&#x5C0F;&#x9F20; Fgf15 &#x57FA;&#x56E0;&#x8868;&#x8FBE;&#x7684;&#x662F;&#x76F4;&#x63A5;&#x53D1;&#x8D77;&#x7684;&#x58F0;&#x6CE2;&#x4FE1;&#x53F7;&#x5728;&#x95F4;&#x8111;&#x548C;&#x4E2D;&#x8111;&#x7684;&#x523A;&#x732C;&#x3002;

Sonic hedgehog (Shh) is a secreted molecule that is thought to regulate tissue growth and patterning in vertebrate embryos. Although it has been reported that Gli transcription factors mediate Shh signaling to the nucleus, little is known about developmental target genes of Gli. In the previous genetic study, we showed that Shh is required for Fgf15 expression in the diencephalon and midbrain. Here, we examined whether Fgf15 is a direct target of Shh signaling through Gli. Shh was expressed in the midline cells and Fgf15 in the medial region of the diencephalon/midbrain by the seven-somite stage. The Fgf15 expression domain coincided with that of Gli1 and overlapped with that of Gli2 at this stage. Fgf15 expression in the diencephalon/midbrain was greatly reduced in the seven-somite Shh mutant embryos. Transgenic analysis showed that the 3.6-kb 5'-flanking region of the Fgf15 gene is sufficient for induction of Fgf15 in the medial/ventral diencephalon/midbrain. Luciferase assay showed that the 3.6-kb Fgf15 enhancer/promoter was activated by Gli2. A Gli-binding site was located 1 kb upstream of the transcription start site and was required for expression in the medial/ventral diencephalon/midbrain in transgenic embryos and for activation in luciferase assay. These findings indicate that Fgf15 is directly regulated by Shh signaling through Gli proteins.

Sonic hedgehog (Shh) est une mol&eacute;cule s&eacute;cr&eacute;t&eacute;e qui est pens&eacute;e pour r&eacute;guler la croissance des tissus et motifs dans des embryons de vert&eacute;br&eacute;s. Bien qu&#39;il a &eacute;t&eacute; signal&eacute; que les facteurs de transcription Gli m&eacute;dient Shh signalisation vers le noyau, on conna&icirc;t les g&egrave;nes cibles du d&eacute;veloppement de Gli. Dans la pr&eacute;c&eacute;dente &eacute;tude g&eacute;n&eacute;tique, nous avons montr&eacute; que Shh est requis pour l&#39;expression de Fgf15 dans le dienc&eacute;phale et le m&eacute;senc&eacute;phale. Ici, nous avons examin&eacute; si la Fgf15 est une cible directe de signalisation de Shh dans Gli. SHH a &eacute;t&eacute; exprim&eacute;e dans les cellules de la ligne m&eacute;diane et la Fgf15 dans la r&eacute;gion m&eacute;diane du dienc&eacute;phale et du m&eacute;senc&eacute;phale par le stade de sept segments. Le domaine d&#39;expression Fgf15 a co&iuml;ncid&eacute; avec celle de Gli1 et chevauche celui de Gli2 &agrave; ce stade. Expression de Fgf15 dans le dienc&eacute;phale et le m&eacute;senc&eacute;phale est fortement r&eacute;duite chez les embryons de mutants de Shh sept segments. Analyse transg&eacute;nique a montr&eacute; que 3,6 Ko 5&#39;-r&eacute;gion du g&egrave;ne Fgf15 est suffisant pour l&#39;induction de Fgf15 dans le dienc&eacute;phale/midbrain m&eacute;dial/ventrale. Lucif&eacute;rase ont montr&eacute; que le 3.6 Ko Fgf15 enhancer/promoteur &eacute;tait activ&eacute; par Gli2. Un site de liaison Gli &eacute;tait situ&eacute; &agrave; 1 kb en amont du site d&#39;initiation de la transcription et requis pour l&#39;expression dans le dienc&eacute;phale/midbrain m&eacute;dial/ventrale chez les embryons transg&eacute;niques et pour l&#39;activation de la lucif&eacute;rase. Ces r&eacute;sultats indiquent que Fgf15 est directement r&eacute;gl&eacute; par Shh de signalisation par l&#39;interm&eacute;diaire de prot&eacute;ines Gli.

