The deletion of microtubule-associated protein stable tubule only polypeptide (STOP) leads to neuroanatomical, biochemical and severe behavioral alterations in mice, partly alleviated by antipsychotics. Therefore, STOP knockout (KO) mice have been proposed as a model of some schizophrenia-like symptoms. Preliminary data showed decreased brain serotonin (5-HT) tissue levels in STOP KO mice. As literature data demonstrate various interactions between microtubule-associated proteins and 5-HT, we characterized some features of the serotonergic neurotransmission in STOP KO mice. In the brainstem, mutant mice displayed higher tissue 5-HT levels and in vivo synthesis rate, together with marked increases in 5-HT transporter densities and 5-HT1A autoreceptor levels and electrophysiological sensitivity, without modification of the serotonergic soma number. Conversely, in projection areas, STOP KO mice exhibited lower 5-HT levels and in vivo synthesis rate, associated with severe decreases in 5-HT transporter densities, possibly related to reduced serotonergic terminals. Mutant mice also displayed a deficit of adult hippocampal neurogenesis, probably related to both STOP deletion and 5-HT depletion. Finally, STOP KO mice exhibited a reduced anxiety- and, probably, an increased helpness-status, that could be because of the strong imbalance of the serotonin neurotransmission between somas and terminals. Altogether, these data suggested that STOP deletion elicited peculiar 5-HT disconnectivity.

The deletion of the microtubule-associated STOP protein affects the serotonergic mouse brain network.

La suppression des prot&eacute;ines associ&eacute;es aux microtubules stables tubule seul polypeptide (STOP) m&egrave;ne &agrave; neuroanatomiques, biochimiques et graves alt&eacute;rations comportementales chez les souris, partiellement att&eacute;nu&eacute;es par antipsychotiques. Par cons&eacute;quent, arr&ecirc;t chez des souris knockout (KO) ont &eacute;t&eacute; propos&eacute;s comme un mod&egrave;le de certains sympt&ocirc;mes de la schizophr&eacute;nie-comme. Donn&eacute;es pr&eacute;liminaires ont montr&eacute; une diminution de cerveau les niveaux de s&eacute;rotonine (5-HT) tissu chez les souris arr&ecirc;ter KO. Souris mutantes a &eacute;galement affichent un d&eacute;ficit de la neurogen&egrave;se hippocampique adulte, probablement li&eacute; &agrave; la fois suppression d&#39;arr&ecirc;t et appauvrissement de la 5-HT. L&#39;eliminazione del microtubule-associated protein tubulo stabile solo polipeptide (STOP) conduce a neuroanatomiche, biochimici e gravi alterazioni comportamentali nei topi, in parte alleviati dai farmaci antipsicotici. Quindi, STOP topi knockout (KO) sono stati proposti come modello di alcuni sintomi di schizofrenia-come. Topi mutanti visualizzato anche un deficit della neurogenesi ippocampale adulta, probabilmente imparentato sia STOP eliminazione e deplezione di 5-HT. Infine, smettere di KO topi esposto una ridotta ansia - e, probabilmente, un&#39;aumentata helpness-status, che potrebbe essere a causa del forte squilibrio della neurotrasmissione della serotonina tra somas e terminali. La supresi&oacute;n de la prote&iacute;na asociada a microt&uacute;bulos estables t&uacute;bulo &uacute;nico polip&eacute;ptido (parada) conduce a neuroanat&oacute;micas, bioqu&iacute;micas y graves alteraciones de comportamiento en ratones, en parte aliviadas por antipsic&oacute;ticos. Por lo tanto, ratones de parada nocaut (KO) se han propuesto como un modelo de algunos s&iacute;ntomas similares a la esquizofrenia. Datos preliminares mostraron disminuci&oacute;n del cerebro los niveles de serotonina (5-HT) tejido en ratones dejar KO. Como datos de la literatura demuestran diversas interacciones entre prote&iacute;nas asociadas a microt&uacute;bulo y 5-HT, caracterizamos algunas caracter&iacute;sticas de la neurotransmisi&oacute;n serotonin&eacute;rgico en ratones dejar KO. En el tronco encef&aacute;lico, ratones mutantes muestran mayores niveles de 5-HT de tejido y tasa de s&iacute;ntesis en vivo, junto con marcados aumentos en densidades de transportador de 5-HT y 5-HT1A sinapsis niveles y electrofisiol&oacute;gico sensibilidad, sin modificaci&oacute;n del n&uacute;mero soma serotonin&eacute;rgicos. Por el contrario, en las &aacute;reas de proyecci&oacute;n, dejar KO ratones exhibidos menores niveles 5-HT y tasa de s&iacute;ntesis en vivo, asociada con severas reducciones en densidades de transportador de 5-HT, posiblemente relacionado con terminales serotonin&eacute;rgicos reducidas. Ratones mutantes tambi&eacute;n muestran un d&eacute;ficit de la neurog&eacute;nesis hipocampal adulta, probablemente relacionados con eliminaci&oacute;n de parada y agotamiento de la 5-HT. Por &uacute;ltimo, dejar KO ratones exhibieron una ansiedad reducida - y, probablemente, un helpness-estado mayor, que podr&iacute;a ser debido al fuerte desequilibrio de la neurotransmisi&oacute;n de serotonina entre Soma y terminales.
