The Yersinia enterocolitica LuxI homologue YenI directs the synthesis of N-3-(oxohexanoyl)homoserine lactone (3-oxo-C6-HSL) and N-hexanoylhomoserine lactone (C6-HSL). In a Y. enterocolitica yenI mutant, swimming motility is temporally delayed while swarming motility is abolished. Since both swimming and swarming are flagellum dependent, we purified the flagellin protein from the parent and yenI mutant. Electrophoresis revealed that in contrast to the parent strain, the yenI mutant grown for 17 h at 26 degrees C lacked the 45-kDa flagellin protein FleB. Reverse transcription-PCR indicated that while mutation of yenI had no effect on yenR, flhDC (the motility master regulator) or fliA (the flagellar sigma factor) expression, fleB (the flagellin structural gene) was down-regulated. Since 3-oxo-C6-HSL and C6-HSL did not restore swimming or swarming in the yenI mutant, we reexamined the N-acylhomoserine lactone (AHL) profile of Y. enterocolitica. Using AHL biosensors and mass spectrometry, we identified three additional AHLs synthesized via YenI: N-(3-oxodecanoyl)homoserine lactone, N-(3-oxododecanoyl)homoserine lactone (3-oxo-C12-HSL), and N-(3-oxotetradecanoyl)homoserine lactone. However, none of the long-chain AHLs either alone or in combination with the short-chain AHLs restored swarming or swimming in the yenI mutant. By investigating the transport of radiolabeled 3-oxo-C12-HSL and by introducing an AHL biosensor into the yenI mutant we demonstrate that the inability of exogenous AHLs to restore motility to the yenI mutant is not related to a lack of AHL uptake. However, both AHL synthesis and motility were restored by complementation of the yenI mutant with a plasmid-borne copy of yenI.

Quorum sensing in Yersinia enterocolitica controls swimming and swarming motility.

Enterocolitica YenI Mutanten wird das Schwimmen Motilit&auml;t zeitlich verz&ouml;gert, w&auml;hrend Ausschw&auml;rmen Motilit&auml;t abgeschafft wird. Da sowohl Schwimmen und schw&auml;rmen Kutscherpeitschennatter angewiesen sind, gereinigt wir das Flagellin-Protein aus den &uuml;bergeordneten und YenI Mutanten. Mit AHL-Biosensoren und Massenspektrometrie, wir identifizierten drei zus&auml;tzliche AHLs synthetisiert &uuml;ber YenI: Homoserin lacton N-(3-Oxodecanoyl), N (3-Oxododecanoyl)-Homoserin lacton (3-Oxo-C12-HSL) und N (3-Oxotetradecanoyl)-Homoserin-lacton. Jedoch wiederhergestellt keines der langkettigen AHLs entweder allein oder in Kombination mit der kurzkettigen AHLs, schw&auml;rmen oder Schwimmen in der YenI-Mutant. Durch die Untersuchung des Transport von radioaktive 3-Oxo-C12-HSL und durch die Einf&uuml;hrung einer AHL-Biosensor in der YenI-Mutant zeigen wir, dass die Unf&auml;higkeit der exogenen AHLs Motilit&auml;t in der YenI-Mutant wiederherstellen nicht mit mangelnder AHL-Aufnahme. Jedoch waren sowohl die AHL-Synthese als auch die Motilit&auml;t durch Erg&auml;nzung von der YenI-Mutant mit einem Plasmid &uuml;bertragene Kopie der YenI wiederhergestellt.

