Currently, there is a clinical need for more effective vaccine for hepatitis B that induces robust cell-mediated immune response capable of viral clearance in chronic hepatitis B infection. In the present study, hepatitis B vaccines formulations were designed by loading the hepatitis B surface antigen into liposomes adjuvanted with rough lipopolysaccharide (Re-LPS) and lpxL1 LPS using conventional rotatory evaporation method and were characterized for various parameters, such as vesicle shape and surface morphology, size and size distribution, entrapment efficiency, turbidity, and in vitro release pattern. The immunoreactivity in mice was evaluated by measuring anti-HBs IgG titer and compared with alum-adsorbed HBsAg solution, plain HBsAg, and liposomal HBsAg formulations. The formulations were also evaluated for cell-mediated immune response by HBsAg specific proliferation of spleenocytes after secondary immunization and re-stimulation in vitro with the same antigen. Simultaneous estimation of cytokines (IL-4, IFN-gamma) was also carried out. Ex vivo cellular uptake study was performed by fluorescence microscopy. Results indicate that the serum IgG titer obtained after i.m administration of Re-LPS- and lpxL1 LPS-adjuvanted liposomal HBsAg formulation was equivalent to alum-adsorbed HBsAg formulation but was more responsive, sustained, and significantly higher than the corresponding liposomal HBsAg and plain HBsAg formulations. Incorporation of lpxL1 LPS into the liposomal HBsAg increased the stimulation index (SI) 6-10 times as compared with plain HBsAg. Re-LPS- and lpxL1 LPS-adjuvanted liposomal HBsAg formulations induced stronger cellular immune response with a predominant Interferon-gamma (IFN-gamma) level than those induced by free HBsAg alone, alum-adsorbed HBsAg, and non-adjuvanted liposomal HBsAg. Probably, the possible mechanism for the enhancement of cellular immunity in addition to humoral immunity by LPS-adjuvanted liposomal HBsAg formulation is due to marked enhancement of immunological presentation and recruitment of antigen via macrophage and antigen-presenting cells (APCs).

Enhancement of T-helper type I immune responses against hepatitis B surface antigen by LPS derivatives adjuvanted liposomes delivery system.

Actuellement, il y a un besoin clinique d&#39;un vaccin plus efficace contre l&#39;h&eacute;patite B qui induit la robuste r&eacute;ponse &agrave; m&eacute;diation cellulaire immunitaire capable de clairance virale de l&#39;h&eacute;patite b chronique. Dans la pr&eacute;sente &eacute;tude, des formulations de vaccins contre l&#39;h&eacute;patite B ont &eacute;t&eacute; con&ccedil;ues en chargeant l&#39;antig&egrave;ne de surface de l&#39;h&eacute;patite B dans les liposomes avec rugueux lipopolysaccharide (LPS-Re) et lpxL1 LPS &agrave; l&#39;aide de la m&eacute;thode conventionnelle d&#39;&eacute;vaporation rotatoire adjuvant et ont &eacute;t&eacute; caract&eacute;ris&eacute;es des diff&eacute;rents param&egrave;tres, tels que la morphologie forme et sa surface de v&eacute;sicule, taille et distribution granulom&eacute;trique, efficacit&eacute; de pi&eacute;geage, turbidit&eacute; et mod&egrave;le lib&eacute;ration in vitro. Incorporation des LPS de lpxL1 dans l&#39;AgHBs liposomale augmente l&#39;indice de stimulation (SI), 6 - 10 fois par rapport aux AgHBs plain. Re-lpxL1 LPS adjuvant LIPOSOMALES AgHBs formulations et le LPS - induite cellulaire r&eacute;ponse immunitaire plus forte avec un niveau d&#39;interf&eacute;ron-gamma (IFN-gamma) pr&eacute;dominant que ceux induits par gratuitement AgHBs seul, alun-adsorb&eacute; AgHBs et HBsAg liposomale sans adjuvant. Sans doute, le m&eacute;canisme possible pour le renforcement de l&#39;immunit&eacute; cellulaire, en plus de l&#39;immunit&eacute; humorale par LPS adjuvant liposomale AgHBs formulation est due &agrave; l&#39;am&eacute;lioration marqu&eacute;e de pr&eacute;sentation immunologique et de recrutement de l&#39;antig&egrave;ne par les macrophages et les cellules pr&eacute;sentatrices d&#39;antig&egrave;ne (APC).

