Pre-existing Ca handling abnormalities constitute the arrhythmogenic substrate in patients developing postoperative atrial fibrillation (POAF), a common complication after cardiac surgery. Postoperative interleukin (IL)-6 levels are associated with atrial fibrosis in several animal models of POAF, contributing to atrial arrhythmias. Here, we hypothesize that IL-6-mediated-Ca handling abnormalities contribute to atrial fibrillation (AF) in sterile pericarditis (SP) rats, an animal model of POAF. SP was induced in rats by dusting atria with sterile talcum powder. Anti-rat-IL-6 antibody (16.7 μg/kg) was administered intraperitoneally at 30 min after the recovery of anesthesia.  electrophysiology,  optical mapping, western blots, and immunohistochemistry were performed to elucidate mechanisms of AF susceptibility. IL-6 neutralization ameliorated atrial inflammation and fibrosis, as well as AF susceptibility  and the frequency of atrial ectopy and AF with a reentrant pattern in SP rats . IL-6 neutralization reversed the prolongation and regional heterogeneity of Ca transient duration, relieved alternans, reduced the incidence of discordant alternans, and prevented the reduction and regional heterogeneity of the recovery ratio of Ca transient. In agreement, western blots showed that IL-6 neutralization reversed the reduction in the expression of ryanodine receptor 2 (RyR2) and phosphorylated phospholamban. Acute IL-6 administration to isolated rat hearts recapitulated partial Ca handling phenotype in SP rats. In addition, intraperitoneal IL-6 administration to rats increased AF susceptibility, independent of fibrosis. Our results reveal that IL-6-mediated-Ca handling abnormalities in SP rats, especially RyR2-dysfunction, independent of IL-6-induced-fibrosis, early contribute to the development of POAF by increasing propensity for arrhythmogenic alternans.

The incidence of atrial fibrillation (AF) increases after surgery and is associated with the activation of NLRP3-inflammation. Our previous studies have found that transient receptor potential vanilloid 4 (TRPV4) blockade reduces the susceptibility to AF, but its molecular mechanisms remains unclear. Therefore, we hypothesized that blockage of TRPV4 reduces the incidence of AF by inhibiting NLRP3-inflammasome in sterile pericarditis (SP) mice. In this study, we established SP mice by dusting talcum powder on atrial surfaces. We first confirmed that genetic or pharmacological TRPV4 inhibition reduced the susceptibility to AF in SP mice. We also found that the expression level of NLRP3-inflammasome and inflammatory cytokines significantly increased in the atria of SP mice, which further increased in application the TRPV4 agonist GSK1016790A (GSK101) and decreased in application the TRPV4 antagonist GSK2193874. More importantly, ERK inhibitor (U0126) or NF-κB inhibitor (Bay11-7082) could partially reverse GSK101-induced NLRP3-inflammasome up-regulation. Interestingly, U0126 can reversed GSK101-induced NF-κB phosphorylation, but Bay11-7082 cannot change GSK101-induced ERK phosphorylation. Finally, we shown that the activation of NLRP3-inflammasome and ERK/NF-κB signaling pathway significantly reduced in TRPV4-knockout SP mice. Collectively, our studies indicate that blockage of TRPV4 prevents AF in SP mice by inhibiting NLRP3-inflammasome through the ERK/NF-κB signaling pathway.

Premature ventricular contractions (PVCs) stemming from the aortic sinus cusp often have preferential conduction to two exits in the outflow tract and exhibited two different morphologies of PVCs, which may render radiofrequency catheter ablation (RFCA) difficult. A 67-year-old male patient underwent RACF for premature ventricular contractions (PVCs) characterizing by bi-morphology (left and right bundle branch block) on electrocardiogram. Dynamic changes in QRS morphology during ablation and evident local voltage potentials during electro-anatomical mapping were critical for identifying the real foci of origin of PVCs. Successful ablation was achieved at the left-right coronary cusp commissure.

Successful ablation of morphology-changing premature ventricular contractions under the guidance of local voltage potentials: A case report.

