The inward rectifier family of potassium (Kir) channels is comprised of at least 16 family members exhibiting broad and often overlapping cellular, tissue, or organ distributions. The discovery of disease-causing mutations in humans and experiments on knockout mice has underscored the importance of Kir channels in physiology and in some cases raised questions about their potential as drug targets. However, the paucity of potent and selective small-molecule modulators targeting specific family members has with few exceptions mired efforts to understand their physiology and assess their therapeutic potential. A growing body of evidence suggests that G protein-coupled inward rectifier K (GIRK) channels of the Kir3.X subfamily may represent novel targets for the treatment of atrial fibrillation. In an effort to expand the molecular pharmacology of GIRK, we performed a thallium (Tl(+)) flux-based high-throughput screen of a Kir1.1 inhibitor library for modulators of GIRK. One compound, termed VU573, exhibited 10-fold selectivity for GIRK over Kir1.1 (IC(50)â€‰=â€‰1.9 and 19â€‰Î¼M, respectively) and was therefore selected for further study. In electrophysiological experiments performed on Xenopus laevis oocytes and mammalian cells, VU573 inhibited Kir3.1/3.2 (neuronal GIRK) and Kir3.1/3.4 (cardiac GIRK) channels with equal potency and preferentially inhibited GIRK, Kir2.3, and Kir7.1 over Kir1.1 and Kir2.1.Tl(+) flux assays were established for Kir2.3 and the M125R pore mutant of Kir7.1 to support medicinal chemistry efforts to develop more potent and selective analogs for these channels. The structure-activity relationships of VU573 revealed few analogs with improved potency, however two compounds retained most of their activity toward GIRK and Kir2.3 and lost activity toward Kir7.1. We anticipate that the VU573 series will be useful for exploring the physiology and structure-function relationships of these Kir channels.

Eine zusammengesetzte, genannt VU573, ausgestellt von 10fach verbesserter Selektivit&auml;t f&uuml;r GIRK &uuml;ber Kir1.1 (IC (50) = 1,9 und 19 &mu;M, bzw.) und war daher f&uuml;r weitere Studie ausgew&auml;hlt. In elektrophysiologischen Experimenten f&uuml;r Xenopus Laevis Eizellen und S&auml;ugetier-Zellen durchgef&uuml;hrt entsprachen VU573 gehemmt Kir3.1/3.2 (neuronale GIRK) und Kir3.1/3.4 (kardiale GIRK) Kan&auml;le mit gleicher Wirksamkeit und bevorzugt gehemmter GIRK, Kir2.3 und Kir7.1 &uuml;ber Kir1.1 und Kir2.1.Tl(+) Fluss Proben f&uuml;r Kir2.3 und den M125R-Pore-Mutanten von Kir7.1 medizinische Chemie Bem&uuml;hungen um potent und selektive Analoga f&uuml;r diese Kan&auml;le zu unterst&uuml;tzen. Die Struktur-Wirkungs-Beziehungen von VU573 ergab einige Analoga mit verbesserten Potenz, aber zwei Verbindungen, die meisten ihrer Aktivit&auml;ten in Richtung GIRK und Kir2.3, und verloren in Richtung Kir7.1 beibehalten. La familia de rectificador interno de canales de potasio (Kir) est&aacute; compuesta de al menos 16 miembros de la familia exhibiendo amplio y a menudo superpuestas distribuciones celular, tejido u &oacute;rgano. El descubrimiento de la enfermedad-causar mutaciones en los seres humanos y los experimentos en ratones knockout ha subrayado la importancia de los canales Kir en fisiolog&iacute;a y en algunos casos plante&oacute; preguntas acerca de su potencial como dianas farmacol&oacute;gicas. Sin embargo, la escasez de potentes y selectivos moduladores de mol&eacute;culas peque&ntilde;as dirigidas a miembros espec&iacute;ficos de la familia ha empantanado con pocas excepciones esfuerzos para comprender su fisiolog&iacute;a y evaluar su potencial terap&eacute;utico. Un creciente cuerpo de evidencia sugiere que los canales de K (GIRK) de rectificador interno acoplado a prote&iacute;nas G de la subfamilia Kir3.X pueden representar nuevas dianas para el tratamiento de la fibrilaci&oacute;n auricular. En un esfuerzo por expandir la farmacolog&iacute;a molecular de GIRK, realizamos un talio (Tl(+)) flujo alto rendimiento pantalla basado en una biblioteca de inhibidor de la Kir1.1 para moduladores de GIRK. Un compuesto, llamado VU573, exhiben diez veces selectividad por GIRK sobre Kir1.1 (IC(50) = &mu;M 1.9 y 19, respectivamente) y por lo tanto fue seleccionado para continuar los estudios. En experimentos electrofisiol&oacute;gicos realizados en ovocitos de Xenopus laevis y c&eacute;lulas de mam&iacute;fero, VU573 inhibe la Kir3.1/3.2 (GIRK neuronal) y Kir3.1/3.4 (GIRK cardiaca) canales con igual potencia y preferencial inhibido GIRK, Kir2.3 y Kir7.1 sobre Kir1.1 y an&aacute;lisis de flujo de Kir2.1.Tl(+) fueron establecidos para que Kir2.3 y el mutante de poro de M125R de Kir7.1 apoyar los esfuerzos de la qu&iacute;mica medicinal para desarrollar m&aacute;s potentes y selectivos an&aacute;logos para estos canales.
