Emphasis has been given to the molecular design of new carriers for the delivery of insulin, calcitonin and various types of interferons for the treatment of diabetes, osteoporosis, multiple sclerosis and cancer. Most popular carriers include advanced designs of swollen hydrogels prepared from neutral or intelligent polymeric networks. The ability to treat cancer by targeting delivery through angiogenesis is also discussed and antiangiogenic drugs in clinical trials are presented. Delivery methods that specifically use nanoparticles are also highlighted, including both degradable and nondegradable polymers.

Human mesenchymal stem cells (hMSCs) were infected with an adenovirus expressing integrin-linked kinase (ILK) to understand the role of cell-ECM signal transduction cascades in suppressing anoikis. Survivability of ILK-infected hMSCs encapsulated in poly(ethylene glycol) (PEG) hydrogels, an anoikis-inducing environment, was sustained at 90% over 7 weeks, and survival was attributed to increased protein kinase B (PKB/Akt) activation. hMSCs encapsulated in RGD-modified hydrogels induced an upregulation in ILK production, PKB/Akt activation, and subsequent survival to the same extent of ILK-infected, encapsulated hMSCs. As negative controls, encapsulated hMSCs were infected with cyclization recombinase (a protein not associated with cell survival)-expressing virus, and uninfected hMSCs exhibited very little ILK production, PKB/Akt activation, and survival ( approximately 55% after 7 weeks). As a measure of cell-matrix interactions, vinculin was also quantified for the encapsulated hMSCs and found to be 30-fold greater for cells encapsulated in RGD-modified hydrogels and fivefold greater for ILK-infected hMSCs than controls, indicating that cell-material interactions are inducing the cell survivability of hMSCs encapsulated in RGD-modified hydrogels. In sum, ILK infection can support cell survival in the absence of matrix interactions and enable fundamental studies of three-dimensional cell function in response to extrinsic signals, independently of matrix-ligand interactions.

Humaner mesenchymaler Stammzellen (hMSCs) waren ein Adenovirus Ausdr&uuml;cken Integrin-linked Kinase (ILK) zum Verst&auml;ndnis der Rolle der Zelle-ECM Signaltransduktion Kaskaden bei der Unterdr&uuml;ckung von Anoikis infiziert. &Uuml;berlebensf&auml;higkeit des ILK-infizierten hMSCs in Poly(ethylene glycol) (PEG) Hydrogele, Anoikis-induzierende Umgebung eingekapselt wurde bei 90 % innerhalb von 7 Wochen aufrechterhalten und &Uuml;berleben war zur&uuml;ckzuf&uuml;hren auf eine erh&ouml;hte Protein-Kinase-B (PKB/Akt)-Aktivierung. hMSCs gekapselt in RGD-modifizierte Hydrogele induziert eine Fehlregulation im ILK-Produktion, PKB/Akt-Aktivierung und anschlie&szlig;ende &Uuml;berleben im gleichen Umfang des ILK-infiziert, gekapselte hMSCs. We have studied the impact of overexpression of an intracellular signaling protein, integrin-linked kinase (ILK), on the survival and function of encapsulated islet tissue used for the treatment of type 1 diabetes. The dimensions of the encapsulated tissue can impact the stresses placed on the tissue and ILK overexpression shows the ability to extend function of dissociated cells as well as intact islets. These results suggest that lost cell-extracellular matrix interactions in cell encapsulation systems can lead to decreased insulin secretion and ILK signaling is a target to overcome this phenomenon.

A common obstacle to the survival of encapsulated tissue is oxygen insufficiency. This appears particularly true of encapsulated pancreatic Î²-cells. Our work investigates a fluorescent hypoxia detection system for early recognition of hypoxic stress in encapsulated pancreatic tissue. Murine insulinoma (MIN6) cells were engineered to produce a red fluorescent protein under the control of hypoxia-inducible-factor-1. Aggregates of these cells were encapsulated in poly(ethylene glycol) hydrogels at densities of 200,000, 600,000, and 1 million cells per capsule then incubated in either a 1% or 20% oxygen environment. Cell function was evaluated by daily measurement of glucose-stimulated insulin secretion. Encapsulated cells were also fluorescently imaged periodically over 72 h for expression of the marker signal. Results indicate that oxygen insufficiency severely impacts insulin release from MIN6 cells, and that large aggregates are especially vulnerable to oxygen limitations. Our marker was found to be successfully indicative of hypoxia and could be used as a predictor of subsequent insulin release. Further work will be required to fully characterize signal dynamics and to evaluate in vivo efficacy. The method presented here represents a unique and valuable approach to detecting hypoxic stress in living tissues which may prove useful to a variety of fields of biological research.

Eine gemeinsame Hindernis f&uuml;r das &Uuml;berleben der gekapselten Gewebe ist Sauerstoff-Insuffizienz. Dies scheint von gekapselten pankreatischer &beta;-Zellen besonders zutreffend. Unsere Arbeit untersucht eine fluoreszierende Hypoxie-Detection-System f&uuml;r Fr&uuml;herkennung von Hypoxie Stress in gekapselten Bauchspeicheldr&uuml;sengewebe. Murine Insulinom (MIN6) Zellen wurden so konstruiert, dass um ein rotes fluoreszierendes Protein unter der Kontrolle der Hypoxie-durch Induktion-Faktor-1 zu produzieren. Aggregate dieser Zellen wurden in Poly(ethylene glycol) Hydrogele bei dichten von 200.000, 600.000 und 1 Million Zellen pro Kapsel, die dann in einem 1 % oder 20 % Sauerstoff Umwelt bebr&uuml;tet gekapselt. Zelle-Funktion wurde durch t&auml;gliche Messung der Glukose stimuliert Insulin-Sekretion ausgewertet. Eingekapselte Zellen wurden auch Leuchtstoff in regelm&auml;&szlig;igen Abst&auml;nden &uuml;ber 72 h f&uuml;r Ausdruck des Signals Markierung abgebildet. Ergebnisse zeigen, dass Sauerstoff Insuffizienz wirkt sich stark Insulin-Freisetzung aus MIN6 Zellen und gro&szlig;e Aggregate sind besonders anf&auml;llig f&uuml;r Sauerstoff-Einschr&auml;nkungen. Unsere Markierung wurde erfolgreich auf Hypoxie hinweisen und als ein Pr&auml;diktor f&uuml;r sp&auml;tere Insulin-Freisetzung eingesetzt werden.
