Kaposi's sarcoma-associated herpesvirus (KSHV) is the etiologic agent for KS tumors, multicentric Castleman's disease, and primary effusion lymphomas. Like other herpesvirus capsids, the KSHV capsid is an icosahedral structure composed of six proteins. The capsid shell is made up of the major capsid protein, two triplex proteins, and the small capsid protein. The scaffold protein and the protease occupy the internal space. The assembly of KSHV capsids is thought to occur in a manner similar to that determined for herpes simplex virus type 1 (HSV-1). Our goal was to assemble KSHV capsids in insect cells using the baculovirus expression vector system. Six KSHV capsid open reading frames were cloned and the proteins expressed in Sf9 cells: pORF25 (major capsid protein), pORF62 (triplex 1), pORF26 (triplex 2), pORF17 (protease), pORF17.5 (scaffold protein), and also pORF65 (small capsid protein). When insect cells were coinfected with these baculoviruses, angular capsids that contained internal core structures were readily observed by conventional electron microscopy of the infected cells. Capsids were also readily isolated from infected cells by using rate velocity sedimentation. With immuno-electron microscopy methods, these capsids were seen to be reactive to antisera to pORF65 as well as to KSHV-positive human sera, indicating the correct conformation of pORF65 in these capsids. When either virus expressing the triplex proteins was omitted from the coinfection, capsids did not assemble; similar to observations made in HSV-1-infected cells. If the virus expressing the scaffold protein was excluded, large open shells that did not attain icosahedral structure were seen in the nuclei of infected cells. The presence of pORF65 was required for capsid assembly, in that capsids did not form if this protein was absent as judged by both by ultrastructural analysis of infected cells and rate velocity sedimentation experiments. Thus, a novel outcome of this study is the finding that the small capsid protein of KSHV, like the major capsid and triplex proteins, is essential for capsid shell assembly.

Small capsid protein pORF65 is essential for assembly of Kaposi's sarcoma-associated herpesvirus capsids.

Herp&egrave;svirus associ&eacute; au sarcome de Kaposi (KSHV) est l&#39;agent &eacute;tiologique de tumeurs KS, la maladie de Castleman multicentrique et lymphomes effusion primaire. Comme autres herp&egrave;svirus capsides, la capside KSHV est une structure icosa&eacute;drique compos&eacute;e de six prot&eacute;ines. La coquille de la capside est compos&eacute;e de la prot&eacute;ine majeure de la capside, deux prot&eacute;ines triplex et la petit de la capside. La prot&eacute;ine d&#39;&eacute;chafaudage et la prot&eacute;ase occupent l&#39;espace interne. L&#39;Assembl&eacute;e des capsides KSHV est cens&eacute;e se produire d&#39;une mani&egrave;re analogue &agrave; celle d&eacute;termin&eacute;e pour le virus herp&egrave;s simplex de type 1 (HSV-1). Notre objectif &eacute;tait de rassembler les capsides KSHV dans des cellules d&#39;insectes en utilisant le syst&egrave;me de vecteur d&#39;expression baculovirus. Six capside KSHV ouvert lecture trames ont &eacute;t&eacute; clon&eacute;s et les prot&eacute;ines exprim&eacute;es dans les cellules Sf9 : pORF25 (major de la capside), pORF62 (1 triplex), pORF26 (2 triplex), pORF17 (prot&eacute;ase), pORF17.5 (prot&eacute;ine d&#39;&eacute;chafaudage) et aussi pORF65 (petit de la capside). Lorsque des cellules d&#39;insecte sont coinfect&eacute;s par ces baculovirus, capsides angulaires qui contenait des structures internes principales ont &eacute;t&eacute; facilement observables en microscopie conventionnelle des cellules infect&eacute;es. Capsides &eacute;taient aussi facilement isol&eacute;s des cellules infect&eacute;es en utilisant la s&eacute;dimentation de la vitesse de taux. Avec les m&eacute;thodes de microscopie immuno-&eacute;lectron, ces capsides &eacute;taient per&ccedil;us comme &eacute;tant r&eacute;actif aux antis&eacute;rums de pORF65 ainsi que les s&eacute;rums humains KSHV-positif, indiquant la bonne conformation de pORF65 dans ces capsides. La pr&eacute;sence de pORF65 &eacute;tait n&eacute;cessaire pour l&#39;assemblage de la capside, que capsides ne se forment pas si cette prot&eacute;ine &eacute;tait absente si l&#39;on en juge par les deux par les analyses ultrastructurales des cellules infect&eacute;es et les exp&eacute;riences de taux vitesse s&eacute;dimentation. Ainsi, un nouveau r&eacute;sultat de cette &eacute;tude est la constatation que la prot&eacute;ine de capside petit de KSHV, comme la majeure de la capside et prot&eacute;ines triplex, est essentielle pour l&#39;assemblage de coquille de capside.