El hom&oacute;logo de LuxI Yersinia enterocolitica YenI dirige la s&iacute;ntesis de N - 3-(oxohexanoyl) Homoserina lactona (3-oxo-C6-HSL) y N-hexanoylhomoserine lactona (C6-HSL). En un mutante de yenI Y. enterocolitica, nataci&oacute;n motilidad temporal se retrasa mientras que pulula la motilidad se suprime. Como nadar y enjambre son dependiente del flagelo, nos purifica la prote&iacute;na flagelina de los padres y yenI mutante. Electroforesis revel&oacute; que a diferencia de la cepa parental, el mutante de yenI crecido h 17 a 26 grados C carec&iacute;a de la prote&iacute;na flagelina de 45 kDa FleB. Transcripci&oacute;n reversa-PCR indicaron que mientras que la mutaci&oacute;n de yenI no tuvo ning&uacute;n efecto en yenR, flhDC (el principal regulador de motilidad) o expresi&oacute;n de fliA (el factor sigma flagelar), fleB (el gen estructural de flagelina) regula. Desde 3-oxo-C6-HSL y C6-HSL no restaur&oacute; nadando o pululando en el mutante de yenI, hemos reexaminado el perfil de la lactona (AHL) de N-acylhomoserine de Y. enterocolitica. Utilizando biosensores AHL y espectrometr&iacute;a de masas, se identificaron tres AHLs adicionales sintetizados v&iacute;a YenI: Homoserina lactona de N-(3-oxodecanoyl), N-(3-oxododecanoyl) Homoserina lactona (3-oxo-C12-HSL) y Homoserina lactona de N-(3-oxotetradecanoyl). Sin embargo, ninguno de las cadena larga AHLs solos o en combinaci&oacute;n con las cadena corta AHLs restaurado enjambre o nadar en el mutante de yenI. Investigando el transporte de radiactivo 3-oxo-C12-HSL y mediante la introducci&oacute;n de un biosensor AHL en el mutante de yenI demostramos que la incapacidad de AHLs ex&oacute;genos para restaurar la movilidad en el mutante de yenI no se relaciona con una falta de absorci&oacute;n AHL. Sin embargo, tanto la s&iacute;ntesis AHL y la motilidad fueron restaurados por la complementaci&oacute;n del mutante yenI una copia transmitida por pl&aacute;smidos de yenI.

Quorum sensing (QS) in Yersinia pseudotuberculosis involves two pairs of LuxRI orthologues (YpsRI and YtbRI) and multiple N-acylhomoserine lactones (AHLs). In a ypsI/ytbI mutant, AHL synthesis was abolished, unaffected in a ypsR/ytbR double mutant and substantially reduced in a ypsI/ytbR mutant, indicating that neither YpsR nor YtbR is essential for AHL synthesis. To determine the interrelationship between YpsRI and YtbRI we constructed chromosomal lux-promoter fusions to ypsR, ypsI, ytbR and ytbI and examined their expression in each of the QS mutant backgrounds. The YpsRI system negatively autoregulates itself but positively regulates the expression of the ytbRI system whereas the ytbRI system is positively autoregulated but only at the level of ytbI expression. YtbRI does not control expression of ypsR or ypsI. This hierarchical QS system controls swimming motility via regulation of flhDC and fliA. The AHLs synthesized via YtbI play a dual role, activating flhDC, in conjunction with YpsR but repressing fliA in conjunction with YtbR and YpsR. In liquid and plate assays, the early onset of motility observed in ypsR and ypsI mutants was abolished in ytbI, ytbR ypsI/ytbI, ypsR/ytbR mutants, indicating that QS regulates motility both positively (via YtbRI) and negatively (via YpsRI).

Functional interplay between the Yersinia pseudotuberculosis YpsRI and YtbRI quorum sensing systems modulates swimming motility by controlling expression of flhDC and fliA.

Quorum sensing (QS) in pseudotuberculosis Yersinia coinvolge due paia di LuxRI orthologues (YpsRI e YtbRI) e pi&ugrave; N-acylhomoserine lattoni (AHLs). En un mutante de ypsI/ytbI, s&iacute;ntesis AHL fue suprimido, afectado en un mutante doble ypsR/ytbR y sustancialmente reducido en un mutante de ypsI/ytbR, indicando que ni YpsR ni YtbR es esencial para la s&iacute;ntesis AHL. Para determinar la relaci&oacute;n entre YpsRI y YtbRI hemos construido fusiones cromos&oacute;micas lux-promotor a ypsR, ypsI, ytbR y ytbI y examin&oacute; su expresi&oacute;n en cada uno de los fondos del mutantes de QS. El sistema YpsRI negativamente autoregulates s&iacute; mismo pero positivamente regula la expresi&oacute;n del sistema ytbRI que el sistema de ytbRI positivamente autoregulated pero s&oacute;lo en el nivel de expresi&oacute;n de la ytbI. YtbRI no controla la expresi&oacute;n de ypsR o ypsI. Este sistema jer&aacute;rquico de QS controla la motilidad de nataci&oacute;n mediante regulaci&oacute;n de flhDC y fliA. Las AHLs sintetizan mediante juego de YtbI un papel dual, flhDC, junto con YpsR de activaci&oacute;n pero reprimiendo fliA junto con YtbR y YpsR. En ensayos de l&iacute;quido y placa, fue suprimido el inicio temprano de la motilidad observada en mutantes ypsR y ypsI en ytbI, ytbR ypsI/ytbI, ypsR/ytbR mutantes, indicando que QS regula la motilidad positivamente (v&iacute;a YtbRI) y negativamente (via YpsRI).

