We previously reported that SadB, a protein of unknown function, is required for an early step in biofilm formation by the opportunistic pathogen Pseudomonas aeruginosa. Here we report that a mutation in sadB also results in increased swarming compared to the wild-type strain. Our data are consistent with a model in which SadB inversely regulates biofilm formation and swarming motility via its ability both to modulate flagellar reversals in a viscosity-dependent fashion and to influence the production of the Pel exopolysaccharide. We also show that SadB is required to properly modulate flagellar reversal rates via chemotaxis cluster IV (CheIV cluster). Mutational analyses of two components of the CheIV cluster, the methyl-accepting chemotaxis protein PilJ and the PilJ demethylase ChpB, support a model wherein this chemotaxis cluster participates in the inverse regulation of biofilm formation and swarming motility. Epistasis analysis indicates that SadB functions upstream of the CheIV cluster. We propose that P. aeruginosa utilizes a SadB-dependent, chemotaxis-like regulatory pathway to inversely regulate two key surface behaviors, biofilm formation and swarming motility.

Analyses mutationnelles de deux composantes de la grappe CheIV, le m&#xE9;thyl-acceptation prot&#xE9;ines chimiotactisme PilJ et le PilJ d&#xE9;m&#xE9;thylase CHPB, soutenir un mod&#xE8;le dans lequel ce groupe chimiotactisme participe &#xE0; la r&#xE9;gulation inverse de la formation de biofilms et la motilit&#xE9; essaimage. L&#x26;#39;analyse indique que les fonctions Epistasis SADB amont de la grappe CheIV. Aqu&#xED; mostramos que una mutaci&#xF3;n en SADB tambi&#xE9;n se traduce en un aumento de enjambraz&#xF3;n en comparaci&#xF3;n con la cepa de tipo salvaje. Nuestros datos son consistentes con un modelo en el que SADB inversamente regula la formaci&#xF3;n de biopel&#xED;culas y la motilidad enjambre a trav&#xE9;s de su capacidad para modular tanto las reversiones de flagelos de una manera dependiente de la viscosidad y de influir en la producci&#xF3;n del exopolisac&#xE1;rido Pel. Tambi&#xE9;n mostramos que SADB est&#xE1; obligado a modular adecuadamente las tasas de inversi&#xF3;n a trav&#xE9;s de la quimiotaxis flagelares grupo IV (grupo CheIV). An&#xE1;lisis mutacional de dos componentes de la agrupaci&#xF3;n CheIV, el metil-prote&#xED;nas aceptar quimiotaxis y la PilJ ChpB PilJ desmetilasa, apoyar un modelo en el que este grupo de la quimiotaxis participa en la regulaci&#xF3;n inversa de la formaci&#xF3;n de biopel&#xED;culas y la motilidad enjambre. The intracellular signaling molecule, cyclic-di-GMP (c-di-GMP), has been shown to influence bacterial behaviors, including motility and biofilm formation. We report the identification and characterization of PA4367, a gene involved in regulating surface-associated behaviors in Pseudomonas aeruginosa. The PA4367 gene encodes a protein with an EAL domain, associated with c-di-GMP phosphodiesterase activity, as well as a GGDEF domain, which is associated with a c-di-GMP-synthesizing diguanylate cyclase activity. Deletion of the PA4367 gene results in a severe defect in swarming motility and a hyperbiofilm phenotype; thus, we designate this gene bifA, for biofilm formation. We show that BifA localizes to the inner membrane and, in biochemical studies, that purified BifA protein exhibits phosphodiesterase activity in vitro but no detectable diguanylate cyclase activity. Furthermore, mutational analyses of the conserved EAL and GGDEF residues of BifA suggest that both domains are important for the observed phosphodiesterase activity. Consistent with these data, the DeltabifA mutant exhibits increased cellular pools of c-di-GMP relative to the wild type and increased synthesis of a polysaccharide produced by the pel locus. This increased polysaccharide production is required for the enhanced biofilm formed by the DeltabifA mutant but does not contribute to the observed swarming defect. The DeltabifA mutation also results in decreased flagellar reversals. Based on epistasis studies with the previously described sadB gene, we propose that BifA functions upstream of SadB in the control of biofilm formation and swarming.

