The suprachiasmatic nucleus (SCN) of the hypothalamus contains the primary circadian pacemaker in both diurnal and nocturnal mammals. The lower subparaventricular zone (LSPV) immediately dorsal to the SCN may also play an important role in the regulation of circadian rhythms. The SCN contains a multitude of oscillator cells that generate circadian rhythms through transcriptional/translational feedback loops involving a set of clock genes including per1 and per2. Little is known about the temporal and spatial features of the proteins encoded by these genes in day-active mammals. The first objective of this study was to characterize the expression of PER1 and PER2 in the SCN of a diurnal rodent, the unstriped Nile grass rat (Arvicanthis niloticus). The second objective was to evaluate the hypothesis that a molecular clock could exist in the LSPV, where endogenous rhythms in Fos expression are seen in grass rats but not in laboratory rats. Animals were kept on a 12:12 light/dark cycle and perfused at 4-h intervals, and their brains were processed for immunohistochemical detection of PER1 and PER2. Both proteins were seen in the SCN where they peaked early in the dark phase, providing further evidence that the differences between diurnal and nocturnal patterns of behavior emerge from mechanisms lying downstream from the pacemaker within the SCN. Rhythmic expression of PER1 and PER2 was also seen in the LSPV providing support for the hypothesis that this region might participate in circadian time keeping in the diurnal grass rat. In addition, rhythms were seen lateral to the LSPV and the SCN. Results of this study are discussed in light of similarities and differences in the circadian time-keeping systems of day- and night-active animals.

Temporal and spatial distribution of immunoreactive PER1 and PER2 proteins in the suprachiasmatic nucleus and peri-suprachiasmatic region of the diurnal grass rat (Arvicanthis niloticus).

Le noyau suprachiasmatique (SCN) de l&#39;hypothalamus contient le pacemaker circadien primaire chez les mammif&egrave;res diurnes et nocturnes. La zone subparaventricular inf&eacute;rieure (LSPV) imm&eacute;diatement dorsale pour le RCS peut &eacute;galement jouer un r&ocirc;le important dans la r&eacute;gulation des rythmes circadiens. Le RCS contient une multitude de cellules d&#39;oscillateur qui g&eacute;n&egrave;rent des rythmes circadiens par des boucles de r&eacute;troaction transcriptionnelle/translationnelle impliquant un ensemble de g&egrave;nes de l&#39;horloge dont PAR1 et per2. On en sait peu sur les caract&eacute;ristiques spatiales et temporelles des prot&eacute;ines cod&eacute;es par ces g&egrave;nes chez les mammif&egrave;res active le jour. Le premier objectif de cette &eacute;tude &eacute;tait de caract&eacute;riser l&#39;expression de PAR1 et PER2 dans le SCN d&#39;un rongeur diurne, le rat d&#39;herbe du Nil xeru (Arvicanthis niloticus). Le second objectif &eacute;tait d&#39;&eacute;valuer l&#39;hypoth&egrave;se qu&#39;une horloge mol&eacute;culaire pourrait exister dans le LSPV, o&ugrave; les rythmes endog&egrave;nes dans l&#39;expression de Fos sont vus chez les rats de l&#39;herbe, mais pas chez les rats de laboratoire. Animaux ont &eacute;t&eacute; gard&eacute;s sur un cycle lumi&egrave;re/obscurit&eacute; de 12:12 et perfus&eacute; &agrave; intervalles de 4 h, et leurs cerveaux ont &eacute;t&eacute; transform&eacute;es pour la d&eacute;tection immunohistochimique de PAR1 et PER2. El n&uacute;cleo supraquiasm&aacute;tico (SCN) del hipot&aacute;lamo contiene al principal marcapasos circadiano en mam&iacute;feros tanto diurnos como nocturnos. La zona subparaventricular m&aacute;s baja (LSPV) inmediatamente dorsal al SCN tambi&eacute;n puede desempe&ntilde;ar un papel importante en la regulaci&oacute;n de los ritmos circadianos. El SCN contiene multitud de c&eacute;lulas del oscilador que generan ritmos circadianos a trav&eacute;s de bucles de retroalimentaci&oacute;n transcripcional/traslacional que implica un conjunto de genes reloj incluyendo per1 y per2. Poco se sabe acerca de las caracter&iacute;sticas temporales y espaciales de las prote&iacute;nas codificadas por estos genes en los mam&iacute;feros de d&iacute;as de actividad. El primer objetivo de este estudio fue caracterizar la expresi&oacute;n de PER1 y PER2 en la escena de un roedor diurno, la rata de hierba del Nilo Xerus (Arvicanthis niloticus). El segundo objetivo fue evaluar la hip&oacute;tesis de que podr&iacute;a existir un reloj molecular en el LSPV, donde los ritmos end&oacute;genos en la expresi&oacute;n de Fos se ven en las ratas de la hierba pero no en ratas de laboratorio. Animales se mantuvieron en un ciclo de luz/oscuridad de 12:12 inundados a intervalos de 4-h y sus cerebros fueron procesados para la detecci&oacute;n de immunohistochemical de PER1 y PER2. Ambas prote&iacute;nas fueron vistos en la escena donde lleg&oacute; temprano en la fase oscura, proporciona evidencia adicional que surgen de las diferencias entre diurnas y nocturnas de patrones de comportamiento de mecanismos mentira aguas abajo de los marcapasos en el SCN. Expresi&oacute;n r&iacute;tmica de PER1 y PER2 tambi&eacute;n se observ&oacute; en el LSPV apoyando la hip&oacute;tesis de que esta regi&oacute;n puede participar en tiempo circadiano en la rata de hierba diurna. Adem&aacute;s, los ritmos eran vista lateral a la LSPV y la escena. Resultados de este estudio se discuten a la luz de las similitudes y diferencias en los sistemas de mantenimiento de tiempo circadianos de animales de d&iacute;a y noche-activa.

