Epithelial cell migration is an essential part of embryogenesis and tissue regeneration, yet their migration is least understood. Using our three-dimensional (3D) motility analysis, migrating epithelial cells formed an atypical polarized cell shape with the nucleus leading the cell front and a contractile cell rear. Migrating epithelial cells exerted traction forces to deform both the anterior and posterior extracellular matrix toward the cell body. The cell leading edge exhibited a myosin II-dependent retrograde flow with the magnitude and direction consistent with surrounding network deformation. Interestingly, on a two-dimensional substrate, myosin IIA-deficient cells migrated faster than wild-type cells, but in a 3D gel, these myosin IIA-deficient cells were unpolarized and immobile. In contrast, the migration rates of myosin IIB-deficient cells were similar to wild-type cells. Therefore, myosin IIA, not myosin IIB, is required for 3D epithelial cell migration.

Cytoskeletal regulation of cell adhesion is vital to the organization of multicellular structures. The focal adhesion protein zyxin emerged as a key regulator of actin assembly because zyxin recruits Enabled/vasodilator-stimulated phospho-proteins (Ena/VASP) to promote actin assembly. Zyxin also localizes to the sites of cell-cell adhesion and is thought to promote actin assembly with Ena/VASP. Using shRNA targeted to zyxin, we analyzed the roles of zyxin at adhesive contacts. In zyxin-deficient cells, the actin assembly at both focal adhesion and cell-cell adhesion was limited, but their migration rate was unchanged. Cell spreading on E-cadherin-coated surfaces and the formation of cell clusters were slower for zyxin-deficient cells than wild type cells. By ablating a single cell within a cell monolayer, we quantified the rate of wound closure driven by a contractile circumferential actin ring. Zyxin-deficient cells failed to recruit VASP to cell-cell junctions at the wound edge and had a slower wound closure rate than wild type cells. Our results suggest that, by recruiting VASP, zyxin regulates actin assembly at the sites of force-bearing cell-cell adhesion.

Regulaci&#xF3;n del citoesqueleto de la adhesi&#xF3;n celular es vital para la organizaci&#xF3;n de las estructuras multicelulares. La prote&#xED;na de adhesi&#xF3;n focal zyxin surgi&#xF3; como un regulador clave de la actina asamblea porque los reclutas zyxin Habilitado / vasodilatador estimulada por fosfo-prote&#xED;nas (ENA / VASP) para promover el ensamblaje de actina. Por la ablaci&#xF3;n de una sola c&#xE9;lula dentro de una monocapa de c&#xE9;lulas, se cuantific&#xF3; la tasa de cierre de la herida impulsado por un anillo circunferencial actina contr&#xE1;ctil. Zyxin las c&#xE9;lulas deficientes en no contratar a la VASP uniones c&#xE9;lula-c&#xE9;lula en el borde de la herida y ten&#xED;a una tasa de cierre de la herida m&#xE1;s lento que las c&#xE9;lulas de tipo salvaje. Nuestros resultados sugieren que, por la VASP de reclutamiento, zyxin regula conjunto de actina en los sitios de la c&#xE9;lula-c&#xE9;lula la fuerza de soporte de la adhesi&#xF3;n.

Maintaining proper adhesion between neighboring cells depends on the ability of cells to mechanically respond to tension at cell-cell junctions through the actin cytoskeleton. Thus, identifying the molecules involved in responding to cell tension would provide insight into the maintenance, regulation, and breakdown of cell-cell junctions during various biological processes. Vinculin, an actin-binding protein that associates with the cadherin complex, is recruited to cell-cell contacts under increased tension in a myosin II-dependent manner. However, the precise role of vinculin at force-bearing cell-cell junctions and how myosin II activity alters the recruitment of vinculin at quiescent cell-cell contacts have not been demonstrated.
