Studies show that sex plays a role in stress-related depression, with women experiencing a higher vulnerability to its effect. Two major targets of antidepressants are brain-derived neurotrophic factor (BDNF) and cyclic adenosine monophosphate response element-binding protein (CREB). The aim of this study was to investigate the levels of CREB, phosphorylation of CREB (pCREB), and BDNF in stress-related brain regions of male and female rats after stress and recovery. CREB and pCREB levels were examined in CA1, CA2, CA3, paraventricular nucleus of the thalamus (PVT), amygdala, anterior cingulate area, dorsal part (ACAd), and infralimbic area of prefrontal cortex (PFC), whereas dentate gyrus (DG) and prelimbic area (PL) of PFC were examined for BDNF levels. Our results demonstrate that levels of CREB and pCREB in male CA1, CA2 and CA3, PVT, amygdala, and ACAd were reduced by stress, whereas the same brain regions of female rats exhibited no change. BDNF levels were decreased by chronic stress in female PL but were increased by acute stress in female DG. BDNF levels in male DG and PL were found not to undergo change in response to stress. Abnormalities in morphology occurred after chronic stress in males but not in females. In all cases, the levels of CREB, pCREB, and BDNF in recovery animals were comparable to the levels of these proteins in control animals. These findings demonstrate a sexual dimorphism in the molecular response to stress and suggest that these differences may have important implications for potential therapeutic treatment of depression.

Sex differences in the effects of acute and chronic stress and recovery after long-term stress on stress-related brain regions of rats.

Des &eacute;tudes montrent que sexe joue un r&ocirc;le dans la d&eacute;pression li&eacute;e au stress, avec des femmes touch&eacute;s par une plus grande vuln&eacute;rabilit&eacute; &agrave; son effet. Deux cibles principales des antid&eacute;presseurs sont brain-derived neurotrophic factor (BDNF) et la prot&eacute;ine de liaison &eacute;l&eacute;ment de r&eacute;ponse ad&eacute;nosine monophosphate cyclique (CREB). Le but de cette &eacute;tude &eacute;tait d&#39;enqu&ecirc;ter sur les niveaux de CREB, la phosphorylation de CREB (pCREB) et le BDNF dans les r&eacute;gions du cerveau li&eacute;es au stress des rats m&acirc;les et femelles apr&egrave;s le stress et la r&eacute;cup&eacute;ration. BDNF dans male DG et PL s&#39;observait ne pas &agrave; subir une transformation en r&eacute;ponse au stress. Les anomalies morphologiques s&#39;est produite apr&egrave;s un stress chronique chez les m&acirc;les, mais pas chez les femelles. Dans tous les cas, les niveaux de BDNF CREB et pCREB chez les animaux de r&eacute;cup&eacute;ration &eacute;taient comparables &agrave; des niveaux de ces prot&eacute;ines chez les animaux t&eacute;moins. BDNF Niveaus wurden durch chronischen Stress in weiblichen PL verringert aber wurden durch akuten Stress in der weiblichen GD erh&ouml;ht. BDNF Niveaus in m&auml;nnlichen DG und PL wurden festgestellt, nicht in Reaktion auf stress ver&auml;ndert werden. Anomalien der Morphologie erfolgte nach chronischer Stress bei M&auml;nnern aber nicht bei Frauen. In allen F&auml;llen waren die Konzentrationen von CREB, pCREB und BDNF in Wiederherstellung Tiere auf das Niveau dieser Proteine in Kontrolltieren vergleichbar. Lo scopo di questo studio era di indagare i livelli di CREB, fosforilazione di CREB (pCREB) e BDNF in regioni del cervello legate allo stress di ratti maschi e femminili dopo lo stress e il recupero. Livelli di CREB e pCREB sono stati esaminati in CA1, CA2, CA3, nucleo paraventricolare il talamo (PVT), amigdala, area cingolato anteriore, parte dorsale (ACAd) e infralimbic zona della corteccia prefrontal (PFC), considerando che la zona prelimbic (PL) di PFC e giro dentato (DG) sono stati esaminati per livelli di BDNF. Di non per subire modifiche in risposta allo stress sono stati trovati livelli di BDNF nel maschio DG e PL. Anomalie nella morfologia si &egrave; verificato dopo lo stress cronico nei maschi ma non nelle femmine. In tutti i casi, i livelli di CREB, pCREB e BDNF in animali di recupero erano paragonabili ai livelli di queste proteine in animali di controllo. Questi risultati dimostrano un dimorfismo sessuale nella risposta molecolare a sottolineare e suggeriscono che queste differenze possono avere importanti implicazioni per il trattamento terapeutico potenziale della depressione.