Diurnal and nocturnal animals differ with respect to the timing of a host of behavioral and physiological events including those associated with neuroendocrine functions, but the neural bases of these differences are poorly understood. In nocturnal species, rhythms in tyrosine hydroxylase-containing (TH+) neurons in the hypothalamus appear to be responsible for rhythms in prolactin secretion. Here we investigated TH+ cells in a diurnal rodent (Arvicanthis niloticus, the unstriped Nile grass rat), and comparing them with those of a nocturnal rodent (Rattus norvegicus, Sprague-Dawley rat). We also examined relationships between TH+ cells and fibers containing vasoactive intestinal polypeptide (VIP) that are thought to originate from cells in the suprachiasmatic nucleus (SCN), the site of the primary circadian clock in mammals. The distribution of TH+ neurons was very similar in the two species except for a population of cells in the basal forebrain that was only present in grass rats. Fibers containing VIP appeared to contact neuroendocrine TH+ cells in both species. These data suggest that, though there may be subtle species differences, temporal information is likely to be carried along the same direct pathways from the SCN to the TH+ neurons in day- and night-active species.

Diurnos y nocturnos animales difieren con respecto a la sincronizaci&oacute;n de un anfitri&oacute;n de eventos conductuales y fisiol&oacute;gicos, incluyendo aquellos asociados con funciones neuroendocrinas, pero las bases neurales de estas diferencias son poco conocidas. En especies nocturnas, ritmos en tirosina hidroxilasa que contienen (TH +) neuronas en el hipot&aacute;lamo parecen ser responsable de ritmos en la secreci&oacute;n de la prolactina. Aqu&iacute; investigamos las c&eacute;lulas TH + en un roedor diurno (Arvicanthis niloticus, la rata Xerus de hierba del Nilo) y compararlos con los de un roedor nocturno (Rattus norvegicus, rata Sprague-Dawley). Tambi&eacute;n examinamos las relaciones entre las c&eacute;lulas TH + y las fibras que contiene el polip&eacute;ptido intestinal vasoactivo (VIP) que se cree que se originan de las c&eacute;lulas en el n&uacute;cleo supraquiasm&aacute;tico (SCN), el sitio del reloj circadiano primario en los mam&iacute;feros. La distribuci&oacute;n de neuronas TH + fue muy similar en las dos especies excepto para una poblaci&oacute;n de c&eacute;lulas en el prosenc&eacute;falo basal que s&oacute;lo estaba presente en las ratas de la hierba. In the diurnal rodent Arvicanthis niloticus (grass rats) the pattern of expression of the clock genes and their proteins in the suprachiasmatic nucleus (SCN) is very similar to that seen in nocturnal rodents. Rhythms in clock gene expression have been also documented in several forebrain regions outside the SCN in nocturnal Ratus norvegicus (lab rats). To investigate the neural basis for differences in the circadian systems of diurnal and nocturnal mammals, we examined PER1 expression in the oval nucleus of the bed nucleus of the stria terminalis (BNST-OV), and in the basolateral (BLA) and the central (CEA) amygdala of male grass rats kept in a 12:12 light/dark cycle. In the BNST-OV, peak levels of PER1 expression were seen early in the light phase of the cycle, 12h out of phase with what has been reported for nocturnal lab rats. In the BLA the pattern of PER1 expression featured sustained high levels during the day and low levels at night. PER1 expression in the CEA was also at its highest early in the light phase, but the effect of sampling time was not statistically significant (p<0.06). The results are consistent with the hypothesis that differences between nocturnal and diurnal species are due to differences in neural systems downstream from the SCN.

