Ixodes didelphilis Fonseca & Aragão was described in Brazil in 1952 as a new tick species that differed from Ixodes loricatus Neumann by the spiracular plate pattern. We have reared four tick colonies from different geographic areas in the laboratory that were started from single engorged females originally identified as I. didelphidis (BMG, colony ) and I. Iocicatus (CSP, PSP, and TRJ colonies). We analyzed the spiracular plate morphology of F1 adult ticks from each tick colony, compared their biological 11th, and performed a molecular analysis of the second internal transcribed rDNA spacer (ITS2) to test the validity of the species I. didelphidis. The spiracular plate analysis of laboratory F1 adult ticks yielded single females from the four colonies showed variations that invalidate morphological parameters for differentiation of loricatus and I. didelphidis. Biological data of the BMC, CSP, and TRJ colonies were similar. The biology ofthe PSP colony was not evaluated. The ITS2 sequence variations observed between the tick colonies ranged from 1.3 to 4.9%, and the similarity tree constructed by the neighbor-joining method with nucleotide distances showerl that the distances between the samples were similar to what is expected for intraspecific variations found in other ticks species. The morphological and biological results, in conjunction with the ITS2 analysis, supported the conspecificity I. loricatus and I. didelphidis.

Although it is well established that patients suffering from malaria experience skeletal muscle problems (contracture, aches, fatigue, weakness), detailed studies have not been performed to investigate changes in the contractile function and biochemical properties of intact and skinned skeletal muscles of mammals infected with malaria. To this end, we investigated such features in the extensor digitorium longus (EDL, fast-twitch, glyocolytic) and in the soleus (SOL, slow-twitch, oxidative) muscles from mice infected with Plasmodium berghei. We first studied maximal tetanic force (T(max)) produced by intact control and malaria-infected muscles before, during and after fatigue. Triton-skinned muscle fibres were isolated from these muscles and used to determine isometric contractile features as well as a basic biochemical profile as analysed by silver-enhanced SDS-PAGE. We found that the T(max) of intact muscles and the maximal Ca2+-activated force (F(max)) of Triton-skinned muscle fibres were reduced by approximately 50% in malarial muscles. In addition, the contractile proteins of Triton-skinned muscle fibres from malarial muscles were significantly less sensitive to Ca2+. Biochemical analysis revealed that there was a significant loss of essential contractile proteins (e.g. troponins and myosin) in Triton-skinned muscle fibres from malarial muscles as compared to controls. The biochemical alterations (i.e., reduction of essential contractile proteins) seem to explain well the functional modifications resolved in both intact muscles and Triton-skinned muscle fibres and may provide a suitable paradigm for the aetiology of muscle symptoms associated with malaria.

