Taste cells have a limited life span and are replaced from a basal cell population, although the specific factors involved in this process are not well known. Short- and long-term cultures of other sensory cells have facilitated efforts to understand the signals involved in proliferation, differentiation, and senescence, yet few studies have reported successful primary culture protocols for taste cells. Furthermore, no studies have demonstrated both proliferation and differentiation in vitro. In this study, we have developed an in vitro culture system to maintain and utilize rat primary taste cells for more than 2 months without losing key molecular and biochemical features. Gustducin, phospholipase C-beta2 (PLC-beta2), T1R3, and T2R5 mRNA were detected in the cultured cells by reverse transcriptase-polymerase chain reaction. Western blot analysis demonstrated gustducin and PLC-beta2 expression in the same samples, which was confirmed by immunocytochemistry. Labeling with bromo-2-deoxyuridine (BrdU) demonstrated proliferation, and a subset of BrdU-labeled cells were also immunoreactive for either gustducin or PLC-beta2, indicating differentiation of newly generated cells in vitro. Cultured cells also exhibited increases in intracellular calcium in response to several taste stimuli. These results indicate that taste cells from adult rats can be generated and maintained under the described conditions for at least 2 months. This system will enable further studies of the processes involved in proliferation, differentiation, and function of mammalian taste receptor cells in an in vitro preparation.

Characterization and long-term maintenance of rat taste cells in culture.

Kultivierte Zellen zeigte auch Erh&ouml;hungen der intrazellul&auml;re Kalzium in Reaktion auf verschiedene Reize der Geschmack. Diese Ergebnisse zeigen, dass Geschmack Zellen von Erwachsenen Ratten erzeugt und unter den beschriebenen Bedingungen f&uuml;r mindestens 2 Monate aufrechterhalten werden k&ouml;nnen. Las c&#xE9;lulas del gusto tienen una vida &#xFA;til limitada y son reemplazadas por una poblaci&#xF3;n de c&#xE9;lulas basales, aunque los factores espec&#xED;ficos que intervienen en este proceso no son bien conocidos. A corto y largo plazo las culturas de otras c&#xE9;lulas sensoriales han facilitado los esfuerzos para comprender las se&#xF1;ales implicadas en la proliferaci&#xF3;n, diferenciaci&#xF3;n y la senectud, sin embargo, pocos estudios han informado de &#xE9;xito los protocolos de cultivo primario de las c&#xE9;lulas gustativas. Por otra parte, ning&#xFA;n estudio ha demostrado tanto en la proliferaci&#xF3;n y diferenciaci&#xF3;n in vitro. En este estudio, hemos desarrollado un sistema de cultivo in vitro para mantener y utilizar las c&#xE9;lulas primarias de rata gusto de m&#xE1;s de 2 meses sin perder las principales caracter&#xED;sticas moleculares y bioqu&#xED;micos. Gustducin, la fosfolipasa C-beta2 (PLC-beta2), T1R3, T2R5 y mRNA se detect&#xF3; en las c&#xE9;lulas cultivadas mediante la reacci&#xF3;n en cadena de la polimerasa-transcriptasa. Western blot demostr&#xF3; la expresi&#xF3;n gustducin y el PLC-beta2 en las mismas muestras, lo cual fue confirmado por inmunocitoqu&#xED;mica. Etiquetado con bromo-2-desoxiuridina (BrdU) demostraron la proliferaci&#xF3;n, y un subconjunto de c&#xE9;lulas BrdU marcado tambi&#xE9;n fueron inmunorreactivas para cualquiera gusducina o PLC beta2, indicando la diferenciaci&#xF3;n de las c&#xE9;lulas reci&#xE9;n generadas in vitro. Las c&#xE9;lulas cultivadas tambi&#xE9;n se presentaron aumento en el calcio intracelular en respuesta a los est&#xED;mulos del gusto de varios. Estos resultados indican que las c&#xE9;lulas gustativas de ratas adultas se puede generar y mantenido bajo las condiciones descritas por lo menos 2 meses. Este sistema permitir&#xE1; realizar m&#xE1;s estudios sobre los procesos implicados en la proliferaci&#xF3;n, diferenciaci&#xF3;n y funci&#xF3;n de mam&#xED;feros, c&#xE9;lulas receptoras del gusto de una preparaci&#xF3;n in vitro.