Dans le BLA, le patron d&#39;expression PER1 vedette soutenues des niveaux &eacute;lev&eacute;s pendant la journ&eacute;e et les faibles niveaux pendant la nuit. Rhythmen im Uhr-Genexpression wurden auch in mehreren Stirnhirn Regionen au&szlig;erhalb der SCN in n&auml;chtlichen Ratus Norvegicus (Laborratten) dokumentiert. Um die neuronale Grundlage f&uuml;r deutlich unterschiedlichen circadian tagaktive und nachtaktive S&auml;ugetiere zu untersuchen, pr&uuml;fen wir PER1 Ausdruck in den ovalen Kern des Kerns Bett der Stria Terminalis (BNST-OV) und Basolateraler (BLA) und die zentrale (CEA) die Amygdala von m&auml;nnlichen Gras Ratten gehalten in einem 12:12-hell-dunkel-Zyklus. In der BNST-OV galten h&ouml;chstem Niveau PER1 Ausdrucks fr&uuml;h in der leichten Phase des Zyklus, 12 h phasenverschoben mit was f&uuml;r n&auml;chtliche Laborratten gemeldet wurde. In der BLA vorgestellten Musters PER1 Ausdruck anhaltend hohe Belastung w&auml;hrend des Tages und niedrigen nachts. PER1 Ausdruck in der CEA war auch am h&ouml;chsten fr&uuml;h in der Licht-Phase, aber die Wirkung der Abtastrate war nicht statistisch signifikant (p &lt; 0.06). Die Ergebnisse stehen im Einklang mit der Hypothese, die Unterschiede zwischen den nachtaktiven und tagaktiven Arten aufgrund von Unterschieden in neuronalen Systemen stromabw&auml;rts von der SCN sind.

Evolutionary transitions between nocturnal and diurnal patterns of adaptation to the day-night cycle must have involved fundamental changes in the neural mechanisms that coordinate the daily patterning of activity, but little is known about how these mechanisms differ. One reason is that information on these systems in very closely related diurnal and nocturnal species is lacking. In this study, we characterize the suprachiasmatic nucleus (SCN), the primary brain structure involved in the generation and coordination of circadian rhythms, in two members of the genus Acomys with very different activity patterns, Acomys russatus (the golden spiny mouse, diurnal) and Acomys cahirinus (the common spiny mouse, nocturnal). Immunohistochemical techniques were used to label cell bodies containing vasoactive intestinal polypeptide (VIP), vasopressin (VP), gastrin-releasing peptide (GRP) and calbindin (CalB) in the SCN, as well as two sets of inputs to it, those containing serotonin (5-HT) and neuropeptide Y (NPY), respectively. All were present in the SCN of both species and no differences between them were seen. On the basis of neuronal phenotype, the SCN was organized into three basic regions that contained VIP-immunoreactive (-ir), CalB-ir and VP-ir cells, in the ventral, middle and dorsal SCN, respectively. In the rostral SCN, GRP-ir cells were in both the VIP and the CalB cell regions, and in the caudal area they were distributed across a portion of each of the other three regions. Fibers containing NPY-ir and serotonin (5-HT)-ir were most concentrated in the areas containing VIP-ir and CalB-ir cells, respectively. The details of the spatial relationships among the labeled cells and fibers seen here are discussed in relation to different approaches investigators have taken to characterize the SCN more generally.