Das intrazellul&auml;re signalisierende Molek&uuml;l, zyklische-di-GMP (c-di-GMP), hat sich gezeigt, bakterielle Verhalten, einschlie&szlig;lich Motilit&auml;t und Biofilm-Bildung zu beeinflussen. Wir berichten &uuml;ber die Identifizierung und Charakterisierung von PA4367, ein Gen Oberfl&auml;che-assoziierte Verhaltensweisen in Pseudomonas Aeruginosa Regulierung beteiligt. Das PA4367-Gen codiert f&uuml;r ein Protein mit einer EAL-Dom&auml;ne zugeordnete c-di-GMP Phosphodiesterase-Aktivit&auml;t, sowie eine GGDEF-Dom&auml;ne, die eine c-di-GMP-Synthese Diguanylate Cyclase-Aktivit&auml;t zugeordnet ist. Dar&uuml;ber hinaus zufolge teilhabende Analysen der konservierten EAL und GGDEF R&uuml;ckst&auml;nde der BifA beide Dom&auml;nen sind wichtig f&uuml;r die beobachteten Phosphodiesterase-Aktivit&auml;t. In &Uuml;bereinstimmung mit diesen Daten, stellt der DeltabifA-Mutanten erh&ouml;hte zellul&auml;re Pools &uuml;ber c-di-GMP im Verh&auml;ltnis zu den Wildtyp und erh&ouml;hte Synthese von einem Polysaccharid, produziert von der Pel-Locus. Diese erh&ouml;hte Polysaccharid ist f&uuml;r die erweiterte Biofilm gebildet von der DeltabifA-mutant-aber nicht zu den beobachteten schw&auml;rmen defekt beitr&auml;gt. Die DeltabifA-Mutation f&uuml;hrt auch verminderte Flimmerband Umkehrungen. La mol&#xE9;cula de se&#xF1;alizaci&#xF3;n intracelular, c&#xED;clico-di-GMP (c-di-GMP), se ha demostrado que influyen en los comportamientos bacterianas, incluyendo la motilidad y la formaci&#xF3;n de biopel&#xED;culas. Se presenta la identificaci&#xF3;n y caracterizaci&#xF3;n de PA4367, un gen implicado en la regulaci&#xF3;n de las conductas asociadas a la superficie en Pseudomonas aeruginosa. El gen PA4367 codifica una prote&#xED;na con un dominio EAL, asociado con c-di-GMP fosfodiesterasa actividad, as&#xED; como un dominio GGDEF, que est&#xE1; asociada con una actividad de la adenilato c-di-GMP-sintetizar diguanilato. Supresi&#xF3;n de las PA4367 gen da lugar a un defecto grave en un enjambre motilidad y un fenotipo hyperbiofilm, por lo que se designa esta Bifa gen, para la formaci&#xF3;n de biopel&#xED;culas. Se demuestra que Bifa se localiza en la membrana interna y, en estudios bioqu&#xED;micos, que proteico purificado Bifa exhibe fosfodiesterasa actividad in vitro, pero sin actividad detectable diguanilato ciclasa. Adem&#xE1;s, los an&#xE1;lisis mutacionales de la EAL conservada y residuos de GGDEF Bifa sugieren que ambos dominios son importantes para la actividad de la fosfodiesterasa observado. De acuerdo con estos datos, las exposiciones DeltabifA mutantes aumentado piscinas celulares de c-di-GMP en relaci&#xF3;n con el tipo salvaje y aumento de la s&#xED;ntesis de un polisac&#xE1;rido producido por el locus de pel. Esta producci&#xF3;n de polisac&#xE1;rido aumentado se requiere para la biopel&#xED;cula reforzada formada por el mutante DeltabifA pero no contribuye al defecto observado enjambre. La mutaci&#xF3;n DeltabifA tambi&#xE9;n se traduce en la disminuci&#xF3;n de inversiones de flagelos. Con base en estudios epistasis con el gen se ha descrito anteriormente SADB, proponemos que las funciones de BIFA aguas arriba de SADB en el control de la formaci&#xF3;n de biopel&#xED;culas y el enjambre.

