Development of new therapeutic approaches for leishmaniasis treatment requires new high throughput screening methodologies for the antileishmanial activity of the new compounds both in vitro and in vivo. Reporter genes as the GFP have become one of the most promissory and widely used tools for drug screening in several models, since it offers live imaging, high sensibility, specificity and flexibility; additionally, the use of GFP as a reporter gene in screening assays eliminates all the drawbacks presented in conventional assays and also those technical problems found using other reporter genes. The utility of the GFP as a reporter gene in drug screening assays with Leishmania parasites depends on the homogeneity and stability of the GFP transfected strains. Stable expression of the GFP in the Old World Leishmania species has been demonstrated using integration vectors; however, no reports exist yet about the success of this methodology in the New World species. Here we report the generation of New World Leishmania strains expressing the GFP protein from an integration vector, which replaces one copy of the 18S RNA in the chromosome with the GFP coding sequence by homologous recombination. We also prove that the expression of the integrated GFP is stable and homogeneous in the transfected parasites after months in culture without selective pressure or during its use in hamster infection assays. The fluorescent strains are useful for in vitro, ex vivo and in vivo drug screening assays since no considerable variations in virulence or infectivity where seen attributable to the genetic manipulation during both in vitro and in vivo infection experiments. The platform described here for drug testing assays based on the use of stable fluorescent Leishmania strains coupled to flow cytometry and fluorescent microscopy is more sensitive, more specific and faster than conventional assays used normally for the evaluation of compounds with potential antileishmanial activity.

D&eacute;veloppement de nouvelles approches th&eacute;rapeutiques pour le traitement de la leishmaniose exige de nouvel m&eacute;thodologies pour l&#39;activit&eacute; antileishmanial des nouveaux compos&eacute;s in vitro et in vivo de criblage haute. Desarrollo de nuevos enfoques terap&eacute;uticos para el tratamiento de la leishmaniasis requiere nuevo alto rendimiento detecci&oacute;n metodolog&iacute;as para la actividad de costosas de los nuevos compuestos tanto in vitro como in vivo. Genes reporteros como la GFP han convertido en una de las herramientas m&aacute;s promisorias y ampliamente utilizadas para la detecci&oacute;n de drogas en varios modelos, ya que ofrece en proyecci&oacute;n de imagen, alta sensibilidad, especificidad y flexibilidad; Adem&aacute;s, el uso de GFP como un gen reportero en ensayos de cribado elimina todos los inconvenientes que se presentaron en los an&aacute;lisis convencionales y tambi&eacute;n esos problemas t&eacute;cnicos encontraron usando otros genes reporteros. La utilidad de la GFP como un gen reportero en ensayos con par&aacute;sitos de Leishmania de la detecci&oacute;n de drogas depende de la homogeneidad y estabilidad de la GFP transfectadas cepas. Se ha demostrado la expresi&oacute;n estable de la GFP en las especies de Leishmania de viejo mundo usando vectores de integraci&oacute;n; sin embargo, ningunos informes existen todav&iacute;a sobre el &eacute;xito de esta metodolog&iacute;a en las especies del nuevo mundo. Aqu&iacute; divulgamos la generaci&oacute;n de cepas de Leishmania de nuevo mundo expresando la prote&iacute;na GFP de un vector de integraci&oacute;n, que sustituye una copia del ARN 18S en el cromosoma con la secuencia de codificaci&oacute;n por recombinaci&oacute;n hom&oacute;loga de GFP. Tambi&eacute;n se demuestra que la expresi&oacute;n de la GFP integrado es estable y homog&eacute;nea en los par&aacute;sitos transfected despu&eacute;s de meses en cultura sin presi&oacute;n selectiva o durante su uso en ensayos de infecci&oacute;n de h&aacute;mster. Las cepas fluorescentes son &uacute;tiles para in vitro, ex vivo y ensayos de detecci&oacute;n de drogas en vivo desde sin variaciones considerables en la virulencia o infectividad donde visto atribuible a la manipulaci&oacute;n gen&eacute;tica en ambos experimentos in vitro e in vivo de la infecci&oacute;n. La plataforma descrita aqu&iacute; para pruebas de an&aacute;lisis basados en el uso de cepas de Leishmania fluorescentes estables de drogas junto a la citometr&iacute;a de flujo y microscopia fluorescente es m&aacute;s sensible, m&aacute;s espec&iacute;fico y m&aacute;s r&aacute;pido que los ensayos convencionales utilizados normalmente para la evaluaci&oacute;n de compuestos con potencial actividad costosas.

Mejorar el sistema de reportero de la prote&iacute;na verde fluorescente en Leishmania spp para la investigaci&oacute;n in vitro e in vivo de drogas costosas.