Los estudios demuestran que el sexo juega un papel en la depresi&oacute;n relacionada con el estr&eacute;s, con las mujeres que experimentan una mayor vulnerabilidad a su efecto. Dos objetivos principales de los antidepresivos son el factor neurotr&oacute;fico derivado del cerebro (FNDC) y prote&iacute;na de uni&oacute;n a elemento de respuesta del monofosfato de adenosina c&iacute;clico (CREB). El objetivo de este estudio fue investigar los niveles de CREB, fosforilaci&oacute;n de CREB (pCREB) y BDNF en regiones del cerebro relacionadas con el estr&eacute;s de machos y hembras despu&eacute;s de estr&eacute;s y recuperaci&oacute;n. Niveles CREB y pCREB fueron examinados en CA1, CA2, CA3, n&uacute;cleo paraventricular del t&aacute;lamo (PVT), Am&iacute;gdala, &aacute;rea del c&iacute;ngulo anterior, parte dorsal (ACAd) y infralimbic &aacute;rea de la corteza prefrontal (PFC), mientras que el giro dentado (DG) y prelimbic (PL) de PFC fueron examinados para los niveles BDNF. Nuestros resultados demuestran que los niveles de CREB y pCREB en hombre CA1, CA2 y CA3, PVT, am&iacute;gdala y ACAd fueron reducidos por el estr&eacute;s, mientras que las mismas regiones del cerebro de ratas hembras no exhibieron ning&uacute;n cambio. Los niveles BDNF fueron disminuidos por estr&eacute;s cr&oacute;nico en PL hembra pero fueron aumentados por estr&eacute;s agudo en DG femenino. Los niveles de BDNF en DG y PL resultaron no para cambiar en respuesta al estr&eacute;s. Women in the reproductive age are more vulnerable to develop affective disorders than men. This difference may attribute to anatomical differences, hormonal influences and environmental factors such as stress. However, the higher prevalence in women normalizes once menopause is established, suggesting that ovarian hormones may play an important role in the development of depression in women. Ovarian hormones such as estrogen can pass the brain-blood barrier and bind to cytoplasmatic estrogen receptor (ER)-alpha and ER-beta in different areas of the limbic system. During stress, estrogen can modulate the behavioral and neurobiological response depending on the concentrations of estrogen. In this review we present evidence for disparate effects of chronic stress on neuroplasticity and brain activity in male and female rats. Furthermore, we will demonstrate that effects of social support on coping with stress can be mimicked by social housing of rats and that this model can be used for identification of underlying neurobiological mechanisms, including behavior, phosphorylation of CREB and ERK1/2, and brain activity changes as measured with fos expression. Using cyclic administration of estrogen in ovariectomized female rats we could specifically address effects of different plasma estrogen levels and antidepressants on stress-induced neuroplasticity and activity changes. In this model we also studied effects of estrogen on recovery after chronic stress. We conclude that the female brain has a different innate strategy to handle stress than the male brain and that female animal models are necessary for studying the underlying mechanisms and options for treatment.

Sex differences in stress responses: focus on ovarian hormones.

Las mujeres en edad f&eacute;rtil son m&aacute;s vulnerables a desarrollar trastornos afectivos que los hombres. Esta diferencia puede atribuir a las diferencias anat&oacute;micas, influencias hormonales y factores ambientales como el estr&eacute;s. Sin embargo, la mayor prevalencia en mujeres normaliza una vez establecida la menopausia, lo que sugiere que las hormonas ov&aacute;ricas pueden desempe&ntilde;ar un papel importante en el desarrollo de la depresi&oacute;n en las mujeres.