Taste loss or alterations can seriously impact health and quality of life due to the resulting negative influence on eating habits and nutrition. Infection and inflammation are thought to be some of the most common causes of taste perception disorders. Supporting this view, neuro-immune interactions in the peripheral gustatory system have been identified, underlying the importance of this tissue in mucosal immunity, but we have little understanding of how these interactions influence taste perception directly or indirectly. This limited understanding is evident by the lack of even a basic knowledge of the resident immune cell populations in or near taste tissues. The present study characterized the distribution and population of the major immune cells and their subsets in healthy human anterior, lingual, fungiform papillae (FP) using immunohistochemistry. Dendritic cells (DCs) were the predominant innate immune cells in this tissue, including four subtypes: CD11c(+) DCs, DC-SIGN+ immature DCs, CD83(+) mature DCs, and CD1a(+) DCs (Langerhans cells). While most DCs were localized beneath the lamina propria and only moderately in the epithelium, CD1a(+) Langerhans cells were exclusively present within the epithelium and not in sub-strata. A small number of macrophages were observed. T lymphocytes were present throughout the FP with CD4(+) T cells more prevalent than CD8(+) T cells. Very few CD19(+) B lymphocytes were detected. The results show that DCs, macrophages, and T lymphocytes are the constitutive guardians of human FP taste tissue, with DCs and CD4 T cells being dominant, while B lymphocytes are rare under normal, healthy conditions. These observations provide a basic anatomical foundation for the immune response in the healthy human tongue as a basis for subsequent disease-related studies, but none of the present data indicate that the immune cell populations identified are, in fact, altered in individuals with abnormal taste perception.

P&eacute;rdida de gusto o alteraciones seriamente pueden afectar la salud y calidad de vida debido a la influencia negativa que resulta en h&aacute;bitos alimentarios y nutrici&oacute;n. Infecci&oacute;n y la inflamaci&oacute;n se cree que algunas de las causas m&aacute;s comunes de trastornos de la percepci&oacute;n del gusto. Few studies have examined the induction of squamous metaplasia in human olfactory nasal tissue caused by tobacco use and the implications it may have for olfaction, particularly when there are pre-existing insults, such as chronic rhinosinusitis (CRS). Quantitative histopathological analyses were performed on Alcian blue- and H&E-stained sections of nasal biopsies taken from the upper aspect of the middle turbinate of CRS patients. Chronic rhinosinusitis patients who were current smokers had a predominance of squamous metaplasia in the olfactory sensory epithelium, whereas CRS patients who were nonsmokers and were not exposed to secondhand cigarette smoke had a prevalence of goblet cell hyperplasia. In spite of this difference, the groups did not differ significantly in olfactory threshold sensitivity. The impact of primary cigarette smoke on olfaction and a possible role of squamous metaplasia in preserving olfactory neurogenesis are discussed.

Smoking-associated squamous metaplasia in olfactory mucosa of patients with chronic rhinosinusitis.

 Cotinine is a principal metabolite of nicotine with a substantially longer half-life, and cotinine levels in saliva, urine or serum are widely used to validate self-reported smoking status. The nasal cavity and olfactory system are directly exposed to tobacco smoke in smokers and in non-smokers who live with or work around smokers. However, despite the potential for a direct impact of tobacco smoke on the nasal epithelium and olfactory neurons, no prior studies have assessed cotinine levels in nasal mucus. We sought to determine whether cotinine levels in nasal lavage fluid (NLF) would provide a reasonable estimate of smoke exposure. We assayed cotinine using a competitive immunoassay in NLF from 23 smokers, 10 non-smokers exposed to tobacco smoke (ETS) and 60 non-smokers who did not report smoke exposure. NLF cotinine levels were significantly higher in smokers than in non-smokers, regardless of their exposure to ambient tobacco smoke. Cotinine levels in this small group of exposed non-smokers were not significantly different than those of non-exposed non-smokers. A cutoff of 1 ng/ml provided a sensitivity of 91% and a specificity of 99% for smoking status in this sample. Data were consistent with self-reported smoking status, and a cutoff of 1.0 ng/ml NLF cotinine may be used to classify smoking status. While saliva is the most easily obtained body fluid, NLF can be used to provide an objective and precise indication of smoking status and more directly reflects smoke exposure in the nasal and olfactory mucosa.

