada por hiperpolarizaci&oacute;n y c&iacute;clicos contribuyen a actividad marcapasos y militar el comportamiento integral de las neuronas y su respuesta a la estimulaci&oacute;n sin&aacute;ptica de la forma. Nuestros datos sugieren que en el procesamiento de informaci&oacute;n visual, acortamiento y conformaci&oacute;n de las respuestas de la luz por la activaci&oacute;n de HCN1 en el nivel de los fotorreceptores es un paso importante en las v&iacute;as tanto escot&oacute;picas y fot&oacute;picas.

Mammalian retinae have rod photoreceptors for night vision and cone photoreceptors for daylight and colour vision. For colour discrimination, most mammals possess two cone populations with two visual pigments (opsins) that have absorption maxima at short wavelengths (blue or ultraviolet light) and long wavelengths (green or red light). Microchiropteran bats, which use echolocation to navigate and forage in complete darkness, have long been considered to have pure rod retinae. Here we use opsin immunohistochemistry to show that two phyllostomid microbats, Glossophaga soricina and Carollia perspicillata, possess a significant population of cones and express two cone opsins, a shortwave-sensitive (S) opsin and a longwave-sensitive (L) opsin. A substantial population of cones expresses S opsin exclusively, whereas the other cones mostly coexpress L and S opsin. S opsin gene analysis suggests ultraviolet (UV, wavelengths <400 nm) sensitivity, and corneal electroretinogram recordings reveal an elevated sensitivity to UV light which is mediated by an S cone visual pigment. Therefore bats have retained the ancestral UV tuning of the S cone pigment. We conclude that bats have the prerequisite for daylight vision, dichromatic colour vision, and UV vision. For bats, the UV-sensitive cones may be advantageous for visual orientation at twilight, predator avoidance, and detection of UV-reflecting flowers for those that feed on nectar.

S-Opsin-gen-Analyse legt nahe Ultraviolett (UV, Wellenl&auml;ngen &lt; 400 nm) Empfindlichkeit und Hornhaut Elektroretinogramm Aufnahmen zeigen eine erh&ouml;hte Empfindlichkeit gegen&uuml;ber UV-Licht, das von einer S-Kegel-Sehpigment vermittelt wird. Daher haben Flederm&auml;use bewahrt das angestammte UV tuning S-Kegel-Pigment. Para la discriminaci&oacute;n de color, mayor&iacute;a de los mam&iacute;feros posee dos poblaciones de cono con dos pigmentos visuales (opsins) que tienen absorci&oacute;n m&aacute;xima en longitudes de onda cortas (azul o luz ultravioleta) y longitudes de onda largas (la luz verde o roja). Murci&eacute;lagos de microchiropteran, que utilizan la ecolocalizaci&oacute;n para navegar y forraje en completa oscuridad, se han considerado durante mucho tiempo tienen retinas de barra puro. Aqu&iacute; utilizamos opsina inmunohistoqu&iacute;mica para demostrar que dos phyllostomid microbats, Glossophaga soricina y Carollia perspicillata, poseen una poblaci&oacute;n significativa de conos y expresar dos cono opsins, una opsina (S) de onda corta sensible y una opsina (L) de onda larga-sensible. Una poblaci&oacute;n considerable de conos expresa opsina S exclusivamente, mientras que los otros conos sobre todo coexpresan opsina L y S. S opsina gene an&aacute;lisis sugiere ULTRAVIOLETA (UV, longitudes de onda &lt; 400 nm) sensibilidad y c&oacute;rnea electroretinograma grabaciones muestran una elevada sensibilidad a la luz UV que es mediada por un pigmento visual del cono S. Por lo tanto murci&eacute;lagos han conservado la afinaci&oacute;n de UV ancestral del pigmento S cono. En la retina de mam&#xED;feros, dos tipos de c&#xE9;lulas amacrinas catecolamin&#xE9;rgicos se han descrito. A pesar de las c&#xE9;lulas dopamin&#xE9;rgicas de tipo 1 est&#xE1;n bien caracterizados, la fisiolog&#xED;a de las c&#xE9;lulas de tipo 2 es, hasta ahora, desconocido. Tipo 2 c&#xE9;lulas son GABA&#xE9;rgicas y tienen un eje dendr&#xED;tica extensa, que estratifica en el medio de la capa plexiforme interna. Nuestros datos sugieren que las c&#xE9;lulas de tipo 2 comprende dos sub-poblaciones con id&#xE9;nticas propiedades fisiol&#xF3;gicas: una tiene su somata encuentra en la capa nuclear interna y la otra en la capa de c&#xE9;lulas ganglionares. In the mammalian retina, two types of catecholaminergic amacrine cells have been described. Although dopaminergic type 1 cells are well characterized, the physiology of type 2 cells is, so far, unknown. To target type 2 cells specifically, we used a transgenic mouse line that expresses green fluorescent protein under the control of the tyrosine hydroxylase promoter. Type 2 cells are GABAergic and have an extensive dendritic arbor, which stratifies in the middle of the inner plexiform layer. Our data suggest that type 2 cells comprise two subpopulations with identical physiological properties: one has its somata located in the inner nuclear layer and the other in the ganglion cell layer. Immunostaining with bipolar cell markers suggested that type 2 cells receive excitatory inputs from type 3 OFF and type 5 ON bipolar cells. Consistently, patch-clamp recordings showed that type 2 cells are ON-OFF amacrine cells. Blocking excitatory inputs revealed that different rod and cone pathways are active under scotopic and mesopic light conditions. Blockade of inhibitory inputs led to membrane potential oscillations in type 2 cells, suggesting that GABAergic and glycinergic amacrine cells strongly influence type 2 cell signaling. Among the glycinergic amacrine cells, we identified the VGluT3-immunoreactive amacrine cell as a likely candidate. Collectively, light responses of type 2 cells were remarkably uniform over a wide range of light intensities. These properties point toward a general function of type 2 cells that is maintained under scotopic and mesopic conditions.

Dans la r&#xE9;tine chez les mammif&#xE8;res, deux types de cellules amacrines cat&#xE9;cholaminergiques ont &#xE9;t&#xE9; d&#xE9;crits. Bien dopaminergiques de type 1 des cellules sont bien caract&#xE9;ris&#xE9;s, la physiologie de cellules de type 2 est, &#xE0; ce jour, inconnue. Pour cibler sp&#xE9;cifiquement les cellules de type 2, nous avons utilis&#xE9; une lign&#xE9;e de souris transg&#xE9;nique qui exprime la prot&#xE9;ine fluorescente verte sous le contr&#xF4;le du promoteur de la tyrosine hydroxylase. De type 2 cellules sont GABAergiques et avoir une tonnelle dendritique vaste, qui stratifie dans le milieu de la couche plexiforme interne. Constamment, de patch-clamp enregistrements ont montr&#xE9; que les cellules de type 2 sont ON-OFF cellules amacrines.