Kaposi-Sarkom-assoziierte Herpesvirus (KSHV) ist das &auml;tiologische Mittel f&uuml;r KS-Tumoren, multizentrischen Castleman-Krankheit und prim&auml;ren Erguss Lymphome. Wie andere Capsids Herpesvirus ist das Kapsid KSHV einer ikosaedrischen Struktur, bestehend aus sechs Proteine. Die Shell Kapsid besteht aus das wichtigste Kapsidprotein, zwei triplex Proteine und das kleine Kapsidprotein. Das Ger&uuml;st-Protein und die Protease einnehmen internen Platz. Die Montage der KSHV Capsids ist vermutlich auftreten, &auml;hnlich wie bei Herpes-Simplex-Virustyp 1 (HSV-1) bestimmt. Unser Ziel war es, KSHV Capsids in Insektenzellen mit dem Baculovirus Ausdruck Vektor-System zusammenstellen. Sechs KSHV Kapsid offen lesen Bilder wurden geklont und die Proteine, die in Sf9 Zellen: pORF25 (wichtigste Kapsidprotein), pORF62 (triplex 1), pORF26 (triplex 2), pORF17 (Protease), pORF17.5 (Ger&uuml;st-Protein) und auch pORF65 (kleine Kapsidprotein). Wenn Insektenzellen mit dieser Baculoviruses coinfected waren, wurden eckig Capsids, die interne Strukturen enthalten leicht von konventionellen Elektronenmikroskopie der infizierten Zellen beobachtet. Capsids wurden auch von infizierten Zellen mit Tarif Geschwindigkeit Ablagerung leicht isoliert. Mit Immuno-Elektronen-Mikroskopie-Methoden galten diese Capsids reaktiv, Antiseren, pORF65 als auch hinsichtlich der menschlichen KSHV-positiven Seren, angibt, die richtige Gestaltung des pORF65 in diesen Capsids sein. Wenn entweder Virus zum Ausdruck der triplex-Proteine in der Coinfection weggelassen wurde, zusammen Capsids nicht; &auml;hnliche Beobachtungen in HSV-1-infizierten Zellen. Wenn das Virus zum Ausdruck bringen des Ger&uuml;st-Proteins ausgeschlossen wurde, galten gro&szlig;e offene Muscheln, die nicht deren Struktur erreichen in den Kernen der infizierten Zellen. Das Vorhandensein von pORF65 war bei der Kapsid Versammlung, in diesem Capsids nicht Formular, wenn dieses Protein nicht vorkam wie von beiden durch ultrastrukturelle Analyse von infizierten Zellen und Tarif Geschwindigkeit Ablagerung Experimente beurteilt. So ist ein neuartiges Ergebnis dieser Studie die Feststellung, dass das kleine Kapsidprotein von KSHV, wie die gro&szlig;en Kapsid und triplex Proteine, f&uuml;r Kapsid Schale Montage wichtig.

Come altri herpesvirus capsidi, il capside KSHV &egrave; una struttura icosaedrica composta da sei proteine. Il guscio del capside &egrave; costituito dalla proteina maggiore del capside, due proteine triplex ed il piccolo capside. La proteina dell&#39;impalcatura e la proteasi occupare lo spazio interno. L&#39;assemblaggio dei capsidi KSHV &egrave; pensato per accadere in un modo simile a quello determinato per l&#39;herpes simplex virus tipo 1 (HSV-1). Il nostro obiettivo era quello di assemblare capsidi KSHV nelle cellule di insetto utilizzando il sistema del vettore di espressione baculovirus. Sei del capside KSHV open reading frames sono stati clonati e le proteine espresse nelle cellule Sf9: pORF25 (le principali proteine del capside), pORF62 (triplex 1), pORF26 (triplex 2), pORF17 (proteasi), pORF17.5 (proteina dell&#39;impalcatura) e anche pORF65 (proteina del capside piccolo). Quando le cellule degli insetti sono stata coinfettate con questi baculoviruses, capsidi angolare che conteneva le strutture del nucleo interno erano facilmente osservabili convenzionale microscopia elettronica delle cellule infette. Capsidi erano anche prontamente isolati da cellule infette tramite sedimentazione velocit&agrave; tasso. Con metodi di microscopia immuno-elettrone, questi capsidi sono stati visti per essere reattivi di antisieri pORF65 pure quanto sieri umani KSHV-positiva, che indica la corretta conformazione del pORF65 in questi capsidi. Quando dal Cytomegalovirus &egrave; stato omesso o virus che esprimono le proteine triplex, capsidi non ha fatto montare; simili alle osservazioni fatte in cellule infettate da HSV-1. Se &egrave; stato escluso il virus che esprimono la proteina dell&#39;impalcatura, grandi conchiglie aperte che non ha fatto raggiungere la struttura icosaedrica erano visti nei nuclei delle cellule infette. La presenza di pORF65 &egrave; stato richiesta per l&#39;assemblaggio del capside, in quanto non ha fatto formare capsidi se questa proteina &egrave; assente come giudicato da entrambi da analisi ultrastrutturale delle cellule infette e tasso velocit&agrave; sedimentazione esperimenti. Cos&igrave;, un romanzo risultato di questo studio &egrave; la constatazione che la proteina del capside piccolo di KSHV, come le principali proteine del capside e proteine triplex, &egrave; essenziale per l&#39;assemblaggio del guscio del capside.