The substructure of the suprachiasmatic nucleus: Similarities between nocturnal and diurnal spiny mice.

Une des raisons est que les informations concernant ces syst&egrave;mes en diurne tr&egrave;s &eacute;troitement apparent&eacute;e et les esp&egrave;ces nocturnes sont absente. Dans cette &eacute;tude, nous caract&eacute;risons le noyau suprachiasmatic (SCN), la structure primaire de cerveau impliqu&eacute;es dans la g&eacute;n&eacute;ration et de la coordination des rythmes circadiens, de deux membres du genre Acomys avec sch&eacute;mas d&#39;activit&eacute; tr&egrave;s diff&eacute;rents, poecilurus Acomys (l&#39;or &eacute;pineuse souris, diurne) et Acomys cahirinus (la commune &eacute;pineuse souris, nocturne). Immunohistochemical Techniken wurden zur Zelle K&ouml;rper mit Vasoaktives intestinales Polypeptid (VIP), Vasopressin (VP), Gastrin-releasing Peptide (GRP) und Calbindin (CalB) in der SCN, sowie zwei Eing&auml;nge und erm&ouml;glicht es, diese enthalten Serotonin (5-HT) und Neuropeptid Y (NPY), bzw. beschriften. Evolutivo transiciones entre patrones nocturnas y diurnas de adaptaci&oacute;n al ciclo d&iacute;a-noche deben haber implicado cambios fundamentales en los mecanismos neuronales que coordinan el patr&oacute;n diario de actividad, pero poco se sabe acerca de c&oacute;mo estos mecanismos son diferentes. Una raz&oacute;n es que la informaci&oacute;n sobre estos sistemas en diurno muy estrechamente relacionado y carecen de especies nocturnas. En este estudio, caracterizamos el n&uacute;cleo supraquiasm&aacute;tico (SCN), la estructura cerebral primario implicado en la generaci&oacute;n y coordinaci&oacute;n de los ritmos circadianos, en dos de los miembros del g&eacute;nero Acomys con patrones de actividad muy diversos, Acomys russatus (el oro Acomys, diurno) y Acomys cahirinus (el com&uacute;n Acomys, nocturna). Se utilizaron t&eacute;cnicas inmunohistoqu&iacute;micas para etiquetar los cuerpos celulares que contienen polip&eacute;ptido intestinal vasoactivo (VIP), vasopresina (VP), liberador de gastrina (GRP) del p&eacute;ptido y calbindin (CalB) en el SCN, as&iacute; como dos juegos de entradas a &eacute;l, &eacute;sos que contienen serotonina (5-HT) y neurop&eacute;ptido Y (NPY), respectivamente. Todos estaban presentes en el SCN de ambas especies y se observaron diferencias entre ellos. Sobre la base de fenotipo neuronal, el SCN se organiz&oacute; en tres regiones b&aacute;sicas que conten&iacute;a VIP-immunoreactive (-ir), CalB-ir y VP-ir c&eacute;lulas, en el SCN ventral y dorsal y medio, respectivamente. En el SCN rostral, GRP-ir c&eacute;lulas eran en el VIP y las regiones de la c&eacute;lula de CalB, y en la zona caudal que se distribuyeron a trav&eacute;s de una porci&oacute;n de cada una de las otras tres regiones. Las fibras que contiene serotonina (5-HT) y el NPY-ir-ir se concentraron m&aacute;s en las &aacute;reas que contienen c&eacute;lulas VIP-ir como CalB-ir, respectivamente. Los detalles de las relaciones espaciales entre el etiquetado de las c&eacute;lulas y fibras vistas aqu&iacute; se discuten en relaci&oacute;n con diferentes enfoques, los investigadores han llevado a caracterizar m&aacute;s generalmente el SCN.