L&#39;analisi di Luciferase ha mostrato che il 3.6 kb Fgf15 enhancer/promotore &egrave; stato attivato da Gli2. Un sito di legame Gli era situato 1 kb a monte del sito di inizio della trascrizione e necessario per l&#39;espressione nel diencefalo/mesencefalo mediale/ventrale in embrioni transgenici e per l&#39;attivazione di analisi di luciferase. Sonic hedgehog (Shh) es una mol&eacute;cula secretada que est&aacute; pensada para regular el crecimiento de tejidos y patrones en embriones de vertebrados. The vertebrate brain is a complex and highly organized structure with numerous neurons and glial cells. During development, undifferentiated progenitor cells proliferate from neural stem/precursor cells and gradually restrict their fates according to their environment. Differentiated cells are arranged precisely to accomplish their function and to maintain integrity as a whole brain. In this respect, cells must receive signals to know where and when they determine their fates. Secreted and membrane molecules convey the information between cells. The secreted glycoprotein Sonic hedgehog (Shh) is one of such signaling molecules. Sonic hedgehog is widely known to specify ventral neuronal types according to the concentration of Shh, whereas differentiation of dorsal neurons is largely independent of Shh. However, in the diencephalon and midbrain, dorsal parts are also affected in Shh-mutant embryos. Detailed analysis demonstrated that Shh signaling indirectly regulates the growth of the dorsal tissue in these regions. One of the fibroblast growth factor (FGF) members, namely FGF15, has been reported to be downstream to Shh signaling in the mouse embryonic brain. Luciferase assays and transgenic analysis revealed that the Fgf15 gene is a direct target of Shh. Downregulation of Tcf4 and upregulation of Bmp4 in Shh mutants suggest that Wnt and BMP signals from the dorsal midline are also involved in the dorsal brain phenotype. These data suggest the coordinating role of the Shh-FGF15-Wnt/BMP signaling cascade between the ventral and dorsal parts of the brain.

El cerebro de vertebrados es una estructura compleja y altamente organizada con numerosas neuronas y c&eacute;lulas gliales. Durante el desarrollo, c&eacute;lulas progenitoras indiferenciadas proliferaci&oacute;n de c&eacute;lulas madre neuronales/precursoras y restringen gradualmente sus destinos seg&uacute;n su entorno. Las c&eacute;lulas diferenciadas se organizan precisamente para cumplir con su funci&oacute;n y para mantener la integridad como un todo el cerebro. En este sentido, las c&eacute;lulas deben recibir se&ntilde;ales para saber d&oacute;nde y Cu&aacute;ndo determinan su destino. Segregada y mol&eacute;culas de membrana transmitan la informaci&oacute;n entre las c&eacute;lulas. El erizo Sonic glicoprote&iacute;na secretada (Shh) es una de estas mol&eacute;culas de se&ntilde;alizaci&oacute;n. Sonic hedgehog es ampliamente conocido para especificar los tipos neuronales ventrales seg&uacute;n la concentraci&oacute;n de Shh, mientras que la diferenciaci&oacute;n de las neuronas dorsales es en gran parte independiente de Shh. Sin embargo, en el dienc&eacute;falo y mesenc&eacute;falo, partes dorsales tambi&eacute;n se ven afectadas en embriones de Shh-mutante. An&aacute;lisis detallado demostr&oacute; que la se&ntilde;alizaci&oacute;n Shh regula indirectamente el crecimiento del tejido dorsal en estas regiones. Uno de los miembros de fibroblastos (FGF) del factor de crecimiento, es decir, FGF15, se ha divulgado para ser descendente para se&ntilde;alizaci&oacute;n Shh en el cerebro embrionario del rat&oacute;n. Prenatal exposure to ethanol induces holoprosencephalic malformations in both humans and laboratory animals. However, its teratogenic window for inducing holoprosencephaly is narrow, and the teratogenic mechanism is not well understood. In the present study, we examined the morphological changes in the craniofacial structures of mouse embryos/fetuses at intervals following ethanol treatment and evaluated gene expression patterns in the embryos.