M&#xEA;me s&#x26;#39;il est bien &#xE9;tabli que les patients souffrant de probl&#xE8;mes musculaires exp&#xE9;rience paludisme squelettiques (contracture, courbatures, fatigue, faiblesse), des &#xE9;tudes d&#xE9;taill&#xE9;es n&#x26;#39;ont pas &#xE9;t&#xE9; r&#xE9;alis&#xE9;es pour &#xE9;tudier les changements dans la fonction contractile et les propri&#xE9;t&#xE9;s biochimiques de la peau intacte et les muscles squelettiques de mammif&#xE8;res infect&#xE9;es par le paludisme. &#xC0; cette fin, nous avons &#xE9;tudi&#xE9; ces caract&#xE9;ristiques dans le muscle long extenseur digitorium (EDL, &#xE0; contraction rapide, glyocolytic) et dans le muscle sol&#xE9;aire (SOL, &#xE0; contraction lente, oxydatif) muscles de souris infect&#xE9;es par Plasmodium berghei. Anche se &egrave; ben stabilito che i pazienti affetti da malaria esperienza problemi muscolari scheletriche (contratture, dolori, stanchezza, debolezza), non sono stati eseguiti studi approfonditi per indagare i cambiamenti nella funzione contrattile e propriet&agrave; biochimiche dei muscoli scheletrici intatti e dalla pelle dei mammiferi infetti con la malaria. A tal fine, abbiamo studiato tali caratteristiche l&#39;estensore digitorium longus (EDL, rapide, glyocolytic) e Soleo (SOL, slow-twitch, ossidativo) muscoli da topi infettati da Plasmodium berghei. Abbiamo studiato prima forza tetanica massima (T(max)) prodotto da controllo intatto e muscoli affetti da malaria, prima, durante e dopo la fatica. Fibre muscolari dalla pelle Tritone sono stati isolati da questi muscoli e utilizzato per determinare la funzionalit&agrave; contrattile isometrica, nonch&eacute; un profilo biochimico di base come analizzati da SDS-PAGE argento-enhanced. Aunque est&#xE1; bien establecido que los pacientes que sufren de problemas de malaria experiencia del m&#xFA;sculo esquel&#xE9;tico (contracturas, dolores, fatiga, debilidad), los estudios detallados no se han realizado para investigar los cambios en la funci&#xF3;n contr&#xE1;ctil y las propiedades bioqu&#xED;micas de los m&#xFA;sculos esquel&#xE9;ticos y la piel intacta de los mam&#xED;feros infectados con el la malaria. Para ello, se investig&#xF3; las caracter&#xED;sticas de este tipo en el largo del dedo digitorium extensor (EDL, de contracci&#xF3;n r&#xE1;pida, glyocolytic) y en el s&#xF3;leo (SOL, de contracci&#xF3;n lenta, oxidativo) los m&#xFA;sculos de los ratones infectados con Plasmodium berghei. En primer lugar, estudi&#xF3; la m&#xE1;xima fuerza tet&#xE1;nica (T (max)) producido por el control y los m&#xFA;sculos intactos infectadas por la malaria antes, durante y despu&#xE9;s de la fatiga. Trit&#xF3;n de piel fibras musculares se aislaron a partir de estos m&#xFA;sculos y se utiliza para determinar las caracter&#xED;sticas isom&#xE9;tricas contr&#xE1;ctiles, as&#xED; como un perfil bioqu&#xED;mico b&#xE1;sico como se analiza por plata-mejorada de SDS-PAGE. Se encontr&#xF3; que el T (m&#xE1;x) de los m&#xFA;sculos intactos y el m&#xE1;ximo de Ca2 +-activado fuerza (F (m&#xE1;x)) de las fibras musculares Trit&#xF3;n de piel se reduce en aproximadamente un 50% en los m&#xFA;sculos malaria. We have described the existence of asymptomatic carriers of Plasmodium vivax and Plasmodium falciparum infections in native Amazon populations. Most of them had low parasitemias, detected only by polymerase chain reaction (PCR). Because they remain symptomless and untreated, we wanted to determine whether they could infect Anopheles darlingi Root, the main Brazilian vector, and act as disease reservoirs. Fifteen adult asymptomatic patients (PCR positive only) were selected, and experimental infections of mosquitoes were performed by direct feeding and by a membrane-feeding system. Seventeen adult symptomatic patients with high parasitemias were used as controls. We found an infection rate in An. darlingi of 1.2% for the asymptomatic carriers and 22% for the symptomatic carriers. Although the asymptomatic group infected mosquitoes at a much lower rate, these patients remain infective longer than treated, symptomatic patients. Also, the prevalence of asymptomatic infections is 4 to 5 times higher than symptomatic infections among natives. These results have implications for the malaria control program in Brazil, which focuses essentially on the treatment of symptomatic patients.

The generation of transgenic mosquitoes with a minimal fitness load is a prerequisite for the success of strategies for controlling mosquito-borne diseases using transgenic insects. It is important to assemble as much information as possible on this subject because realistic estimates of transgene fitness costs are essential for modeling and planning release strategies. Transgenic mosquitoes must have minimal fitness costs, because such costs would reduce the effectiveness of the genetic drive mechanisms that are used to introduce the transgenes into field mosquito populations. Several factors affect fitness of transgenic mosquitoes, including the potential negative effect of transgene products and insertional mutagenesis. Studies to assess fitness of transgenic mosquitoes in the field (as opposed to the laboratory) are still needed.

The introduction of genes that impair Plasmodium development into mosquito populations is a strategy being considered for malaria control. The effect of the transgene on mosquito fitness is a crucial parameter influencing the success of this approach. We have previously shown that anopheline mosquitoes expressing the SM1 peptide in the midgut lumen are impaired for transmission of Plasmodium berghei. Moreover, the transgenic mosquitoes had no noticeable fitness load compared with nontransgenic mosquitoes when fed on noninfected mice. Here we show that when fed on mice infected with P. berghei, these transgenic mosquitoes are more fit (higher fecundity and lower mortality) than sibling nontransgenic mosquitoes. In cage experiments, transgenic mosquitoes gradually replaced nontransgenics when mosquitoes were maintained on mice infected with gametocyte-producing parasites (strain ANKA 2.34) but not when maintained on mice infected with gametocyte-deficient parasites (strain ANKA 2.33). These findings suggest that when feeding on Plasmodium-infected blood, transgenic malaria-resistant mosquitoes have a selective advantage over nontransgenic mosquitoes. This fitness advantage has important implications for devising malaria control strategies by means of genetic modification of mosquitoes.