Derzeit gibt es eine klinische Notwendigkeit wirksamer Impfstoff gegen Hepatitis B, die robuste Zelle &ndash; Mediated Immune Reaktion f&auml;hig der virale Clearance bei chronischer Hepatitis B-Infektion bedingt. In der vorliegenden Studie Hepatitis-B-Impfstoffe Formulierungen wurden die Hepatitis-B-Oberfl&auml;chenantigen in Liposomen adjuvantierte mit groben Lipopolysaccharid (Re-LPS) und lpxL1 LPS mit konventionellen Dreh-Verdunstung-Methode laden entworfen und zeichneten sich f&uuml;r verschiedene Parameter, wie Vesikel Form und Oberfl&auml;che Morphologie, Gr&ouml;&szlig;e und Gr&ouml;&szlig;enverteilung, Verlockende Falle Effizienz, Tr&uuml;bung und in-vitro-Freigabe-Muster. Die Immunoreactivity in den M&auml;usen wurde durch Messung der Anti-HBs-IgG-Titer ausgewertet und verglichen mit Alaun adsorbiert HBsAg-L&ouml;sung, einfache HBsAg und liposomal HBsAg-Formulierungen. Die Formulierungen wurden auch f&uuml;r Zelle &ndash; Mediated Immune Reaktion von HBsAg bestimmte Verbreitung von Spleenocytes nach sekund&auml;ren Immunisierung und Re-stimulation in-vitro mit dem gleichen Antigen bewertet. Gleichzeitige Absch&auml;tzung der Zytokine (IL-4, IFN-Gamma) wurde auch durchgef&uuml;hrt. Ex Vivo erfolgte Aufnahme Studie durch Fluoreszenzmikroskopie. R&uuml;ck-LPS und lpxL1 LPS-adjuvantierte liposomalen HBsAg Formulierungen induzierte st&auml;rker zellul&auml;re Immunantwort mit einem vorherrschenden Interferon-Gamma (IFN-Gamma) als diese induzierten von HBsAg allein frei, Alaun adsorbiert HBsAg und nicht-adjuvantierte liposomalen HBsAg. Wahrscheinlich, der m&ouml;gliche Mechanismus zur Verbesserung der zellul&auml;ren Immunit&auml;t neben der humoralen Immunit&auml;t von LPS-adjuvantierte liposomalen HBsAg-Formulierung ist aufgrund der deutlichen Verbesserung der immunologischen Pr&auml;sentation und Rekrutierung von Antigen &uuml;ber Makrophagen und Antigen-pr&auml;sentierende Zellen (APC).

Verbesserung der T-Helfer Typ I Immunantworten gegen Hepatitis-B-Oberfl&auml;chenantigen von LPS-Derivate-adjuvantierte Liposomen-Delivery-System.

