Antifungal resistance represents a major clinical challenge to clinicians responsible for treating invasive fungal infections due to the limited arsenal of systemically available antifungal agents. Currently, fungal invasive infections are related to antifungal resistance by pathogenic fungi. The current study assumed to use of natural ingredients such as the fruiting bodies of large fungi, specifically Agaricus gennadii. This study is also a new initial step in the field of producing an antifungal that has a promising and safe future. However, most of the used antifungals such as polyenes, azoles, echinocandins, and flucytosine have side effects and may lead to damage to the human body. To achieve the goal of this study, 120 mid-stream urine samples were collected from urinary tract infection (UTI) patients, who attended Samarra General Hospital, the primary care sector and some medical clinics in the city of Samarra/Salah al-Din during the period from 11-1-2020 to 1-2-2021. Many laboratory examinations were performed including: microbiological, biochemical, and molecular tests of 70 samples of UTI patients, which develop yeast colonies on culture plates and were considered as positive results. Our results showed that the extract of the Agaricus gennadii fruit bodies contained a number of organic compounds including phenolics, flavonoids, saponins, terpenoids and alkaloids. Regarding the susceptibility of the isolated yeast species, many concentrations (100%, 75%, 50% and 25%) of the fungal extract were investigated. Data analysis of the obtained results showed that among all tested yeasts, Trichosporon mucoides and Candida parapsilosis were susceptible to the fungal extract at all concentrations, however, no effect of the fungal extract on the rest of the studied yeasts. Also, our results demonstrated that the susceptibility was increased with the increase of the fungal extract concentration. More studies are needed to separate and test the exact role of these compounds in the inhibition of fungal growth.

Isolation and identification of yeasts from UTI patient's exhibit susceptibility to Agaricus gennadii extract.

The current project was designed to develop piperine-loaded solid lipid microparticles (SLMs) to assess the anti-arthritic potential of piperine (PIP). Variable proportions of carnauba wax, beeswax, and tween 80 were employed for preparing SLMs by using the solvent evaporation technique. The developed formulations were subjected to particle size measurements, entrapment efficiency (EE), and zeta potential (ZP) determination. Microparticles were also investigated for piperine-lipid compatibility, thermal analysis, surface morphology, piperine (PIP) release trend, and anti-rheumatic activity in rats. The network's grafting was confirmed by FTIR and XRD results. The thermal stability of the constructed network was confirmed by the DSC and TGA results. SEM findings confirm porous surface morphology. The dissolution experiments on SLMs confirmed the sustained release profile, delivering 87.82% to 94.92% of piperine at 7.4 pH for 24 h. All developed formulations followed a zero-order kinetic model and the Korsmeyer-Peppas model. Furthermore, the anti-rheumatic potentials of piperine from SLMs were also investigated and compared with diclofenac sodium (the standard treatment) in a rat model. The analysis revealed that PIP significantly reduced the severity of arthritis, as confirmed by the findings of multiple arthritic assessment parameters.

Development and In-Vitro Tuning of Piperine Containing Solid Lipid Microparticles for the Treatment of Rheumatoid Arthritis.

Talaromyces marneffei is a pathogenic fungus that causes fatal health complications for patients who are infected with HIV. For the current investigation, sputum samples were collected from 19 immunosuppressed patients from two hospitals located in Baghdad, Iraq by which they were inoculated onto both Sabouraud dextrose agar (SDA) medium at 25 °C and BHI (brain heart infusion) agar at 36±1 °C for growth before being identified using single and nested PCR methods. The 18S rRNA gene sequence of T. marneffei was used to create two sets of oligonucleotide primers, RRF1 and RRH1 which are considered fungus-specific outer primers were employed. Both nested and solo PCRs using the T. marneffei-specific inner primers (Pm1 and Pm2) were carried out. To define the phylogenetic relatedness of this isolate, the MEGA X program was used to align the nuclear ribosomal DNA (rDNA) sequences of T. marneffei. Results showed that the wine-colored pigmented isolates in agar were dimorphic, exhibited bloom-like twigs and spore chains characteristic under the microscope, and were filamentous type colonies with light yellow villi. Finally, immuno-compromised patients in Iraq have T. marneffei in their blood cultures that will be induced to pathogenicity, and the PCR assay is valuable for T. marneffe identifying. Other results from nested PCR revealed that 8 human isolates, from 19, have specific fragments of about 400 bp on the agarose gel.