Predatory Bdellovibrio bacteriovorus bacteria are remarkable in that they attach to, penetrate and digest other Gram-negative bacteria, living and replicating within them until all resources are exhausted, when they escape the prey ghost to invade fresh prey. Remarkable remodeling of both predator and prey cell occurs during this process to allow the Bdellovibrio to exploit the intracellular niche they have worked so hard to enter, keeping the prey "bdelloplast" intact until the end of predatory growth. If one views motile non-predatory bacteria in a light microscope, one is immediately struck by how rare it is for bacteria to collide. This highlights how the cell surface of Bdellovibrio must be specialized and adapted to allow productive collisions and further to allow entry into the prey periplasm and subsequent secretion of hydrolytic enzymes to digest it. Bdellovibrio can, however, also be made to grow artificially without prey; thus, they have a large genome containing both predatory genes and genes for saprophytic heterotrophic growth. Thus, the membrane and outer surface layers are a patchwork of proteins encompassing not only those that have a sole purpose in heterotrophic growth but also many more that are specialized or employed to attach to, enter, remodel, kill and ultimately digest prey cells. There is much that is as yet not understood, but molecular genetic and post-genomic approaches to microbial physiology have enhanced the pioneering biochemical work of four decades ago in characterizing some of the key events and surface protein requirements for prey attack.

Bdellovibrio bacteriovorus is a Gram-negative bacterium that is a pathogen of other Gram-negative bacteria, including many bacteria which are pathogens of humans, animals and plants. As such Bdellovibrio has potential as a biocontrol agent, or living antibiotic. B. bacteriovorus HD100 has a large genome and it is not yet known which of it encodes the molecular machinery and genetic control of predatory processes. We have tried to fill this knowledge-gap using mixtures of predator and prey mRNAs to monitor changes in Bdellovibrio gene expression at a timepoint of early-stage prey infection and prey killing in comparison to control cultures of predator and prey alone and also in comparison to Bdellovibrio growing axenically (in a prey-or host independent "HI" manner) on artificial media containing peptone and tryptone. From this we have highlighted genes of the early predatosome with predicted roles in prey killing and digestion and have gained insights into possible regulatory mechanisms as Bdellovibrio enter and establish within the prey bdelloplast. Approximately seven percent of all Bdellovibrio genes were significantly up-regulated at 30 minutes of infection--but not in HI growth--implicating the role of these genes in prey digestion. Five percent were down-regulated significantly, implicating their role in free-swimming, attack-phase physiology. This study gives the first post-genomic insight into the predatory process and reveals some of the important genes that Bdellovibrio expresses inside the prey bacterium during the initial attack.

Yersinia pseudotuberculosis forms biofilms on Caenorhabditis elegans which block nematode feeding. This genetically amenable host-pathogen model has important implications for biofilm development on living, motile surfaces. Here we show that Y. pseudotuberculosis biofilm development on C. elegans is governed by N-acylhomoserine lactone (AHL)-mediated quorum sensing (QS) since (i) AHLs are produced in nematode associated biofilms and (ii) Y. pseudotuberculosis strains expressing an AHL-degrading enzyme or in which the AHL synthase (ypsI and ytbI) or response regulator (ypsR and ytbR) genes have been mutated, are attenuated. Although biofilm formation is also attenuated in Y. pseudotuberculosis strains carrying mutations in the QS-controlled motility regulator genes, flhDC and fliA, and the flagellin export gene, flhA, flagella are not required since fliC mutants form normal biofilms. However, in contrast to the parent and fliC mutant, Yop virulon proteins are up-regulated in flhDC, fliA and flhA mutants in a temperature and calcium independent manner. Similar observations were found for the Y. pseudotuberculosis QS mutants, indicating that the Yop virulon is repressed by QS via the master motility regulator, flhDC. By curing the pYV virulence plasmid from the ypsI/ytbI mutant, by growing YpIII under conditions permissive for type III needle formation but not Yop secretion and by mutating the type III secretion apparatus gene, yscJ, we show that biofilm formation can be restored in flhDC and ypsI/ytbI mutants. These data demonstrate that type III secretion blocks biofilm formation and is reciprocally regulated with motility via QS.