Attualmente, ci &egrave; una necessit&agrave; clinica di vaccino pi&ugrave; efficace per l&#39;epatite B che induce la robusta cellula &ndash; mediata risposta immunitaria in grado di clearance virale nell&#39;infezione da epatite B cronica. Nel presente studio, formulazioni di vaccini contro l&#39;epatite B sono stati progettati caricando l&#39;antigene di superficie dell&#39;epatite B in liposomi adiuvati con ruvida lipopolisaccaride (LPS-Re) e LPS lpxL1 utilizzando il metodo convenzionale evaporazione rotatorio e sono stati caratterizzati per vari parametri, come la morfologia di superficie e forma di vescicola, dimensioni e distribuzione granulometrica, intrappolamento efficienza, torbidit&agrave; e modello di rilascio in vitro. Immunoreattivit&agrave; nei topi &egrave; stato valutato misurando il titolo di IgG anti-HBs e confrontato con soluzione di allume-adsorbita HBsAg, pianura HBsAg e formulazioni liposomiale di HBsAg. Le formulazioni sono state valutate anche per la risposta immunitaria delle cellule &ndash; mediata da HBsAg specifiche proliferazione di spleenocytes dopo immunizzazione secondaria e re-stimulation in vitro con l&#39;antigene stesso. Stima simultanea di citochine (IL-4, IFN-gamma) inoltre &egrave; stato effettuato. Ex vivo uptake cellulare studio &egrave; stato condotto da microscopia di fluorescenza. I risultati indicano che il titolo di IgG del siero ottenuto dopo la somministrazione im di Re-LPS - ed lpxL1 LPS-adiuvati formulazione liposomiale di HBsAg era equivalente alla formulazione di HBsAg adsorbita allume ma era pi&ugrave; reattivo, sostenuta e significativamente superiore al corrispondente liposomiale HBsAg e la pianura HBsAg formulazioni. Incorporazione di LPS lpxL1 l&#39;HBsAg liposomiale aumentato l&#39;indice di stimolazione (SI) 6 - 10 volte rispetto al normale HBsAg. Re-lpxL1 LPS-adiuvati liposomiale HBsAg formulazioni e LPS - indussero pi&ugrave; forte cellular risposta immunitaria con un livello di interferone-gamma (IFN-gamma) predominante rispetto quelle indotte da libero solo HBsAg, allume adsorbita HBsAg e HBsAg liposomiale non adiuvati. Actualmente, existe una necesidad cl&iacute;nica de vacuna m&aacute;s eficaz contra la hepatitis b que induce robusta transmitida por inmunorespuesta c&eacute;lulas capaces de separaci&oacute;n viral en la infecci&oacute;n de hepatitis B cr&oacute;nica. En el presente estudio, formulaciones de vacunas de hepatitis B fueron dise&ntilde;adas por cargar el ant&iacute;geno de superficie de hepatitis B en liposomas adyuvados con &aacute;spero lipopolisac&aacute;rido (LPS-Re) y LPS lpxL1 m&eacute;todo de evaporaci&oacute;n rotatorios convencionales y se caracterizaban por diversos par&aacute;metros, como la morfolog&iacute;a de forma y superficie de ves&iacute;cula, tama&ntilde;o y distribuci&oacute;n de tama&ntilde;o, eficiencia de captura, turbidez y patr&oacute;n de liberaci&oacute;n in vitro. La inmunoreactividad en ratones fue evaluada midiendo t&iacute;tulo de IgG anti-HBs y comparada con soluci&oacute;n de HBsAg alumbre-adsorbidos, llano HBsAg y liposomal formulaciones de HBsAg. Transmitida por inmunorespuesta c&eacute;lulas tambi&eacute;n evaluaron las formulaciones por HBsAg proliferaci&oacute;n espec&iacute;fica de spleenocytes despu&eacute;s de la inmunizaci&oacute;n secundaria y REESTIMULACI&Oacute;N in vitro con el mismo ant&iacute;geno. Tambi&eacute;n se realiz&oacute; una estimaci&oacute;n simult&aacute;nea de citoquinas (IL-4, IFN-gamma). Ex vivo se realiz&oacute; el estudio de la absorci&oacute;n celular por microscop&iacute;a de fluorescencia. Los resultados indican que el t&iacute;tulo de IgG del suero obtenido despu&eacute;s de la administraci&oacute;n i.m de Re-LPS - y lpxL1 LPS-adyuvada formulaci&oacute;n lipos&oacute;mica de HBsAg equival&iacute;a a formulaci&oacute;n de HBsAg alumbre-adsorbidos pero era m&aacute;s sensible, sostenida y significativamente mayor que el correspondiente lipos&oacute;mica HBsAg y formulaciones de HBsAg llanas. Incorporaci&oacute;n de lpxL1 LPS en el HBsAg lipos&oacute;mico aument&oacute; el &iacute;ndice de estimulaci&oacute;n (SI) de 6 a 10 veces en comparaci&oacute;n con HBsAg llano. Re-LPS - y lpxL1 LPS-adyuvada lipos&oacute;micos HBsAg formulaciones indujeron m&aacute;s fuerte respuesta inmune celular con un nivel de interfer&oacute;n-gamma (IFN-gamma) predominante que &eacute;sos inducida por libre HBsAg solo HBsAg alumbre-adsorbidos y no con HBsAg lipos&oacute;mica. Probablemente, el mecanismo posible para la mejora de la inmunidad celular, adem&aacute;s de la inmunidad humoral por LPS-adyuvada formulaci&oacute;n lipos&oacute;mica de HBsAg es debido a la marcada mejora de presentaci&oacute;n inmunol&oacute;gico y reclutamiento de ant&iacute;geno por los macr&oacute;fagos y presentadoras de ant&iacute;geno (APC) de las c&eacute;lulas.

