We have evaluated the performance of two three-dimensional (3-D) reconstruction algorithms with data acquired from microPET, a high resolution tomograph dedicated to small animal imaging. The first was a linear filtered-backprojection algorithm (FBP) with reprojection of the missing data, and the second was a statistical maximum a posteriori probability algorithm (MAP). The two algorithms were evaluated in terms of their resolution performance, both in phantoms and in vivo. Sixty independent realizations of a phantom simulating the brain of a baby monkey were acquired, each containing three million counts. Each of these realizations was reconstructed independently with both algorithms. The ensemble of the 60 reconstructed realizations was used to estimate the standard deviation as a measure of the noise for each reconstruction algorithm. More detail was recovered in the MAP reconstruction without an increase in noise relative to FBP. Studies in a simple cylindrical compartment phantom demonstrated improved recovery of known activity ratios with MAP. Finally, in vivo studies also demonstrated a clear improvement in spatial resolution using the MAP algorithm. The quantitative accuracy of the MAP reconstruction was also evaluated by comparison with autoradiography and direct well counting of tissue samples and was shown to be superior.

Comparison of 3-D maximum a posteriori and filtered backprojection algorithms for high-resolution animal imaging with microPET.

We are currently developing a small animal positron emission tomography (PET) scanner with a design goal of 1 microlitre (1 mm3) image resolution. The detectors consist of a 12 x 12 array of 1 x 1 x 10 mm lutetium oxyorthosilicate (LSO) scintillator crystals coupled to a 64-channel photomultiplier tube (PMT) via 5 cm long optical fibre bundles. The optical fibre connection allows a high detector packing fraction despite the dead space surrounding the active region of the PMT. Optical fibre bundles made from different types of glass were tested for light transmission, and also their effects on crystal identification and energy resolution, and compared to direct coupling of the LSO arrays to the PMTs. We also investigated the effects of extramural absorber (EMA) in the fibre bundles. Based on these results, fibre bundles manufactured from F2 glass were selected. We built three pairs of prototype detectors (directly coupled LSO array, fibre bundle without EMA and fibre bundle with EMA) and measured flood histograms, energy resolution, intrinsic spatial resolution and timing resolution. The results demonstrated an intrinsic spatial resolution (FWHM) of 1.12 mm (directly coupled), 1.23 mm (fibre bundle without EMA coupling) and 1.27 mm (fibre bundle with EMA coupling) using an approximately 500 microm diameter Na-22 point source. Using a 330 microm outer diameter steel needle line source filled with F-18, spatial resolution for the detector with the EMA optical fibre bundle improved to 1.05 mm. The respective timing and energy FWHM values were 1.96 ns, 21% (directly coupled), 2.20 ns, 23% (fibre bundle without EMA) and 2.99 ns, 30% (fibre bundle with EMA). The peak-to-valley ratio in the flood histograms was better with EMA (5:1) compared to the optical fibre bundle without EMA (2.5:1), due to the decreased optical cross-talk. In comparison to the detectors used in our current generation microPET scanner, these detectors substantially improve on the spatial resolution, preserve the timing resolution and provide adequate energy resolution for a modern high-resolution animal PET tomograph.

Detector development for microPET II: a 1 microl resolution PET scanner for small animal imaging.

Biological discovery has moved at an accelerated pace in recent years, with a considerable focus on the transition from in vitro to in vivo models. As a result, there has been a significant increase in the need to adapt clinical imaging methods, as well as for novel imaging technologies for biological research. Positron emission tomography (PET) is a clinical imaging modality that permits the use of positron-labeled molecular imaging probes for non-invasive assays of biochemical processes. The imaging procedure can be repeatedly performed before and after interventions, thereby allowing each animal to be used as its own control. Positron-labeled compounds that target a range of molecular targets have been and continue to be synthesized, with examples of biological processes ranging from receptors and synthesis of transmitters in cell communication, to metabolic processes and gene expression. In animal research, PET has been used extensively in the past for studies of non-human primates and other larger animals. New detector technology has improved spatial resolution, and has made possible PET scanning for the study of the most important modern molecular biology model, the laboratory mouse. This paper presents the challenges facing PET technology as applied to small animal imaging, provides a historical overview of the development of small animal PET systems, and discusses the current state of the art in small animal PET technology.

Molecular imaging of small animals with dedicated PET tomographs.

