Rapamycin increases lifespan in mice, but whether this represents merely inhibition of lethal neoplastic diseases, or an overall slowing in multiple aspects of aging is currently unclear. We report here that many forms of age-dependent change, including alterations in heart, liver, adrenal glands, endometrium, and tendon, as well as age-dependent decline in spontaneous activity, occur more slowly in rapamycin-treated mice, suggesting strongly that rapamycin retards multiple aspects of aging in mice, in addition to any beneficial effects it may have on neoplastic disease. We also note, however, that mice treated with rapamycin starting at 9 months of age have significantly higher incidence of testicular degeneration and cataracts; harmful effects of this kind will guide further studies on timing, dosage, and tissue-specific actions of rapamycin relevant to the development of clinically useful inhibitors of TOR action.

Rapamycine augmente la dur&eacute;e de vie chez les souris, mais si il s&#39;agit simplement de l&#39;inhibition de maladies n&eacute;oplasiques mortelles, ou un ralentissement global dans plusieurs aspects du vieillissement est actuellement incertaine. Nous rapportons ici que plusieurs formes de changement de fonction de l&#39;&acirc;ge, y compris toute modification dans le c&oelig;ur, foie, glandes surr&eacute;nales, endom&egrave;tre et tendon, ainsi que fonction de l&#39;&acirc;ge d&eacute;clin de l&#39;activit&eacute; spontan&eacute;e, se produisent plus lentement chez les souris trait&eacute;es &agrave; la rapamycine, sugg&eacute;rant fortement que la rapamycine retarde les multiples aspects du vieillissement chez la souris, en plus des effets b&eacute;n&eacute;fiques, elle pourrait avoir sur la maladie n&eacute;oplasique. Segnaliamo qui che molte forme di cambiamento di et&agrave;-dipendente, tra cui alterazioni nel cuore, fegato, ghiandole surrenali, dell&#39;endometrio e tendine, cos&igrave; come il declino et&agrave;-dipendente nell&#39;attivit&agrave; spontanea, si verificano pi&ugrave; lentamente nei topi trattati con rapamicina, suggerisce fortemente che ritarda la rapamicina molteplici aspetti dell&#39;invecchiamento nei topi, oltre a eventuali effetti benefici pu&ograve; avere sulla malattia neoplastica. Notiamo anche, tuttavia, che i topi trattati con rapamicina a partire da 9 mesi di et&agrave; hanno significativamente pi&ugrave; alta incidenza di degenerazione testicolare e cataratta; gli effetti nocivi di questo tipo guida ulteriori studi sul temporizzazione, dosaggio e tessuto-specifiche azioni di rapamicina rilevante per lo sviluppo di inibitori clinicamente utili di azione TOR.