Pseudomonas aeruginosa has served as an important organism in the study of biofilm formation; however, we still lack an understanding of the mechanisms by which this microbe transitions to a surface lifestyle. A recent study of the early stages of biofilm formation implicated the control of flagellar reversals and production of an exopolysaccharide (EPS) as factors in the establishment of a stable association with the substratum and swarming motility. Here we present evidence that SadC (PA4332), an inner membrane-localized diguanylate cyclase, plays a role in controlling these cellular functions. Deletion of the sadC gene results in a strain that is defective in biofilm formation and a hyperswarmer, while multicopy expression of this gene promotes sessility. A DeltasadC mutant was additionally found to be deficient in EPS production and display altered reversal behavior while swimming in high-viscosity medium, two behaviors proposed to influence biofilm formation and swarming motility. Epistasis analysis suggests that the sadC gene is part of a genetic pathway that allows for the concomitant regulation of these aspects of P. aeruginosa surface behavior. We propose that SadC and the phosphodiesterase BifA (S. L. Kuchma et al., J. Bacteriol. 189:8165-8178, 2007), via modulating levels of the signaling molecule cyclic-di-GMP, coregulate swarming motility and biofilm formation as P. aeruginosa transitions from a planktonic to a surface-associated lifestyle.

We reported previously that low concentrations of sodium citrate strongly promote biofilm formation by Staphylococcus aureus laboratory strains and clinical isolates. Here, we show that citrate promotes biofilm formation via stimulating both cell-to-surface and cell-to-cell interactions. Citrate-stimulated biofilm formation is independent of the ica locus, and in fact, citrate represses polysaccharide adhesin production. We show that fibronectin binding proteins FnbA and FnbB and the global regulator SarA, which positively regulates fnbA and fnbB gene expression, are required for citrate's positive effects on biofilm formation, and citrate also stimulates fnbA and fnbB gene expression. Biofilm formation is also stimulated by several other tricarboxylic acid (TCA) cycle intermediates in an FnbA-dependent fashion. While aconitase contributes to biofilm formation in the absence of TCA cycle intermediates, it is not required for biofilm stimulation by these compounds. Furthermore, the GraRS two-component regulator and the GraRS-regulated efflux pump VraFG, identified for their roles in intermediate vancomycin resistance, are required for citrate-stimulated cell-to-cell interactions, but the GraRS regulatory system does not impact the expression of the fnbA and fnbB genes. Our data suggest that distinct genetic factors are required for the early steps in citrate-stimulated biofilm formation. Given the role of FnbA/FnbB and SarA in virulence in vivo and the lack of a role for ica-mediated biofilm formation in S. aureus catheter models of infection, we propose that the citrate-stimulated biofilm formation pathway may represent a clinically relevant pathway for the formation of these bacterial communities on medical implants.

Genetic evidence for an alternative citrate-dependent biofilm formation pathway in Staphylococcus aureus that is dependent on fibronectin binding proteins and the GraRS two-component regulatory system.

Abbiamo segnalato in precedenza che basse concentrazioni di sodio citrato fortemente promuovano la formazione di biofilm da Staphylococcus aureus laboratorio ceppi e di isolati clinici. La formaci&#xF3;n de biopel&#xED;culas tambi&#xE9;n es estimulado por el &#xE1;cido tricarbox&#xED;lico varias otras (TCA), intermediarios del ciclo de una manera fnbA-dependiente. Mientras aconitasa contribuye a la formaci&#xF3;n de biopel&#xED;culas en la ausencia de intermediarios del ciclo TCA, no se requiere para la estimulaci&#xF3;n del biofilm por estos compuestos. Por otra parte, los GraRS de dos componentes del regulador y el regulado VraFG GraRS flujo de salida de la bomba, identificada por su papel en la resistencia a vancomicina intermedia, son necesarios para la citrato-estimulado la c&#xE9;lula a c&#xE9;lula interacciones, pero el sistema GraRS regulatorio no afecta a la expresi&#xF3;n de la fnbA y los genes fnbB.
