Guard cells are highly specialized cells that form tiny pores called stomata on the leaf surface. The opening and closing of stomata control leaf gas exchange and water transpiration as well as allow plants to quickly respond and adjust to new environmental conditions. Mesophyll cells are specialized for photosynthesis. Despite the phenotypic and obvious functional differences between the two types of cells, the full protein components and their functions have not been explored but are addressed here through a global comparative proteomics analysis of purified guard cells and mesophyll cells. With the use of isobaric tags for relative and absolute quantification (iTRAQ) tagging and two-dimensional liquid chromatography mass spectrometry, we identified 1458 non-redundant proteins in both guard cells and mesophyll cells of Brassica napus leaves. Based on stringent statistical criteria, a total of 427 proteins were quantified, and 74 proteins were found to be enriched in guard cells. Proteins involved in energy (respiration), transport, transcription (nucleosome), cell structure, and signaling are preferentially expressed in guard cells. We observed several well characterized guard cell proteins. By contrast, proteins involved in photosynthesis, starch synthesis, disease/defense/stress, and other metabolisms are preferentially represented in mesophyll cells. Of the identified proteins, 110 have corresponding microarray data obtained from Arabidopsis guard cells and mesophyll cells. About 72% of these proteins follow the same trend of expression at the transcript and protein levels. For the rest of proteins, the correlation between proteomics data and the microarray data is poor. This highlights the importance of quantitative profiling at the protein level. Collectively this work represents the most extensive proteomic description of B. napus guard cells and has improved our knowledge of the functional specification of guard cells and mesophyll cells.

Functional differentiation of Brassica napus guard cells and mesophyll cells revealed by comparative proteomics.

L&#x26;#39;ouverture et la fermeture des stomates des feuilles de contr&#xF4;le du gaz et de l&#x26;#39;eau de la transpiration de change ainsi que permettre aux plantes de r&#xE9;pondre rapidement et de s&#x26;#39;adapter aux nouvelles conditions environnementales. Les cellules du m&#xE9;sophylle sont sp&#xE9;cialis&#xE9;s pour la photosynth&#xE8;se. Malgr&#xE9; les diff&#xE9;rences ph&#xE9;notypiques et fonctionnels &#xE9;vidents entre les deux types de cellules, les composants prot&#xE9;iques complets et leurs fonctions n&#x26;#39;ont pas &#xE9;t&#xE9; explor&#xE9;es, mais sont abord&#xE9;es ici &#xE0; travers une analyse globale comparative de la prot&#xE9;omique purifi&#xE9; des cellules de garde et les cellules du m&#xE9;sophylle. Avec l&#x26;#39;utilisation d&#x26;#39;&#xE9;tiquettes isobares pour la quantification relative et absolue (iTRAQ) le marquage et &#xE0; deux dimensions spectrom&#xE9;trie de masse chromatographie en phase liquide, nous avons identifi&#xE9; 1458 non-redondants prot&#xE9;ines dans les cellules de garde et les cellules du m&#xE9;sophylle de feuilles de Brassica napus. Sur la base de crit&#xE8;res stricts statistiques, un total de 427 prot&#xE9;ines ont &#xE9;t&#xE9; quantifi&#xE9;es, et 74 prot&#xE9;ines ont &#xE9;t&#xE9; trouv&#xE9;s &#xE0; &#xEA;tre enrichi en cellules de garde. Les prot&#xE9;ines impliqu&#xE9;es dans l&#x26;#39;&#xE9;nergie (la respiration), le transport, la transcription (nucl&#xE9;osome), structure de la cellule, et la