Eine Strategie zur Bek&auml;mpfung der Malaria ist die Einf&uuml;hrung von Genen, die Plasmodium Entwicklung in Moskito-Populationen beeintr&auml;chtigen. Die Transgene wirkt auf Moskito Fitness ein entscheidender Parameter beeinflussen den Erfolg dieses Ansatzes. Wir haben bereits gezeigt, dass Anopheline Moskitos zum Ausdruck bringen des SM1-Peptids in den Mitteldarm Lumen f&uuml;r &Uuml;bertragung von Plasmodium Berghei wertgemindert sind. Dar&uuml;ber hinaus hatte die transgenen M&uuml;cken keine sp&uuml;rbaren Fitness-Last gegen&uuml;ber nontransgenic M&uuml;cken bei noninfected M&auml;usen ern&auml;hrt. Hier zeigen wir, dass wenn M&auml;use infiziert mit p. Berghei gef&uuml;ttert, diese transgenen M&uuml;cken mehr passen (h&ouml;here Fruchtbarkeit und geringere Kindersterblichkeit) als Geschwister nontransgenic M&uuml;cken. Im K&auml;fig Experimente, transgene M&uuml;cken allm&auml;hlich ersetzt Nontransgenics wenn M&uuml;cken M&auml;use infiziert mit Gametocyte erzeugenden gepflegt wurden Parasiten (Stamm ANKA 2,34) aber nicht, wenn M&auml;use infiziert mit Gametocyte-defizienten Parasiten (Stamm ANKA 2,33) gepflegt. El efecto de los transgenes en el gimnasio del mosquito es un par&#xE1;metro crucial influir en el &#xE9;xito de este enfoque. Hemos demostrado anteriormente que los mosquitos anofeles que expresan el p&#xE9;ptido SM1 en el lumen del intestino medio se vean afectados por la transmisi&#xF3;n de Plasmodium berghei. Por otra parte, los mosquitos transg&#xE9;nicos no ten&#xED;a la carga de fitness notable en comparaci&#xF3;n con los mosquitos transg&#xE9;nicos cuando se alimentan con ratones no infectados. Anopheline species of the subgenus Kerteszia, including Anopheles (Kerteszia) cruzii Dyar & Knab (Diptera: Culicidae), are bromeliad-malaria vectors in the Atlantic rain forest of Brazil. Morphological, genetic, and molecular polymorphisms among different populations of An. cruzii have been reported, and it has been suggested that this taxon includes a complex of cryptic species. Specimens of An. cruzii were collected in the states of SHo Paulo and Santa Catarina, from locations where autochthonous malaria cases have been reported during the last decade. The second internal transcribed spacers (ITS2) of nuclear ribosomal DNA of the captured specimens were sequenced and compared with each other. Intraspecific ITS2 sequence polymorphisms were identified, and nucleotide divergence among specimens varied from 0.3 to 0.9%. The number of nucleotides in the ITS2 sequences of these mosquitoes varied from 327 to 334, and the CG contents varied from 61.7 to 62%. The data provide further indication of An. cruzii being a complex of cryptic species.

Intraspecific variation of second internal transcribed spacer of nuclear ribosomal DNA among populations of Anopheles (Kerteszia) cruzii (Diptera: Culicidae).

Three transgenic Anopheles stephensi lines were established that strongly inhibit transmission of the mouse malaria parasite Plasmodium berghei. Fitness of the transgenic mosquitoes was assessed based on life table analysis and competition experiments between transgenic and wild-type mosquitoes. Life table analysis indicated low fitness load for the 2 single-insertion transgenic mosquito lines VD35 and VD26 and no load for the double-insertion transgenic mosquito line VD9. However, in cage experiments, where each of the 3 homozygous transgenic mosquitoes was mixed with nontransgenic mosquitoes, transgene frequency of all 3 lines decreased with time. Further experiments suggested that reduction of transgene frequency is a consequence of reduced mating success, reduced reproductive capacity, and/or insertional mutagenesis, rather than expression of the transgene itself. Thus, for transgenic mosquitoes released in the field to be effective in reducing malaria transmission, a driving mechanism will be required.

Rickettsial spotted fever is common in southeastern Brazil. Differential diagnosis of pathogens can be performed with proper laboratory methods. A traveler arriving from Portugal developed a fatal febrile hemorrhagic syndrome diagnosed as spotted fever rickettsiosis. We isolated the agent, which was identified as Rickettsia conorii conorii by sequencing rickettsial genes.