Holoprosencephaly (HPE) is a malformation of the human brain caused primarily by incomplete division of the prosencephalon into two halves and is often associated with various facial anomalies. Although HPE is rather rare in newborns (1/10,000-15,000 births), it is frequently encountered in therapeutic abortuses (>1/250). To date, nine gene mutations responsible for human HPE have been identified, but the pathogenetic mechanisms of the craniofacial anomalies in HPE have just begun to be understood. Here, we summarize our studies on human embryos with HPE and discuss the embryogenesis and the underlying molecular mechanisms of HPE malformations under the following headings: pathology, pathogenesis, and critical period of development.

Fibroblast growth factor 15 (Fgf15) is expressed in the medial region of diencephalon and midbrain by the seven-somite stage. In the previous studies, we showed that Sonic hedgehog signaling through Gli protein is required for Fgf15 expression in this region. The Fgf15 expression domain overlapped with that of Gli2 and the Gli-binding site (GliBs) is located in the 3.6-kb 5'-flanking enhancer/promoter region of the Fgf15 gene. In this study, we identified the two additional Gli-binding sites in row, called Gli-responsive elements (GliREs). Chromatin immunoprecipitation assay indicated that Gli2 directly binds to GliREs. The results from luciferase assays indicated that the Gli2 activator form binds to the GliBS and that the Gli2 repressor form binds to the GliREs. These findings suggest that the repressor form of Gli2 preferentially binds to the GliREs to control Fgf15 expression.

Expression of Fgf15 is regulated by both activator and repressor forms of Gli2 in vitro.

Fibroblasten-Wachstumsfaktor 15 (Fgf15) wird durch die sieben-Somiten-B&uuml;hne in der medialen Region von Zwischenhirn und Mittelhirn ausgedr&uuml;ckt. Sonic hedgehog (Shh) function is essential for patterning and cell fate specification, particularly in ventral regions of the central nervous system. It is also a crucial mitogen for cerebellar granule neuron precursors and is important in maintenance of the stem cell niche in the postnatal telencephalon. Although it has been reported that Shh is expressed in the developing dorsal telencephalon, functions of Shh in this region are unclear, and detailed characterization of Shh mRNA transcripts in situ has not been demonstrated. To clarify the roles of Shh signaling in dorsal pallium (neocortex primordium) development, we have knocked out the Shh and Smo genes specifically in the early developing dorsal telencephalon by using Emx1cre mice. The mutants showed a smaller dorsal telencephalon at E18.5, which was caused by cell cycle kinetics defects of the neural progenitor/stem cells. The cell cycle length of the progenitor/stem cells was prolonged, and the number of cycle-exiting cells and neurogenesis were decreased. Birth-date analysis revealed abnormal positioning of neurons in the mutants. The characteristics of the subventricular zone, ventricular zone and subplate cells were also affected. Weak immunoreactivity of Shh was detected in the dorsal telencephalon of wild types. Reduced Shh immunoreactivity in mutant dorsal telencephalons supports the above phenotypes. Our data indicate that Shh signaling plays an important role in development of the neocortex.

Funci&oacute;n de Sonic hedgehog (Shh) es esencial para modelar y c&eacute;lula especificaci&oacute;n de destino, especialmente en las regiones ventrales del sistema nervioso central. Tambi&eacute;n es un mit&oacute;geno crucial para precursores de neuronas gr&aacute;nulo cerebeloso y es importante en el mantenimiento del nicho de c&eacute;lulas madre en el telenc&eacute;falo postnatal. Aunque se ha reportado que Shh se expresa en el telenc&eacute;falo dorsal en desarrollo, funciones de Shh en esta regi&oacute;n no son claros, y no se ha demostrado la caracterizaci&oacute;n detallada de transcripciones de Shh mRNA in situ. The developing avian lung is formed mainly by branching morphogenesis, but there is also a unique cystic structure, the air sac, in the ventral region. It has been shown that mesenchymal tissue is responsible for the differential development of a cystic or branched structure, and that the transcription factor Hoxb may be involved in determining this regional difference. We have previously developed two scenarios for branch-cyst transition, both experimentally and theoretically: increased production or increased diffusion of FGF. The aim of the present study was to discover whether one of these scenarios actually operates in the ventral region of the chick lung. We found that the FGF10 level was lower while the diffusion of FGF10 was more rapid in the ventral lung, indicating that the second scenario is more plausible. There are two possibilities as to why the diffusion of FGF10 differs between the two regions: (1) diffusion is facilitated by the looser tissue organisation of the ventral lung mesenchyme; (2) stronger expression of heparan sulphate proteoglycan (HSPG) in the dorsal lung traps FGF and decreases the effective diffusion coefficient. Mathematical analysis showed that the dorsal-ventral difference in the amount of HSPG is not sufficient to generate the observed difference in pattern, indicating that both extracellular matrix and tissue architecture play a role in this system. These results suggest that the regional cystic-branched difference within the developing chick lung results from a difference in the rate of diffusion of morphogen between the ventral and dorsal regions due to differential levels of HSPG and a different mesenchymal structure.

