Methods that allow monitoring of individual cells over time, using live cell imaging, are essential for studying dynamical cellular processes in heterogeneous cell populations such as primary T lymphocytes. However, applying single cell time-lapse microscopy to study activation and differentiation of these cells was limited due to a number of reasons. First, primary naÃ¯ve T cells are non-adherent and become highly motile upon activation through their antigen receptor. Second, CD4(+) T cell differentiation is a relatively slow process which takes 3-4 days. As a result, long-term dynamic monitoring of individual cells during the course of activation and differentiation is challenging as cells rapidly escape out of the microscope field of view. Here we present and characterize a platform which enables capture and growth of primary T lymphocytes with minimal perturbation, allowing for long-term monitoring of cell activation and differentiation. We use standard cell culture plates combined with PDMS based arrays containing thousands of deep microwells in which primary CD4(+) T cells are trapped and activated by antigen coated microbeads. We demonstrate that this system allows for live cell imaging of individual T cells for up to 72 h, providing quantitative data on cell proliferation and death times. In addition, we continuously monitor dynamics of gene expression in those cells, of either intracellular proteins using cells from transgenic mice expressing fluorescent reporter proteins, or cell surface proteins using fluorescently labeled antibodies. Finally, we show how intercellular interactions between different cell types can be investigated using our device. This system provides a new platform in which dynamical processes and intercellular interactions within heterogeneous populations of primary T cells can be studied at the single cell level.

Les m&eacute;thodes qui permettent de surveiller des cellules individuelles au fil du temps, &agrave; l&#39;aide de l&#39;imagerie de cellules vivantes, sont essentiels pour l&#39;&eacute;tude des processus dynamiques cellulaires dans les populations de cellules h&eacute;t&eacute;rog&egrave;nes comme les lymphocytes T primaires. Toutefois, appliquant la microscopie time-lapse unicellulaire pour &eacute;tudier l&#39;activation et la diff&eacute;renciation de ces cellules a &eacute;t&eacute; limit&eacute;e en raison d&#39;un certain nombre de raisons. Tout d&#39;abord, primaire na&iuml;ve T cellules sont non adh&eacute;rents et devient hautement mobiles lors de l&#39;activation par le biais de leur r&eacute;cepteur d&#39;antig&egrave;ne. En second lieu, la diff&eacute;renciation des cellules T CD4 + est un processus relativement lent qui prend 3-4 jours. En cons&eacute;quence, surveillance dynamique &agrave; long terme des cellules individuelles au cours de l&#39;activation et la diff&eacute;renciation est difficile car les cellules s&#39;&eacute;chapper rapidement hors du champ de vision de microscope. Ici nous pr&eacute;sentons et caract&eacute;riser une plateforme qui permet la capture et la croissance des lymphocytes T primaires avec une perturbation minime, ce qui permet une surveillance &agrave; long terme de l&#39;activation des cellules et la diff&eacute;renciation. Methoden, mit denen &Uuml;berwachung einzelner Zellen im Laufe der Zeit mit Leben-Zelle Darstellung, sind wesentlich f&uuml;r die Untersuchung dynamischer zellul&auml;rer Prozesse in heterogenen Zelle Bev&ouml;lkerungen wie prim&auml;re T-Lymphozyten. Anwenden von einzelligen Zeitraffer Mikroskopie Aktivierung und Differenzierung der Zellen zu studieren war jedoch begrenzt durch eine Reihe von Gr&uuml;nden. Erste, prim&auml;re naiven T-Zellen sind nicht adh&auml;rent und werden nach der Aktivierung durch den Antigen-Rezeptor stark gekr&uuml;mmter. Zweitens