A common obstacle to the survival of encapsulated tissue is oxygen insufficiency. This appears particularly true of encapsulated pancreatic Î²-cells. Our work investigates a fluorescent hypoxia detection system for early recognition of hypoxic stress in encapsulated pancreatic tissue. Murine insulinoma (MIN6) cells were engineered to produce a red fluorescent protein under the control of hypoxia-inducible-factor-1. Aggregates of these cells were encapsulated in poly(ethylene glycol) hydrogels at densities of 200,000, 600,000, and 1 million cells per capsule then incubated in either a 1% or 20% oxygen environment. Cell function was evaluated by daily measurement of glucose-stimulated insulin secretion. Encapsulated cells were also fluorescently imaged periodically over 72 h for expression of the marker signal. Results indicate that oxygen insufficiency severely impacts insulin release from MIN6 cells, and that large aggregates are especially vulnerable to oxygen limitations. Our marker was found to be successfully indicative of hypoxia and could be used as a predictor of subsequent insulin release. Further work will be required to fully characterize signal dynamics and to evaluate in vivo efficacy. The method presented here represents a unique and valuable approach to detecting hypoxic stress in living tissues which may prove useful to a variety of fields of biological research.

Un obstacle commun &#xE0; la survie du tissu encapsul&#xE9; est l&#x26;#39;insuffisance d&#x26;#39;oxyg&#xE8;ne. Cela semble particuli&#xE8;rement vrai du pancr&#xE9;as encapsul&#xE9; les cellules &#xDF;. Notre travail &#xE9;tudie un syst&#xE8;me de d&#xE9;tection fluorescente hypoxie pour la reconnaissance pr&#xE9;coce de stress hypoxique dans le tissu pancr&#xE9;atique encapsul&#xE9;. Murins insulinome (MIN6) les cellules ont &#xE9;t&#xE9; modifi&#xE9;es pour produire une prot&#xE9;ine fluorescente rouge sous la commande d&#x26;#39;inductible en hypoxie-facteur-1. Granulats de ces cellules ont &#xE9;t&#xE9; encapsul&#xE9;es dans du poly (&#xE9;thyl&#xE8;ne glycol) hydrogels &#xE0; des densit&#xE9;s de 200.000, 600.000 et 1 million de cellules par capsule, puis incub&#xE9;s dans un 1% ou 20% d&#x26;#39;oxyg&#xE8;ne environnement. Des travaux compl&#xE9;mentaires seront n&#xE9;cessaires pour caract&#xE9;riser compl&#xE8;tement la dynamique du signal et d&#x26;#39;&#xE9;valuer l&#x26;#39;efficacit&#xE9; in vivo. La m&#xE9;thode pr&#xE9;sent&#xE9;e ici repr&#xE9;sente une approche unique et pr&#xE9;cieuse &#xE0; la d&#xE9;tection de stress hypoxique dans les tissus vivants qui peuvent se r&#xE9;v&#xE9;ler utiles pour une vari&#xE9;t&#xE9; de domaines de la recherche biologique.