The suprachiasmatic nucleus (SCN) of the hypothalamus is the central pacemaker that controls circadian rhythms in mammals. In diurnal grass rats (Arvicanthis niloticus), many functional aspects of the SCN are similar to those of nocturnal rodents, making it likely that the difference in the circadian system of diurnal and nocturnal animals lies downstream from the SCN. Rhythms in clock genes expression occur in several brain regions outside the SCN that may function as extra-SCN oscillators. In male grass rats PER1 is expressed in the oval nucleus of the bed nucleus of the stria terminalis (BNST-ov) and in the central and basolateral amygdala (CEA and BLA, respectively); several features of PER1 expression in these regions of the grass rat brain differ substantially from those of nocturnal species. Here we describe PER2 rhythms in the same three brain regions of the grass rat. In the BNST-ov and CEA PER2 expression peaked early in the light period Zeitgeber time (ZT) 2 and was low during the early night, which is the reverse of the pattern of nocturnal rodents. In the BLA, PER2 expression was relatively low for most of the 24-h cycle, but showed an acute elevation late in the light period (ZT10). This pattern is also different from that of nocturnal rodents that show elevated PER2 expression in the mid to late night and into the early day. These results are consistent with the hypothesis that diurnal behavior is associated with a phase change between the SCN and extra-SCN oscillators.

La BNST-ov et le CEA PER2 expression a atteint un sommet au d&eacute;but de la p&eacute;riode lumineuse Zeitgeber temps (ZT) 2 et faible pendant le d&eacute;but de la nuit, qui est l&#39;inverse de la tendance de rongeurs nocturnes. Ausdruck war f&uuml;r den Gro&szlig;teil der 24-h-Zyklus relativ gering, aber zeigte eine akute H&ouml;he sp&auml;t in der Licht-Zeit (ZT10). El n&uacute;cleo supraquiasm&aacute;tico (SCN) del hipot&aacute;lamo es el marcapasos central que controla los ritmos circadianos en mam&iacute;feros. En ratas de hierba diurna (Arvicanthis niloticus), muchos aspectos funcionales de la SCN son similares a los de roedores nocturnos, lo que es probable que la diferencia en el sistema circadiano de animales diurnos y nocturnos se encuentra aguas abajo de la SCN. Ritmos en la expresi&oacute;n de genes reloj ocurren en varias regiones del cerebro fuera de la escena que podr&iacute;a funcionar como osciladores extra-SCN. En ratas macho hierba PER1 se expresa en el n&uacute;cleo oval del n&uacute;cleo de la cama de la estr&iacute;a terminal (BNST-ov) y en la am&iacute;gdala central y basolateral (CEA y BLA, respectivamente); varias caracter&iacute;sticas de expresi&oacute;n PER1 en estas regiones del cerebro de la rata de la hierba difieren sustancialmente de los especies nocturnas. Aqu&iacute; describimos PER2 ritmos en las mismas regiones de tres cerebro de la rata de la hierba. En el BNST-ov y CEA PER2 expresi&oacute;n lleg&oacute; temprano en el per&iacute;odo de luz Zeitgeber tiempo (ZT) 2 y fue baja durante la noche temprana, que es la inversa de la pauta de roedores nocturnos. En el BLA, PER2 expresi&oacute;n era relativamente baja durante la mayor parte del ciclo de 24 horas, pero mostr&oacute; una elevaci&oacute;n aguda en el periodo de luz (ZT10). Este patr&oacute;n es tambi&eacute;n diferente de la de roedores nocturnos que muestran elevada expresi&oacute;n de PER2 en el mediados de a tarde en la noche y el d&iacute;a temprano. Estos resultados son consistentes con la hip&oacute;tesis de que comportamiento diurna se asocia con un cambio de fase entre los osciladores SCN y extra-SCN.