The impact of chronic rhinosinusitis (CRS) on the olfactory mucosa (OM) is dramatic. Cellular profiles and epithelial integrity in OM biopsies were evaluated using histological and immunohistochemical methods to define a strategy for future histological studies of CRS. We have examined nasal biopsies of 54 CRS patients (18-63 years old) and have defined specific histopathological patterns of the OM: normal pseudostratified, goblet cell hyperplasia, squamous metaplasia, and erosion. Goblet cell hyperplasia was most similar to a normal pseudostratified OM pattern but with goblet cells intermixed in the apical layers. Squamous metaplasia exhibited an absence of olfactory supporting cells and had olfactory sensory neurons that were morphologically abnormal. It is unknown if these neurons would be functional in this type of tissue transformation. The pattern of erosion exhibited a severe loss of epithelial layers and a higher prevalence of infiltrating inflammatory cells within the olfactory epithelium when compared to the other OM patterns. Although it is not known if the OM patterns we have noted correspond to specific stages or distinct pathways of the disease, the template proposed here can be used in further studies to understand how the histopathological progression of CRS relates to olfactory loss and the response to treatment.

Aqu&#xED; se describe un m&#xE9;todo para aislar c&#xE9;lulas humanas enzim&#xE1;tica de las papilas fungiformes obtenidas por biopsia y mantener en cultivo por m&#xE1;s de 7 pasos (7 meses) sin p&#xE9;rdida de viabilidad y manteniendo al mismo tiempo muchas de las propiedades funcionales de las c&#xE9;lulas del gusto sumamente aisladas. Las c&#xE9;lulas en estos cultivos exhiben aumentos en el calcio intracelular cuando se estimulan con concentraciones apropiadas de est&#xED;mulos perceptualmente gustativas varias, indicando que al menos algunas de las v&#xED;as de se&#xF1;alizaci&#xF3;n nativas estaban presentes. The ability to maintain human fungiform papillae cells in culture for multiple cell cycles would be of considerable utility for characterizing the molecular, regenerative, and functional properties of these unique sensory cells. Here we describe a method for enzymatically isolating human cells from fungiform papillae obtained by biopsy and maintaining them in culture for more than 7 passages (7 months) without loss of viability and while retaining many of the functional properties of acutely isolated taste cells. Cells in these cultures exhibited increases in intracellular calcium when stimulated with perceptually appropriate concentrations of several taste stimuli, indicating that at least some of the native signaling pathways were present. This system can provide a useful model for molecular studies of the proliferation, differentiation, and physiological function of human fungiform papillae cells.

Nous d&#xE9;crivons ici une m&#xE9;thode pour isoler les cellules humaines par voie enzymatique &#xE0; partir de papilles fongiformes obtenus par biopsie et de les maintenir en culture pendant plus de 7 passages (7 mois) sans perte de viabilit&#xE9; et tout en conservant un grand nombre des propri&#xE9;t&#xE9;s fonctionnelles des cellules gustatives isol&#xE9;es aigu&#xEB;. Cellules en culture expos&#xE9; ces augmentations du calcium intracellulaire lorsqu&#x26;#39;il est stimul&#xE9; avec des concentrations appropri&#xE9;es de stimuli perceptuellement go&#xFB;t plusieurs, indiquant qu&#x26;#39;au moins une partie des voies de signalisation natifs sont pr&#xE9;sents. We report a cross-sectional study of olfactory impairment with age based on both odorant-stimulated responses of human olfactory sensory neurons (OSNs) and tests of olfactory threshold sensitivity. A total of 621 OSNs from 440 subjects in 2 age groups of younger (≤ 45 years) and older (≥ 60 years) subjects were investigated using fluorescence intensity ratio fura-2 imaging. OSNs were tested for responses to 2 odorant mixtures, as well as to subsets of and individual odors in those mixtures. Whereas cells from younger donors were highly selective in the odorants to which they responded, cells from older donors were more likely to respond to multiple odor stimuli, despite a loss in these subjects' absolute olfactory sensitivity, suggesting a loss of specificity. This degradation in peripheral cellular specificity may impact odor discrimination and olfactory adaptation in the elderly. It is also possible that chronic adaptation as a result of reduced specificity contributes to observed declines in absolute sensitivity.