The dramatic advances of biological research in recent years that have focused on the molecular basis of how systems of the body (e.g. cells, organs and the whole organism) function, have increased the need for molecular imaging instrumentation. Of the several imaging modalities available today applied for in vivo studies of research animals, positron emission tomography (PET) is a technique that permits non-invasive use of positron labeled molecular imaging probes to image and assay biochemical processes of cellular function in the living subject. Imaging can be performed repeatedly before and after interventions and therefore allows the use of each animal as its own control. Many different positron labeled compounds have been and continue to be synthesized as probes that target a range of molecular targets within specific biochemical pathways. These molecular imaging probes are used in extremely low mass amounts, such that biological processes involving compounds in nanomolar concentration or lower can be imaged without disturbing the process. Biological processes from receptors and synthesis of transmitters in cell communication pathways, to metabolic processes and gene expression can be imaged. In the past, PET in animal research has been used extensively for studies of primates and larger animals. In recent years, the development of new detector technology has lowered the limits of spatial resolution. This has made it possible to use PET scanning for the study of the most important modern molecular biology model, the laboratory mouse. This paper presents some of the challenges facing small animal PET technology, provides an overview of the development of small animal PET systems, and discusses the current state of the art technology, some of its applications, as well as some future directions.

PET scanners dedicated to molecular imaging of small animal models.

We explored the feasibility of imaging myocardial perfusion and of demonstrating the flow changes in response to reduction of cardiac work non-invasively in anesthetized mice using high spatial resolution, dedicated small-animal positron emission tomography (microPET). In 31 C57BL/6 mice anesthetized with pentobarbital or isoflurane, (13)N-ammonia was injected intravenously and images were recorded with microPET from 4 to 20 min. Fifteen mice (group 1) were studied consecutively at baseline (BL) and after reduction of heart rate (HR) with intraperitoneal injection of clonidine (CLN) to investigate effects of CLN-induced reduction of cardiac work on myocardial (13)N-ammonia uptake. Eight mice (group 2) were imaged repeatedly at BL and eight mice (group 3) twice after CLN to examine reproducibility. Total myocardial (13)N-ammonia accumulation was determined from the transaxial images and normalized for injected dose (%ID). HR was 412+/-97 beats/min at BL and 212+/-44 beats/min after CLN (P<0.0001). In group 1, the %ID significantly decreased from 1.50%+/-0.27% at BL to 1.29%+/-0.28% after CLN (P<0.0001). In groups 2 and 3, reproducibility of %ID was good (y=0.96x+0.105, SEE=0.212%, r(2)=0.749, P<0.0001). In conclusion, (13)N-ammonia microPET imaging demonstrated non-invasively a reduction of myocardial perfusion induced by clonidine in mice. We believe this study is of importance as the first report on myocardial perfusion imaging and flow validation in in vivo mouse hearts with a left ventricular size of only 5 mm using (13)N-ammonia and PET. MicroPET will aid in elucidating cardiac pathophysiology in transgenic mice and monitoring effects of gene therapies on myocardial perfusion.

Nitrogen-13 ammonia cardiac positron emission tomography in mice: effects of clonidine-induced changes in cardiac work on myocardial perfusion.

Noninvasive imaging of molecular events and interactions in living small animal models has gained increasing importance in preclinical research. Two of the imaging modalities available for this research with potential for translation to the clinic are dedicated small animal positron emission tomography and single-photon emission computed tomography. This brief review introduces the fundamental principles behind these imaging technologies and instrumentation, and discusses the limitations in terms of their spatial resolution and sensitivity. In addition, it provides a perspective regarding the research and commercial development of these systems and presents examples of biological applications. Finally, it discusses the major challenges facing these technologies, advantages and limitations with respect to other technologies, and some future prospects.

Instrumentation for molecular imaging in preclinical research: Micro-PET and Micro-SPECT.

The design of an imaging system capable of detecting both high-energy γ-rays and optical wavelength photons is underway at the Crump Institute for Molecular Imaging. This system will noninvasively image small animal models in vivo for the presence of positron emission tomographic (PET) and optical signals. The detector will consist of modules of multichannel photomultiplier tubes (MC-PMT) coupled to arrays of scintillator crystals. The MC-PMT will detect both the photons produced due to bioluminescence and the photons generated by the interaction of γ-rays within the crystals. The long wavelength photons produced through bioluminescence are only slightly attenuated by these crystals and are detected directly at the photocathode of the MC-PMT, resulting in signals of small (5-10 mV) short (~15 ns) pulses. In contrast, annihilation (511 keV) γ-rays interacting in the scintillator crystal send large bursts of photons to the PMT, and result in pulses that can be as large as 500 mV and > 200 ns duration. The processing of pulses with such different characteristics in a single circuit requires significant alteration of the standard pulse processing circuitry used in PET scanners. In this paper, we discuss the requirements of such a circuit and show the results of implementation of one design using single and multiple channel PMTs.

