Legionellales-infected water is a frequent cause of local outbreaks of Legionnaires' disease and Pontiac fever. Decontaminations are difficult because Legionellales reproduce in eukaryotic microorganisms (protists). Most often, Legionellales have been isolated from amoebae; however, the culture-based sampling methods are taxonomically biased. Sequencing studies show that amoebae in the cercozoan class Thecofilosea are dominant in soils and wastewater treatment plants, prompting us to screen their capability to serve as potential hosts of endosymbiotic bacteria. Environmental isolates of Thecofilosea contained a surprising richness of endosymbiotic Legionellales, including . Considering the widespread dispersal of Legionellales in apparently unrelated amoeboid protist taxa, it appears that the morphotype and not the evolutionary origin of amoebae determines their suitability as hosts for Legionellales. We further provide a protocol for gnotobiotic cultivation of Legionellales and their respective hosts, facilitating future genomic and transcriptomic research of host-symbiont relationships.

Novel Endosymbionts in Rhizarian Amoebae Imply Universal Infection of Unrelated Free-Living Amoebae by Legionellales.

Wastewater is treated by concerted actions of the microbial communities within bioreactors. Although protists (unicellular eukaryotes) are good bioindicators and important players influencing denitrification, nitrification, and flocculation, they are the least known organisms in WWTPs. The few recent environmental surveys of the protistan diversity in WWTPs show that the most abundant protistan sequences in WWTPs belong to Thecofilosea (Rhizaria). We re-investigated previously published environmental sequencing data and gathered strains from seven WWTPs to determine which species dominate WWTPs worldwide. We found that all highly abundant thecofilosean sequences represent a single species - Rhogostoma minus. Considering that Thecofilosea are frequent hosts for Legionellales, i.e. bacteria linked to waterborne diseases, we confirm that Rhogostoma minus functions as a host for Legionellales in WWTPs. Whether the highly abundant Rhogostoma minus also serves as a host for known human pathogenic Legionellales requires further attention.

Recent studies on root traits have shown that there are two axes explaining trait variation belowground: the collaboration axis with mycorrhizal partners and the conservation ('fast - slow') axis. However, it is yet unknown whether these trait axes affect the assembly of soilborne fungi. We expect saprotrophic fungi to link to the conservation axis of root traits, whereas pathogenic and arbuscular mycorrhizal fungi link to the collaboration axis, but in opposite directions, as arbuscular mycorrhizal fungi might provide pathogen protection. To test these hypotheses, we sequenced rhizosphere fungal communities and measured root traits in monocultures of 25 grassland plant species, differing in age. Within the fungal guilds, we evaluated fungal species richness, relative abundance and community composition. Contrary to our hypotheses, fungal diversity and relative abundance were not strongly related to the root trait axes. However, saprotrophic fungal community composition was affected by the conservation gradient and pathogenic community composition by the collaboration gradient. The rhizosphere AMF community composition did not change along the collaboration gradient, even though the root trait axis was in line with the root mycorrhizal colonization rate. Overall, our results indicate that in the long term, the root trait axes are linked with fungal community composition.

Responses of rhizosphere fungi to the root economics space in grassland monocultures of different age.

In a pot experiment, we investigated synergistic interaction of N and P fertilisation on barley biomass () on both shoot and root level with the aim to determine whether N-P interaction would be the same for all levels of N and P fertilisation. We further aimed to determine whether there was a critical level of N and/or P fertilisation rate, above which, a decrease in resource allocation to roots (as nutrient availability increased) could be demonstrated. Barley plants were grown from seed on a nutrient poor substrate and subjected to a two-way NxP fertilisation gradient using a modified Hoagland fertilisation solution. We observed N-P interactions in shoot and root biomass, and N and P use-efficiencies. A synergistic response in biomass was observed only above a critical level of P fertilisation when P was not limiting growth. Furthermore, we found that the same incremental increase in N:P ratio of applied fertiliser elicited different responses in shoot and root biomass depending on P treatment and concluded that barley plants were less able to cope with increasing stoichiometric imbalance when P was deficient. We provide, for the first time, stoichiometric evidence that critical levels for synergistic interactions between N-P may exist in crop plants.

Thecate amoebae play important roles in terrestrial and aquatic ecosystems. This study introduces a novel thecofilosean amoeba from Arctic and Antarctic sea sediments. Phylogenetic analysis based on the 18S rDNA sequence places it in the family Chlamydophryidae (order Tectofilosida, class Thecofilosea). However, the novel organism exhibits a significant genetic divergence and distinct morphology from its closest relatives, prompting us to erect the novel genus Katarium with its type species Katarium polorum. K. polorum is a consumer of diatoms and prokaryotes, indicating an important role in nutrient cycling in the polar marine food webs.

Trait-based approaches have been increasingly used to relate plants to soil microbial communities. Using the recently described root economics space as an approach to explain the structure of soil-borne fungal communities, our study in a grassland diversity experiment reveals distinct root trait strategies at the plant community level. In addition to significant effects of plant species richness, we show that the collaboration and conservation gradient are strong drivers of the composition of the different guilds of soil fungi. Saprotrophic fungi are most diverse in species-rich plant communities with 'slow' root traits, whereas plant pathogenic fungi are most diverse and abundant in communities with 'fast' and 'DIY' root traits. Fungal biomass is strongly driven by plant species richness. Our results illustrate that the root economics space and plant species richness jointly determine the effects of plants on soil fungal communities and their potential role in plant fitness and ecosystem functioning.

Plant Species Richness and the Root Economics Space Drive Soil Fungal Communities.

The increased positive impact of plant diversity on ecosystem functioning is often attributed to the accumulation of mutualists and dilution of antagonists in diverse plant communities. While increased plant diversity alters traits related to resource acquisition, it remains unclear whether it reduces defence allocation, whether this reduction differs between roots and leaves, or varies among species. To answer these questions, we assessed the effect of plant species richness, plant species identity and their interaction on the expression of 23 physical and chemical leaf and fine root defence traits of 16 plant species in a 19-yr-old biodiversity experiment. Only leaf mass per area, leaf and root dry matter content and root nitrogen, traits associated with both, resource acquisition and defence, responded consistently to species richness. However, species richness promoted a decoupling of these defences in leaves and fine roots, possibly in response to resource limitations in diverse communities. Species-specific responses were rare and related to chemical defence and mutualist collaboration, likely responding to species-specific antagonists' dilution and mutualists' accumulation. Overall, our study suggests that resource limitation in diverse communities might mediate the relationship between plant defence traits and antagonist dilution.