signalisation sont pr&#xE9;f&#xE9;rentiellement exprim&#xE9; dans les cellules de garde. Nous avons observ&#xE9; plusieurs prot&#xE9;ines bien caract&#xE9;ris&#xE9;es des cellules de garde. En revanche, les prot&#xE9;ines impliqu&#xE9;es dans la photosynth&#xE8;se, la synth&#xE8;se de l&#x26;#39;amidon, de la maladie / d&#xE9;fense / stress, et d&#x26;#39;autres m&#xE9;tabolismes sont pr&#xE9;f&#xE9;rentiellement repr&#xE9;sent&#xE9;es dans les cellules du m&#xE9;sophylle. Parmi les prot&#xE9;ines identifi&#xE9;es, 110 ont donn&#xE9;es de biopuces correspondants obtenus &#xE0; partir de cellules de garde d&#x26;#39;Arabidopsis et les cellules du m&#xE9;sophylle. Environ 72% de ces prot&#xE9;ines suivent la m&#xEA;me tendance d&#x26;#39;expression &#xE0; des niveaux de transcription et des prot&#xE9;ines. Pour le reste des prot&#xE9;ines, la corr&#xE9;lation entre les donn&#xE9;es prot&#xE9;omiques et les donn&#xE9;es microarray est pauvre. Cela souligne l&#x26;#39;importance quantitative du profilage au niveau prot&#xE9;ique. Collectivement, ce travail repr&#xE9;sente la description la plus compl&#xE8;te des cellules B. prot&#xE9;omique garde napus et a am&#xE9;lior&#xE9; notre connaissance de la sp&#xE9;cification fonctionnelle des cellules de garde et les cellules du m&#xE9;sophylle.

Diff&#xE9;renciation fonctionnelle des cellules de Brassica napus et de la Garde cellules du m&#xE9;sophylle R&#xE9;v&#xE9;l&#xE9; par prot&#xE9;omique comparative

Guard Zellen sind hoch spezialisierte Zellen, die winzige Poren genannt Spalt&ouml;ffnungen auf der Blattoberfl&auml;che zu bilden. Das &Ouml;ffnen und Schlie&szlig;en der Spalt&ouml;ffnungen Blatt Gas-Austausch und Wasser Transpiration zu steuern sowie Pflanzen schnell reagieren und Anpassung an neue Umweltbedingungen zu erm&ouml;glichen. Mesophyll Zellen sind f&uuml;r die Photosynthese spezialisiert. Trotz die ph&auml;notypische und offensichtlichen funktionalen Unterschiede zwischen den zwei Arten von Zellen die vollst&auml;ndige Proteinkomponenten und deren Funktionen nicht erforscht, sondern richten sich hier durch eine globale vergleichende Proteomics-Analyse des gereinigten Guard und Mesophyll Zellen. Mit dem Einsatz der Isobare Tags f&uuml;r relative und absolute Quantifizierung (iTRAQ) tagging und zweidimensionalen Fl&uuml;ssigchromatographie-Massenspektrometrie haben wir festgestellt, 1458 nicht-redundanten Proteine in sowohl Guard und Mesophyll Zellen Brassica Napus Bl&auml;tter. Auf der Grundlage der strenger statistischer Kriterien insgesamt 427 Proteine wurden quantifiziert und 74 Proteine wurden gefunden, um in Guard Zellen angereichert werden. Energie (Atmung), Transport, Transkription (Nukleosom), Zellstruktur und Signalisierung Proteine werden bevorzugt in Guard Zellen ausgedr&uuml;ckt. Wir beobachteten mehrere gut gekennzeichnete Wache Zelle Proteine. Im Gegensatz dazu werden Proteine beteiligt, Photosynthese, St&auml;rke-Synthese, Krankheit/Verteidigung/Stress und anderen Stoffwechsel bevorzugt im Mesophyll Zellen dargestellt. Der identifizierte Proteine haben 110 entsprechende Microarray Daten aus Arabidopsis Guard und Mesophyll Zellen. Ungef&auml;hr 72 % dieser Proteine folgen den gleichen Trend des Ausdrucks auf der Abschrift und Protein. F&uuml;r den Rest der Proteine ist die Korrelation zwischen Proteomics und den Microarray Daten schlecht. Dies unterstreicht die Bedeutung des quantitativen profiling auf Protein-Ebene. Gemeinsam diese Arbeit stellt die umfangreichste Proteomic Beschreibung B. Cellule di guardia sono cellule altamente specializzate che formano minuscoli