Entwickelnde Vogelgrippe Lunge entsteht haupts&auml;chlich durch Verzweigung Morphogenese, aber es gibt auch eine einzigartige zystische Struktur, die Luft-Sac in der ventralen Region. Es hat sich gezeigt, dass mesenchymale Gewebe f&uuml;r die differenzielle Entwicklung einer zystische oder verzweigte Struktur verantwortlich ist und der Transkriptionsfaktor Hoxb beteiligt sein kann, bei der Bestimmung dieser regionalen Unterschied. Wir haben zwei Szenarien f&uuml;r die Branch-Zyste &Uuml;bergang, zuvor entwickelt, sowohl theoretisch als auch experimentell: Produktion erh&ouml;ht oder erh&ouml;ht Verbreitung der FGF. El desarrollo pulmonar aviar est&aacute; formado principalmente por la ramificaci&oacute;n de la morfog&eacute;nesis, pero tambi&eacute;n existe una &uacute;nica estructura qu&iacute;stica, el saco de aire, en la regi&oacute;n ventral. Se ha demostrado que tejido mesenquimal es responsable por el desarrollo diferencial de una estructura enquistada o ramificado, y que el factor de transcripci&oacute;n Hoxb podr&iacute;a estar implicado en la determinaci&oacute;n de esta diferencia regional. Previamente hemos desarrollado dos escenarios para la transici&oacute;n de la rama-quiste, tanto experimental como te&oacute;ricamente: aumento de la producci&oacute;n o mayor difusi&oacute;n de FGF. El objetivo del presente estudio era descubrir si uno de estos escenarios realmente funciona en la regi&oacute;n ventral del pulm&oacute;n polluelo. Encontramos que el nivel de FGF10 era inferior, mientras que la difusi&oacute;n de FGF10 fue m&aacute;s r&aacute;pida en el pulm&oacute;n ventral, indicando que la segunda hip&oacute;tesis es m&aacute;s plausible. Hay dos posibilidades en cuanto a por qu&eacute; la difusi&oacute;n de FGF10 diferencia entre las dos regiones: (1) difusi&oacute;n es facilitado por la organizaci&oacute;n de tejido m&aacute;s floja de la mes&eacute;nquima pulmonar ventral; (2) m&aacute;s fuerte expresi&oacute;n de hepar&aacute;n sulfato proteoglicano (HSPG) en el pulm&oacute;n dorsal atrapa FGF y disminuye el coeficiente de difusi&oacute;n efectiva. An&aacute;lisis matem&aacute;tico demostraron que la diferencia de dorsal-ventral en la cantidad de HSPG no es suficiente para generar la diferencia observada en patr&oacute;n, indicando que tanto la matriz extracelular y la arquitectura de tejido desempe&ntilde;an un papel en este sistema. Estos resultados sugieren que la diferencia regional ramificado enquistado en el pulm&oacute;n de pollo en desarrollo resulta de una diferencia en la tasa de difusi&oacute;n de morf&oacute;geno entre las regiones ventrales y dorsales debido a niveles diferenciales de HSPG y una estructura diferente mesenquimal.

La diferencia del quiste-rama en pulm&oacute;n de pollo en desarrollo resulta de un coeficiente de difusi&oacute;n de morf&oacute;genos diferentes.