Herpesvirus asociado sarcoma de Kaposi (KSHV) es el agente etiol&oacute;gico para tumores de KS, enfermedad de Castleman multic&eacute;ntrico y linfomas primario de la efusi&oacute;n. Como otro herpesvirus icosah&eacute;drica, la c&aacute;pside KSHV es una estructura icosa&eacute;drica compuesta por seis prote&iacute;nas. La c&aacute;scara de la c&aacute;pside est&aacute; compuesta por la prote&iacute;na de la c&aacute;pside principales, dos prote&iacute;nas triples y la prote&iacute;na de la c&aacute;pside peque&ntilde;o. La prote&iacute;na y la proteasa ocupan el espacio interno. La Asamblea de la c&aacute;psida KSHV se piensa para ocurrir en forma similar a la que se determin&oacute; para el virus de herpes simple tipo 1 (HSV-1). Nuestro objetivo era montar icosah&eacute;drica KSHV en c&eacute;lulas de los insectos utilizando el sistema de vector de expresi&oacute;n de baculovirus. Seis c&aacute;pside KSHV se clonaron marcos de lectura abierta y las prote&iacute;nas expresadas en c&eacute;lulas Sf9: pORF25 (importante prote&iacute;na de la c&aacute;pside), pORF62 (1 triple), pORF26 (2 triples), pORF17 (proteasa), pORF17.5 (prote&iacute;na) y tambi&eacute;n pORF65 (prote&iacute;na de la c&aacute;pside peque&ntilde;o). Cuando las c&eacute;lulas de los insectos estaban coinfectadas por estos baculovirus, c&aacute;psida angular que conten&iacute;an estructuras internas principales f&aacute;cilmente fueron observada por microscop&iacute;a convencional de las c&eacute;lulas infectadas. Tambi&eacute;n fueron aislado f&aacute;cilmente c&aacute;psida de c&eacute;lulas infectadas mediante el uso de sedimentaci&oacute;n de velocidad de tasa. Con m&eacute;todos de microscop&iacute;a de electrones inmuno, estos icosah&eacute;drica parec&iacute;an reactiva a antisueros a pORF65 as&iacute; como suero humano KSHV-positivo, indicando la conformaci&oacute;n correcta de pORF65 en estos icosah&eacute;drica. Cuando cualquiera de los dos virus que expresa las prote&iacute;nas triples fue omitido de la coinfecci&oacute;n, no montar c&aacute;psida; similar a las observaciones en c&eacute;lulas infectadas con HSV-1. Si el virus que expresa la prote&iacute;na fue excluido, grandes conchas abiertas que no logr&oacute; estructura icosa&eacute;drica fueron vistos en los n&uacute;cleos de las c&eacute;lulas infectadas. La presencia de pORF65 era necesaria para el montaje de la c&aacute;pside, en eso icosah&eacute;drica no form&oacute; si esta prote&iacute;na era ausente seg&uacute;n lo juzgado por tanto por an&aacute;lisis ultraestructural de las c&eacute;lulas infectadas y velocidad velocidad sedimentaci&oacute;n experimentos. As&iacute;, un resultado novedoso de este estudio es la conclusi&oacute;n que la c&aacute;pside peque&ntilde;o de KSHV, como los principal c&aacute;pside y triples prote&iacute;nas, es esencial para la Asamblea de c&aacute;scara de c&aacute;pside.

C&aacute;pside peque&ntilde;a prote&iacute;na pORF65 es esencial para el montaje de la c&aacute;psida de herpesvirus asociado sarcoma de Kaposi.

Staphylococcal peptidoglycan hydrolases are a potential new source of antimicrobials. A large subset harbors C-terminal SH3b_5 cell wall binding domains. These C-terminal domains have been shown to be necessary for accurate cell wall recognition and subsequent staphylolytic activity for some endolysins. Over fifty proteins of staphylococcal or phage origin containing SH3b domains were aligned, yielding five highly repetitive groups of proteins. Representative C-termini from these five groups, and six staphylococcal proteins for which no homologues have been identified, were aligned, revealing two distinct SH3b_5 subgroups with overlapping but differentially conserved residues. A premise behind this research is that there may be unique cell wall binding properties conferred by these staphylococcal domains that could be exploited to specifically enhance anti-staphylococcal efficacy in heterologous protein fusion constructs. To identify functional differences between the two subgroups, the native Cpl-7 cell wall binding domains of the streptococcal LambdaSa2 endolysin were replaced by staphylococcal SH3b domains from both subgroups. SH3b domains from either lysostaphin (bacteriocin) or LysK (phage endolysin) resulted in a approximately 5x increase in staphylolytic activity conferred on the streptococcal endopeptidase domain, and surprisingly these same fusions maintained significant streptolytic activity suggesting that the staphylococcal SH3b domains are not always staphylococcal-specific. A comparison of the differences in lytic activity conferred on the LambdaSa2 endopeptidase domain by either LysK or lysostaphin SH3b domain differed by no more than a factor of two. Through the collection of peptidoglycan hydrolase sequences, three new putative intron-containing phage endolysin genes were identified in public data sets for the phages G1, X2 and 85.