pori chiamati stomi sulla superficie della foglia. Le cellule del mesofillo sono specializzate per la fotosintesi. Nonostante le differenze fenotipiche e ovvie funzionale tra i due tipi di cellule, la proteina completa componenti e le loro funzioni non sono state esplorate ma sono qui affrontate attraverso un&#39;analisi proteomica comparativa globale delle cellule di guardia purificate e le cellule del mesofillo. Con l&#39;utilizzo di tag isobarico per tagging quantificazione relativa e assoluta (iTRAQ) e spettrometria di massa di cromatografia liquida bidimensionale, abbiamo identificato 1458 non ridondante proteine in cellule di guardia e le cellule del mesofillo delle foglie di Brassica napus. Basato su criteri statistici rigorosi, un totale di 427 proteine sono stati quantificati, e 74 proteine sono state trovate per essere arricchito in cellule di guardia. Proteine coinvolte in energia (respirazione), trasporto, trascrizione (nucleosoma), struttura cellulare e segnalazione sono preferenzialmente espressi in cellule di guardia. Abbiamo osservato diverse proteine delle cellule di guardia ben caratterizzati. Al contrario, le proteine coinvolte nella fotosintesi, la sintesi di amido, stress o malattia/difesa e altri metabolismi preferenzialmente sono rappresentate nelle cellule del mesofillo. Delle proteine identificate, 110 hanno corrispondenti dati di microarray ottenuti da cellule di guardia di Arabidopsis e cellule del mesofillo. Circa il 72% di queste proteine seguono la stessa tendenza di espressione a livello di trascrizione e proteine. Per il resto delle proteine, la correlazione tra dati di proteomica e i dati di microarray &egrave; povera. Ci&ograve; evidenzia l&#39;importanza dell&#39;analisi quantitativa a livello della proteina. Collettivamente, questo lavoro rappresenta la pi&ugrave; ampia descrizione di proteomic delle cellule di b. napus guardia e ha migliorato la nostra conoscenza della specifica funzionale delle cellule di guardia e le cellule del mesofillo.

Las c&#xE9;lulas de la Guardia son c&#xE9;lulas altamente especializadas que forman peque&#xF1;os poros llamados estomas en la superficie de la hoja. La apertura y cierre de los estomas de las hojas de control de intercambio de gases y el agua de la transpiraci&#xF3;n, as&#xED; como permitir que las plantas para responder r&#xE1;pidamente y adaptarse a nuevas condiciones ambientales. C&#xE9;lulas del mes&#xF3;filo est&#xE1;n especializados para la fotos&#xED;ntesis. A pesar de las diferencias fenot&#xED;picas y funcionales evidentes entre los dos tipos de c&#xE9;lulas, los componentes de las prote&#xED;nas completas y sus funciones no han sido exploradas, pero se abordan a trav&#xE9;s de un an&#xE1;lisis global de la prote&#xF3;mica comparativa de purificar las c&#xE9;lulas oclusivas y las c&#xE9;lulas del mes&#xF3;filo. Con el uso de marcadores isob&#xE1;ricos para cuantificaci&#xF3;n absoluta y relativa (iTRAQ) el etiquetado y en dos dimensiones espectrometr&#xED;a de cromatograf&#xED;a l&#xED;quida de masas, se identificaron 1458 no redundante prote&#xED;nas en las c&#xE9;lulas de guardia y las c&#xE9;lulas del mes&#xF3;filo de Brassica napus hojas. Sobre la base de estrictos criterios estad&#xED;sticos, un total de 427 prote&#xED;nas se cuantificaron y se encontraron 74 prote&#xED;nas que se enriquece en c&#xE9;lulas de guarda. Las prote&#xED;nas que intervienen en la energ&#xED;a (respiraci&#xF3;n), el transporte, la transcripci&#xF3;n (nucleosomas), la estructura celular, y la se&#xF1;alizaci&#xF3;n se expresa preferentemente en c&#xE9;lulas de