Conduction velocity (CV) of myelinated axons has been shown to be regulated by oligodendrocytes even after myelination has been completed. However, how myelinating oligodendrocytes regulate CV, and what the significance of this regulation is for normal brain function remain unknown. To address these questions, we analyzed a transgenic mouse line harboring extra copies of the myelin proteolipid protein 1 (plp1) gene (plp1(tg/-) mice) at 2 months of age. At this stage, the plp1(tg/-) mice have an unaffected myelin structure with a normally appearing ion channel distribution, but the CV in all axonal tracts tested in the CNS is greatly reduced. We also found decreased axonal diameters and slightly abnormal paranodal structures, both of which can be a cause for the reduced CV. Interestingly the plp1(tg/-) mice showed altered anxiety-like behaviors, reduced prepulse inhibitions, spatial learning deficits and working memory deficit, all of which are schizophrenia-related behaviors. Our results implicate that abnormalities in the neuron-glia interactions at the paranodal junctions can result in reduced CV in the CNS, which then induces behavioral abnormalities related to schizophrenia.

We here describe a case of congenital leukemia that ended in intrauterine fetal demise at 30 weeks of gestation. Acute enlargement of the fetal trunk, elevated pulsatility index of the umbilical artery with concomitant decline of pulsatility index of the middle cerebral artery, pleural effusion, and polyhydramnios preceded the fetal death. Diagnosis of congenital myeloid leukemia was suggested by microscopic examination of the placental tissue, revealing immature myeloid precursors filling the lumina of fetal vessels in the umbilical cord and chorionic villi. Extensive vascular involvement of the placenta by leukemic cells was considered to be a primary cause of the fetal death.

Nous d&eacute;crivons ici un cas de leuc&eacute;mie cong&eacute;nitale qui se termina par une mort f&oelig;tale intra-ut&eacute;rine &agrave; 30 semaines de gestation. &Eacute;largissement aigu&euml; du tronc foetal, indice &eacute;lev&eacute; pulsatilit&eacute; de l&#39;art&egrave;re ombilicale avec une diminution concomitante des index de pulsatilit&eacute; de l&#39;art&egrave;re c&eacute;r&eacute;brale moyenne, &eacute;panchement pleural et hydramnios ont pr&eacute;c&eacute;d&eacute; la mort foetale. Diagnostic de la leuc&eacute;mie my&eacute;lo&iuml;de cong&eacute;nitale a &eacute;t&eacute; sugg&eacute;r&eacute; par un examen microscopique du tissu placentaire, r&eacute;v&eacute;lant des pr&eacute;curseurs my&eacute;lo&iuml;des immatures remplissant les lumi&egrave;res des bateaux f&oelig;tales dans le cordon ombilical et de villosit&eacute;s choriales. Une large participation vasculaire du placenta par des cellules leuc&eacute;miques &eacute;tait consid&eacute;r&eacute;e comme des principales causes de la mort foetale.

Wir beschreiben hier einen Fall von kongenitaler Leuk&auml;mie, die intrauterine fetale Niedergang bei 30 Wochen der Schwangerschaft endete. Akute Erweiterung des fetalen Trunks, erh&ouml;hten Pulsatility Index der Arteria umbilicalis mit gleichzeitigen R&uuml;ckgang des Pulsatility Index der mittleren Hirnarterie, Pleuraerguss und Polyhydramnion voraus den fetalen Tod. Diagnostik der angeborene myeloischen Leuk&auml;mie wurde durch mikroskopische Untersuchung der Plazenta Gewebe, offenbart unreife myeloische Vorstufen, die F&uuml;llung der Lumina fetalen Gef&auml;&szlig;e der Nabelschnur und Chorionzotten vorgeschlagen. Umfassende vaskul&auml;rer Beteiligung der Plazenta von leuk&auml;mischen Zellen galt als eine prim&auml;re Ursache der fetalen Tod.

Descriviamo qui un caso di leucemia congenita che si concluse con la morte fetale intrauterina a 30 settimane di gestazione. Allargamento acuta del tronco fetale, indice di elevata pulsatilit&agrave; della arteria ombelicale con concomitante diminuzione dell&#39;indice di pulsatilit&agrave; della arteria cerebrale media, versamento pleurico e polidramnios preceduto la morte fetale. La diagnosi di leucemia mieloide congenita &egrave; stata suggerita da un esame microscopico del tessuto placenta, rivelando precursori mieloidi immaturi riempiendo i lumi dei vasi fetali nel cordone ombelicale e villi coriali. Ampio coinvolgimento vascolare della placenta da parte delle cellule leucemiche &egrave; stato considerato una causa primaria di morte fetale.
