Differential detergent fractionation (DDF) is frequently used to partition fresh cells and tissues into distinct compartments. We have tested whether DDF can reproducibly extract and fractionate cellular protein components from frozen tissues. Frozen kidneys were sequentially extracted with three different buffer systems. Analysis of the three fractions with liquid chromatography-tandem mass spectrometry (LC-MS/MS) identified 1693 proteins, some of which were common to all fractions and others of which were unique to specific fractions. Normalized spectral index (SI(N)) values obtained from these data were compared to evaluate both the reproducibility of the method and the efficiency of enrichment. SI(N) values between replicate fractions demonstrated a high correlation, confirming the reproducibility of the method. Correlation coefficients across the three fractions were significantly lower than those for the replicates, supporting the capability of DDF to differentially fractionate proteins into separate compartments. Subcellular annotation of the proteins identified in each fraction demonstrated a significant enrichment of cytoplasmic, cell membrane, and nuclear proteins in the three respective buffer system fractions. We conclude that DDF can be applied to frozen tissue to generate reproducible proteome coverage discriminating subcellular compartments. This demonstrates the feasibility of analyzing cellular compartment-specific proteins in archived tissue samples with the simple DDF method.

Hydrogen production from Saccharina Japonica by simultaneous dark fermentation (DF) and microbial electrolysis cell (MEC), called sDFMEC, was studied. In the novel sDFMEC process, substrates were converted to H and volatile fatty acids (VFAs) by DF in the bulk phase, and VFAs are simultaneously oxidized by the exoelectrogens in the microbial film on anode electrode with further production of H at the cathode. The sDFMEC process was compared with DF and a combined process of DF and MEC in series (DF-MEC) in terms of H production. The overall H production from S. Japonica in sDFMEC process was higher (438.7 ± 13.3 mL/g-TS), than DF (54.6 ± 0.8 mL/g-TS) and DF-MEC (403.5 ± 7.9 mL/g-TS) process, respectively, which is approximately 3-times higher than those reported in the literature.

Hydrogen production from macroalgae by simultaneous dark fermentation and microbial electrolysis cell.

In this study, a hybrid process of dark fermentation (DF) and microbial fuel cell (MFC), sDFMFC, was investigated for simultaneous H and electricity production from Saccharina japonica in a single reactor. The sDFMFC exhibited a considerably enhanced energy recovery owing to simultaneous H/carboxylic acids (CAs) production by DF and electricity production by MFC consuming CAs. The co-production of H and electricity was confirmed by a time course of CAs concentration in sDFMFC. An excellent energy recovery of 17.3% was obtained from S. japonica with H yield of 110 mL/g-VS and maximum power density of 1.82 W/m. The sDFMFC showed a diverse microbial community for a desirable microbial conversion of organic substrates. The results indicate that sDFMFC can be a promising single reactor process to produce H and electricity from various biomass feedstock with considerable cost savings while ensuring the strength of the individual DF and MFC process.