guarda. Hemos observado varios bien caracterizado prote&#xED;nas de las c&#xE9;lulas de guardia. Por el contrario, las prote&#xED;nas implicadas en la fotos&#xED;ntesis, la s&#xED;ntesis de almid&#xF3;n, la enfermedad / defensa / estr&#xE9;s, y otros metabolismos son preferentemente representado en las c&#xE9;lulas del mes&#xF3;filo. De las prote&#xED;nas identificadas, 110 disponen de datos correspondientes de microarrays obtenidos a partir de c&#xE9;lulas de guarda de Arabidopsis y las c&#xE9;lulas del mes&#xF3;filo. Alrededor del 72% de estas prote&#xED;nas siguen la misma tendencia de expresi&#xF3;n en los niveles de transcripci&#xF3;n y las prote&#xED;nas. Para el resto de las prote&#xED;nas, la correlaci&#xF3;n entre los datos de la prote&#xF3;mica y la de datos de microarrays es pobre. Esto pone de relieve la importancia cuantitativa de los perfiles a nivel de prote&#xED;nas. En conjunto, este trabajo representa la descripci&#xF3;n prote&#xF3;mico m&#xE1;s extenso de B. napus c&#xE9;lulas de guarda y ha mejorado nuestro conocimiento de la especificaci&#xF3;n funcional de las c&#xE9;lulas de guardia y las c&#xE9;lulas del mes&#xF3;filo.

Protein redox regulation is increasingly recognized as an important switch of protein activity in yeast, bacteria, mammals and plants. In this study, we identified proteins with potential thiol switches involved in jasmonate signaling, which is essential for plant defense. Methyl jasmonate (MeJA) treatment led to enhanced production of hydrogen peroxide in Arabidopsis leaves and roots, indicating in vivo oxidative stress. With monobromobimane (mBBr) labeling to capture oxidized sulfhydryl groups and 2D gel separation, a total of 35 protein spots that displayed significant redox and/or total protein expression changes were isolated. Using LC-MS/MS, the proteins in 33 spots were identified in both control and MeJA-treated samples. By comparative analysis of mBBr and SyproRuby gel images, we were able to determine many proteins that were redox responsive and proteins that displayed abundance changes in response to MeJA. Interestingly, stress and defense proteins constitute a large group that responded to MeJA. In addition, many cysteine residues involved in the disulfide dynamics were mapped based on tandem MS data. Identification of redox proteins and their cysteine residues involved in the redox regulation allows for a deeper understanding of the jasmonate signaling networks.

R&#xE9;gulation des prot&#xE9;ines redox est de plus en plus reconnu comme un changement important de l&#x26;#39;activit&#xE9; des prot&#xE9;ines dans la levure, les bact&#xE9;ries, les mammif&#xE8;res et les plantes. Dans cette &#xE9;tude, nous avons identifi&#xE9; des prot&#xE9;ines avec des commutateurs thiols potentiels impliqu&#xE9;s dans la signalisation jasmonate, qui est essentiel pour la d&#xE9;fense des plantes. Jasmonate de m&#xE9;thyle (MeJA) traitement a conduit &#xE0; une production accrue de peroxyde d&#x26;#39;hydrog&#xE8;ne dans des feuilles de Arabidopsis et les racines, en indiquant dans le stress oxydatif in vivo. Avec monobromobimane (MBBR) l&#x26;#39;&#xE9;tiquetage de capturer oxyd&#xE9;s groupes sulfhydryle et la s&#xE9;paration de gel 2D, un total de 35 spots prot&#xE9;iques qui pr&#xE9;sentaient d&#x26;#39;importantes oxydo-r&#xE9;duction et / ou totale des changements d&#x26;#39;expression de prot&#xE9;ines ont &#xE9;t&#xE9; isol&#xE9;es. En outre, de nombreux r&#xE9;sidus cyst&#xE9;ine impliqu&#xE9;s dans la dynamique disulfure ont &#xE9;t&#xE9; cartographi&#xE9;s sur la base de donn&#xE9;es MS