Fabrication of magnetic particles (MPs) with high magnetization and large surface area simultaneously is critical for the application of MPs in bioseparation and drug delivery but remains a challenge. In this article, we describe an unprecedented approach to synthesize mesoporous magnetic colloidal nanocrystal clusters (MCNCs) stabilized by poly(Î³-glutamic acid) (PGA) with high magnetization, large surface area (136 m(2)/g) and pore volume (0.57 cm(3)/g), excellent colloidal stability, prominent biocompatibility, and acid degradability. This result provides the important step toward the construction of a new family of MCNCs and demonstrates its capacity in a "magnetic motor" drug delivery system. Here, as an example, we explore the applicability of as-prepared mesoporous MCNCs as hydrophobic drug delivery vehicles (paclitaxel as model drug), and the resultant loading capacity is as high as 35.0 wt %. The antitumor efficacy measured by MTT assay is significantly enhanced, compared with free drugs. Thus, combined with their inherent high magnetization, the mesoporous MCNCs pave the way for applying magnetic targeting drug carriers in antitumor therapeutics.

Fabricaci&oacute;n de part&iacute;culas magn&eacute;ticas (MPs) con alta magnetizaci&oacute;n y gran superficie al mismo tiempo es fundamental para la aplicaci&oacute;n de MPs en la entrega de bioseparaci&oacute;n y drogas pero sigue siendo un desaf&iacute;o. En este art&iacute;culo, describimos un acercamiento sin precedentes para sintetizar mesoporosos magn&eacute;tico nanocrystal coloidal racimos (MCNCs) estabilizados por poli (&aacute;cido &gamma;-glut&aacute;mico) (PGA) con alta magnetizaci&oacute;n, &aacute;rea de gran superficie (136 m(2)/g) y poro volumen (0,57 cm(3)/g), excelente estabilidad coloidal, prominente biocompatibilidad y degradabilidad &aacute;cida. Este resultado proporciona el paso importante hacia la construcci&oacute;n de una nueva familia de MCNCs y demuestra su capacidad en un &quot;motor magn&eacute;tico&quot; sistema de entrega de drogas. Aqu&iacute;, como ejemplo, exploramos la aplicabilidad de los preparados como mesoporosos MCNCs como veh&iacute;culos de entrega de drogas hidrof&oacute;bico (paclitaxel como droga de modelo), y el resultante de capacidad de carga es tan alto como el 35,0% de peso. An effective method was developed for synthesizing magnetite/polymer colloidal composite microspheres with controllable variations in size and shape of the nanostructures and desirable interfacial chemical functionalities, using surfactant-free seeded emulsion polymerization with magnetite (Fe(3)O(4)) colloidal nanocrystal clusters (CNCs) as the seed, styrene (St) as the monomer, and potassium persulfate (KPS) as the initiator. The sub-micrometer-sized citrate-acid-stabilized Fe(3)O(4) CNCs were first obtained via ethylene glycol (EG)-mediated solvothermal synthesis, followed by 3-(trimethoxysilyl)propyl methacrylate (MPS) modification to immobilize the active vinyl groups onto the surfaces, and then the hydrophobic St monomers were polymerized at the interfaces to form the polymer shells by seeded emulsion radical polymerization. The morphology of the composite microspheres could be controlled from raspberry- and flower-like shapes, to eccentric structures by simply adjusting the feeding weight ratio of the seed to the monomer (Fe(3)O(4)/St) and varying the amount of cross-linker divinyl benzene (DVB). The morphological transition was rationalized by considering the viscosity of monomer-swollen polymer matrix and interfacial tension between the seeds and polymer matrix. Functional groups, such as carboxyl, hydroxyl, and epoxy, can be facilely introduced onto the composite microspheres through copolymerization of St with other functional monomers. The resultant microspheres displayed a high saturation magnetization (46 emu/g), well-defined core-shell nanostructures, and surface chemical functionalities, as well as a sustained colloidal stability, promising for further biomedical applications.

La morphologie des microsph&egrave;res composites peut &ecirc;tre contr&ocirc;l&eacute;e &agrave; partir de formes de type framboise et fleur, &agrave; structures excentriques en ajustant simplement l&#39;alimentation poids ratio des semences pour le monom&egrave;re (Fe(3)O(4)/St) et en variant la quantit&eacute; de r&eacute;ticulant divinyl benz&egrave;ne (DVB). La transition morphologique a &eacute;t&eacute; rationalis&eacute;e en tenant compte de la viscosit&eacute; de la matrice polym&egrave;re gonfl&eacute; de monom&egrave;re et de la tension interfaciale entre les graines et la matrice polym&egrave;re. Die sub-micrometer mittlere Citrat-S&auml;ure-stabilisierte Fe(3)O(4) CNCs wurden zun&auml;chst &uuml;ber Ethylenglykol (EG) - vermittelte Solvothermal-Synthese, gefolgt von 3 - erzielt (Trimethoxysilyl) Propyl Methacrylat (MPS) &Auml;nderungen an die aktive Vinyl zu immobilisieren gruppiert auf den Oberfl&auml;chen und dann wurden die hydrophobe St-Monomere an den Schnittstellen die Polymer-Schalen bilden durch radikale ges&auml;ten Emulsionspolymerisation polymerisiert. Die Morphologie des zusammengesetzten Mikrosph&auml;ren k&ouml;nnten von Himbeere und Blume-artigen gestalten, zu kontrolliert werden exzentrische Strukturen durch einfach anpassen der F&uuml;tterung Kraftverh&auml;ltnis der Samen, das Monomer (Fe(3)O(4)/St) und variiert die H&ouml;he der Cross-Linker Divinyl Benzol (DVB). Der morphologische &Uuml;bergang wurde rationalisiert, unter Ber&uuml;cksichtigung der Viskosit&auml;t des Monomer-geschwollen Polymermatrix und Grenzfl&auml;chenspannung zwischen der Samen und der Polymermatrix. Funktionsgruppen, wie Karboxyl-Hydroxyl und Epoxy, k&ouml;nnen facilely auf die zusammengesetzten Mikrosph&auml;ren durch Copolymerisation von St mit anderen funktionalen Monomere eingef&uuml;hrt werden. Die resultierenden Mikrosph&auml;ren angezeigt ein hoher S&auml;ttigung Magnetisierung (46 emu/g), klar definierte Core-Shell-Nanostrukturen und Oberfl&auml;che chemische Funktionalit&auml;ten sowie eine nachhaltige kolloidale Stabilit&auml;t, vielversprechende f&uuml;r weitere biomedizinische Anwendungen.