Readout of the Optical PET (OPET) Detector.

The design of an imaging system capable of detecting both high-energy γ-rays and optical wavelength photons is underway at the UCLA Crump Institute for Molecular Imaging. This system, which we call optical PET (OPET), will be capable of non-invasively and repeatedly imaging small animal models in vivo for the presence of PET and optical signals. In this study, we describe the physical principles behind the operation of the OPET imaging system and discuss the design concept for one of the detector modules. Additionally, we demonstrate the operation of an initial prototype detector module for simultaneous detection and imaging of annihilation radiation and single optical photons emanating from separate sources. These results indicate that the construction of an imaging system based on this detector technology is feasible.

Detector Concept for OPET-A Combined PET and Optical Imaging System.

The accurate assessment of nodal involvement in prostate cancer is crucial to planning treatment, yet there is a shortage of noninvasive imaging techniques capable of visualizing nodal lesions directly. This study demonstrates the feasibility of using recombinant human adenoviral vectors to detect nodal metastases in a human prostate cancer model. This was achieved by the prostate-restricted expression of optical and positron emission tomography (PET) imaging reporter genes by the viral vector coupled with the innate lymphotropic properties of adenovirus. We show that peritumoral administration of these vectors results in the direct detection of reporter gene expression in metastatic lesions within sentinel lymph nodes. Notably, this approach parallels the current lymphoscintigraphy method but enables the direct PET visualization of sentinel lymph node metastases, eliminating the need for invasive lymphadenectomy. These findings may lead to more effective diagnostic and therapeutic strategies for individuals with advanced-stage prostate cancer.

Adenovirus-mediated gene expression imaging to directly detect sentinel lymph node metastasis of prostate cancer.

Laboratory mice routinely are housed at 20 to 22 °C-well below the murine thermoneutral zone of 29 to 34 °C. Chronic cold stress requires greater energy expenditure to maintain core body temperature and can lead to the failure of mouse models to emulate human physiology. We hypothesized that mice housed at ambient temperatures of 20 to 22 °C are chronically cold-stressed, have greater energy expenditure, and have high glucose utilization in brown adipose tissue. To test our hypotheses, we used indirect calorimetry to measure energy expenditure and substrate utilization in C57BL/6J and Crl:NU-Foxn1(nu) nude mice at routine vivarium (21 °C), intermediate (26 °C), and heated (31 °C) housing temperatures. We also examined the activation of interscapular brown adipose tissue, the primary site of nonshivering thermogenesis, via thermography and glucose uptake in this region by using positron emission tomography. Energy expenditure of mice was significantly higher at routine vivarium temperatures compared with intermediate and heated temperatures and was associated with a shift in metabolism toward glucose utilization. Brown adipose tissue showed significant activation at routine vivarium and intermediate temperatures in both hirsuite and nude mice. Crl:NU-Foxn1(nu) mice experienced greater cold stress than did C57BL/6J mice. Our data indicate mice housed under routine vivarium conditions are chronically cold stress. This novel use of thermography can measure cold stress in laboratory mice housed in vivaria, a key advantage over classic metabolic measurement tools. Therefore, thermography is an ideal tool to evaluate novel husbandry practices designed to alleviate murine cold stress.

The hidden cost of housing practices: using noninvasive imaging to quantify the metabolic demands of chronic cold stress of laboratory mice.