en tandem. Protein-Redox-Verordnung wird zunehmend als wichtige Schalter Protein Aktivit&auml;t in Hefe, Bakterien, S&auml;ugetiere und Pflanzen erkannt. In dieser Studie wurden Proteine mit potenziellen Thiol-Switches beteiligt Jasmonate signalisieren, die f&uuml;r die Verteidigung der Pflanze erforderlich ist. Methyl Jasmonate (MeJA) Behandlung f&uuml;hrten zu erh&ouml;hte Produktion von Wasserstoffperoxid in Arabidopsis Bl&auml;tter und Wurzeln, die angibt, in-vivo oxidativen Stress. Mit Monobromobimane (mBBr) etikettieren zu erfassen, die oxidiert Sulfhydryl-Gruppen und 2D Gel Trennung, insgesamt 35 Protein-Spots, die erhebliche Redox und/oder Gesamtprotein Ausdruck &Auml;nderungen angezeigt wurden isoliert. Mittels LC-MS/MS, wurden die Proteine in 33 Pl&auml;tze in Kontrolle und MeJA-behandelten Proben identifiziert. Durch vergleichende Analyse der mBBr und SyproRuby Gelbildern konnten wir bestimmen viele Proteine, die Redox entgegenkommend waren und Proteine, die als Reaktion auf MeJA F&uuml;lle &Auml;nderungen angezeigt. Interessant ist, bilden Stress und Verteidigung Proteine eine gro&szlig;e Gruppe, die MeJA beantwortet hat. Dar&uuml;ber hinaus wurden viele Cystein R&uuml;ckst&auml;nde die Disulfid-Dynamik beteiligt zugeordnet, basierend auf Tandem-MS-Daten. Identifizierung von Redox-Proteine und ihre Cystein R&uuml;ckst&auml;nde bei der Redox-Verordnung erm&ouml;glicht ein tieferes Verst&auml;ndnis f&uuml;r die Jasmonate signalisieren Netze.

In questo studio, abbiamo individuato le proteine con potenziale tiolo interruttori coinvolti in Jasmonato di segnalazione, che &egrave; essenziale per la difesa della pianta. Metil jasmonato (MeJA) trattamento ha portato a una maggiore produzione di perossido di idrogeno in Arabidopsis foglie e radici, che indica lo stress ossidativo in vivo. Interessante, lo stress e la difesa di proteine costituiscono un grande gruppo che ha risposto alla MeJA. Inoltre, molti residui di cisteina coinvolti nella dinamica del disolfuro sono stati mappati sulla base dei dati di tandem MS. Identificazione di proteine redox e loro residui di cisteina coinvolti nella regolazione redox permette una pi&ugrave; profonda comprensione di jasmonato reti di segnalazione.

Guard cells, which form stomata on the leaf epidermis, play important roles in plant gas exchange and defense against pathogens. Abscisic acid (ABA) is a phytohormone that can be induced by drought and leads to stomatal closure. Guard cells have been a premier model system for studying ABA signal transduction. Despite significant progress on the identification of molecular components in the ABA signaling pathway, our knowledge of the protein components is very limited. Here, we employ a recently developed multiplexed isobaric tagging technology to identify ABA-responsive proteins in Brassica napus guard cells. A total of 431 unique proteins were identified with relative quantitative information in control and ABA-treated samples. Proteins involved in stress and defense constituted a major group among the 66 proteins with increased abundance. Thirty-eight proteins were decreased in abundance and fell into several functional groups including metabolism and protein synthesis. Many of the proteins have not been reported as being ABA responsive or involved in stomatal movement. A large percentage of the protein-coding genes contained ABA-responsive elements. This study not only established a comprehensive inventory of ABA-responsive proteins, but also identified new proteins for further investigation of their functions in guard cell ABA signaling.

