The specific phenotypes and progression to maturity of primary cortical neurons in long-term culture correlate well with neurons in vivo. Utilizing a model of neuronal injury in long-term cultures at 21 days in vitro (DIV), we have identified a distinct population of neurons that translocate into the injury site. 5-bromo-2'-deoxyUridine (BrdU) incorporation studies demonstrated that neurons with the capacity to translocate were 21 days old. However, this motile ability is not consistent with the traditional view of the maturation and structural stability of neurons in long-term culture. Therefore, we examined the neurons' cytoskeletal profile using immunocytochemistry, to establish relative stage of maturation and phenotype. Expression of marker proteins including beta-III-tubulin, alpha-internexin, NF-L and NF-M, tau and L1 indicated the neurons were differentiated, and in some cases polarized. The neurons did not immunolabel with NF-H or MAP2, which might suggest they had not reached the level of maturity of other neurons in culture. They did not express the microtubule-associated migration marker doublecortin (DCX). Cytoskeletal disrupting agents were used to further investigate the role of the microtubule cytoskeleton in translocation, and microtubule destabilization significantly enhanced aspects of their motility. Finally, molecular guidance cues affected their motility in a similar manner to that reported for both axon guidance and early neuron migration. Therefore, this study has identified and characterized a population of motile neurons in vitro that have the capacity to migrate into a site of injury. These studies provide new information on the structurally dynamic features of subsets of neurons.

The cytoplasmic dynein complex is fundamentally important to all eukaryotic cells for transporting a variety of essential cargoes along microtubules within the cell. This complex also plays more specialized roles in neurons. The complex consists of 11 types of protein that interact with each other and with external adaptors, regulators and cargoes. Despite the importance of the cytoplasmic dynein complex, we know comparatively little of the roles of each component protein, and in mammals few mutants exist that allow us to explore the effects of defects in dynein-controlled processes in the context of the whole organism. Here we have taken a genotype-driven approach in mouse (Mus musculus) to analyze the role of one subunit, the dynein light intermediate chain 1 (Dync1li1). We find that, surprisingly, an N235Y point mutation in this protein results in altered neuronal development, as shown from in vivo studies in the developing cortex, and analyses of electrophysiological function. Moreover, mutant mice display increased anxiety, thus linking dynein functions to a behavioral phenotype in mammals for the first time. These results demonstrate the important role that dynein-controlled processes play in the correct development and function of the mammalian nervous system.

Nous constatons que, de fa&#xE7;on surprenante, une mutation ponctuelle dans cette N235Y r&#xE9;sultats de prot&#xE9;ines dans le d&#xE9;veloppement neuronal modifi&#xE9;, comme indiqu&#xE9; par des &#xE9;tudes in vivo dans le cortex en d&#xE9;veloppement, et les analyses de la fonction &#xE9;lectrophysiologique. Trotz der Bedeutung der komplexen cytoplasmatische Dynein vergleichsweise wenig der Rollen der einzelnen Komponenten-Protein bekannt und bei S&auml;ugetieren existieren einige Mutanten, die uns erm&ouml;glichen, die Auswirkungen von M&auml;ngeln Dynein-gesteuerte Prozesse im Kontext des gesamten Organismus zu erkunden. Hier haben wir eine Genotyp-gesteuerte Maus (Mus Musculus) analysieren die Rolle der eine Untereinheit, die Dynein Mittelstufe Leichtketten 1 (Dync1li1) Ansatz. Wir finden, dass &uuml;berraschenderweise eine Punktmutation N235Y in dieses Protein in ver&auml;nderte neuronale Entwicklung, Ergebnisse, wie von in-vivo-Studien in die Entwicklung der Hirnrinde, und Analysen der elektrophysiologischen Funktion gezeigt. El complejo dine&#xED;na citoplasm&#xE1;tica es de importancia fundamental para todas las c&#xE9;lulas eucariotas para el transporte de una variedad de cargas esenciales a lo largo de los microt&#xFA;bulos de la c&#xE9;lula. Este complejo tambi&#xE9;n desempe&#xF1;a funciones m&#xE1;s especializadas en las neuronas. El complejo consta de 11 tipos de prote&#xED;nas que interact&#xFA;an entre s&#xED; y con los adaptadores externos, reguladores y las cargas. A pesar de la importancia del complejo dine&#xED;na citoplasm&#xE1;tica, sabemos relativamente poco de los roles de cada componente proteico, y en los mam&#xED;feros existen pocos mutantes que nos permiten explorar los efectos de los defectos de dine&#xED;na procesos controlados en el contexto de todo el organismo. Aqu&#xED; hemos adoptado un enfoque basado en el genotipo del rat&#xF3;n (Mus musculus) para analizar el papel de una subunidad, la cadena ligera de dine&#xED;na intermedio 1 (Dync1li1). Nos parece que, sorprendentemente, una mutaci&#xF3;n puntual en el N235Y esta prote&#xED;na resulta en el desarrollo neuronal alterada, como se muestra en los estudios in vivo en el desarrollo de la corteza, y el an&#xE1;lisis de la funci&#xF3;n electrofisiol&#xF3;gica. Por otra parte, los ratones mutantes muestran aumento de la ansiedad, vinculando as&#xED; las funciones de dine&#xED;na a un fenotipo conductual en los mam&#xED;feros, por primera vez. Estos resultados demuestran el importante papel que dine&#xED;na procesos controlados por jugar en el correcto desarrollo y funcionamiento del sistema nervioso de los mam&#xED;feros.