The development of a prototype dual-modality optical and PET (OPET) small animal imaging tomograph is underway in the Crump Institute for Molecular Imaging at the University of California Los Angeles. OPET consists of a single ring of six detector modules with a diameter of 3.5 cm. Each detector has an 8 × 8 array of optically isolated BGO scintillators which are coupled to multichannel photomultiplier tubes and open on the front end. The system operates in either PET or optical mode and reconstructs the data sets as 3D tomograms. The detectors are capable of detecting both annihilation events (511 keV) from PET tracers as well as Single Photon Events (SPEs) (2-3 eV) from bioluminescence. Detector channels are readout using a custom multiplex readout scheme and then filtered in analog circuitry using either a γ-ray or SPE specific filter. Shaped pulses are sent to a Digital Signal Processing (DSP) unit for event processing. The DSP unit has 100 MHz Analog-to-Digital Converters on the front-end which send digitized samples to Field Programmable Gate Arrays which are programmed via user configurable algorithms to process PET coincidence events or bioluminescence SPEs. Information determined using DSP includes: event timing, energy determination-discrimination, position determination-lookup, and coincidence processing. Coincidence or SPE events are recorded to an external disk and minimal post processing is required prior to image reconstruction. Initial imaging results from a phantom filled with FDG solution and an optical pattern placed on the front end of a detector module in the vicinity of a SPE source are shown.

FPGA Electronics for OPET: A Dual-Modality Optical and Positron Emission Tomograph.

Automated protocols for measuring and dispensing solutions containing radioisotopes are essential not only for providing a safe environment for radiation workers but also to ensure accuracy of dispensed radioactivity and an efficient workflow. For this purpose, we have designed ARAS, an automated radioactivity aliquoting system for dispensing solutions containing positron-emitting radioisotopes with particular focus on fluorine-18 ((18)F).

We have used simulations and measurements to investigate the feasibility of using slanted scintillator crystal geometries as means to provide depth-of-interaction (DOI) information for a pixelated gamma ray imaging detector. The simulations were performed to estimate the fraction of scintillation light detected by the photodetector as a function of interaction location along the height of crystals with different geometries. In addition, physical measurements of the light output for these crystal geometries were obtained from individual crystals coupled to a solid state photodetector (Philips digital-SiPM DPC-3200). In agreement with previous work, we found a change in light output in the slanted region of the crystals compared to the rectangular region. The results from this study indicate the potential of using slanted crystals to gather DOI information based on light output changes as a function of the location of interaction. An examination of the measured energy spectra for the geometries evaluated here, suggests that for BGO crystals somewhere between 2 or 3 DOI bins could be implemented. Based on these results, we conceived a design for a DOI detector module that consists of two slanted crystals, each read-out by separate SiPM pixels.

Feasibility of Using Crystal Geometry for a DOI Scintillation Detector.

Radio thin layer chromatography (radio-TLC) is commonly used to analyze purity of radiopharmaceuticals or to determine the reaction conversion when optimizing radiosynthesis processes. In applications where there are few radioactive species, radio-TLC is preferred over radio-high-performance liquid chromatography due to its simplicity and relatively quick analysis time. However, with current radio-TLC methods, it remains cumbersome to analyze a large number of samples during reaction optimization. In a couple of studies, Cerenkov luminescence imaging (CLI) has been used for reading radio-TLC plates spotted with a variety of isotopes. We show that this approach can be extended to develop a high-throughput approach for radio-TLC analysis of many samples.

High-throughput radio-TLC analysis.

For decades, there has been immense progress in miniaturizing analytical methods based on electrophoresis to improve sensitivity and to reduce sample volumes, separation times, and/or equipment cost and space requirements, in applications ranging from analysis of biological samples to environmental analysis to forensics. In the field of radiochemistry, where radiation-shielded laboratory space is limited, there has been great interest in harnessing the compactness, high efficiency, and speed of microfluidics to synthesize short-lived radiolabeled compounds. We recently proposed that analysis of these compounds could also benefit from miniaturization and have been investigating capillary electrophoresis (CE) and hybrid microchip electrophoresis (hybrid-MCE) as alternatives to the typically used high-performance liquid chromatography (HPLC). We previously showed separation of the positron-emission tomography (PET) imaging tracer 3'-deoxy-3'-fluorothymidine (FLT) from its impurities in a hybrid-MCE device with UV detection, with similar separation performance to HPLC, but with improved speed and lower sample volumes. In this paper, we have developed an integrated radiation detector to enable measurement of the emitted radiation from radiolabeled compounds. Though conventional radiation detectors have been incorporated into CE systems in the past, these approaches cannot be readily integrated into a compact hybrid-MCE device. We instead employed a solid-state avalanche photodiode (APD)-based detector for real-time, high-sensitivity β particle detection. The integrated system can reliably separate [F]FLT from its impurities and perform chemical identification via coinjection with nonradioactive reference standard. This system can quantitate samples with radioactivity concentrations as low as 114 MBq/mL (3.1 mCi/mL), which is sufficient for analysis of radiochemical purity of radiopharmaceuticals.