Las c&#xE9;lulas de la Guardia, que forman los estomas en la epidermis de las hojas, juegan un papel importante en el intercambio de planta de gas y la defensa contra los pat&#xF3;genos. &#xC1;cido absc&#xED;sico (ABA) es una fitohormona que puede ser inducida por la sequ&#xED;a y conduce a cierre de los estomas. Las c&#xE9;lulas de la Guardia han sido un modelo de sistema principal para el estudio de la transducci&#xF3;n de la se&#xF1;al de ABA. A pesar de progresos significativos en la identificaci&#xF3;n de componentes moleculares en la v&#xED;a de se&#xF1;alizaci&#xF3;n de ABA, nuestro conocimiento de los componentes de las prote&#xED;nas es muy limitada. En este caso, se emplea una tecnolog&#xED;a de multiplexado desarrollado recientemente marcado isob&#xE1;rica para identificar las prote&#xED;nas que responden a ABA en Brassica napus c&#xE9;lulas de guarda. Un total de 431 prote&#xED;nas &#xFA;nicas fueron identificadas con informaci&#xF3;n cuantitativa relativa en el control y las muestras tratadas con ABA. Las prote&#xED;nas que intervienen en el estr&#xE9;s y la defensa constituyen un grupo importante entre los 66 prote&#xED;nas con mayor abundancia. Treinta y ocho prote&#xED;nas se redujeron en abundancia y cay&#xF3; en varios grupos funcionales, incluyendo el metabolismo y la s&#xED;ntesis de prote&#xED;nas. Muchas de las prote&#xED;nas no se han reportado como ABA de respuesta o que participan en el movimiento de los estomas. Un gran porcentaje de los genes que codifican prote&#xED;nas contenidas ABA elementos de respuesta. Este estudio no s&#xF3;lo establece un inventario completo de ABA que responden a prote&#xED;nas, sino tambi&#xE9;n identificar nuevas prote&#xED;nas para una mayor investigaci&#xF3;n de sus funciones en la guardia de la se&#xF1;alizaci&#xF3;n celular ABA.

Juvenile idiopathic arthritis reflects a group of clinically heterogeneous arthritides hallmarked by elevated concentrations of circulating immune complexes. In this study, the circulating immune complex proteome was examined to elucidate disease-associated proteins that are overexpressed in patients with an aggressive, and at times destructive, disease phenotype. To solve this proteome, circulating immune complexes were isolated from the sera of patients with chronic, erosive or early-onset, aggressive disease and from patients in medical remission or healthy controls subsequent to protein separation by 2-DE. Thirty-seven protein spots were overexpressed in the circulating immune complexes of the aggressive disease groups as compared to controls, 28 of which have been confidently identified to date. Proteolytic fragments of glyceraldehyde-3-phosphate dehydrogenase, serotransferrin, and Î±-1-antitrypsin have been identified among others. In total, these 28 putative disease-associated proteins most definitely contribute to immune complex formation and likely have a significant role in disease etiology and pathogenesis. Moreover, these proteins represent markers of aggressive disease, which could aid in diagnosis and management strategies, and potential therapeutic targets to prevent or control disease outcome. This is the first in-depth analysis of the circulating immune complex proteome in juvenile idiopathic arthritis.