 For a long time, aspirin has been recommended for the prevention of cardiovascular disease (CVD). However, results of long-term effects of aspirin use on the risk of CVD and all-cause death as well as cause-specific mortality are not consistent. This study aims to investigate the relationship between low- or high-dose preventive aspirin use and the risk of death from all-cause, CVD, and cancer among US adults aged 40 years and older.  A prospective cohort study was conducted by utilizing four cycles of the National Health and Nutrition Examination Survey (NHANES) and linked 2019 mortality files. Cox proportional hazard models accounting for multiple covariates were used to calculate hazard ratio (HR) and 95% confidence interval (CI) for the associations between low- or high-dose aspirin use and risk of death.  A total of 10,854 individuals (5,364 men and 5,490 women) were enrolled in the study. During a median follow-up of 4.8 years, 924 death events including 294 CVD death and 223 cancer death were documented. We found no evidence that taking low-dose aspirin decreased the chance of dying from any cause (HR: 0.92, 95% CI: 0.79-1.06), CVD (HR: 1.03, 95% CI: 0.79-1.33), or cancer (HR: 0.80, 95% CI: 0.60-1.08). High-dose aspirin users had a higher risk of CVD death compared to participants who had never used aspirin (HR: 1.63, 95% CI: 1.11-2.41).  Using low-dose aspirin has no effect on the risk of death from any causes, whereas taking high doses of aspirin increases the risk of CVD death.

ING5, initially identified as a tumor-suppressor, serves as a chromatin regulator with a diverse and extensive range of biological functions. This review undertakes an in-depth exploration of the structural characteristics and domain organization of ING family proteins, with a specific emphasis on ING5. The functional characteristics of ING5 are highly intricate and multi-dimensional. In the context of chromatin regulation and gene expression control, ING5 engages in interactions with diverse protein complexes through its conserved domains. It actively participates in the fine-tuning of chromatin structure and gene expression within tumor cells. Moreover, ING5 plays a pivotal and indispensable role in the regulation of DNA replication, cell cycle, and apoptosis, thereby exerting a profound influence on the fundamental biological processes of cells. Additionally, the binding properties and genomic associations of ING5 contribute significantly to its wide-ranging functions. ING5 exerts multiple and intricate action mechanisms in the processes of tumorigenesis, tumor development, and cancer treatment. It has substantial impacts on the biological behaviors of tumor cells, including proliferation, migration, and invasion. Furthermore, ING5 has emerged as a highly promising target for cancer therapy, presenting novel opportunities for the development of tumor-specific treatment strategies. Beyond its well-established role in tumor suppression, ING5 exhibits a diverse array of physiological functions. In the context of stem cell differentiation, ING5 regulates gene expression patterns, which are of utmost importance for determining cell fate. During embryonic development, it ensures the normal expression of genes associated with cell proliferation and differentiation, thereby being essential for the proper morphogenesis of the embryo. ING5 is also involved in metabolic regulation, particularly lipid metabolism, by modulating relevant genes to influence lipid levels. Additionally, it participates in the maintenance of vascular function by regulating the activities of vascular endothelial cells and angiogenesis, which are crucial for vascular homeostasis. This review comprehensively summarizes the extensive functions of ING5 as an epigenetic regulator in maintaining physiological homeostasis. By delving into its roles beyond tumor suppression, we aspire to attain a more comprehensive and in-depth understanding of its significance and potential implications in various biological processes and medical applications.

Reproductive lifespan (RLS), defined as the interval between menarche and menopause, reflects the trajectory of ovarian aging. While high-density lipoprotein cholesterol (HDL-C) plays a key role in reproductive health, the association between HDL-related inflammatory indices and RLS remains unclear. This study investigated the relationship between HDL-related inflammatory indices-including lymphocyte-to-HDL-C ratio (LHR), monocyte-to-HDL-C ratio (MHR), neutrophil-to-HDL-C ratio (NHR), and platelet-to-HDL-C ratio (PHR)-and RLS among women.