Eine gemeinsame Hindernis f&uuml;r das &Uuml;berleben der gekapselten Gewebe ist Sauerstoff-Insuffizienz. Dies scheint von gekapselten pankreatischer &beta;-Zellen besonders zutreffend. Unsere Arbeit untersucht eine fluoreszierende Hypoxie-Detection-System f&uuml;r Fr&uuml;herkennung von Hypoxie Stress in gekapselten Bauchspeicheldr&uuml;sengewebe. Murine Insulinom (MIN6) Zellen wurden so konstruiert, dass um ein rotes fluoreszierendes Protein unter der Kontrolle der Hypoxie-durch Induktion-Faktor-1 zu produzieren. Aggregate dieser Zellen wurden in Poly(ethylene glycol) Hydrogele bei dichten von 200.000, 600.000 und 1 Million Zellen pro Kapsel, die dann in einem 1 % oder 20 % Sauerstoff Umwelt bebr&uuml;tet gekapselt. Zelle-Funktion wurde durch t&auml;gliche Messung der Glukose stimuliert Insulin-Sekretion ausgewertet. Eingekapselte Zellen wurden auch Leuchtstoff in regelm&auml;&szlig;igen Abst&auml;nden &uuml;ber 72 h f&uuml;r Ausdruck des Signals Markierung abgebildet. Ergebnisse zeigen, dass Sauerstoff Insuffizienz wirkt sich stark Insulin-Freisetzung aus MIN6 Zellen und gro&szlig;e Aggregate sind besonders anf&auml;llig f&uuml;r Sauerstoff-Einschr&auml;nkungen. Unsere Markierung wurde erfolgreich auf Hypoxie hinweisen und als ein Pr&auml;diktor f&uuml;r sp&auml;tere Insulin-Freisetzung eingesetzt werden. Weiterer arbeiten werden erforderlich, um Signal Dynamik vollst&auml;ndig zu charakterisieren und in-vivo Wirksamkeit zu bewerten. Die hier vorgestellte Methode stellt einen einzigartigen und wertvollen Ansatz die Aufdeckung von Hypoxie Spannung im lebenden Gewebe, die zu einer Vielzahl von Bereichen der biologischen Forschung n&uuml;tzlich erweisen k&ouml;nnen.

Un ostacolo comune per la sopravvivenza del tessuto incapsulato &egrave; insufficienza di ossigeno. Questo sembra particolarmente vero delle &beta;-cellule pancreatiche incapsulate. Il nostro lavoro indaga su un sistema di rilevazione di ipossia fluorescente per il riconoscimento precoce di stress ipossico nel tessuto pancreatico incapsulato. Cellule murine insulinoma (MIN6) sono state progettate per produrre una proteina fluorescente rossa sotto il controllo di ipossia-inducible factor-1. Aggregati di queste cellule sono stati incapsulati in idrogel poly(ethylene glycol) a densit&agrave; di 1 milione di cellule per capsula poi incubati in un 1% o 20% ossigeno ambiente, 600.000 e 200.000. Il nostro marcatore &egrave; stato trovato per essere correttamente indicativo di ipossia e potrebbe essere usato come un predittore di rilascio di insulina successivi. Ulteriore lavoro sar&agrave; richiesto per caratterizzare completamente dinamica del segnale e per valutare l&#39;efficacia in vivo. Un obst&#xE1;culo com&#xFA;n para la supervivencia del tejido encapsulado es la insuficiencia de ox&#xED;geno. Esto parece particularmente cierto en p&#xE1;ncreas encapsulado de las c&#xE9;lulas &#xDF;. Nuestro trabajo investiga un sistema de detecci&#xF3;n de la hipoxia fluorescente para la detecci&#xF3;n temprana del estr&#xE9;s hip&#xF3;xico en el tejido pancre&#xE1;tico encapsulado. Murino insulinoma (MIN6) las c&#xE9;lulas fueron dise&#xF1;ados para producir una prote&#xED;na fluorescente de color rojo bajo el control de la hipoxia-inducible factor 1. Los agregados de estas c&#xE9;lulas se encapsulan en poli (etileno glicol) hidrogeles a densidades de 200.000, 600.000, y 1 mill&#xF3;n de c&#xE9;lulas por c&#xE1;psula luego incubadas en un entorno de ox&#xED;geno ya sea 1% o 20%. Funci&#xF3;n de la c&#xE9;lula se evalu&#xF3; mediante la medici&#xF3;n diaria de glucosa estimulada por la secreci&#xF3;n de insulina. Las c&#xE9;lulas encapsuladas tambi&#xE9;n fueron fluorescentemente fotografiada peri&#xF3;dicamente durante 72 h para la expresi&#xF3;n de la se&#xF1;al de marcador. Los resultados indican que la insuficiencia de ox&#xED;geno afecta gravemente la liberaci&#xF3;n de insulina de las c&#xE9;lulas MIN6, y que los grandes agregados son especialmente vulnerables a las limitaciones de ox&#xED;geno. Nuestro marcador se encontr&#xF3; que era indicativo de la hipoxia con &#xE9;xito y podr&#xED;a ser utilizado como un predictor de la liberaci&#xF3;n de insulina posterior.