Juveniler idiopathischer Arthritis spiegelt eine Gruppe von klinisch heterogene Arthritiden gekennzeichnet durch erh&ouml;hte Konzentrationen von zirkulierenden Immunkomplexen. In dieser Studie wurde das zirkulierende immune komplexe Proteome untersucht, um die Krankheit-assoziierte Proteine zu erhellen, die bei Patienten mit einer aggressiven und manchmal zerst&ouml;rerische, Krankheit-Ph&auml;notyp Pleuramesotheliom sind. Um diese Proteome zu l&ouml;sen, wurden zirkulierende Immunkomplexe isoliert von den Seren von Patienten mit chronischen, erosive oder fr&uuml;h beginnende, aggressive Krankheit und von Patienten in der medizinischen Remission oder gesunden Kontrollen nach Protein Trennung von 2-DE. Insgesamt diese 28 vermeintliche Krankheit-assoziierte Proteine definitiv immun Komplexbildung und wahrscheinlich eine bedeutende Rolle in der Krankheit &Auml;tiologie und Pathogenese. Dar&uuml;ber hinaus stellen diese Proteine Marker der aggressiven Krankheit, die helfen k&ouml;nnten, Diagnose und Management-Strategien und m&ouml;gliche therapeutische Ziele zu verhindern oder zu kontrollieren Krankheit Ergebnis dar. Artrite idiopatica giovanile riflette un gruppo di artriti clinicamente eterogeneo caratterizzato da elevate concentrazioni di immunocomplessi circolanti. In questo studio, &egrave; stato esaminato il proteoma di complesso immune circolo per delucidare le proteine associate a malattie che sono overexpressed in pazienti con un fenotipo malattia aggressiva e a volte distruttivo. Per risolvere questa proteome, immunocomplessi circolanti sono stati isolati dai sieri dei pazienti con malattia cronica, erosiva o ad esordio precoce, aggressivo e da pazienti in remissione medica o sani controlli dopo separazione di proteine da 2-DE. Trenta-sette macchie proteiche erano overexpressed in immunocomplessi circolanti dei gruppi rispetto ai controlli, 28 delle quali sono state identificate con certezza alla data malattia aggressiva. Frammenti proteolitici della gliceraldeide-3-fosfato deidrogenasi, &alpha;-1-antitripsina e serotransferrin sono stati identificati tra gli altri. In totale, questi 28 putativo malattia-associated proteins sicuramente contribuire alla formazione dei complessi immuni e probabilmente hanno un ruolo significativo nella malattia eziologia e patogenesi. La artritis idiop&#xE1;tica juvenil refleja un grupo de artritis cl&#xED;nicamente heterog&#xE9;neos caracterizaron por altas concentraciones de complejos inmunes circulantes. En este estudio, el proteoma de circulaci&#xF3;n de complejos inmunes se examin&#xF3; para dilucidar la enfermedad asociada a las prote&#xED;nas que est&#xE1;n sobreexpresados &#x200B;&#x200B;en los pacientes con un agresivo y, a veces destructiva fenotipo, la enfermedad. Para resolver este proteoma, los complejos inmunes circulantes se aislaron de los sueros de pacientes con infecci&#xF3;n cr&#xF3;nica, erosiva o de inicio temprano, enfermedad agresiva y de pacientes en remisi&#xF3;n m&#xE9;dica o controles sanos posteriores a la separaci&#xF3;n de prote&#xED;nas por 2-DE. Treinta y siete puntos de la prote&#xED;na se sobreexpresa en los complejos inmunes circulantes de los grupos de enfermedades agresivas en comparaci&#xF3;n con los controles, de las cuales 28 han sido identificados hasta la fecha con confianza. Fragmentos proteol&#xED;ticos de deshidrogenasa de gliceraldehido-3-fosfato, serotransferrin, y &#x3B1;-1-antitripsina se han identificado entre otros. En total, estos 28 supuestos enfermedades asociadas a las prote&#xED;nas sin duda contribuyen a la formaci&#xF3;n de complejos inmunes y que probablemente tienen un papel importante en la etiolog&#xED;a de la enfermedad y la patogenia. Por otra parte, estas prote&#xED;nas representan los marcadores de la enfermedad agresiva, lo que podr&#xED;a ayudar en el diagn&#xF3;stico y las estrategias de gesti&#xF3;n, y posibles objetivos terap&#xE9;uticos para prevenir o controlar la enfermedad resultados. Este es el primer an&#xE1;lisis en profundidad de la circulaci&#xF3;n de proteoma de complejos inmunes en la artritis idiop&#xE1;tica juvenil.