Par polym&eacute;risation du plasmide pYV de virulence du mutant ypsI/ytbI, en cultivant des YpIII dans des conditions permissives pour type III aiguille formation mais pas la s&eacute;cr&eacute;tion de Yop et par mutation du g&egrave;ne appareils de type III s&eacute;cr&eacute;tion, yscJ, nous montrons que la formation de biofilms peut &ecirc;tre restaur&eacute;e chez les mutants flhDC et ypsI/ytbI. Ces r&eacute;sultats d&eacute;montrent que le type de s&eacute;cr&eacute;tion III bloque la formation de biofilm et r&eacute;ciproquement rel&egrave;ve de la mobilit&eacute; par l&#39;interm&eacute;diaire de QS.

Yersinia Pseudotuberculosis bildet Biofilmen auf Caenorhabditis Elegans Fadenwurm F&uuml;tterung blockieren. Pseudotuberculosis St&auml;mme auszudr&uuml;cken Enzym AHL-erniedrigende oder in dem der AHL-Synthase (YpsI und YtbI) oder Antwort Regler (YpsR und YtbR) Gene mutiert worden sind, sind abgeschw&auml;cht. Obwohl Biofilm-Bildung auch in Y. Pseudotuberculosis St&auml;mme mit Mutationen in der QS-gesteuerte Motilit&auml;t Regulator Gene, FlhDC und FliA und das Flagellin-Export-gen, FlhA, abgeschw&auml;cht ist sind Flagellen nicht erforderlich, da FliC Mutanten normale Biofilme bilden. Allerdings sind im Gegensatz zu den &uuml;bergeordneten und FliC Mutant, Yop Virulon Proteine oben-geregelt im FlhDC, FliA und FlhA Mutanten auf unabh&auml;ngige Weise Temperatur und Kalzium. &Auml;hnliche Beobachtungen wurden f&uuml;r die Y. Pseudotuberculosis QS-Mutanten, die darauf hinweist, dass die Yop Virulon &uuml;ber den Meister Motilit&auml;t Spannungsregler, FlhDC von QS unterdr&uuml;ckt ist gefunden. Indem man heilt die pYV Virulenz Plasmid aus der YpsI/YtbI-Mutant, durch den Anbau von YpIII unter Bedingungen f&uuml;r Typ III permissive Nadel, Bildung aber nicht Yop Sekretion und mutiert das Typ III-Sekretion Apparat gen, YscJ, zeigen wir, dass Biofilm-Bildung in FlhDC und YpsI/YtbI Mutanten wiederhergestellt werden kann. Diese Daten zeigen, dass Typ-III-Sekretion Biofilm-Bildung blockiert und gegenseitig mit Motilit&auml;t &uuml;ber QS geregelt ist.

Yersinia pseudotuberculosis forma biopel&iacute;culas en Caenorhabditis elegans, que bloquean la alimentaci&oacute;n del nematodo. Gen&eacute;ticamente susceptibles hospedante-pat&oacute;geno tiene implicaciones importantes para el desarrollo de la biopel&iacute;cula sobre superficies m&oacute;viles vivas. Aqu&iacute; mostramos que el desarrollo de biofilms Y. pseudotuberculosis en C. elegans se rige por N-acylhomoserine lactona (AHL)-mediada qu&oacute;rum sensing (QS) puesto que (i) AHLs se produjeron en nematodos asociados biofilms y cepas de pseudotuberculosis (ii) y expresando una enzima AHL-degradantes o en que el AHL synthase (ypsI y ytbI) o genes de respuesta regulador (ypsR y ytbR) han sido transformados, se aten&uacute;an. Aunque la formaci&oacute;n de biopel&iacute;culas tambi&eacute;n se aten&uacute;a en las cepas de Y. pseudotuberculosis que portan mutaciones en los genes de regulador de motilidad QS-controlado, flhDC y fliA y el gen de exportaci&oacute;n de flagelina, flhA, flagelos no son necesarios ya que mutantes de fliC forman biofilms normales. Sin embargo, en contraste con el padre y fliC mutante, Yop virulon prote&iacute;nas son regulados por arriba en mutantes flhDC, fliA y flhA de una manera independiente de temperatura y calcio. Observaciones similares se encontraron la Y. pseudotuberculosis mutantes de QS, indicando que la virulon Yop es reprimido por QS mediante el regulador principal de la motilidad, flhDC. Por el curado el pl&aacute;smido de virulencia de pYV del mutante de ypsI/ytbI, creciendo YpIII en condiciones permisivas para tipo III aguja formaci&oacute;n pero no Yop secreci&oacute;n y por mutaci&oacute;n en el gene de aparato secreci&oacute;n de tipo III, yscJ, demostramos que la formaci&oacute;n del biofilm puede restaurarse en mutantes flhDC y ypsI/ytbI. Estos datos demuestran que tipo de secreci&oacute;n III bloquea la formaci&oacute;n de biopel&iacute;culas y rec&iacute;procamente se regula con movilidad via QS.