ist die CD4(+) T-Zelldifferenzierung ein relativ langsamer Prozess, der dauert 3-4 Tage. Infolgedessen ist langfristige dynamische &Uuml;berwachung einzelner Zellen im Verlauf der Aktivierung und Differenzierung anspruchsvoll, da Zellen rasch aus dem Mikroskop Blickfeld zu entkommen. Hier stellen wir und charakterisieren eine Plattform, die erm&ouml;glicht die Erfassung und das Wachstum von prim&auml;r-T-Lymphozyten mit minimaler St&ouml;rung zulassend Langzeit&uuml;berwachung von ZELLAKTIVIERUNG und Differenzierung. Wir verwenden Standardzellen-Zellkultur-Platten kombiniert mit PDMS basierte Arrays mit Tausenden von tiefen Vertiefungen in denen prim&auml;re CD4(+) T-Zellen sind gefangen und durch Antigen beschichtet Microbeads aktiviert. Wir zeigen, dass dieses System f&uuml;r live Cell imaging des einzelnen T-Zellen f&uuml;r bis zu 72 Stunden, mit quantitative Daten &uuml;ber die Zellproliferation und Tod Mal erlaubt. Dar&uuml;ber hinaus &uuml;berwachen wir kontinuierlich Dynamik der Genexpression in diese Zellen entweder intrazellul&auml;re Proteine mit Zellen von transgenen M&auml;usen Reporter fluoreszierende Proteine oder Zelle Oberfl&auml;che Proteine mit fluorescently beschriftete Antik&ouml;rper zum Ausdruck zu bringen. M&eacute;todos que permiten el seguimiento de c&eacute;lulas individuales con el tiempo, usando proyecci&oacute;n de imagen de c&eacute;lulas vivas, son esenciales para el estudio de los procesos celulares din&aacute;micos en poblaciones heterog&eacute;neas de la c&eacute;lula como primarias linfocitos T. Sin embargo, aplicar la microscopia de time-lapse de unicelular para estudiar la activaci&oacute;n y diferenciaci&oacute;n de estas c&eacute;lulas fue limitado debido a varias razones. Primer, primario ingenuo T c&eacute;lulas son no adherente y convertirse en altamente m&oacute;viles despu&eacute;s de la activaci&oacute;n a trav&eacute;s de su receptor de ant&iacute;geno. En segundo lugar, la diferenciaci&oacute;n de las c&eacute;lulas T CD4 es un proceso relativamente lento que toma 3-4 d&iacute;as. Como resultado, monitorizaci&oacute;n din&aacute;mica a largo plazo de las c&eacute;lulas individuales en el curso de la activaci&oacute;n y diferenciaci&oacute;n es dif&iacute;cil como c&eacute;lulas salir r&aacute;pidamente del microscopio campo de visi&oacute;n. Aqu&iacute; presentamos y caracterizar una plataforma que permite la captura y el crecimiento de los linfocitos de T primarios con m&iacute;nima perturbaci&oacute;n, lo que permite el monitoreo a largo plazo de la diferenciaci&oacute;n y activaci&oacute;n de la c&eacute;lula. Utilizamos placas est&aacute;ndar celular combinadas con matrices PDMS base que contiene miles de micropocillos profundos en el que las c&eacute;lulas T CD4 primarias son atrapadas y activadas por microesferas recubiertas de ant&iacute;genos. Demostramos que este sistema permite c&eacute;lulas vivas im&aacute;genes de c&eacute;lulas T para hasta 72 h, proporcionando datos cuantitativos sobre la proliferaci&oacute;n celular y los tiempos de la muerte. Adem&aacute;s, supervisamos continuamente din&aacute;mica de expresi&oacute;n g&eacute;nica en las c&eacute;lulas, ya sea prote&iacute;nas intracelulares con c&eacute;lulas de ratones transg&eacute;nicos que expresan prote&iacute;nas fluorescentes reportero, o prote&iacute;nas de la superficie de la c&eacute;lula utilizando anticuerpos etiquetados fluorescencia. Por &uacute;ltimo, os mostramos c&oacute;mo intercelulares interacciones entre c&eacute;lulas diferentes tipos pueden ser investigados utilizando nuestro dispositivo. Este sistema ofrece una nueva plataforma en la que procesos din&aacute;micos e interacciones intercelulares dentro de poblaciones heterog&eacute;neas de las c&eacute;lulas T primarias pueden estudiarse a nivel unicelular.