In Richtung Designer Magnetit/Polystyrol kolloidale zusammengesetzte Mikrosph&auml;ren mit steuerbaren Nanostrukturen und w&uuml;nschenswert Oberfl&auml;che Funktionalit&auml;ten.

&Egrave; stato sviluppato un metodo efficace per la sintesi di magnetite/polimero colloidale microsfere composito con variazioni controllabile in dimensione e forma delle nanostrutture e auspicabile funzionalit&agrave; Chimica interfacciale, mediante polimerizzazione in emulsione seminato privo di tensioattivi con magnetite (Fe(3)O(4)) colloidale nanocristallo cluster (CNCs) come il seme, stirene (St) come il monomero e potassio persolfato (KPS) come l&#39;iniziatore. Il citrato-acido-stabilizzato Fe(3)O(4) CNC di dimensioni sub-micrometer prima sono stati ottenuti tramite glicole etilenico (EG) - sintesi mediata solvothermal, seguita da 3 - modifica di metacrilato (MPS) (trimetossisilil) propil per immobilizzare il vinile attivi gruppi sulle superfici, e quindi i monomeri St idrofobo erano polimerizzati alle interfacce per formare i gusci di polimero polimerizzazione radicale di emulsione di teste di serie. La morfologia delle microsfere di composito pu&ograve; essere controllata da lampone e fiore-come forme, a strutture eccentrici modificando semplicemente l&#39;alimentazione peso rapporto del seme per il monomero (Fe(3)O(4)/St) e variando la quantit&agrave; di benzene di divinil cross-linker (DVB). Se desarroll&oacute; un m&eacute;todo eficaz para la s&iacute;ntesis de microesferas compuesto coloidal de magnetita/pol&iacute;mero con variaciones controlables en tama&ntilde;o y forma de las nanoestructuras y deseables interfaciales funcionalidades de la qu&iacute;micas, mediante la polimerizaci&oacute;n de emulsiones de semillas libres de surfactante con Magnetita (Fe(3)O(4)) coloidal nanocrystal racimos (CNCs) como la semilla, estireno (St) como el mon&oacute;mero y persulfato de potasio (KPS) como iniciador. Las CNCs sub-micrometer-tama&ntilde;o-estabilizado con &aacute;cido citrato Fe(3)O(4) primero se obtuvieron mediante etilenglicol (EG) - mediada solvothermal s&iacute;ntesis, seguida por 3 - modificaci&oacute;n de metacrilato (MPS) (trimetoxisilil) propil para inmovilizar el vinilo activo grupos en las superficies, y luego los mon&oacute;meros hidrof&oacute;bicos de St fueron polimerizados a las interfaces para la formaci&oacute;n de los pol&iacute;meros por polimerizaci&oacute;n radical sembrados de la emulsi&oacute;n. An ultrafast, facile, and efficient microwave hydrothermal approach was designed to fabricate magnetic Fe(3)O(4)/phenol-formaldehyde (PF) core-shell microspheres for the first time. The structure of the Fe(3)O(4)/PF core-shell microspheres could be well controlled by the in situ polycondensation of phenol and formaldehyde with magnetic Fe(3)O(4) clusters as the seeds in an aqueous solution without any surfactants. The effect of synthetic parameters, such as the feeding amounts of phenol, the dosages of formaldehyde, the reaction temperatures, and the microwave heating time, on the morphologies and sizes of the Fe(3)O(4)/PF microspheres were investigated in details. The phenol-formaldehyde shell is found to be evenly coated on Fe(3)O(4) clusters within 10 min of the irradiation. The as-prepared microspheres were highly uniform in morphology, and the method was found to allow the shell thickness to be finely controlled in the range of 10-200 nm. The properties of the composite microspheres were characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), thermogravimetic analysis (TGA), Fourier transform infrared (FT-IR) spectra, X-ray diffraction (XRD), and vibrating sample magnetometer (VSM). The as-prepared Fe(3)O(4)/PF microspheres were monodisperse and highly dispersible in water, ethanol, N,N-dimethyformamide, and acetone, a beneficial quality for the further functionalization and applications of the Fe(3)O(4)/PF microspheres.