Represi&oacute;n dependiente de la detecci&oacute;n de qu&oacute;rum de tipo secreci&oacute;n III facilita el desarrollo de biofilms en Caenorhabditis elegans por Yersinia.

Bdellovibrio bacteriovorus grows in one of two ways: either (i) predatorily [in a host-dependent (HD) manner], when it invades the periplasm of another Gram-negative bacterium, exporting into the prey co-ordinated waves of soluble enzymes using the prey cell contents for growth; or (ii) in a host-independent (HI) manner, when it grows (slowly) axenically in rich media. Periplasmic invasion potentially exposes B. bacteriovorus to extremes of pH and exposes the need to scavenge electron donors from prey electron transport components by synthesis of metalloenzymes. The twin-arginine transport system (Tat) in other bacteria transports folded metalloenzymes and the B. bacteriovorus genome encodes 21 potential Tat-transported substrates and Tat transporter proteins TatA1, TatA2 and TatBC. GFP tagging of the Tat signal peptide from Bd1802, a high-potential iron-sulfur protein (HiPIP), revealed it to be exported into the prey bacterium during predatory growth. Mutagenesis showed that the B. bacteriovorus tatA2 and tatC gene products are essential for both HI and HD growth, despite the fact that they partially complement (in SDS resistance assays) the corresponding mutations in Escherichia coli where neither TatA nor TatC are essential for life. The essentiality of B. bacteriovorus TatA2 was surprising given that the B. bacteriovorus genome encodes a second tatA homologue, tatA1. Transcription of tatA1 was found to be induced upon entry to the bdelloplast, and insertional inactivation of tatA1 showed that it significantly slowed the rates of both HI and HD growth. B. bacteriovorus is one of a few bacterial species that are reliant on a functional Tat system and where deletion of a single tatA1 gene causes a significant growth defect(s), despite the presence of its tatA2 homologue.

Bdellovibrio bacteriovorus &#xD2B8;&#xC708; &#xC544;&#xB974;&#xAE30;&#xB2CC; &#xC804;&#xC1A1; &#xC2DC;&#xC2A4;&#xD15C; &#xC57D;&#xD0C8; &#xBC0F; &#xBA39;&#xC774; &#xB3C5;&#xB9BD;&#xC801;&#xC778; &#xC131;&#xC7A5;&#xC5D0; &#xC5ED;&#xD560;&#xC5D0;.

Bdellovibrio bacteri&oacute;vora crece en uno de dos maneras: (i) predatorily [en forma dependiente anfitri&oacute;n (HD)], cuando invade el periplasma de otra bacteria gramnegativa, exportar a las olas coordinada de la presa de enzimas solubles utilizando el contenido de la celda de presa para el crecimiento; o (ii) de manera independiente del host (HI), cuando axenically (lentamente) crece en medios enriquecidos. Invasi&oacute;n periplasmic potencialmente expone B. bacteri&oacute;vora a los extremos de pH y expone la necesidad de buscar a donantes del electr&oacute;n de componentes de transporte de electrones de presa por s&iacute;ntesis de metaloenzimas. El sistema de transporte de doble-arginina (Tat) en transportes de otros tipo de bacterias doblado metaloenzimas y el genoma B. bacteri&oacute;vora codifica 21 posibles sustratos transportado por el Tat y prote&iacute;nas transportadoras de Tat TatA1, TatA2 y TatBC. GFP marcado del p&eacute;ptido se&ntilde;al Tat de Bd1802, una prote&iacute;na hierro-azufre de alto potencial (HiPIP), revel&oacute; a la exportaci&oacute;n en la bacteria de la presa durante el crecimiento del depredador. Mutag&eacute;nesis demostr&oacute; que los productos del gene bacteri&oacute;vora B. tatA2 y tatC son esenciales para el crecimiento tanto en HI y en HD, a pesar del hecho que parcialmente complementan (en ensayos de resistencia de SDS) las correspondientes mutaciones en Escherichia coli donde TatA ni TatC son esenciales para la vida. La esencialidad del B. bacteri&oacute;vora TatA2 fue sorprendente, dado que el genoma B. bacteri&oacute;vora codifica un segundo hom&oacute;logo de tatA, tatA1. Transcripci&oacute;n de tatA1 fue encontrado para ser inducido al entrar a la bdelloplast, y insertional inactivaci&oacute;n de tatA1 demostr&oacute; que desaceler&oacute; significativamente las tasas de crecimiento tanto HI y HD.