Primaquine, an 8-aminoquinoline, is the drug of choice for radical cure of relapsing malaria. Use of primaquine is limited due to its hemotoxicity, particularly in populations with glucose-6-phosphate dehydrogenase deficiency [G6PD(-)]. Biotransformation appears to be central to the anti-infective and hematological toxicities of primaquine, but the mechanisms are still not well understood. Metabolic studies with primaquine have been hampered due to the reactive nature of potential hemotoxic metabolites. An in vitro metabolism-linked hemotoxicity assay has been developed. Co-incubation of the drug with normal or G6PD(-) erythrocytes, microsomes or recombinant cytochrome P(450) (CYP) isoforms has allowed in situ generation of potential hemotoxic metabolite(s), which interact with the erythrocytes to generate hemotoxicity. Methemoglobin formation, real-time generation of reactive oxygen intermediates (ROIs) and depletion of reactive thiols were monitored as multiple biochemical end points for hemotoxicity. Primaquine alone did not produce any hemotoxicity, while a robust increase was observed in methemoglobin formation and generation of ROIs by primaquine in the presence of human or mouse liver microsomes. Multiple CYP isoforms (CYP2E1, CYP2B6, CYP1A2, CYP2D6 and CYP3A4) variably contributed to the hemotoxicity of primaquine. This was further confirmed by significant inhibition of primaquine hemotoxicity by the selective CYP inhibitors, namely thiotepa (CYP2B6), fluoxetine (CYP2D6) and troleandomycin (CYP3A4). Primaquine caused similar methemoglobin formation in G6PD(-) and normal human erythrocytes. However, G6PD(-) erythrocytes suffered higher oxidative stress and depletion of thiols than normal erythrocytes due to primaquine toxicity. The results provide significant insights regarding CYP isoforms contributing to hemotoxicity and may be useful in controlling toxicity of primaquine to increase its therapeutic utility.

Biotransformaci&oacute;n parece ser central a la toxicidad hematol&oacute;gica y antiinfecciosa de primaquina, pero los mecanismos son todav&iacute;a no bien entendidos. The most prominent adverse effects seen during treatment with dapsone, an antibacterial and antiprotozoal agent, are hemolysis and methemoglobinemia. An in vitro microsomal/cytochrome P(450) (CYP)-linked assay, which allows reactive metabolites generated in situ to react with the co-incubated human erythrocytes, was employed to profile CYP isoforms responsible for hemotoxicity of dapsone. Dapsone caused a robust generation of methemoglobin in human erythrocytes in the presence of human/mouse liver microsomes, which indicates contribution of CYP-mediated metabolism for hemotoxicity. The highest methemoglobin formation with dapsone was observed with CYP2C19, with minor contributions from CYP2B6, CYP2D6 and CYP3A4. Cimetidine and chloramphenicol completely abrogated methemoglobin generation by dapsone, thus confirming a predominant contribution of CYP2C19. The results provide useful insights into CYP-dependent hemotoxicity of dapsone in human erythrocytes.

A new indolizidine alkaloid, named Î”Â¹,â¶-juliprosopine (1), together with previously known indolizidine analogs (2- 6), was isolated from the leaves of Prosopis glandulosa var. glandulosa, collected from Nevada, USA; while two other known indolizidines, juliprosopine (6) and juliprosine (7), were isolated from P. glandulosa leaves collected in Texas, USA. The structures of compound 1 and 7 were determined using a combination of NMR and MS techniques. Compound 7 exhibited potent antiplasmodial activity against Plasmodium falciparum D6 and W2 strains with IC (50) values of 170 and 150 ng/mL, respectively, while 1 was found to be less active (ICâ‚…â‚€ values 560 and 600â€‰ng/mL, respectively). Both compounds were devoid of VERO cells toxicity up to a concentration of 23â€Š800â€‰ng/mL. The antileishmanial activity of indolizidines was evaluated against Leishmania donovani promastigotes, axenic amastigotes, and amastigotes in THP1 macrophage cultures. When tested against macrophage cultures, the tertiary bases (1, 3, 6) were found to be more potent than quaternary salts (2, 5, 7), displaying ICâ‚…â‚€ values between 0.8-1.7â€‰Âµg/mL and 3.1-6.0â€‰Âµg/mL, respectively. In addition, compound 7 showed potent antifungal activity against Cryptococcus neoformans and antibacterial activity against Mycobacterium intracellulare, while 1 was potent only against C. neoformans and weakly active against other organisms.

Un nuevo alcaloide indolizid&iacute;nicos, llamado &Delta;&sup1;, Cartago-juliprosopine (1), junto con el conocido indolizid&iacute;nicos an&aacute;logos (2-6), fue aislado de las hojas de Prosopis glandulosa var. glandulosa, de Nevada, Estados Unidos; mientras que otros dos indolizidines conocidos, juliprosopine (6) y juliprosine (7), fueron aislados de p. glandulosa hojas recolectadas en Texas, Estados Unidos. Las estructuras de los compuestos 1 y 7 fueron determinadas usando una combinaci&oacute;n de t&eacute;cnicas de RMN y MS. Compuesto 7 mostraron antiplasmodial potente actividad contra Plasmodium falciparum D6 y W2 cepas con valores de IC (50) de 170 y 150 ng/mL, respectivamente, mientras que 1 result&oacute; para ser menos activo (IC&#x2085;&#x2080; valores 560 y 600 ng/mL, respectivamente). Ambos compuestos fueron desprovisto de toxicidad de c&eacute;lulas VERO hasta una concentraci&oacute;n de 23 800 ng/mL. Se evalu&oacute; la actividad de costosas de indolizidines contra Leishmania donovani promastigotes, amastigotes ax&eacute;nicos y amastigotes en culturas de macr&oacute;fagos THP1. Cuando se prueban contra culturas de macr&oacute;fagos, las bases terciarias (1, 3, 6) fueron encontradas para ser m&aacute;s potente que sales cuaternarias (2, 5, 7), muestran valores IC&#x2085;&#x2080; entre 0.8-1.7 &micro;g/mL y 3.1-6.0 &micro;g/mL, respectivamente. Adem&aacute;s, 7 compuesto demostr&oacute; potente actividad antif&uacute;ngica contra Cryptococcus neoformans y anti-bacteriano actividad contra Mycobacterium intracellulare, mientras que 1 era potente s&oacute;lo contra C. neoformans y d&eacute;bil activo contra otros organismos.

Utilidad terap&eacute;utica de primaquina, un f&aacute;rmaco antimal&aacute;rico 8-aminoquinoline, ha sido limitado debido a su toxicidad hemol&iacute;tico en poblaci&oacute;n con deficiencia de glucosa 6-fosfato deshidrogenasa. Investigaciones recientes en nuestro laboratorio han demostrado que los metabolitos generados a trav&eacute;s de reacciones metab&oacute;licas de citocromo P 450-dependientes son responsables de efectos hemot&oacute;xicos de primaquina, que podr&iacute;a controlarse con acumulaci&oacute;n de metahemoglobina y aumenta el estr&eacute;s oxidativo. Los marcadores moleculares para la posterior cascada de eventos asociados con la separaci&oacute;n temprana de los eritrocitos de la circulaci&oacute;n se evaluaron para comprender el mecanismo de toxicidad hemol&iacute;tico de primaquina. Primaquina sola aunque no indujeron acumulaci&oacute;n de metahemoglobina sensible, pero produce estr&eacute;s oxidativo significativo, que fue mayor en deficiencia de G6PD que en eritrocitos normales. Primaquina, presumiblemente a trav&eacute;s de metabolitos de redox hemot&oacute;xico activos generados in situ en ensayo ligado a metabolismo microsomal hep&aacute;tica humana, inducida por una acumulaci&oacute;n de metahemoglobina dependiente de la dosis y el estr&eacute;s oxidativo, que eran casi similares en eritrocitos normales y deficiencia de G6PD. Primaquina sola o en presencia de los microsomas de h&iacute;gado humanos combinados produjo un efecto significativo en los niveles de calcio intraerythrocytic ni afect&oacute; la asimetr&iacute;a de fosfatidil serina normal los eritrocitos humanos deficiencia G6PD como flowcytometrically vigilada con ensayo de enlace de anexina V. Therapeutic utility of primaquine, an 8-aminoquinoline antimalarial drug, has been limited due to its hemolytic toxicity in population with glucose 6-phosphate dehydrogenase deficiency. Recent investigations at our lab have shown that the metabolites generated through cytochrome P(450)-dependent metabolic reactions are responsible for hemotoxic effects of primaquine, which could be monitored with accumulation of methemoglobin and increased oxidative stress. The molecular markers for succeeding cascade of events associated with early clearance of the erythrocytes from the circulation were evaluated for understanding the mechanism for hemolytic toxicity of primaquine.
